Characterization of Airborne Bioaerosol Concentration in Public Facilities (다중이용시설내 공기중 바이오에어로졸 농도분포 특성에 관한 연구)

  • This study was conducted to evaluate the characterization of airborne bioaerosol in public facilities in Seoul. A total of 17 public facilities were investigated from December, 2002 to February, 2003. As results of the survey, the mean concentrations of bacteria and fungi in indoor air of public facilities were $378.08\pm296.33$ CFU/㎥ by RCS and $106.38\pm171.63$ CFU/㎥ and $347.46\pm335.32$ CFU/㎥ and $95.23\pm62.61$ CFU/㎥, by Six-stage cascade air sampler respectively. The mean concentrations of bacteria in indoor air (by ventilation method) were $517.14\pm343.93$ CFU/㎥ of natural ventilation and $215,83\pm100.71$ CFU/㎥ of mechanical ventilation. The mean concentrations of fungi in indoor air (by ventilation method) were $83.14\pm79.16$ CFU/㎥ of natural ventilation and $133.50\pm248.07$ CFU/㎥ of mechanical ventilation. The mean concentrations of bacteria in indoor air were 449.44 CFU/㎥ for the ground and $217.50\pm103.68$ CFU/㎥ for the underground. The mean concentrations of fungi in indoor air were $63.89\pm77.66$ CFU/㎥ for the ground and $202.00\pm290.08$ CFU/㎥ for the underground.

Enrichment of Electrochemically Active Bacteria Using a Three-Electrode Electrochemical Cell

  • Electrochemically active bacteria were successfully enriched in an electrochemical cell using a positively poised working electrode. The positively poised working electrode (+0.7 V vs. Ag/AgCl) was used as an electron acceptor for enrichment and growth of electrochemically active bacteria. When activated sludge and synthetic wastewater were fed to the electrochemical cell, a gradual increase in amperometric current was observed. After a period of time in which the amperometric current was stabilized (generally 8 days), linear correlations between the amperometric signals from the electrochemical cell and added BOD (biochemical oxygen demand) concentrations were established. Cyclic voltammetry of the enriched electrode also showed prominent electrochemical activity. When the enriched electrodes were examined with electron microscopy and confocal scanning laser microscopy, a biofilm on the enriched electrode surface and bacterium-like particles were observed. These experimental results indicate that the electrochemical system in this study is a useful tool for the enrichment of an electrochemically active bacterial consortium and could be used as a novel microbial biosensor.

Comparison of Antibacterial Activities of Garlic Juice and Heat-treated Garlic Juice (생마늘즙과 열처리 마늘즙의 항균활성 비교)

  • Antibacterial activities of garlic and heat-treated garlic juices on food-poisoning and lactic acid bacterial were examined. Aqueous extract of garlic juice showed bacteriocidal effect against both types of bacteria. Food-poisoning and lactic acid bacterial counts decreased at over 0.5 and 1.5% (w/v) garlic juice. Heat-treated garlic juice, which showed lower antibacterial effect than garlic juice against food-poisoning bacteria, had no significant antibacterial effect against Bifidobacterium, but instead increased Bifidobacterium count.

PCR-based Identification of Microorganisms in a Kefir Grain

  • Nowadays many people are concerned about being healthy, and many dairy products are taken as health supplementary foods. Among dairy products, kefir, also called as Tibet mushroom, is a yogurt fermented by kefir grain, which is a mixture of lactic acid bacteria and yeasts. Although there are many empirical evidences that kefir is very influential for human body, the exact reason is not definitively discovered. Therefore, it would be useful to understand characteristics of a kefir grain and to categorize bacteria in a kefir grain. In this paper, molecular biological apparatus such as PCR, electrophoresis, PCR purification, DNA sequencing were used to identify and classify the species of lactic acid bacteria and yeast in a kefir grain. We used PCR-based identification method using 16S rRNA primer and Internal Transcribed Spacer (ITS) primer. We identified 6 different species which were selected on different medium. In addition, observation with scanning electron microscope (SEM) enabled us to grasp an external shape of the kefir grain. Although we found a limited number of microbial species, more intensive research are needed for extensive identification of microorganism species in Korean kefir grain.

Analysis of Erythromycin Resistance Gene in Pathogenic Bacteria Isolates from Cultured Olive flounder Paralichthys olivaceus in Jeju (제주지역 양식 넙치(Paralichthys olivaceus)에서 분리한 어병세균 내 Erythromycin 내성 유전자 분석)

  • We determined the resistance rates of pathogenic bacteria isolated from cultured olive flounder Paralichthys olivaceus to erythromycin (Em), antibiotic typically used in aquaculture and analyzed the genotypes of resistant bacteria using polymerase chain reaction (PCR). We isolated and utilized 160 isolates of Streptococcus parauberis, 1 of S. iniae, 66 of Edwardsiella tarda, 56 of Vibrio sp. and 23 of unidentified bacteria from presumed infected olive flounder from Jeju Island from March 2016 to October 2017. Of the 306 isolated strains, Em-resistant strains included 33 of S. parauberis, 39 of E. tarda and 2 of Vibrio sp. We conducted PCR to assess the resistance determination of Em-resistant strains. Five different types of Em-resistance genes were detected in the 74 Em-resistant strains: erm (A), erm (B), erm (C), mef (A) and mef (E); erm (A) and erm (B) were detected in 1 (3%) and 24 (72.7%) S. parauberis isolates, respectively. In E. tarda, erm (B) was detected in five isolates (12.8 %) and no Em-resistance genes were detected in the two Vibrio sp. isolates.

Functional Characteristics of Kombucha Fermented with Lactic Acid Bacteria, Yeast, and Acetic Acid Bacteria Derived from Korea Traditional Foods

  • In this study, to determine the importance of lactic acid bacteria (LAB) in Kombucha fermentation, biological functions, such as organic acid production and anti-inflammatory and antibacterial activities, of Kombucha, with or without LAB inoculation, were evaluated. Lactobacillus paracasei DK215, Saccharomyces cerevisiae C3, and Acetobacter pasteurianus P2 were selected as the inoculants. Organic acids were measured every 3 days from the end of fermentation using HPLC; the organic acid content of LAB-inoculated Kombucha was relatively high. Samples with or without LAB inoculation showed high antibacterial activity against Escherichia coli. The MTT assay results indicated no significant difference in concentration difference and cell death. In the NO production test, compared with the uninoculated Kombucha sample, the LAB-inoculated Kombucha sample exhibited a value similar to that of the group without LPS treatment. The levels of cytokine (IL-1α, IL-6, TNF-α) production were significantly lower than those of the LPS(+) group, indicating the anti-inflammatory activity potential of the Kombucha sample. This improvement in the biological function of the LAB-inoculated Kombucha further verifies the value of LAB in the fermented food and beverage industry.

Quality Analysis According to Nutritional Content and Storage Period of Pickled Cabbage by Cultivation Region (재배지역별 절임배추의 영양성분 및 저장기간에 따른 품질 분석)

  • This study analyzed the changes in the nutritional components, properties, physicochemical properties, general bacteria, and lactic acid bacteria of pickled cabbage depending on the cultivation area. An analysis of the nutritional composition on the first day of storage showed no significant difference in the calories, carbohydrates, protein, fat, and dietary fiber contents regardless of the cultivation region, but regional differences were observed in the vitamin C content in the order of Andong > Pyeongchang > Haenam. The total free amino acid content appeared in the order Andong > Pyeongchang > Haenam, and the GABA (γ-amino-n-butyric acid) content appeared in the order Haenam > Andong > Pyeongchang. In addition, pickled cabbage was observed on the 1st, 5th, and 10th days of storage at 4℃. No significant change in appearance was observed on the 1st and 5th days, but changes were observed on the 10th day. Regardless of the cultivation area, the salinity (p<0.05) and pH (p<0.05) decreased significantly as the storage period increased, while the sugar content (p<0.05) and acidity (p<0.05) increased significantly. The general bacteria (p<0.05) and lactic acid bacteria (p<0.05) counts increased significantly as the storage period increased, irrespective of the cultivation region.

Quality Characteristics of Beef by Different Cooking Methods for Frozen Home Meal Replacements

  • Blanching beef for use in home meal replacements (HMR) is an important process that determines the final quality of the beef after the cooking process. Thermal pretreatment also minimizes the change in quality during the main cooking process or storage. In this study, beef samples were washed and sliced, then treated by immersion in boiling water (1-10 min), steaming (1-10 min), or pan-frying in oil (30-240 s). The color after each thermal treatment showed higher L* and b* values and lower a* values compared with the raw beef, except for the pan-frying thermal treatment. The total color difference (∆E) and pH value were significantly increased by panfrying (p<0.05). There was no significant difference in the shear force of the beef samples, except for the sample pan-fried for 210 s. The nutritional content of beef was measured as the moisture, protein, fat, and ash contents, which were 69.96, 16.64, 3.49, and 1.13%, respectively, in raw beef. After thermal treatment, the crude protein and fat contents were increased, whereas the moisture and ash contents decreased. The mineral content, including Na, Mg, Fe, and Ca was highest after pan-frying. The heat treatment decreased microorganisms in all the samples. The total bacteria count in raw beef was 4.5-4.7 Log CFU/g, whereas the bacteria count decreased to 2.2-2.8 Log CFU/g after blanching. Thermophilic bacteria, coliform, mold, and yeast not detected in any thermally treated sample.

Antioxidant and Cholesterol-lowering Effects of Lactic Acid Bacteria Isolated from Kelp Saccharina japonica Kimchi (다시마(Saccharina japonica)김치에서 분리한 유산균의 항산화 및 콜레스테롤 감소 효과)

  • Previous studies have suggested that microbial fermentation is an attractive process to develop food products using seaweed. The objective of this study was to isolate and characterize lactic acid bacteria (LAB), which are used as starters for seaweed fermentation. The isolation of LAB strains was conducted using kelp Saccharina japonica kimchi, a well-known fermented seaweed in southeastern Korea. Based on the assay of acid tolerance, bile tolerance and antioxidant activity, 15 strains of LAB were selected for further study. The LABs exhibited cholesterol lowering activity in the range of 15.50 to 94.77%. Among the LABs suitable for food production, Lactobacillus plantarum D-11 had the highest antioxidant and cholesterol lowering activities. This probiotic strain will be applied to develop various kelp fermentation products.

Antioxidant Activity of Kelp Saccharina japonica Extract Fermented by Probiotic Lactic Acid Bacteria (Probiotic 유산균 발효에 의한 다시마(Saccharina japonica) 추출액의 항산화 활성)

  • The objective of this study was to investigate the effect of lactic acid bacteria (LAB) fermentation on the antioxidant activity of kelp Saccharina japonica water extract. Three LAB strains that had exhibited superior antioxidant activity in a previous study were selected for the kelp fermentation starter. The antioxidant activity of the fermented extracts was analyzed during fermentation. After 48 h of fermentation, the extract-fermented Lactobacillus plantarum D-11 strains showed the highest antioxidant activity in terms of DPPH (2,2-diphenyl-2-picryl hydrazyl) radical scavenging, ABTS [2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)] radical scavenging, oxygen radical absorbance capacity (ORAC) and fluorescence recovery after photobleaching (FRAP) assay. Furthermore, the analysis of total phenolic and flavonoid contents revealed that the enhanced antioxidant activity was mainly due to the increased antioxidant content from fermentation. Thus, this study suggests that probiotic LAB fermentation is an attractive approach for the development of various kelp fermentation products.
