MTT 방법에 의한 항진균성 활성효과의 측정

  • In this study, we show a convenient MTT assay for detect the susceptibility of yeast-like form of Trichosporon beigelii against antifungal agents. This assay was developed based on mitocondrial respiration by determining reduction of 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) to formazan. Cells of T beigelii are seeded into 96-well microtiter plates, and antifungal agents, amphotericin B, magainin and CA-ME hybrid peptide were added with various concentrations. After 24 hr incubation, MTT was added, then incubations were continued for 4 hr. Formazan formation was quantified photometrically after extraction of the formazan with acid sodium dodesyl sulfate (SDS). From this assay, we could obtained MICs of antifungal agents against T. beigelii. The presented method can easily be used as an effective methods to assess the antiftingal action of various agents on yeasts with minimal amounts of antifungal agents.

  • This paper presents a tuning-free conditional integration anti-windup strategy for induction machine with Proportional-Integral (PI) type speed controller. The on/off condition of integral action is determined by the frequency domain analysis of machine torque command without a prior knowledge of set-point changes. There are no tuning parameters to be selected by users for anti-windup scheme. In addition, the dynamic performance of the proposed scheme assures a desired tracking response curve with minimal oscillation and settling time even in the change of operating conditions. This algorithm is useful in many high performance induction machine applications not to allow the oscillation and overshoot of speed/torque responses. The main idea can be extended to general applications such as chemical processes and industrial robots.

  • Using the isolated perfused rat heart this study investigated 1) the cardiac uptake of idarubicin (IDA), 2) the role of P-glycoprotein (P-gp) in the uptake process, 3) the formation of IDOL from IDA in the heart, and 4) the effect of P-gp inhibitors (verapamil, amiodarone, PSC 833), doxorubicin, hypothermia, xanthine derivatives (caffeine, theophylline) and metabolism inhibitors (rutin, phenobarbital) on the pharmacokinetics and pharmacodynamics of IDA using a mathematical modeling approach. A minimal model was constructed; the differential equations were numerically solved and fitted to the data using the ADAPT II-software package using maximum likelihood estimation assuming that the measurement error has a standard deviation which is a linear function of the measured quantity[1].   • Approximately equivalent to character disorder or character neuroses or behavioral reaction. For a discussion of the term as used currently, in the 1968 revision of psychiatric nomenclature(DSM-II). In DSM-I(1952 nomenclature), this term referred to those cases in which the personality, in its struggle for adjustment to internal and external stresses, utilized primarily a pattern of action or behavior rather than symptoms in the mental, somatic or emotional spheres. There is minimal subjective anxiety and little or no sense of distress. As thus defined, there are three main groups of personality disorder, personality pattern disturbance, personality trait disturbance and sociopathic, personality disturbance. Morton Prince asks whether a subconscious process can perform the same function as are ordinarily performed by conscious intelligence that is to say memory, perception, reasoning, imagination, volition, affectivity, etc? He cites clinical material at great length, concluding that the quality of the functions performed they frequently exhibit, that which is characteristic of intelligence.

  • Engineers are always concerned with life cycle costs for making important economic decisions through engineering action like reliability of products. Decisions during the reliability growth development of products involve trade-offs between invested costs and its returns. In order to find minimal LCC containing the reliability improvement cost, production cost, repair and replacement costs, and holding cost of spare parts for failure items we suggest in this paper relationship between development cost and sustaining cost in values of growth parameter $\beta$ of AMSAA model. This model is applied to the reliability growth program based on AMSAA model during R&D phase, the warranty activities of items and the block replacement policy for maintenance of items in avionic equipment.

  • Recently combining regimen of minimal doses of chloral hydrate, hydroxyzine and midazolam is good in sedation of children. Midazoalm, water soluble benzdiazepine, has rapid onset and relatively short duration of action. And midazolam has prospective amnesic effect. With these advantages midazolam is popular in conscious sedation for children. This study was to reveal the dose-dependency of behavior and physiologic effects of submucosal midazolam. Sedation records were surveyed retrospectively, of which the patients admitted from April, 2005 to July, 2007. we assigned three groups according the dose of midazolam, 0.1 mg/kg, 0.2 mg/kg and 0.3 mg/kg, respectively and the behavioral evaluation was analyzed with Houpt scale statistically. Combined submucosal midazolam increased the success rate in sedation and the vital signs were stable during sedation.

  • This study evaluates user experience of streaming stick service through usability testing. Streaming stick services such as Roku, Chromecast, and Everoncast were tested in ten levels belonging to five usability variables that are composed of efficiency of use, accuracy, meaningfulness, flexibility, and consistency. Each variable has two levels respectively that are responsiveness, minimal action, error prevention, error detection, error recovery, understandability, learnability, substitutivity, predictability, and familiarity in order. Twenty five participants volunteerly joined this research, and the results showed that Roku is superior to another two OTTs in most levels and variables. It implies that participants prefer to remote control, many channels, and personalized service during experiencing the service.

  • Antimicrobial peptides are widely used in various organisms as a defense system against infection. The peptides are lethal towards bacteria and fungi, however have minimal toxicity in mammalian and plant cells. In this aspect, it is considered that antimicrobial peptides are new alternative materials for defensing against microbial infection. Here, we describe overall characteristics of antimicrobial peptides based on the mechanism of action, classification of the peptides, report detection/screening methods and chemical/biological production. It is expected that understanding of innate immune system based on antimicrobial peptides tends to develop novel natural antimicrobial agents, which might be applied for defensing pathogenic microorganisms resistant to conventional antibiotics.

Anticariogenic activity of piceatannol isolated from Callistemon citrinus fruit against Streptococcus mutans

  • Dental caries is the destruction of the enamel of teeth by Streptoccus mutans. S. mutans has been isolated from human dental plaque and is associated with the initial development of enamel lesions. We have studied the antibacterial action of the fruit of Callistemon citrin us against a cariogenic bacterium, S. mutans. From the fruit of C. citrinus, piceatannol (3,3',4',5-tetrahydroxystilbene) was isolated by repeated column chromatography with $SiO_2$ and Sephadex LH-20. Its structure was elucidated by instrumental analysis using 1D-NMR, 2D-NMR and EI-MS. This compound was isolated from the fruit of C. citrin us for the first time. The anticarcinogenic activity of this compound was determined by using agar well-diffusion method and minimal inhibition concentration (MIC).

  • A criterion and a method for solving a problem on the prevention of mine working fracture under the action of tectonic and gravitational forces are offered. Based on minimal criterion, theoretical analysis of the definition of the optimal shape of working in the rock mass weakened by arbitrarily located rectilinear cracks was carried out. A closed system of algebraic equations allowing to minimize the stress state and stress intensity factors depending on mechanical and geometrical characteristics of the rock, is constructed. The relation between the shape of the working and the stress intensity factors and also location and sizes of the cracks is obtained. The found optimal shape of working increases load-bearing capacity of the rock.
