Comparison of preservation in poultry carcasses processed by different chilling systems (도계육 냉각공정 방식에 따른 보존성 비교)

  • Cold air or ice water are used to chill poultry carcasses after slaughter. In order to compare the microbial reduction effect of two different chilling systems in poultry carcasses, Pre-and post-chilled carcass samples were tested for contamination agents such as aerobic bacteria counts, E. coli counts and Salmonella spp. counts. Water chilling system showed higher reduction ratio of bacteria than air chilling system during the three seasons. Also, aging of slaughter facility was related with bacterial contamination of pre-chilld carcasses. And additional volatile basic nitrogen (VBN) test were conducted in poultry packing meats and it increased steadily during 15 days. VBN at 8~9 days were over 13 mg% with putrid smell. Poultry packing meats by water chilling system were fresher than air chilling system during early storage time. But those decayed faster after 9 days.

  • The elevator is needed healthy and comfortable indoor air quality (IAQ) for using many people, but we found nothing about IAQ studies of an elevator. In general, air in the elevator car is sucked from the elevator's hoistway straight into the car using a fan. The air sucked into the hoistway may be filled with dust, mold and bacteria. This study was performed to measure of characteristics of indoor air quality (PM10, falling bacteria, $CO_2$, Rn and HCHO) in elevator's hoistway, CAR and lobby of 8 sites (4 apartments and 4 commercial buildings) in Gyeongnam from May, 2010 to January, 2011. With regards to the differences of pollutant distribution among hoistway, CAR, and lobby, the concentration of Rn and HCHO were the highest in hoistway followed by CAR and lobby, and PM10, falling bacteria and $CO_2$ were the highest in CAR followed by hoistway and lobby. Mean concentrations of PM10 were 104.9 ${\mu}g/m^3$ in CAR, 92.3 ${\mu}g/m^3$ in hoistway and 68.2 ${\mu}g/m^3$ in lobby, respectively.

  • Methanotrophs are microorganisms that possess the unique ability to utilize methane as their sole source of carbon and energy. A novel culture system, known as the compulsory circulation diffusion system, was developed for rapid growth of methanotrophic bacteria. Methanol dehydrogenase (MDH, EC 1.1.99.8) from Methylomicrobium sp. HG-1, which belongs to the type I group of methanotrophic bacteria, can catalyze the oxidation of methanol directly into formaldehyde. This enzyme was purified 8-fold to electrophoretic homogeneity by means of a 4 step procedure and was found in the soluble fraction. The relative molecular weight of the native enzyme was estimated by gel filtration to be 120 kDa. The enzyme consisted of two identical dimers which, in turn, consisted of large and small subunits in an ${\alpha}_2{\beta}_2$ conformation. The isoelectric point was 5.4. The enzymatic activity of purified MDH was optimum at pH 9.0 and $60^{\circ}C$, and remained stable at that temperature for 20 min. MDH was able to oxidize primary alcohols from methanol to octanol and formaldehyde.

A STUDY ON THE RELATIONSHIP BETWEEN THE CLINICAL SYMPTOMS AND THE PREDOMINENT PATHOGENIC MICROFLORA IN INFECTED ROOT CANALS (감염 근관에서의 주요 병인균과 임상증상간의 관계에 관한 연구)

  • In the Infected root canals, the majority of the isolated bacteria are either strict anaerobes or microaerophilic organisms. Among thease, Black-pigmented Bacteroides species are the most important and have relation with clinical symptoms. The purpose of this study were to investigate on the 7 different types of bacteria which include Black-pigmented Bacteroides - Black--pigmented Bacteroides, Actinobacillus actinomycetemcomitans, Streptococcus, Capnocytophaga, Eiknella corrodens, Fusobacterium, Actinomyces - and the interrelationship between these 7 bacterical species, and to compare Black-pigmented Bacteroides with the clinical symptoms in infected root canals. The canal contents of 15 necrotized teeth with 8 clinical symptoms were sampled and cultured aerobically and anaerobically for growth in 7 selective agar plates for 7 bacterial species. Black-pigmented Bacteroides and Streptococcus subspecies were identified by biochemical tests. The results were as follows; 1. 70.51% of the bacteria isolated were anaerobes and 29.49% were aerobes. 2. B. loescheii, B. intermedius, B. denticola were isolated in 8 cases, Streptococcus was isolated in 8 cases, Fusobacterium was isolated in 6 cases, Actinomyces was isolated in 5 cases and Eiknella corrodens, Actinobacillus actinomycetemcomitans, Capnocytophaga were not isolated. 3. There were mutual inhibition of growth between Black-pigmented Bacteroides and Streptococcus. (P<0.01) But Actinomyces and Fusobacterium showed mutual aids for growth. (P<0.05) 4. Black-Pigmented Bacteroides was found to be related both to sinus tract formation and to apical sensitivity to palpation.(P < 0.05).

Effects of Temperature in Anaerobic Nitrogen Removal Process from Piggery Waste : Activities in Ranges of Low Field-temperature (돈사폐수의 혐기성 질소제거에 있어서 온도의 영향 : 낮은 현장 온도범위에서의 활성)

  • ANAMMOX (Anaerobic ammonium oxidation) reactor, which was cultivated ANAMMOX bacteria in mesophilic condition ($35^{\circ}C$), was operated to investigate the effects of temperature. In $20{\sim}30^{\circ}C$ of operation condition, which was assumed as field-temperature, total N removal and $NH_4-N$ removal rate were declined from about 2.50 and $1.27kg\;N/{m^3}_{reactor}-day$ (0.06 and 0.03 kg N/kgVSS/day) to 1.62 and $0.41kg\;N/{m^3}_{reactor}-day$ (0.04 and 0.01 kg N/kgVSS/day), In this range of temperature, ANAMMOX had very low activities but acid fermentation bacteria and denitrifiers, which were competitors of substrates, had high activities relatively. Though operation temperature was higher than inhibition condition for two months, ANAMMOX activities could not been recovered once they were inhibited by low temperature. This fact was resulted from very slow doubling time of ANAMMOX bacteria. This study shows that maintenance device of optimal temperature is necessary required in field application of ANAMMOX.

Lactobacillus plantarum Improves the Nutritional Quality of Italian Ryegrass with Alfalfa Mediated Silage

  • The present study was planned to analyze the nutritional quality, microbial counts and fermentative acids in Italian ryegrass (IRG) 80% and alfalfa 20% (IRG-HV) mediated silage inoculated with lactic acid bacteria (LAB) as a probiotic strain for 3 months. Crude protein (CP), acid detergent fiber (ADF), and neutral detergent fiber (NDF), total digestible nutrient (TDN) and In-vitro dry matter digestibility (IVDMD), lactic acid bacteria (LAB), yeast and fungi counts and fermentation metabolites such as lactic acid, acetic acid and butyric acids were analyzed. The result shows that the nutritional quality and metabolite profiles of silage were significantly improved with LAB. For microbial counts, LAB showed dominant followed by yeast as compared with control silage. The pH of the silage also reduced significantly when silage inoculated with LAB. The result confirmed that silage preparation using different crops with L. plantarum inoculation is most beneficial for the farmers.

Garlic Fermentation by Lactic Acid Bacteria

  • Garlic has been used for condiments and also for medicines to cure various diseases since ancient times. Many studies on the processing of garlic have been published, however, few of them were related with fermentation because of the antimicrobial action of the garlic. In this study, to conduct garlic fermentation, 4 lactic acid bacteria (LAB) strains with growth abilities in garlic medium were selected. Addition of various nitrogen, carbon, and mineral sources generally did not improve the growth of experimental strains during garlic fermentation except for Lactobacillus casei KFRI 704 by yeast extract and Lactococcus lactis subsp. cremoris ATCC 19257 by mineral sources. High performance liquid chromatography (HPLC) analysis of 32 phenolic compounds during fermentation showed that formononetin was decreased time dependently. The concentrations of volatile compounds and alliin did not change during fermentation. The results of this study would provide the basic understanding of garlic fermentation by selected strains of LAB.

Isolation of Lactococci Inhibiting Listeria monocytogenes from Kimchi Habitat and Its Identification by 16S rDNA Analysis (김치 서식처에서 Listeria monocytogenes를 억제하는 lactococci의 분리와 16S rDNA분석에 의한 동정)

  • A bacteriocin-producing strain was isolated from kimchi at the early stage of kimchi fermentation. It was identified as Lactococcus lactis by morphological, cultural and physiological characteristics and partial sequence of 16S rDNA. The bacteriocin from isolate had antimicrobial activity against gram positive pathogenic bacteria, such as Listeria monocytogenes. Staphylococcus aureus and several strains of lactic acid bacteria but not to gram negative bacteria, Yersinia enterocolitica. The bacteriocin was sensitive to protease, protease ⅩⅣ, a-chymotrypsin and pepsin but not to lipase, trypsin and lysozyme. The bacteriocin activity was stable at pH 2-11 and temperature of 100 for 10 min. Thus, Listeria monocytogenes could be inhibited by Lactococcus lactis at early stage of fermentation.

Antibacterial Activity of Caryophylli Flos on the Growth of Dental Caries Bacteria, Streptococcus mutans KDJ-50 (구강균 Streptococcus mutans KDJ-50에 대한 정향의 향균효과)

  • This study was conducted to find antibacterial agent against growth of dental caries bacteria. St. mutans KDJ-50 from various medicinal plants of which safety was indetified. Medicinal plants used in this study was dried and grinded after purchased at Daegu yakryung sijang and Kyungsan jungang sijang. Medicinal plants extracted with 80% ethanol at boiling point for 3 hrs was used as antibacterial agent after freeze dried. This study was conducted to find antibacterial agent against growth of dental caries bacteria. St. mutans KDJ-50 from 32 medicinal plants of which safety was indetified. The result of using paper disc method. Caryophylli Flos. Coptidis Rhizoma. Phellodendri Cortex. Schizamdrae Fructs. Myristicae Semen, Crataegi Fructus and Acori Graminei Rhizoma was selected as antibacterial agent. The result of viable cell counting method. the antibacterial activity of Caryophylli Flos was highest among tested 7 medicinal plants followed by Phellodendri Cortex and Coptidis Rhizoma. The extinction effect of the St. mutans by Caryophylli Flos was shown with the addition of 0.5 (w/v) in the medium. The high antibacterial activity was acquired at high extraction temperature and long extraction temperature. The antibacterial activity of Caryophylli Flos was not effected by the concentration of ethanol.

The Effects of Lactic Acid Bacteria on the Growth and Aflatoxin $B_1$ Production of Aspergillus parasiticus (유산균이 Aspergillus parasiticus의 성장과 Aflatoxin $B_1$ 생성에 미치는 영향)

  • This study was performed to investigate the competitive effect of Leuconostoc mesenteroides and Lactobacillus plantarum on the growth and aflatoxin $B_1$ production of Aspergillus parasiticus. Leu. mesenteroides and Lac. plantarum were grown with A. parasiticus in a modified APT broth at $28^{\circ}C$ for 9 days. It was observed that the growth of A. parasiticus got lower when the two bacteria were inoculated simultaneously than when the mold grew alone. The pH of mixed culture of Leu. mesenteroides and Lac. plantarum showed significantly lower value than the pH of the pure culture of A. parasiticus (p<0.05). The acidity of the mixed culture group significantly increased compared with the control group (A. parasiticus alone group) (p<0.05). The aflatoxin $B_1$ production was significantly decreased in the mixed culture group than in the A. parasiticus alone group (p<0.05). Leu. mesenteroides showed the more efficient effect than Lac. plantarum. These results indicate that the two lactic acid bacteria have inhibitory effect on the growth and aflatoxin $B_1$ production of Aspergillus parasiticus.
