휘발성 Allyl Isothiocyanate계 화합물의 항균 활성에 관한 연구 (Antimicrobial Effects of Allyl Isothiocyanates on Several Microorganisms)

  • 천연에서 발견되는 휘발성 물질인 isothiocyanate계(ITCs) 물질을 대상으로 Listeria monocytogenes, Bacillus subtilis, Pseudomonas fluorescens, Escherichia coli, Erwinia carotovora, Saccharomyces cerevisiae, Candida albicans, Aspergillus oryzae and Penicillium roqueforti 등 9종의 균주에 대한 증식 억제 효과를 측정하였으며 초산과 혼합하여 사용할 때 상승효과를 비교하였다. Allyl isothiocyanate (AIT), benzyl isothiocyanate (BIT) 및 ethyl isothiocyanate (EIT)가 대상 균주 모두에 $100{\sim}200\;{\mu}g/dish$ 수준의 최소증식저해농도(MIC)를 보였고 증식 저해 정도는 곰팡이>효모>Gram 음성 세균>Gram 양성 세균 순 이었다. 초산은 $50{\sim}500\;{\mu}g/dish$로 ITCs 보다 MIC가 낮았다. ITCs와 초산을 혼합하여 사용하는 경우 대부분의 실험 대상 균주에 대하여 증식 저해 상승효과가 인정되었고 AIT의 경우 AIT 단독 또는 초산 단독으로 사용할 때보다 $2{\sim}10$배 이상 효과가 상승하였으며 전체적으로 증식 억제 경향은 ITCs 단독 사용할 때와 같은 경향이었다.

Antibacterial and therapeutic effects of a combination of Sophora flavescens and Glycyrrhiza uralensis Fischer ethanol extracts on mice infected with Streptococcus pyogenes

  • This study evaluated the antibacterial effects of a mixture of Sophorae radix and Glycyrrhiza uralensis Fischer (1 : 1) ethanol extracts (SGE) on mice infected with Streptococcus (S.) pyogenes. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration of SGE necessary for antibacterial effects against S. pyogenes were $20{\mu}g/mL$. Based on the time-kill curves for S. pyogenes, SGE was effective at $4{\times}$ MIC after 16 h. On Day 12 after challenge, the survival rate of mice treated with 2.0 mg/kg SGE was 60%. In conclusion, SGE had potent in vitro and in vivo antibacterial activities against S. pyogenes.

Antibacterial activity of Dioscorea batatas, Morus alba, and Tagetes erecta against E. coli and S. aureus

  • In this study, the anti-bacterial activity of three medicinal herbs such as Dioscorea batatas, Morus alba, and Tagetes erecta, have been applied in oriental medicine against Escherichia coli and Staphylococcus aureus. The antibacterial activity of Dioscorea batatas ethanol extract (DBEE), Morus alba ethanol extract (MAEE), and Tagetes erecta ethanol extract (TEEE) was analyzed by disk diffusion, minimum inhibitory concentration (MIC), and minimum bactericidal concentration (MBC) assays. As a result, three medicinal herbs exhibited antibacterial activity in a dose-dependent manner as well as MAEE and TEEE showed the most potent antibacterial activity followed by disk diffusion assay against E. coli and S. aureus, respectively. MIC against E. coli and S. aureus was 1.6 mg/mL of MAEE and TEEE as well as 0.2 mg/mL of TEEE. MBC against both strains was 5 mg/mL of DBEE and 2.5 mg/mL of TEEE. Consequently, three medicinal herbs in this study showed potent antibacterial activity through the inhibited growth of E. coli and S. aureus, which is considered as a potential candidate for cosmeceutic agents to attenuate the pruritus. In addition, antibacterial and antihistamine effects followed by the extraction solvents should be analyzed in future studies.

송이버섯 추출액의 항균력 확인과 저염젓갈 생산에 적용 가능성 (Determination of Antibacterial Activity from Tricholoma matsutake Extract and Its Application to Low Salted Jeot-gal)

  • 송이버섯 추출액의 박테리아에 대한 항균력을 측정하기 위하여 최소저해농도를 확인한 결과 젓갈 제조시 빈번하게 발생하는 부패균, 어병원인 균등 9가지 박테리아에 대해 성장이 저해되는 것을 확인하였다. 확인된 송이버섯 추출액의 항균력을 저염 젓갈의 제조 공정에 적용하기 위해 식품보존제인 사과산나트륨과 아질산나트륨을 함께 첨가하여 S. aureus에 대한 최소저해농도를 확인하였다. 그 결과 사과산나트륨과 아질산나트륨의 최소저해농도가 각각 0.25 g/L와 0.025 g/L로 크게 낮아지는 것을 확인할 수 있었다. 본 연구를 통해 항균력을 갖는 송이버섯 추출액을 첨가함으로써 일반적으로 첨가되는 식품보존제의 농도를 크게 낮출 수 있었을 뿐 아니라 송이버섯의 다양한 기능성이 추가된 고급화된 저염 젓갈을 개발할 수 있을 것으로 예상된다.

Restoring Ampicillin Sensitivity in Multidrug-Resistant Escherichia coli Following Treatment in Combination with Coffee Pulp Extracts

  • Escherichia coli, particularly multidrug-resistant (MDR) strains, is a serious cause of healthcare-associated infections. Development of novel antimicrobial agents or restoration of drug efficiency is required to treat MDR bacteria, and the use of natural products to solve this problem is promising. We investigated the antimicrobial activity of dried green coffee (DGC) beans, coffee pulp (CP), and arabica leaf (AL) crude extracts against 28 isolated MDR E. coli strains and restoration of ampicillin (AMP) efficiency with a combination test. DGC, CP, and AL extracts were effective against all 28 strains, with a minimum inhibitory concentration (MIC) of 12.5-50 mg/ml and minimum bactericidal concentration of 25-100 mg/ml. The CP-AMP combination was more effective than CP or AMP alone, with a fractional inhibitory concentration index value of 0.01. In the combination, the MIC of CP was 0.2 mg/ml (compared to 25 mg/ml of CP alone) and that of AMP was 0.1 mg/ml (compared to 50 mg/ml of AMP alone), or a 125-fold and 500-fold reduction, respectively, against 13-drug resistant MDR E. coli strains. Time-kill kinetics showed that the bactericidal effect of the CP-AMP combination occurred within 3 h through disruption of membrane permeability and biofilm eradication, as verified by scanning electron microscopy. This is the first report indicating that CP-AMP combination therapy could be employed to treat MDR E. coli by repurposing AMP.

수종 한약재 추출물의 Staphylococcus aureus에 대한 항균활성 (Antibacterial activity from medicinal plant extracts on the Staphylococcus aureus)

  • 수종의 한약계를 70% 메탄올로 추출하여 식중독 세균인 Staphylococcus aureus에 대한 항균활성으로 생균수의 측정, 생육저해환의 측정, 최소저해농도와 최소사멸농도를 조사한 결과 이들 추출물의 Staphylococcus aureus에 대한 항균활성의 크기는 Rubus coreanus > Sanguisorba officinalis > Eriobotrya japonica > Prunus mume > Crataegus pinnatifida로 나타났다. 생육저해환은 복분자가 16.5 mm, 지유 14.3 mm, 오매 14.0 mm, 산사 12.7 mm로 측정되었다. MIC는 지유가 5mg / ml로 가장 낮았으며, MBC는 복분자가 10mg / ml로 나타났다. 실험에 사용된 한약재 추출물중 복분자, 비파엽, 산사, 오매, 지유는 Staphylococcus aureus에 대해서 강한 항균활성을 나타내었다. 따라서 이들 추출물을 이용한 천연보존료를 개발하여 식품에 적용한다면 그 기대효과가 상당히 클 것으로 생각된다.

Inhibitory Effect of Corn Silk Extract on Growth of Food-Borne Bacterial Pathogens

  • Various levels of antibacterial activity have been identified for water and ethanol extracts of corn silk, particularly against Salmonella typhimurium KCTC 2515. In general, the water extract was more effective than the ethanol extract. The minimum inhibitory concentration (MIC) for the water extract was 7.5 mg/disc for S. typhimurium KCTC 2515 and B. cereus KCTC 1092, as well as for the ethanol extract against S. typhimurium KCTC 2515 and S. typhimurium KCTC 1925. However, the MICs for the water extract were lower than those for the ethanol extract against all bacteria tested, except S. typhimurium KCTC 1925 and B. cereus KCTC 1014. The growth of the tested organisms in the synthesized broth medium was inhibited with the addition of 5-fold levels of MIC. Using sterilized milk as the model food system, we found that the lag phase for these microorganisms was extended up to 3 days at $20^{\circ}C$, but was not affected at $4^{\circ}C$. These results indicate that bacterial growth was strongly inhibited by corn silk extract at $20^{\circ}C$.

One Pot Synthesis of Bioactive Novel Cyanopyridones

  • Cyanopyridone was prepared by the condensation of cyanoacetamide, substituted arylaldehydes and malononitrile in presence of pipyridine. The structure of the synthesized compound CP 1-20 was assigned on the basis of elemental analysis, IR, $^1H$-NMR and mass spectroscopy. These compounds were also screened for antimicrobial activity. The Minimum Inhibitory Concentration (MIC) of all the synthesized compounds was compared with standard drugs.

  • Acanthamoeba strains were photoirradiated in the presence of light-sensitive organic peroxides (hydroperoxynaphthalimide derivatives) including a Photo-Fenton reagent at 366 nm. The survival rates of Acantharnoeba strains determined after 20 h culture showed a significant decrease only upon photoirradiated conditions. The most effective hydroperoxynaphthalimides among these compounds was the bromohydroperoxynaphthalimide (Br-HPO II). The minimum inhibitory concentration (MIC) of Br-HPO II is 100 times lower than that of hydrogen peroxide.

Fungistatic Activity of Kojic Acid Against Human Pathogenic Fungi and Inhibition of Melanin-production in Cryptococcus neoformans

  • Kojic acid was investigated for its antifungal activity against the human pathogenic fungi including Candida albicans, Cryptococcus neoformans and Trichophyton rubrum. For C. albicans, C. neoformans and T. rubrum, the MIC(minimum inhibitory concentration) of kojic acid was 640, 80 and 160 ${\mu}g/ml$, respectively. In C. neoformans, melanin-producing yeast, kojic acid-treated nonmelanized cell was more susceptible to magainin than melanized cell, suggesting melanin give a protective function against microbial peptide.
