Adiponectin Gene Cloning and Its Expression in Insect Cell Expression System

  • This study was to examine expression of the recombinant full-length adiponectin (recombinant adiponectin) in insect ovarian cell culture system and to characterize structural properties of the recombinant adiponectin secreted in medium. Gene construct encoding the recombinant adiponectin contained N-terminal collagen-like domain (110 Amino Acids, AAs), C-terminal globular domain (137 AAs) and C-terminal peptides for detection with V5 antibody (26 AAs included adaptor peptide) and purification using the 6xHis tag (6 AAs). The approximate molecular weight of the product (monomer) was 35 kDa. Molecular mass species of the expressed recombinant adiponectin were monomer (~35 kDa), dimer (~70 kDa), trimer (~105 kDa) and hexamer (~210 kDa). The major secreted species were the LMW forms, such as monomer, dimer, and trimer. There was MMW of hexamer as minor form. HMW multimers (~300 kDa) were shown as a tracer or not detected on the SDS-PAGE in several experiments (data not shown). The multimer forms in this study were not compatible to those in animal or human serum and adipose tissue by other researcher's study in which the major multimer forms were HMW. By protein denaturing experiments with reducing reagent (${\beta}$-MeOH), anionic detergent (SDS) and heat ($95^{\circ}C$) on the SDS-PAGE, not all adiponectin multimers seemed to have disulfide bond linked structure to form multimers. The recombinant adiponectin which expressed in insect ovarian cell culture system seemed to have the limitation as full physiological regulator for the application to animal and human study.

Chiari 1.5 Malformation : An Advanced Form of Chiari I Malformation

  • The Chiari 1.5 malformation is defined as a tonsillar hemiation within a Chiari I malformation with additional caudal descent of the brainstem through the foramen magnum. We describe a patient with Chiari I malformation who evolved to Chiari 1.5 malformation during longitudinal follow-up. A 15-year-old girl presented with neck pain during exercise for two years. She had been diagnosed with Chiari I malformation with mild hydrocephalus after minor cervical trauma at the age of six years. At that time, she was asymptomatic. After she complained of aggravated neck pain, neuroimaging (nine years after first imaging) revealed caudal descent of the brainstem and syringomyelia in addition to progression of tonsillar hemiation. Posterior fossa decompressive surgery resulted in complete resolution of neck pain. Based on neuroimaging and operative findings, she was diagnosed as Chiari 1.5 malformation. Neuroimaging performed seven months after surgery showed an increased anterior-posterior diameter of the medulla oblongata and markedly decreased syringomyelia. This case demonstrates progressive developmental process of the Chiari 1.5 malformation as an advanced form of the Chiari I malformation.

A Study on the Plane Spatial Characteristics of Modern Hanok in the Jeonju Hanok Village using Space Syntax (Space Syntax를 응용한 전주한옥마을 근대한옥의 평면공간특성에 관한 연구)

  • The residential space of humans has continuously developed to the most suitable form considering the natural and social environment. Based on this, it has become a unique residential architectural culture of an area. In the architectural field, the space of residential structures is being categorized topological aspect and the quantitative indicators are being calculated to conduct an objective comparative analysis of the characteristics of space by regional groups and individual rooms. Hence, the purpose of this study is to investigate the characteristics of the spatial composition of Hanok which a modern residential structure connecting traditional residential architecture with modern residential architecture. Hanok generally had a similar spatial composition to that of the traditional Hanok. However, a minor change was spotted due to it being a modern Hanok. It was objectively determined through the environmental characteristics that this form of spatial composition is due to the influence of the Japanese colonial era as the Western lifestyle of inner space centered lifestyle was introduced in the same period that the Jeonju Hanok was mostly created. The sequence of topological spatial gene of Hanok in Jeonju Hanok Village appeared in the order of Toetmaru - Madang - Anbang - Geonneonbang - Meorit bang - Kitchen.

Patterns of Leaf and Leaf Growth in Arisaema robustum Population (넓은잎천남성 (Arisaema robustum) 개체군의 잎과 잎생장 패턴)

  • The Arisaema robustum, perennial non-stem herb, population in the temperate forest of Sanseong-ri, Joongbu-myeon, Kyonggi Province was studied for leaf pattern and leaf growth from 1993 to 1995. The area of each side leaflet was larger than that of the apical leaflet in 3-leaflet form plants. But there was no significant difference among the leaflets (apical, 1st side and 2nd side leaflet) in t-testing of the 5-leaflet form plants. The specific leaf area (SLA) of the 3-leaflet form plant was greater than that of the 5-leaflet form plant, and the difference was significant at 0.1%. But differences of SLA value among the leafleats of the same leaflet form plants were minor. The area of leaflets or total leaf area was inversely proportioned to the their SLA values. Especially, the correlation coefficients (CC) between leaf area and SLA in apical leaflets and side leaflets of the 3-leaflet form plant compared to the 1st side leaflets of the 5-leaflet form plants were significant at the level of 1%. The differences between two sides in the dry weight and area of leaflets starting from the apical leaflets were about 5%. The CC values between attributes of the petiole (length and dry weight) and those of leaf blades (dry weight, leaf area and SLA) were high and significant at a 1% level. Especially, The CC value between the dry weight of the petioles and that of the leaf blades was higher than any others (r=0.952). The morphologies of the leaf margins were entire, medium serrate and serrate, and the same plant showed the same type from year to year. The plants usually had one leaf and occasionally 2 leaves. 1-leaf plants usually had 3 or 5 leaflets and occasionally 2 or 4 leaflets. Each year, the ration of individuals having the same number of leaflets was 72%, and that of individuals having a different number of leaflets was 27.54% (an increase of 20.4% and a decrease of 7.1% respectively). In the individuals increasing in the number of leaflets, the ration of individuals shifting from the 3-leaflet from to the 5-leaflet form was 14%. The increase ration of leaf area per individual a year was 37%, and the increase ratios of the plants increasing in leaflets were 60-70% regardiness of leaflet increase. The ratios of leaflet length/breadth were about 1-2, and the variations of those were 0.182-0.286 each year (12-20%).

  • Child abuse is defined by a recent act or failure to act that results in death, serious physical or emotional harm, sexual abuse or exploitation, or imminent risk of serious harm; involved a child; and is carried out by a parent or caregiver. This report provides guidance in the clinical approach to the evaluation of suspected physical abuse in children, and role of pediatrician. The medical assessment is outlined with obtaining a medical history, physical examination, and diagnostic testing. A minor form of child abuse which only involves skin injury is most frequently seen by a pediatrician. This kind of child abuse can be followed by more severe forms of child abuse, which have high mortality rates and cause serious physical and mental sequelae to the survivor. Therefore, a pediatrician's role in an early detection and prevention of child abuse is very important.

  • In summary, progesterone is probably the only naturallly occurring progestational agent of any significance. Small amounts may be synthesized by the cells of the follicle in the provulatory swelling phase; however, the major production is by the corpus luteum cells of the ovary during the luteal phase of the cycle. It is constantly produced in small amounts by the adrenal gland and by the testis in the male. In the adrenal and the testis, it probably serves as the precursor for corticoids and androgens. It is transported in the blood by a specific binding protein and metabolized and conjugated in the liver into sodium pregnanediol glucuronide which also circulates in the blood. Approximately 20% is excreted in the urine as sodium pregnanediol glucronide; pregnanolone represents a minor metabolic product. The pregnanediol which is excreted in the bile is enzymically hydrolyzed by the gut so that the pregnanediol recovered in the feces is in the free form.

AN OBSERVATION ON THE FRACTURE SYSTEMS OF THE SOUTHERN VIETNAM

  • A study of the fracture systems in outcrops of southern onshore Vietnam revealed two kinds of fracture groups according to their origin: cooling fractures and deformation related fractures. Cooling of magma introduced extensive fractures in the batholiths with wide spacing and narrow aperture. They are found widespread in all magmatic bodies, but result in poor reservoir quality due to low bulk porosity and narrow aperture. Cooling fractures are often reactivated during later stress regimes. Deformation related fractures, especially 'fault damage zones' and 'hanging wall deformation' is thought to form the most important reservoir type in the fractured basement rock. The porosity formed by intense fracturing and fault breccia along minor fault zones is thought to be the producing zones in the producing fields of Cuu Long basin. They are found along major faults and widespread in hanging wall blocks.

Measurement of Physical Properties of Korean Garlic for Grade Standard

  • Garlic is one of the major seasoning vegetables in Korea and consumed mostly in a form of peeled cloves. Conventional Korean standards for garlic grading consist of four classes according to the size of bulb and its shape. Sorting and grading of garlic are manually done but could be in the process of automated mechanization using machine vision system in the near future. The proportion of mass of cloves in a garlic bulb to the volume of the bulb (g/ml) was determined to find out the best way of representing both the quantity and quality of cloves in each bulb. Garlic bulb was assumed as an ellipsoid and its major and minor axis and its height were measured to calculate its volume. The mass proportions and density of a garlic bulb and cloves were measured for four domestic varieties of garlic to propose it as a standard for Korean garlic grading machine.

Recurrent Plasmacytoid Myoepithelioma of the Lung - A case Report - (재발성 형질세포양 세포성 폐 근상피종 -1례 보고-)

  • A recurrent myoepithelioma of the lung in a 36-year-old man is reported. The neoplasm showed histologic features identical to those described in myoepitheliomas of major and minor salivary glands on the basis of Dardick's morphological classification of Myoepitheliomas. He was treated totally with surgical en-bloc resection including the chest wall. The tumor was found to be well encapsulated, and it appeared to be mainly composed of plasmacytoid cells and clear cells with occasional microcystic spaces in a solid growth form by light microscopy. Immunocytochemical, ultrastructural and flow-cytometrical studies supported myoepithelioma differentiation.

A Case of Serious Caustic Injury on Gastrointestinal Tract after Ingestion of Sodium Carbonate Peroxyhydrate (과탄산 나트륨 음독 후 발생한 심한 위장관 손상 1례)

  • Laundery detergent ingestions are generally considered to have minor consequences. But some detergent ingestions have severe consequences. Unlike household bleaches contain sodium hypochlorite, bleaching agents that contain sodium carbonate have the potential to cause significant mucosal damage to the gastrointestinal tract if ingested. Especially, when ingested in solid form of Sodium carbonate, corrosive injury is much heavier. Therefore, patient who ingest sodium carbonate peroxyhydrate need more intensive management than patient who ingest other bleaches. We experienced a case of serious caustic injury after ingestion of Sodium Carbonate Peroxyhydrate. We report this case with review of literature.
