Correlation between Plate Count Agar and Dry Rehydratable Film Method for Enumeration of Total Aerobic Bacteria in Water (물 중 일반세균 정량에 대한 배지법과 건조필름법의 상관관계)

  • The quantitative accuracy of dry rehydratable film method was compared with the standard culture method to analyze the applicability of dry rehydratable film in the total aerobic bacteria test for water. The materials used in this study were 500 cases of water and were tested according to the Korean official test method for drinking water. The mean value of the total aerobic bacteria in plate count agar(PCA) and MC-Media Pad AC(MAC) were 2.6 ± 1.1 Log cfu/mL and 2.6 ± 1.2 Log cfu/mL. The p value and correlation coefficient(R2) were 0.933 and 0.9985 between the PCA and MAC, respectively, indicating no significant difference and very high correlation. According to the water types, the p value and R2 were 0.887 and 0.9911 in the PCA and MAC. In the purified water, the p value was 0.973 and R2 was 0.9934 in the PCA and MAC of other water. In each type of water, there were no significant differences between the PCA and MAC, and the correlation was very high. Thus, it is suggested that the MAC would be available for total aerobic bacteria test for water.

Microbial Differentiation on Chemical Properties of Paddy Soils in Reclaimed Tidal Lands at Western-coast Area of Korea

  • The scientific information on the microbial differentiation according to the changes in chemical properties of paddy soil in reclaimed tidal lands is not enough to understand the reclamation processes. The changes in microflora based on the chemical properties of paddy soils at the same sites of reclaimed tidal lands (21 samples from Nampo, Ewon, Sukmoon and Shihwa sites) were investigated in 2013 and 2015. In general, organic matter in paddy soils increased whereas pH decreased with the reclamation time. The electrical conductivities (EC) of soil samples were closely related to the exchangeable $Na^+$. With an increases in EC of paddy soils from 0.39 to $48.9dS\;m^{-1}$, the ratios (%) of halotolerant and halophilic bacteria to mesophilic bacteria proportionally increased from 0.2% to 102,000%. The population of halotolerant and halophilic bacteria in total microflora was also differentiated with the changes in EC of the same sites from reclaimed tidal soils within 2 years. The population of mesophilic bacteria decreased with an increase in EC above $5dS\;m^{-1}$. The microflora including halotolerant and halophilic bacteria could be a candidate as a biological parameter in evaluating the reclamation processes in addition to the chemical index of EC.

Profile of airborne microorganisms distributed in general offices (일반 사무실 실내공기 내 부유미생물의 분포 양상)

  • Mean levels of airborne bacteria, airborne fungi, temperature, relative humidity and carbon dioxide in total 69 general offices were $426({\pm}83)\;cfu/m^3$, $234({\pm}125)\;cfu/m^3$, $25.9({\pm}1.3)\;^{\circ}C$, $57.7({\pm}8.6)\;%$, $422({\pm}38)\;ppm$, respectively. The I/O ratio of airborne bacteria and fungi was over 1 and there was no significant difference among temperature, relative humidity and carbon dioxide in total 69 general offices. In construction period, a concentration of airborne bacteria and fungi was significantly highest in general offices constructed under one year and over three years since construction, respectively (p<0.05). The concentration of airborne fungi in general offices located at basement was significantly higher than those located at ground (p<0.05). No significant difference of airborne bacteria and fungi in general offices was found regardless of installation of HVAC system (p>0.05). The dominant bacterial genera identified in general offices was Staphylococcus, followed by Micrococcus, Bacillus, and Corynebacterium while usarium, Penicillium, Aspergillus, Alternaria, Rhizopus and Mucor were identified as dominant fungal genera in general offices.

Quality Characteristics of Sourdough Bread with Lactic Acid Bacteria in the Antibacterial Activity (항균 활성이 있는 유산균을 이용한 발효빵의 제조 및 품질 특성)

  • This study was performed to evaluate the quality characteristics of sourdough bread with addition of lactic acid bacteria (LAB) in the antibacterial activity. The starter cultures were used for sourdough bread to increase bread shelf-life: Lactobacillus fermentum, Lactobacillus paracasei KB28, and Pediococcus pentosaceus MJK7. The results showed that the viable counts of lactic acid bacteria were increased in 8.1~9.2 log $cfu/m{\ell}$, respectively. The pH of the three sourdoughs was lower than that of control. In the texture characteristics, hardness, gumminess, and chewiness were tended to increase on the sourdough bread. Among the three sourdough breads, bread volume and retrogradation of bread were not significantly different. According to the results from preservation period, the sourdough breads with addition of Lb. paracasei KB28, Ped. pentosaceus MJK7 were showed the appearance of mold and bacteria growth in 5~6 days, whereas the control were showed in 3 days. As a result, a sensory evaluation indicated the highest overall acceptance in order of Lb. fermentum, Lb. paracasei KB28, Ped. pentosaceus MJK7. Based on these results, Lb. paracasei KB28 can be applicable to improve the quality of sourdough bread.

A study of Swine Wastewater Treatment using Photosynthetic Bacteria (광합성 세균을 이용한 돈분 폐수 처리에 관한 연구)

  • Photosynthetic bacteria, strains KN 1-1, KN 2-1 and KN 2-3 were isolated from nature, and were studied for swine wastewater treatment. Growth of those photosynthetic bacteria were increased to 2~3 fold in organic-acid added medium(sodium acetate 1g, sodium propionate 1g and sodium butyrate 1g in Lascelles basal medium $1{\ell}$) than cultivation in Lascelles basal medium, and amount of bacteriochlorophyll a were increased to 1.5~2 fold. Chemical Oxygen Demand(COD) in swine wastewater using photosynthetic bacteria, strains KN 1-1, KN 2-1 and KN 2-3 were reduced 80%, 89% and 75%, respectively.

Identification and Characterization of Bacteriocin-Producing Lactic Acid Bacteria Isolated from Kimchi

  • Lactic acid bacteria were isolated from Kimchi and screened for bacteriocin. A total of 99 strains showed antimicrobial activity when grown on solid media, yet only 10 showed antimicrobial activity in liquid media. Strain H-559, identified as Lactococcus lactis subsp. lactis, exhibited the strongest inhibitory activity and was active against pathogenic bacteria including Listeria monocytogenes, Staphylococcus aureus, and Bacillus cereus as well as other lactic acid bacteria. The antimicrobial substance produced by L. lactis subsp. lactis H-559 was confirmed to be a bacteriocin by the treatment of $\alpha$-chymotrypsin, and protease type Ⅸ and ⅩIV. The bacteriocin activity remained stable between pH 2.0 and pH 11.0 and during heating for 10 min at $100^{\circ}C$. The bacteriocin production started in the exponential phase and stopped in the stationary phase. L. lactis subsp. lactis H-559 showed the highest bacteriocin activity at a culture temperature of $25^{\circ}C$, and an inverse relationship between the bacteriocin productivity and mean growth rate at different culture temperatures was observed. The mean growth rate and bacteriocin productivity of L. lactis subsp. lactis H-559 increased as the initial pH of the media increased.

Bacterial strains isolated from Jeotgal (salted seafood) induce maturation and cytokine production in mouse bone marrow-derived dendritic cells (마우스 골수 유래 수지상세포의 성숙과 사이토카인 생산에 대한 젓갈 분리균의 효과 연구)

  • Jeotgal (salted seafood) has been one of major fermented foods in Korea for long time. Although there are many studies about Jeotgal in various aspects of food, its immunological importance on hosts has not been elucidated yet. In this study, we investigated if several bacteria isolated from Jeotgal may modulate the function of dendritic cells (DCs), powerful antigen-presenting cells equipped with special immunological capabilities. 4 Jeotgal bacteria were selected as representatives and used for experiments. To treat viable DCs, those bacteria were killed at $60^{\circ}C$ for 30 min. The viability of DCs treated with Jeotgal bacteria was verified and two isolates significantly induced high production of interleukin-12, a representative cell-mediated cytokine of DCs. Surface activation and maturation markers (MHC class II, CD40, CD86) of DCs were analyzed by flow cytometer. In addition, the treated DCs showed significantly high lymphocyte stimulatory capability compared to control DCs based on allogeneic mixed lymphocyte reactions. These observations suggest that Jeotgal isolates can function as immunostimulating bacteria in hosts, like Lactobacillus. Taken together, these experimental evidences may broaden the use of Jeotgal isolates in immunological fields in addition to as a fermented food.

Characterization of the Biodiversity of the Spoilage Microbiota in Chicken Meat Using Next Generation Sequencing and Culture Dependent Approach

  • This study investigated the psychrotrophic bacteria isolated from chicken meat to characterize their microbial composition during refrigerated storage. The bacterial community was identified by the Illumina MiSeq method based on bacterial DNA extracted from spoiled chicken meat. Molecular identification of the isolated psychrotrophic bacteria was carried out using 16S rDNA sequencing and their putrefactive potential was investigated by the growth at low temperature as well as their proteolytic activities in chicken meat. From the Illumina sequencing, a total of 187,671 reads were obtained from 12 chicken samples. Regardless of the type of chicken meat (i.e., whole meat and chicken breast) and storage temperatures ($4^{\circ}C$ and $10^{\circ}C$), Pseudomonas weihenstephanensis and Pseudomonas congelans were the most prominent bacterial species. Serratia spp. and Acinetobacter spp. were prominent in chicken breast and whole chicken meat, respectively. The 118 isolated strains of psychrotrophic bacteria comprised Pseudomonas spp. (58.48%), Serratia spp. (10.17%), and Morganella spp. (6.78%). All isolates grew well at $10^{\circ}C$ and they induced different proteolytic activities depending on the species and strains. Parallel analysis of the next generation sequencing and culture dependent approach provides in-depth information on the biodiversity of the spoilage microbiota in chicken meat. Further study is needed to develop better preservation methods against these spoilage bacteria.

Colonization and Population Changes of a Biocontrol Agent, Paenibacillus polymyxa E681, in Seeds and Roots

  • Paenibacillus polymyxa E681, with its plant growth promotion and root colonization ability, has been proven to be a promising biocontrol agent of cucumber and barley. This study investigated the attributes related to the movement of bacteria from the seed to the radicle and to the whole root system. It also illustrated the existing form and population changes of the bacteria on seed and root using the scanning electron microscope and confocal laser scanning microscopy. The bacteria invaded and colonized the inside of the seed coat while the seeds were soaked in bacterial suspension. Almost the same number of bacteria on seed surface invaded the inside of the seed coat right after seed soaking. The population densities of E681 increased greatly inside as well as on the surface of the seed before the radicle emerged. The bacteria attached on the emerging radicle directly affected the initial population of newly emerg-ing root. The colonized cells on the root were arranged linearly toward the elongation of the root axis. In addition to colonizing the root surface, strain E681 was found inside the roots, where cells colonized the inter-cellular space between certain epidermal and cortical cells. When the cucumber seeds were soaked in bacterial suspension and sown in pot, the bacterial populations attached on both the surface and inside of the root were sustained up to harvesting time. This means that E681 successfully colonized the root of cucumber and sustained its population density up to harvesting time through seed treatment.

Ubiquitous Presence and Activity of Thiosulfate Oxidizing Bacteria in Rhizosphere of Economically Important Crop Plants of Korea (국내 작물 근권에 서식하는 황산화세균의 분포와 합성)

  • The presence of thiosulfate oxidizing bacteria was examined in rhizosphere soils of 19 economically important plant species belonging to 10 different families. The results showed that the thiosulfate oxidizing bacteria were present in all the tested rhizosphere soils, and the total 32 thiosulfate oxidizing bacteria were recovered. Furthermore, the biochemical characterization revealed that 56% and 44% of the isolates belonged to the obligate chemolithoautotrophs and facultative heterotrophs, respectively. The isolates ATSR15P utilized 19.17 mM of thiosulfate and accumulated 11.65 mM of sulfate in the medium. Concurrently, the decrease in pH of the medium was observed. This study comprehensively demonstrates that the active sulfur oxidation is a ubiquitous phenomenon in the rhizosphere of crop plants in Korea.
