Highly Branched Glucooligosaccharide and Mannitol Production by Mixed Cultrue Fermentation of Leuconostoc mesenteroides and Lipomyces starkeyi

  • The influence of process conditions on highly branched glucooligosaccharides production by mixed culture of Leuconostoc mesenteroides ATCC 13146 and Lipomyces starkeyi ATCC 74054 was studied. We divided the batch culture fermentations into two groups according to inoculation method. One-point inoculation was performed by coinoculation of L. mesenteroides and L. starkeyi at the ration of 10 to 1, and two-point inoculation by L. mesenteroides inoculation first and L. starkeyi inoculation after L. mesenteroides grew to the end of the log phase of growth. Two-point inoculation improved the yield of oligosaccharide by 1.5 to 20 fold more than one-point inoculation. In this process, the highest yield of oligosaccharides (48% of theoretical yield) and productivity (0.85 g/l/h) were obtained with starch as an initial substrate for L. starkeyi growth. The estimated composition of the end product consisted of 31.5% oligosaccharides, 17.6% dextran, and 46.5% mannitol.

  • The spatial and temporal distribution of petroleum-degrading bacteria(PDB) was studied at six sampling sites in Kyeonggi Bay of the Yellow Sea fiom March 1990 to October 1991. In addition, petroleum-degrading potcntial of natural ~iiai-ineb acterial population was studied at different culturc contlitions. During the period o f stutly. thc heterotrophic bacterial number and PDB number were n1e;rsured in the range of 7 000-108.400 CFU/nil. 0-2.800 MPN1100 mi. respectively. The spatial tlistribution of PDB wa\ highly affected by presence of petroleum hydrocarbon. In laboratory cxperirncnt. petrolcu~n biodegradation wac enhanced hy addition of yeast cxtracl. cell free cxtr:~ct. anti rnixctl culture of PI)B.

Optimal Conditions of Co-Immobilized Mixed Culture System with Aspergillus awamori and Zymomonas mobilis (Aspergillus awamori와 Zymomonas mobilis로 구성된 혼합고정화 배양계의 최적 조건)

  • Co-immobilized mixed culture system(A-Z system) composed of two different oxygen-demanding strains, aerobic(Aspergillus awamori) and anaerobic(Zymomonas mobilis) strains, in a Ca-alginate gel beads was developed to increase ethanol production from raw starch as a carbon source. Optimal mixture ratio of A. awamori and Z. mobilis was $1.25{\times}10^{9}\;spores/L-gel$ and 0.5g cells/L-gel, respectively. After 120 hours of cultivation, gel beads distinguished oxygen-rich surface for A. awamori from oxygen-deficient central part for Z. mobilis. At A-Z culture system, yield of ethanol on glucose, $Y_{p/s}=0.18$, was very low and there was high leakage of cells from surface of gel beads. At A-Z 36 cultrue system with changing silicon check valve for cotton plug at 36 hours in A-Z culture system, there was no cell leakage from gel beads, pH was maintained at around 4.3 during cultivation, and yield of ethanol on glucose, $Y_{p/s}=0.36$, showed 2 times higher than that of control culture system(cotton plug culture).
