Olibanum Extract Inhibits Vascular Smooth Muscle Cell Migration and Proliferation in Response to Platelet-Derived Growth Factor

  • Olibanum (Boswellia serrata) has been shown to have anti-inflammatory, anti-arthritic and anticancer effects. This study determined the role of a water extract of olibanum in platelet-derived growth factor (PDGF)-stimulated proliferation and migration of rat aortic smooth muscle cells (RASMCs). PDGF-BB induced the migration and proliferation of RASMCs that were inhibited by olibanum extract in a dose-dependent manner. The PDGF-BB-increased phosphorylation of p38 mitogen-activated protein kinase (MAPK); the heat shock protein (Hsp) 27 was significantly inhibited by the olibanum extract. The effects of PDGF-BB-induced extracellular signal-regulated kinase1/2 was not altered by the olibanum extract. Treatment with olibanum extract inhibited PDGF-BB-stimulated sprout out growth of aortic rings. These results suggest that the water extract of olibanum inhibits PDGF-BB-stimulated migration and proliferation in RASMCs as well as sprout out growth, which may be mediated by the inhibition of the p38 MAPK and Hsp27 pathways.

지역 간 인구이동과 지역경제성장의 동태적 특징에 관한 실증 분석 (Analysis on the Dynamic Characteristics of Migration and Regional Economic Growth between Regions)

  • 지역 간 인구이동요인은 개인의 라이프코스에 기인하여 세대별·연령별로 차이가 존재하므로 지역경제성장이 인구이동에 미치는 영향도 연령별로 다를 수 있다. 이에 본 연구는 지역경제성장의 변화가 지역 간 인구이동에 미치는 영향을 연령별로 구분하여 실증 분석하였다. 분석결과를 요약하자면, 첫째, 연령층과 상관없이 지역의 경제성장률과 일자리 증가율의 상승은 순이동률을 증가시키는 것으로 나타났다. 둘째, 청년층은 중장년층에 비해 지역경제성장률과 일자리 증가율이 순이동률에 미치는 영향이 큰 것으로 나타났다. 셋째, 25-29세 연령층은 개인소득수준, 40-64세 중장년층은 지가변동수준의 지역 간 격차가 순이동률에 영향을 주는 것으로 나타났다. 이와 같은 분석결과를 볼 때, 지역 간 인구이동에서 지역경제성장은 여전히 중요한 요인으로 작용하고 있으며, 특히 청년층에게 영향력이 큰 것을 알 수 있었다.

Plasminogen kringle 5 재조합 단백질에 의한 ERK1/2 활성화 및 세포골격 재배열 억제 (Inhibition of ERK1/2 Activation and Cytoskeleton Rearrangement by the Recombinant Protein of Plasminogen Kringle 5)

  • Plasminogen kringle 5는 plasminogen kringles 1-4로 구성된 내생의 혈관 신생 억제제인 angiostatin과 같이 내피세포의 분열을 강력하게 억제한다고 알려져 있다. 본 연구에서는 plasminogen kringle 5의 재조합 단백질을 효모 발현 체계에서 생산하여 내피세포의 이동에 대한 저해 효과와 이에 대한 작용기전을 조사하였다 재조합 단백질 PK5는 plasminogen의 Thr456에서 Phe546까지 이르는 cDNA 부분을 ${\alpha}-factor$ prepro-peptide의 분비 신호 서열 뒤에 도입하여 Pichia pastoris GS115에서 발현시켰다. 메탄올 유도 후 얻은 배양액을 S-spin column을 이용하여 정제하였다. 정제된 단백질을 SDS-PACE하였을 때 약 10kDa의 단일 밴드를 나타냄을 확인할 수 있었다. 정제된 PK5는 bFGF나 VEGF에 의해 유도된 인간의 제대 유래 내피 세포의 이동을 약 500nM의 $IC_{50}$ 값으로 농도 의존적으로 감소시켰다. 내피 세포에 PK5 500M을 처리한 결과 bFGF에 의해 유도된 ERK1/2의 인산화를 감소시켰다 또한, PK5는 bFGF에 의해 유도된 내피세포의 골격 재형성을 강력하게 억제하는 것으로 관찰되었다. 따라서, 이러한 결과들은 효모 생산 PK5가 내피세포의 이동을 효과적으로 억제하며, 이는 ERK1/2의 활성과 세포골격의 재배열을 억제함으로써 나타나는 것으로 부분적으로 설명될 수 있다.

Role of TAZ in Lysophosphatidic Acid-Induced Migration and Proliferation of Human Adipose-Derived Mesenchymal Stem Cells

  • Transcriptional co-activator with a PDZ-binding motif (TAZ) is an important factor in lysophosphatidic acid (LPA)-induced promotion of migration and proliferation of human mesenchymal stem cells (MSCs). The expression of TAZ significantly increased at 6 h after LPA treatment, and TAZ knockdown inhibited the LPA-induced migration and proliferation of MSCs. In addition, embryonic fibroblasts from TAZ knockout mice exhibited the reduction in LPA-induced migration and proliferation. The LPA1 receptor inhibitor Ki16425 blocked LPA responses in MSCs. Although TAZ knockdown or knockout did not reduce LPA-induced phosphorylation of ERK and AKT, the MEK inhibitor U0126 or the ROCK inhibitor Y27632 blocked LPA-induced TAZ expression along with the reduction in the proliferation and migration of MSCs. Our data suggest that TAZ is an important mediator of LPA signaling in MSCs in the downstream of MEK and ROCK signaling.

도시민의 귀촌행태 유형화와 시사점 (The Typology of Urban-Rural Migration and Its Implications)

  • 귀농 귀촌자의 동기, 태도, 행태에 대한 이해를 위해 설문조사를 통해 귀농 귀촌자를 귀농 귀촌동기에 따라 유형화하였다. 요인분석 결과 귀농 귀촌자는 5개의 귀농 귀촌동기 차원을 가지고 있었다. 5개의 귀농 귀촌동기 차원은 '경제적 성공 추구', '전원적 삶 추구', '가족생활 추구', '도시생활회피 추구', '영농승계 추구'이다. 이 중에서 전체변량을 가장 많이 설명하는 귀농동기 차원은 '경제적 성공 추구'와 '전원적 삶 추구'로 나타났다. 이 같은 결과는 귀농자들은 '전원적 삶 추구'를 가장 중요한 귀농동기로 고려하고 있음을 나타낸다. 귀농 귀촌동기에 따라 귀농 귀촌자를 유형화하기 위하여 귀농 귀촌동기 차원에 기초하여 군집분석을 실시하였다. 군집분석결과 4개의 군집으로 구분되었다. 4개의 군집은 '기업가형', '소규모창업형', '전원형', '영농승계형' 이다.

다공성 매질에서의 착화하는 방사성핵종의 이동에 대한 이론적 접근 (A Theoretical Approach on the Migration of a Chelating Radionuclide in Porous Medium)

  • 이온 또는 착화된 상태의 방사성핵종의 이동에 대한 착화제의 영향을 조사하기 위하여 새로운 모델이 제시되었다. 착화된 방사성핵종의 이동거동은 착화제와 착화된 방사성핵종의 열화를 포함하는 대류-확산 이동방정식에 의해 해석되었다. 이 수학적 모델은 해석적인 방법 에 의해 구해졌으며 지연요소를 조사함으로써 분석되었다. 계산결과들은 감소된 지연요소에 의해 착화된 방사성핵종의 이동속도가 이온형태의 방사성핵종보다 매우 빠름을 보여주었다. 따라서 원자력발전소로부터의 착화제의 발생과 환경으로의 유출을 감소시킬 수 있는 새로운 구제책이 필요하다고 하겠다.

Development of a model to forecast the external migration rate in development projects reflecting city characteristics

  • In planning public service systems such as waterworks, the design population is very important factor. Owing to the limitations of the indirect method, two new models, which take into consideration urban characteristics, were developed to accurately predict external migration rate (EMR), which is an essential component in estimating reliably the design population. The root mean square error (RMSE) between the model values and observed values were 10.12 and 15.58 for the metropolitan cities and counties respectively and were lower compared to RMSE values of 27.31 and 28.79 obtained by the indirect method. Thus, the developed models provide a more accurate estimate of EMR than the indirect method. In addition, the major influencing factors for external migration in counties were development type, ageing index, number of businesses. On the other hand, the major influencing migration factors for cities were project scale, distance to city center, manufacturing size, population growth rate and residential environment. Future medium and long-term studies would be done to identify emerging trends to appropriately inform policy making.

Chloroquine Exerts Anti-metastatic Activities Under Hypoxic Conditions in Cholangiocarcinoma Cells

  • Intra-tumoral hypoxia is an environment that promotes tumor cell migration, angiogenesis and epithelial-mesenchymal transition that accounts for a major mechanism of metastasis. Chloroquine potentially offers a new therapeutic approach with an 'old' drug for effective and safe cancer therapies, as it exerts anti-metastatic activity. We investigated the inhibitory effect of chloroquine on cholangiocarcinoma (CCA) cell migration under cobalt chloride ($CoCl_2$)-stimulated hypoxia. We showed that chloroquine suppressed CCA cell migration under hypoxic-mimicking conditions on exposure to $100{\mu}M$ $CoCl_2$. Moreover, chloroquine stabilized the protein level of prolyl hydroxylase domain proteins (PHD-2) but reduced the levels of hypoxic responsive proteins such as hypoxia-inducible factor (HIF-$1{\alpha}$) and vascular endothelial growth factor (VEGF). It also suppressed epithelial mesenchymal transition (EMT) by increasing the ratio of E-cadherin to N-cadherin under hypoxic conditions. In conclusion, chloroquine can inhibit hypoxia-stimulated metastasis via HIF-$1{\alpha}$/VEGF/EMT which may serve as a useful additional strategy for CCA therapy.

Decorin: a multifunctional proteoglycan involved in oocyte maturation and trophoblast migration

  • Decorin (DCN) is a proteoglycan belonging to the small leucine-rich proteoglycan family. It is composed of a protein core containing leucine repeats with a glycosaminoglycan chain consisting of either chondroitin sulfate or dermatan sulfate. DCN is a structural component of connective tissues that can bind to type I collagen. It plays a role in the assembly of the extracellular matrix (ECM), and it is related to fibrillogenesis. It can interact with fibronectin, thrombospondin, complement component C1, transforming growth factor (TGF), and epidermal growth factor receptor. Normal DCN expression regulates a wide range of cellular processes, including proliferation, migration, apoptosis, and autophagy, through interactions with various molecules. However, its aberrant expression is associated with oocyte maturation, oocyte quality, and poor extravillous trophoblast invasion of the uterus, which underlies the occurrence of preeclampsia and intrauterine growth restriction. Spatiotemporal hormonal control of successful pregnancy should regulate the concentration and activity of specific proteins such as proteoglycan participating in the ECM remodeling of trophoblastic and uterine cells in fetal membranes and uterus. At the human feto-maternal interface, TGF-β and DCN play crucial roles in the regulation of trophoblast invasion of the uterus. This review summarizes the role of the proteoglycan DCN as an important and multifunctional molecule in the physiological regulation of oocyte maturation and trophoblast migration. This review also shows that recombinant DCN proteins might be useful for substantiating diverse functions in both animal and in vitro models of oogenesis and implantation.

혈관내피성장인자의 섬유아세포 증식과 Notch 1 발현에 대한 영향 (Vascular Endothelial Growth Factor Effect on Notch 1 Expression and Proliferation of Fibroblast)

  • Purpose: Vascular endothelial growth factor (VEGF) is known as a growth factor of endothelium and fibroblast. The purpose is to know the VEGF effects on fibroblast proliferation and fibroblast's notch receptor expression. Methods: CCD-986sk fibroblast was purchased from the Korean Cell Bank and was used in XTT assay for proliferation and wound healing assay for migration. Immunofluorescent (IF) staining and western blotting were used in testing notch expression of fibroblast. Semiquantitative RT-PCR was used in checking notch 1 mRNA production by fibroblast. Student-t test was used for analyzing results. Results: Cell proliferation assay using XTT showed significant higher proliferation in VEGF treated fibroblast, $2.324{\pm}0.0026$ vs. $2.463{\pm}0.017$ (p=0.002). Wound healing assay showed longer migration in VEGF treated fibroblast (p=0.062). The fluorescence was brighter in VEGF treated cells of notch 1 IF staining. Notch 1 expressions and mRNA productions increased more in VEGF treated cells. Conclusion: VEGF stimulates fibroblast to proliferate, migrate and to express Notch 1 simultaneously. Notch receptor could be related to VEGF mediated wound healing.
