Cell migration plays a role in many physiological and pathological processes. Reactive oxygen species (ROS) produced in mammalian cells influence intracellular signaling processes which in turn regulate various biological activities. Here, we investigated whether melanoma cell migration could be controlled by ROS production under normoxia condition. Cell migration was measured by wound healing assay after scratching confluent monolayer of B16F10 mouse melanoma cells. Cell migration was enhanced over 12 h after scratching cells. In addition, we found that ROS production was increased by scratching cells. ERK phosphorylation was also increased by scratching cells but it was decreased by the treatment with ROS scavengers, N-acetylcysteine (NAC). Tumor cell migration was inhibited by the treatment with PD98059, ERK inhibitor, NAC or DPI, well-known ROS scavengers. Tumor cell growth as judged by succinate dehydrogenase activity was inhibited by NAC treatment. When mice were intraperitoneally administered with NAC, the intracellular ROS production was reduced in peripheral blood mononuclear cells. In addition, B16F10 tumor growth was significantly inhibited by in vivo treatment with NAC. Collectively, these findings suggest that tumor cell migration and growth could be controlled by ROS production and its downstream signaling pathways, in vitro and in vivo.
Due to their stem-like characteristics and immunosuppressive properties, Mesenchymal stem cells (MSCs) offer remarkable potential in regenerative medicine. Much effort has been devoted to enhancing the efficacy of MSC therapy by enhancing MSC migration. In this study, we identified deubiquitinase BRCA1-associated protein 1 (BAP1) as an inhibitor of MSC migration. Using deubiquitinase siRNA library screening based on an in vitro wound healing assay, we found that silencing BAP1 significantly augmented MSC migration. Conversely, BAP1 overexpression reduced the migration and invasion capabilities of MSCs. BAP1 depletion in MSCs upregulates ERK phosphorylation, thereby increasing the expression of the migration factor, osteopontin. Further examination revealed that BAP1 interacts with phosphorylated ERK1/2, deubiquitinating their ubiquitins, and thus attenuating the ERK signaling pathway. Overall, our study highlights the critical role of BAP1 in regulating MSC migration through its deubiquitinase activity, and suggests a novel approach to improve the therapeutic potential of MSCs in regenerative medicine.
Two kinds of polymer coating (acrylic polymer and vinyl acetate ethylene copolymer) added with 5% nisin were fabricated on the paperboards and tested in their antimicrobial activity against Micrococcus flavus ATCC 10240 inoculated into water contacting the coating at $10^{\circ}C$. Vinyl acetate ethylene copolymer giving faster and higher nisin migration presented the greater reduction in the microbial count than the other coating, which endorsed that the migrated nisin is mainly responsible for the microbial inhibition or destruction. There was also a slight effect of the coating polymer itself in the microbial inhibition.
Wheat gluten-chitosan composite film (WGCCF) can prevent moisture migration and enhance the antimicrobial properties of gluten in intermediate-moisture foods like sandwiches. To mimic the structure of actual sandwich-type products we developed multi-layer food models, where moisture content and water activity differ. Water activity gradients direct moisture migration and therefore determine product characteristics and product stability. A 10% wheat gluten film-forming solution was mixed with chitosan film-forming solution (0-3%, w/w) and evaporated to generate WGCCF. Addition of 3% chitosan enhanced the mechanical properties of the film composite, lowered its water vapor permeability, and improved its ability to protect against both, Streptococcus faecalis and Escherichia coli, in a 24 hr sandwich test (reduction of 1.3 and 2.7 log cycles, respectively, compared to controls). Best barrier and antimicrobial performance was found for 3% chitosan WGCCF at pH 5.1. Film of this type may find application as barrier film for intermediate-moisture foods.
Harmine, a beta-carboline alkaloid isolated from the seeds of Peganum harmala has been reported as a promising drug candidate for cancer therapy. However, the effect of harmine on breast cancer remains still unclear. In this study, the effect of harmine on the cell proliferation, migration, and invasion of breast cancer MDA-MB231 cells and the underlying mechanism were investigated. The results indicated that harmine inhibited the proliferation MDA-MB231 cells in a dose-dependent manner and markedly suppressed migration and invasion of MDA-MB231 cells. The mechanism involved in part through Notch signaling. The Notch activity was significantly inhibited by harmine treatment and harmine suppressed the expression of Jagged1 which is a key ligand to activate Notch signaling. These findings suggest a novel mechanism of harmine on anti-cancer activity and harmine may act as a potential therapeutic drug for breast cancer treatment.
A powerful tool for chicken transgenesis could be established by employing a germline chimera production through primordial germ cell transplantation. This study was conducted to examine whether foreign gene-transfected gonadal primordial germ cells (gPGCs) have a migration activity into the gonad after transfer to recipient embryos. In Experiment 1, gPGCs of Korean Ogol Chicken were retrieved from 5.5-day-old embryos and subsequently transferred to the dorsal aorta of 2.5-day-old White Leghorn embryos after being labeled with PKH26 fluorescent dye. To confirm migration activity after transplantation, recipient embryos were sacrificed and examined on 3 days after transfer. Sex determination was concomitantly undertaken to examine whether sex of recipient embryos could affect the migration activity of gPGCs. All of embryonic gonads examined showed positive signals with PKH26 fluorescence and W-chromosome specific band by polymerase chain reaction (PCR) was detected in male embryos when gPGCs with ZW chromosome were transferred to recipient embryos. In Experiment 2, retrieved gPGCs were transfected with LacZ gene-containing cytomegalovirus promoter ($pCMV{\beta}$) by electroporation and subsequently transferred to recipient embryos. LacZ gene expression was identified in the gonads of 6 or 10-day-old recipient embryos and hatched-chicks. A total of 20 embryos and 12 hatched-chicks were examined and 11 of them (10 embryos and one hatched chicken; 11/32=34.4%) expressed $\beta$-galactosidase, a marker substance of LacZ gene. The results of this study demonstrated that foreign gene-transfected gPGCs can migrate and settle down into the gonad after being transferred into the blood vessel of the recipient embryos. This established technique will contribute to developing a peer biotechnology for transgenic chicken.
In this study, the immunomodulating effects of levamisole, selenium and tocopherol on blood neutrophils and peritoneal macrophages of goat were evaluated in vitro and in vivo. The functions of blood neutrophils and peritoneal macrophages were assayed by random and direct migration, phagocytosis of S aureus, production of superoxide and hydrogen peroxide. The results obtained were summarized as follows: In vitro trials 1. Levamisole treatment enhanced the random and direct migration of goat blood neutrophils when compared with untreated cell, and a significant (p<0.01) enhancement was noticed at the concentration of $100{\mu}g$ for direct migration and $50{\mu}g{\sim}1,000{\mu}g\;per\;ml$ of culture medium for random migration. There was no influence of selenium and tocopherol on random and direct migration of neutrophil at all of treatment concentration. 2. Neutrophils produced higher levels of superoxide by lcvamisole treatment at the concentration of $100{\mu}g$ and by selenium treatment at the concentration of $1.0{\mu}g$, but the production of hydrogen peroxide was not increased. Tocopherol had no effect on the production of antimicrobicidal oxygen metabolites of neutrophils at various concentrations. 3. No differences of phagocytic activity were observed when neutrophils were treated with three substances. In vivo trials 1. Blood neutrophils of goats orally administered levaraisole showed significantly (p<0.05) higher random migration from 2 to 24 hours after feeding (2.5mg/kg of body weight). Augmentation of random migration of neutrophil from goats orally administered selenium-tocopherol mixture (selenium $100{\mu}g$-tocopherol 200IU/head/day) was observed at 10 days and the significant (p<0.05) increase was shown from 30 days after feeding and continued throughout the feeding periods. 2. There was no effect on phagocytic activity and production of antimicrobicidal oxygen metabolites of neutrophils from goats administered levamisole or selenium-tocopherol mixture. 3. Random migration, production of superoxide and hydrogen peroxide and S aureus phagocytic activity of peritoneal macrophages of goats administered 300ml of levamisole-thioglycollatc medium mixture $(2.5{\mu}g/ml)$ into peritoneal cavity increased significantly (p<0.01 or p<0.05) when compared with those of goats administered thioglycollate medium alone.
Aralia elata is an edible mountain vegetable. Angiogenesis, the formation of new blood vessels, is a process involving migration, proliferation and cell differentiation, as well as the formation of new capillary structures. Matrix metalloproteinases (MMPs) plays an important role in angiogenesis. The development of a functional vascular system requires a variety of growth factors, their receptors, and intracellular signals. This study examines the effects of water extracts from: (i) A. elata root bark (Aralia extracts); (ii) a combination of Aralia extracts and fibroblast growth factors (FGF-2) on cultured porcine coronary artery endothelial cells (PCAECs). Aralia extracts induced the migration of PCAECs, which was inhibited by MMPs inhibitors. Combining Aralia extracts and FGF-2 enhanced the migration and the secretion of MMP-2 and MMP9 from PCAECs. We postulated that the Aralia extracts, which induced migrating activity in PCAECs, may be accomplished by increased secretion levels of MMP-2 and MMP-9.

Vascular endothelial cells release proteinases that degrade the extracellular matrix (ECM), thus enabling cell migration during angiogenesis and vasculogenesis. Sildenafil citrate stimulates the nitric oxide-cyclic guanosine monophosphate pathway through inhibition of phosphodiesterase type V (PDE5). In this report, we examined the mechanisms underlying sildenafil citrate-induced cell migration using cultured mouse aortic endothelial cells (MAECs). Sildenafil citrate induced migration and proteinase secretion by murine endothelial cells. Sildenafil citrate induced the secretion of matrix metalloproteinase-2 (MMP-2) and MMP-9, which is inhibited by $NF-{\kappa}B$ inhibitors. Sildenafil citrate also induced the secretion of plasmin, which is inhibited by PI 3'-kinase inhibitors. It is suggested that sildenafil citrate-induced migrating activity in endothelial cells may be accomplished by increased secretion of proteinases.
Membrane-type 1 matrix metalloproteinase (MT1-MMP) is a zinc-dependent proteinase found in cholesterol-rich lipid rafts on the plasma membrane. MT1-MMP hydrolyzes extracellular matrix (ECM) proteins, activates pro-matrix metalloproteinase-2 (proMMP-2) and plays an important role in ECM remodeling, cancer cell migration and metastasis. The role of caveolin-1, an integral protein of caveolae, in the activation of MT1-MMP remains largely unknown. Here, we show that the expression of caveolin-1 attenuates the activation of proMMP-2, reduces proteolytic cleavage of ECM and inhibits cell migration. We utilized the cytoplasmic tail domain deletion (${\Delta}CT$) or the E240A mutant of MT1-MMP. Co-expression of caveolin-1 with the wild-type or the ${\Delta}CT$ MT1-MMP decreased the proMMP-2 activation and inhibited collagen degradation and cell migration. Caveolin-1 had no effect on the catalytically inert E240A MT1-MMP. Our findings suggest that caveolin-1 is essential in the down-regulation of MT1-MMP activity by promoting internalization from the cell surface.
