Seed Disinfectant Effect of Pleurotus ostreatus (Heuktari) Extract on Fusarium fujikuroi Nirenberg (흑타리버섯 추출물의 벼 키다리병에 대한 종자소독 효과)

  • This study was carried out to investigate antimicrobial activities of water extracted Pleurotus ostreatus var Heuktari (PO), and their application to F. fujikuroi Nirenberg growth inhibition material. Various organic solvents (chloroform, hexane, ethyl acetate, ethanol) were tested to investigate the antimicrobial activities of Pleurotus ostreatus var Heuktari against F. fujikuroi Nirenberg. Chloroform, hexane, ethyl acetate and hot water extracts had no antimicrobial activities, but butanol extract showed 2.5% strong activities in order of F. fujikuroi Nirenberg disk diffusion test. Then we observe Antifungal activity using green house. Bed soil and mushroom extract mixed at 10%, 7.5%, 5%, 2.5%, 1% total bed soil mass. Screening of Antifungal activity was tested two periode 18day and 25day. The Antifungal activity rate of each period and extract density was 7.5% mixed bedsoil has investigated 80% Antifungal activity rate. The result indicates that the Pleurotus ostreatus var Heuktari extract using F. fujikuroi Nirenberg registance, which shows the development of organic agriculture seed disinfectant.

Enhanced Permeation of Leucine Enkephalin and [D-Ala2]-leucine Enkephalinamide across Nasal, Rectal and Vaginal Mucosae of Rabbit (토끼의 비강, 직장 및 질 점막을 통한 로이신엔케팔린과 [D-알라2]-로이신엔케팔린아미드의 투과 증진)

  • The effects of enzyme inhibitors and penetration enhancers on the permeation of leucine enkephalin (Leu-Enk) and its synthetic analog, [${D-ala}^2$]-leucine enkephalinamide (YAGFL) across the nasal, rectal and vaginal mucosae were evaluated. Enzyme inhibitors and penetration enhancers employed for Leu-Enk permeation study were amastatin(AM), thimerosal(TM) and ethylenediaminetetraacetic acid disodium salt(EDTA), and sodium taurodihydrofusidate (STDHF). Those for YAGFL permeation study were TM, benzalkonium chloride(BC) and EDTA, and STDHF, sodium deoxycholate(SDC), sodium glycholate(SGC), glycyrrhizic acid ammonium salt (GAA), L-$\alpha$-Iysophosphatidylcholine(LPC) and mixed micelle (MM, STDHF: linoleic acid = 15 mM : 5 mM). The addition of TM alone on the donor and receptor solutions for Leu-Enk permeation study across all the three kinds of mucosae failed to inhibit the degradation; it completely degraded in 6 hrs, and no permeation occurred. However, with addition of three kinds of inhibitors together, the fluxes across nasal, rectal and vaginal mucosae were $\20.7{pm}2.5$>/TEX>,$\0.3{pm}0.05$>/TEX> and $\1.4{pm}0.5$ $\mu$\mid$textrm{m}$/$\textrm{cm}^2$/hr, respectively. Moreover, the addition of STDHF in the presence of the above three inhibitors enhanced permeation across nasal, rectal and vaginal mucosae 1.3, 15 and 1.3 times, respectively. YhGFL also degraded in the donor and receptor solutions rapidly as time went. With mixed inhibitors of TM and EDTA, the percents of YAGFL remaining in the donor solutions facing nasal, rectal and vaginal mucosae were 69.7, 69.8 and 79.8%, respectively; the percent permeated increased to 10, 2.1 and 5.7%, respectively. The addition of STDHF in the presence of either BC/EDTA or TW/EDTA increased the permeation 2.2, 11.0 and 2.9 times, and 2.21, 14.0 and 2.7 times for nasal, rectal and vaginal mucosae, respectively. With SDC, SGC, GAA, LPC ud MM in the presence of TM/EDTA increased permeation; especially, they increased permeation across vaginal mucosae effectively, and the enhancement factors were 12.5, 7.6, 8.7, 5.7 and 5.5, respectively. The degradation extent of YAGFL was correlated with protein concentrations in the epidermal and serosal extracts. The flux of YAGFL across nasal mucosa increased dose-dependently.

Characteristics of Crude Polysaccharide Separated from the Herbal Medium of Trichloloma Matsutake Mycelium and its Anti-diabetic Effect

  • As part of studies to develop new materials to lower blood glucose levels using crude polysaccharide, this study was attempted to analyze the characteristics of crude polysaccharide obtained from the extracts of a mixed herbal medium(OCM) where Trichloloma matsutake mycelium and Cordyceps militaris mycelium were cultured together and to look into the influence of administering these by concentration upon the blood glucose and serum lipid levels of rats with diabetes which was induced by STZ(Streptozotosin). Experimental group was divided into 6 groups: first, it was divided into normal control group(NC group) and diabetes-induced group, and diabetes-induced group was subdivided into diabetic control group(DC group), acarbose-treated group(PC group), 100 mg/kg/body weight-treated by crude polysaccharide of OCM(UE) group(UE100 group), 200 mg/kg/body weight-treated group(UE200 group), and 300 mg/kg/body weight-treated group(UE300 group). In diabetic-induced groups, after streptozotocin was melted in 0.01M citrate buffer at 50 mg/kg/body weight, when the non-fasting blood glucose level not on an empty stomach was 300 mg/dl or more in blood collected from the tail vein, it was regarded as diabetic induction and then such diabetic-induced experimental animals were used in this experiment. The yield of crude polysaccharide obtained from OCM was found to be 0.31% and the ${\beta}$-glucan content 39.40%. As a result of analyzing NO on immune function, which is known as major physiological activity of crude polysaccharide, high NO viability was shown; when 1 mg/ml LPS was treated at 1 ug/ml, it was found to be 50.77 uM, and when LPS was treated at 10 ug/m, it was found to be 53.78 uM. Also, regarding cancer cells, cell count was decreased by about 26% in proportion to sample concentration, while for normal cells, it was a little decreased in proportion to concentration, however, cell count was maintained in the range of $81.92{\sim}98.16%$ at all concentrations. In case of blood glucose level, it was decreased in all extract-treated groups compared to DC group and in the cases of ALT and AST, they were found to be lower in extract-treated groups compared to PC group and for serum lipid, it was found to be lower in UE100 group compared to PC group. Thus this study tried to utilize these results as fundamental data for development of preventive and therapeutic agents against diabetes as well as functional foods using the crude polysaccharide of mushrooms.

Effects of Enzyme Treatments on Quality and Yields of Barley Tea (보리의 효소처리가 보리차의 품질 및 수율에 미치는 영향)

  • In order to improve the quality and yields of barley tea(water extracts), enzymatic hydrolysis was carried out with using three kinds of mired enzymes of ${\alpha}-amylase,\;{\beta}-amylase$ and protease. The barley treated with enzymes were the crushed , soaked and slightly roasted(light brown) Youngsanbori(hulless) and the physical, chemical and sensory characteristics were investigated. The results showed that enzymatic hydrolysis caused a signigicant increase in solid yields (80%) of barley tea, particularly with the mixed enzyme 5,000 which has higher activity of ${\alpha}-amylase$ than the other enzymes. The intrinsic viscosity of barley tea was reduced by enzymatic hydrolysis as the value of dextrose equivalent increased The Hunter L, a, b values was lower for the enzyme treated barley tea but showed not significant difference with reaction time. The organoleptic evaluation clearly showed that both intensity and acceptability of odor and taste was markedly Increased. The roasted nutty, and sweety odor and taste were particular in increase by mixed enzyme 5,000.

Neutralization of Rice Hull Charcoal with Nitric Acid Solution and its Neutritional Effect on Tobacco Seedling (연초용(煙草用) 상토재료(床土材料)로서의 왕겨숯(燻炭)의 질산중화효과(窒酸中和效果))

  • Rice hull was reduced to ash by carbonization grades to illcuidate alkalinity increase and extract of inorganic nutrients in the rice hull charcoal. Alkaline reaction of water extraction was neutral at less carbonized charcoal, but much carbonized ash from 65% weight loss reached over 10 of pH value, also origin shape of rice hull was maintained until near 65% carbonized grade. Therefore, physical properties sustained good condition for seedling bed. The more charcoal carbonized to ash, the pH value and concentration of inorganic nutrient in their extracts were increased gradually. Nitric acid concentrations for neutralizing extract from charcoal were stronger in proportion to the carbonized grade but 0.1 N nitric acid solution was very reasonable to neutralize the 65% carbonized charcoal for mixing with heavy texture acidy soil(pH 5.3) of uncultivated deep horizon to transplant the tobacco seedlings. Volume ratio mixing for seedling bed is adequate at five of ash to one of acid solution. Neutralization with nitric acid solution also accelerated extraction of the inorganic nutrient in rice hull ash. Tobacco seedlings grown on bed mixed with neutralized rice hull charcoal and soil had shown better results on the agronomic measurement than alkaline ash bed, and phosphorus and cations were uptaken more amounts.

  • PCBs had numerous uses such as hydraulic fluid, heat exchange fluid, sealant, lubricant, and carbonless copy paper. They are most likely found in electric utilities, power stations, industrial facilities, electronic manufacturing plants, petrochemical plants, railroad systems, electric equipment repair facilities, mining sites (active or abandoned), and military camps. Due to its outstanding chemical and thermal stabilities and electrical insulation properties, the commercial and industrial products of PCBs, such as Aroclors, Kaneclors, Clophens, Phenaclors etc., had been widely used as thermal oil and transformer oil from 1930s until the 1970s. The transformer oils were analyzed as a main source of polychlorinated biphenyls (PCBs) emission into the environment. Qualitative estimation of oil extracts as carried out with Aroclor 1242, 1248, 1254, 1260. The transformer oils contained the pure and mixed of Aroclor 1242, Aroclor 1254, and Aroclor 1260. Also, commercial screening kit of 20 ppm and 50 ppm were applied to the transformer oil samples.

Double-talk Control using Blind Signal Separation based on Geometric Concept in Acoustic Echo Canceller (음향반향제거기에서 기하학적 개념의 BSS를 이용한 동시통화 제어)

  • This paper describes an acoustic echo canceller with double-talk using BSS(: Blind Signal Separation) based on the geometric concept. The acoustic echo canceller may be deteriorated or diverged during the double-talk period. So we use the blind signal separation to detect the double talking by separating the near-end speech signal from the mixed microphone signal. In the closed reverberation environment, the blind signal separation extracts the near-end signal from unknown signals with the transformation and rotation based on the geometric concept. By this method, the acoustic echo canceller operates irrespective of double-talking. We verified performances of the proposed acoustic echo canceller by computer simulations. The results show that the acoustic echo canceller with this algorithm detects the double-talk periods thoroughly, and operates stably in the normal state without diverging of coefficients after ending the double-talking.

  • Android malware steals users' private information, and embedded unsafe advertisement (ad) libraries, which execute unsafe code causing damage to users. The majority of such traffic is HTTP and is mixed with other normal traffic, which makes the detection of malware and unsafe ad libraries a challenging problem. To address this problem, this work describes a novel HTTP traffic flow mining approach to detect and categorize Android malware and unsafe ad library. This work designed AndroCollector, which can automatically execute the Android application (app) and collect the network traffic traces. From these traces, this work extracts HTTP traffic features along three important dimensions: quantitative, timing, and semantic and use these features for characterizing malware and unsafe ad libraries. Based on these HTTP traffic features, this work describes a supervised classification scheme for detecting malware and unsafe ad libraries. In addition, to help network operators, this work describes a fine-grained categorization method by generating fingerprints from HTTP request methods for each malware family and unsafe ad libraries. This work evaluated the scheme using HTTP traffic traces collected from 10778 Android apps. The experimental results show that the scheme can detect malware with 97% accuracy and unsafe ad libraries with 95% accuracy when tested on the popular third-party Android markets.

Studies on Antimutagenic and Lipotropic Action of Flavonoids of Buckwheats -Desmutagenic Activity of Buckwheat Leaf Extracts (메밀 Flavonoids의 항돌연변이원성 및 지질대사 조절기능에 관한 연구 -메밀 잎 에탄올 추출물의 항돌연변이원성 연구-)

  • In spore rec-assay using B. subtillus H17(rec) and M 45(rec) , the ethanol extract of buckwheat leaves showed antimutagenicity in condition of low concentrations, but its did comutagenicity in condition of high concentrations. In Ames test, the ethanol extract of buckwheat leaves reduced the mtabenicity of N-methyl-N' -nitro-N-nitrosoguaidine (MNNG), benzo (a) pyrene(B($\alpha$)P), 2-amino-fluorene(2AF), and 3-amino -1, 4-dime-thyl-5-H-pyrido(4, 3-b) indol(Trp-P-1) in Salmonella typhimurium TA98 and TA100. The ethanol extract was fractionated by hexane, ethylacetate, butanol and water. Among Them hexane fraction showed the highest inhibition rate on the mutagenicity of B($\alpha$)P, and so did chloroform fraction on the mutagenicity of MNNG in S. typhimurium Ta98 and TA100. To elucidate the antimutagenic mechanism of the ethanol extract, it was mixed and co-incubated with various metagens, S9 mix, and the bacteria with different experimental orders and different reaction times. The ethanol extract did not affect reversion rate of pre-mutated. S.typhimurium. However, when the ethanol extract was added to the mutagens before their interaction with S.typhimurium , it reduced the mutation rate to 152$\pm$12-273$\pm$18 colonies/plates in case of MNNG, and 135$\pm$13-195$\pm$10 colonies/ plates in case of B($\alpha$)P), showing strong desmutagenic activity.

Effects of Various Salt Concentrations on Physicochermical Properties of Brined Cucumbers for Pickle Process (피클제조를 위한 취청오이의 염농도에 따른 염장중 이화학적 특성)

  • Changes of physicochemical properties of brined cucumber were investigated in the samples mixed cucumber and water at 1 : 1.2 ratio in various salt concentrations from 15 to 30%. As salt concentrations in brine solutions were increased, cucumber was getting constricted and decreased in the size of diameter. Salt concentrations of brined cucumber were reached an equilibrium after 30 days of brine storage. When cucumber was preserved at 30% salt concentration, pH was gradually decreased until 60 days of brine storage. When cucumber was preserved in 15% salt solution, the yellowness on the surface of cucumber peels was the most intense among the groups. Among the groups preserved by more than 20% of salt, no difference was observed in intensity of yellowness after 30 days of brine storage. The absorbances of isopropyl alcohol extracts at 410, 505, 607, and 665 nm were different from those of raw cucumbers. Regardless of salt concentration, the hardness of cucumber was increased as preservation was prolonged except the group preserved in 15% salt solution.
