Tigripus japonicus의 세균 섭이 (Feeding of Bacteria by Copepod Tigriopus japonicus)

  • Tigriopus japonicus를 대량생산하는데 필요한 기초적인 자료를 얻기 위해 무균 배양을 통한 nauplius 유생 단계와 성체의 식성에 대하여 조사하였다. 무균 상태의 성체는 먹이로 공급한 RT 박테리아주를 적극적으로 이용하여, 순조로운 증식을 보였다. 먹이로 공급한 RT 균주의 증식은 포식자 T. japonicus의 섭식에 의해서 그 증식이 크게 억제되었다. RT 균주만을 먹이로 배양한 T. japonicus의 성체가 가지고 있는 난낭안에는 oil bead로 충만해 있었다. 그러나, 무균 상태의 N. oculata만을 먹이로 공급한 경우에는 oil bead를 전혀 관찰할 수 없었다. 무균 조건의 nauplius 유생의 경우에는 먹이로서 공급한 RT 박테리아주를 적극적으로 이용하여 RT 박테리아주의 증식을 크게 억제하였다. 이 결과는 지금까지 T. japonicus가 잡식성인 것만으로 알려진 것과는 달리, 최소한 섭식기구가 미발달된 nauplius의 초기 유생은 사육수중에 세균이 존재하지 않는 경우에는 정상적인 발생이 불가능하였다. 즉, 부화 직후 또는 발생 초기의 T. japonicus의 nauplius 유생은 박테리아식성 (bacterivorous)이었다.

시중에서 유통되는 가쓰오부시의 미생물학적·화학적 위해요소분석 및 안전성 평가 (Risk Analysis and Safety Assessment of Microbiological and Chemical Hazards in Katsuobushi Products Distributed in the Market)

  • For the safety assessment of microbiological and chemical hazards in katsuobushi, fifteen samples of katsuobushi were purchased from supermarkets. The contamination levels of total viable bacteria, coliforms, Escherichia coli, and nine pathogenic bacteria [Staphylococcus aureus, Salmonella spp., Listeria monocytogenes, Bacillus cereus, Vibrio parahaemolyticus, Clostridium perfringens, Enterohemorrhagic E. coli (EHEC), Yersinia enterocolitica and Campylobacter jejuni/coli] were quantitatively or qualitatively assessed. Additionally, the heavy metals (total and methyl mercury) content, radioactivity (131 I, 134 Cs+ and 137 Cs) were quantitatively assessed. Microbial and chemical analyses were performed using standard methods in Korean food code. The contamination level of total viable bacteria was 2.70 (1.18-4.42) log CFU/g. Coliforms, E. coli and S. aureus were not detected in any samples. Other eight pathogenic bacteria were negative in all samples. The contamination levels of total and methyl mercury were 0.366 (0.227-0.481) and 0.120 (0.002-0.241) mg/kg, respectively. In addition, radioactivity was not detected in any samples. The results will be helpful in revitalizing domestic use and boosting exports of katsuobushi because the microbiological and chemical safety of katsuobushi has been assured. Furthermore, the results may be used as a basis for performing chemical and microbial risk assessments of katsuobushi.

Relationship between Obesity, Gingival Inflammation, and Periodontal Bacteria after 4-Week Weight Control Program in 20's

  • Background: Obesity weakens acquired immunity and causes infection. This study aimed to investigate the relationship between the inflammatory markers in the gingival crevicular fluid and serum and periodontal bacteria in saliva through obesity control for 4 weeks. Methods: Forty-six subjects with a body mass index (BMI) of ≥23 kg/m2 stayed in the camp for 4 weeks, followed by exercise and a low salt-low fat diet. Body size measurements, oral examinations, blood, saliva, and gingival crevicular fluid were collected before and after the program. C-reactive protein (CRP) in serum, matrix metalloproteinase (MMP)-8, MMP-9, and interleukin (IL)-1β in the gingival sulcus fluid were measured. After extracting bacterial genomic DNA from saliva, the presence of periodontal bacteria were detected using Taq probe. The relationship of each index before and after the program was analyzed through paired t-test and partial correlation analysis. Results: Campylobacter rectus (Cr) increased after the program, and there was no significant change in other bacteria. Serum CRP and Fusobacterium nucleatum (Fn), Aggregatibacter actinomycetemcomitans, Cr, ratio of Fn, and ratio of Cr had a positive relationship at baseline; however, the relationship was not significant after the program. Ratio of Prevotella intermedia had a positive relationship with MMP-9, MMP-8, IL-1β at baseline. Moreover, the ratio of Treponema denticola and the ratio of Tannerella forsythia showed a positive relationship with MMP-8, MMP-9, and IL-1β. The relationship between the ratio of Porphyromonas gingivalis and IL-1β showed a constant positive relationship at baseline and after the program. Conclusion: Obesity control program in subjects with a BMI of ≥23 kg/m2 accompanied by diet and exercise did not affect the changes in periodontal bacteria itself, but changes in the relationship between periodontal bacteria and serum CRP, the relationship between the inflammatory index in the gingival crevicular fluid and periodontal bacteria was observed.

Bacterial Contamination of Digital Panoramic Dental X-Ray Equipment

  • Background: Digital panoramic dental X-ray equipment (PDX) is frequently used by patients and dental workers for diagnosis and examination in dental institutions; however, infection control has not been properly implemented. Therefore, in this study, we aimed to systematically review the potential risk of cross-infection in the dental environment by investigating the contamination level of general aerobic bacteria and Staphylococcus aureus, which are important in hospital infections, in PDX areas that people mainly contact. Methods: This survey was conducted from March to May 2023 and covered one general hospital, three dental hospitals, and nine dental clinics equipped with PDX. Bacteria samples were collected from the left-handle, right-handle, forehead support, and head side support as the patient's contact areas, as well as the X-ray exposure switch and left-click mouse button as the dental hygienist's contact areas of the PDX. The collected bacteria were spread on Petrifilm, and colonies formed after 48 hours of culture were counted. Results: General aerobic bacteria and S. aureus were detected in all areas investigated. Significant differences in bacterial counts between different regions of the PDX were observed in both groups (p<0.001). The detection rates of general aerobic bacteria (p<0.001) and S. aureus (p<0.001) were significantly higher in the contact areas of patients than those of dental hygienists. A positive correlation was observed between the forehead and the temple region in terms of general aerobic bacteria and S. aureus detection (r=1) (p<0.01). Conclusion: Taken together, the presence of many bacteria, including S. aureus, detected in PDX indicates that PDX has a potential cross-infection risk. Our results therefore highlight the need for the development of appropriate disinfection protocols for reusable medical devices such as PDX and periodic infection prevention training for hospital-related workers, including dental hygienists.

쌀가루 제분용 백미 내 잠재적 위해 세균에 대한 살균 및 세척 효과 (Antibacterial and Rinsing Activities Against Potentially Harmful Bacteria in Rice during Rice Flour Production)

  • 습식 쌀가루의 제분과정 중에서 유해 미생물의 증식을 억제하기 위하여 침지공정이나 세척공정에 사용되는 물에 천연살균제인 쌀뜨물발효액 혹은 화학살균제인 chlorine dioxide (CD)와 sodium benzoate (SB)를 각각 첨가하여 침지공정이나 세척공정 중 쌀에 잔류 미생물의 증식억제 정도를 파악하였다. 또한 원료 쌀의 잔류 미생물을 제거하기 위하여 침지공정 전과 후 세척 횟수별 잔류 미생물의 제거 정도를 파악하였다. 쌀뜨물발효액, CD, 혹은 SB는 백미 내 잔류 가능성이 있는 잠재적 위해 세균들로 선정된 S. typhimurium, E. coli, 그리고 B. cereus 표준균주들에 대하여 액체배양액 내에서 각각 첨가된 농도가 증가함에 따라서 이들 살균제들의 항균력이 비교적 급격히 증가하는 경향을 나타내었으며, 쌀뜨물발효액이나 SB와 비교하였을 때 CD가 액체배양액 내에서 가장 높은 항균 효과를 나타내었다. 또한 쌀뜨물발효액, CD, 혹은 SB는 침지과정 중에 각각 첨가된 농도가 증가하면서 백미 내 잔류세균에 대하여 이들 살균제들의 항균력이 증가하는 경향을 나타내었다. 그러나 이러한 현상은 위에서 언급한 S. typhimurium, E. coli, 그리고 B. cereus 표준균주들에 대하여 액체배양액 내에서 이들 천연살균제와 화학살균제들이 나타낸 항균력보다 낮게 나타났다. 한편 백미 검체의 침지 전과 후 세척 횟수에 따른 백미 잔류 세균의 제거효과는 백미 검체를 침지하지 않고 세척하였을 때가 가장 높은 효과를 나타내었다. 그리고 세척을 한 후 침지를 시켰을 때는 세척 횟수가 증가하여도 잔류 세균의 수가 비슷한 정도로 나타났다. 그러므로 결론적으로 백미의 살균 및 제균 방법으로는 살균제에 의한 살균방법보다는 물 등으로 세척하여 잔류 세균을 제거하는 것이 보다 안전하고 효율적인 방법이 될 수 있을 것으로 사료된다.

A Study on the Rapid Hydrolysis of Fish Using Proteolytic Bacteria Isolated from Anchovy Jeotkal

  • A study on the hydrolysis of anchovy using proteolytic bacteria isolated from anchovy jeotkal (a salt-fermented fish) was carried out to develop a rapid process of liquefied anchovy jeotkal. Five kinds of proteolytic bacteria, such as Staphylococcus sp.-l, Photobacterium sp., Volcaniella sp., Staphylococcus sp.-2 and Bacillus sp., were isolated from the anchovy jeotka1 that fermented with $20\%$ NaCl at room temperature for 2 months. Those grew well at $40^{\circ}C$, pH 7.0 on TPY broth with $2.0\%$ NaCl. The optimal hydrolysis temperature, pH, time and proteolytic bacteria densities for hydrolysis of minced anchovy were$40^{\circ}C$, 7.0, 6 hours and $1.8\times10^8$ cells/g raw anchovy, respectively.

Inhibition of Lactic Acid Bacteria in Kimchi Fermentation by Nisin

  • Sixty isolates of lactic acid bacteria found in kimchi, a traditional Korean dish of fermented vegetables, were tested for nisin sensitivity. Of the sixty isolates, all belonging to the genera Leuconostoc, Lactobacillus, and Pediococcus, fifty isolates were sensitive to nisin at a concentration of 100 IU/$m\ell$, and four isolates appeared to be resistant to nisin. This demonstrated that the nisin sensitivity of lactic acid bacteria found in kimchi varied considerably among isolates. In MRS broth containing nisin at concentrations of 100 to 300 IV/$m\ell$, the growth of sensitive isolates of Leuconostoc mesenteroides and Lactobacillus plantarum was inhibited for two to three days at 2$0^{\circ}C$. When nisin was added to kimchi preparations at a concentration of 100 IU/$m\ell$, the growth of lactic acid bacteria was delayed and reached a maximum two days later than that in kimchi without nisin. These results suggest the possible use of nisin in kimchi preparation, at recommended levels, to control the lactic acid fermentation. Scanning electron micrographs of a sensitive isolate L. plantarum revealed the formation of pores on cell surfaces followed by rapid cell wall destruction 1 h after the addition of nisin.

Assessment of Bacteria Removal Using Silver Ion Absorbed Ceramic Filter

  • The objective of this study was to evaluate bacteria removal ability of the metallic silver which was baked silver ion impregnated ceramic filter at heating condition. Silver leaking from baked ceramic filter was tested to sustain bacteria removal for a long time. Silver impregnated ceramic filter could remove E. coli completely at $10^{12}$ MPN/100ml to $10^{13}$ MPN/100ml of influent. However, ceramic filter without silver did not remove E. coli completely under the same condition. After baking, the silver impregnated ceramic filter almost didn't leak out the silver ion from filter. Photo of TEM (Transmission Electron Microscopy) showed that absorbed silver ions remained in ceramic filter after baking process and most of silver were less than 10 nm. According to the increase in the amount of silver in the ceramic filter, removal efficiencies of E. coli were increased but turbidity removal was decreased. It can be accounted that increased removal efficiency of E. coli was from disinfection of silver that is in the ceramic filter. Simulated concentrations of bacteria agree well with the observed experimental effluent concentration data. Moreover, first-order decay coefficients increased to 0.0034/min after silver was added in the ceramic filter. Increase of first-order decay coefficient proves that silver-added ceramic filter can remove bacteria easily.

The Role of AiiA, a Quorum-Quenching Enzyme from Bacillus thuringiensis, on the Rhizosphere Competence

  • Bacteria sense their population density and coordinate the expression of target genes, including virulence factors in Gram-negative bacteria, by the N-acylhomoserine lactones (AHLs)-dependent quorum sensing (QS) mechanism. In contrast, several soil bacteria are able to interfere with QS by enzymatic degradation of AHLs, referred to as quorum quenching. A potent AHL-degrading enzyme, AiiA, from Bacillus thuringiensis has been reported to effectively attenuate the virulence of bacteria by quorum quenching. However, little is known about the role of AiiA in B. thuringiensis itself. In the present study, an aiiA-defective mutant was generated to investigate the role of AHA in rhizosphere competence in the root system of pepper. The aiiA mutant showed no detectable AHL¬-egrading activity and was less effective for suppression of soft-rot symptom caused by Erwinia carotovora on the potato slice. On the pepper root, the survival rate of the aiiA mutant significantly decreased over time compared with that of wild type. Interestingly, viable cell count analysis revealed that the bacterial number and composition of E. carotovora were not different between treatments of wild type and the aiiA mutant. These results provide evidence that AHA can play an important role in rhizosphere competentce of B. thuringiensis and bacterial quorum quenching to Gram-negative bacteria without changing bacterial number or composition.

소목 추출물의 항균력 및 살균소독력 (Antimicrobial Activity and Bactericidal Activity of Caesalpinia sappan L. Extract)

  • Antimicrobial activity and bactericidal activity of Caesalpinia sappan L. extracts were investigated against five food-borne pathogens, E. coli, S. aureus, S. typhimurium, B. cereus and L. monocytogenes. Methanol extract of Caesalpinia sappan L. revealed antimicrobial activities against five pathogens. In particular, by paper disc diffusion the highest activity was shown against L. monocytogenes. Antimicrobial activities of methanol extracts showed the most potent activities, but hexane fraction had no activity. Fractions of ethyl acetate and butanol turned out to have higher antimicrobial activities against Gram(+) bacteria than Gram(-) bacteria. The minimum inhibitory concentration against five food-borne pathogens was 1.563 mg/ml on Gram(+) bacteria and 3.125 mg/ml on Gram(-) bacteria. The result of antimicrobial activity in a shaking flask method showed that bacterial growth rate fell by more than 99.999% at 3.125 mg/ml of methanol extract. The highest rate of viable reduction (99.998%) was shown at 0.781 mg/ml of methanol extract against L. monocytogenes. After five minutes of reaction between test strains and methanol extracts, the growth rates of five kinds of bacteria were reduced by more than 99.999% at a concentration of 100 mg/ml. Therefore, it is suggested that methanol extracts of Caesalpinia sappan L. can be developed as a natural sanitizer or disinfectant.
