This study was undertaken in order to compare reflux extraction(RE) and microwave-assisted extraction(MAE) in extraction efficiency and establish optimum microwave extraction conditions in obtaining Ligularia fischeri extracts. A considerable reduction in extraction time was accomplished by MAE. When 70% methanol 50% methanol 70% ethanol, or 50% ethanol was used, MAE extract contained equal levels of soluble solid and total polyphenol as obtained by RE. The optimum microwave-assisted extraction conditions for Ligularia fischeri were achieved by 120∼150 watts of microwave energy and 4∼8 minutes of extraction time. No significant changes were found in antioxidant activity with DPPH scavenging method over the variation of microwave energy or extraction time. The use of diluted methanol or ethanol improved soluble solid content(30%), total polyphenol content(2.7%) and antioxidant activity(68%).
This study was conducted in order to compare reflux extraction(RE) and microwave-assisted extraction(MAE) in extraction efficiency and establish optimum microwave extraction conditions in obtaining Aster scaber Thunb extracts. Extraction time was reduced considerably in MAE. When 70% methanol, 50% methanol, 70% ethanol, or 50% ethanol was used, no difference was found in the amount of soluble solid and total phenol between MAE and RE. The optimum microwave-assisted extraction conditions for Aster scaber Thunb were achieved by 120-150 watts of microwave energy and 4∼8 minutes of extraction time. No significant changes were found in antioxidant activity using DPPH scavenging method over the variation of microwave power or extraction time. The use of diluted methanol or ethanol improved soluble solid content(30.8%), total polyphenol content(2.9%) and antioxidant activity(69% ).
In this study, conventional solvent extraction (CSE), microwave-assisted extraction (MAE), and ultrasound-assisted extraction (UAE) were compared for the recovery of paclitaxel from biomass. As a result of investigating the effect of the extraction solvent type (acetone, chloroform, ethanol, methanol, methylene chloride), methanol was the most suitable for all extraction methods. In the case of MAE and UAE using methanol, most of the paclitaxel (> 95%) was recovered by only one extraction. The recovery rate of paclitaxel increased with the increase of extraction temperature (25-45 ℃), microwave power (50-150 W), and ultrasonic power (180-380 W) for MAE and UAE. In addition, SEM analysis showed that the biomass surface structure was slightly corrugated in CSE, while in the MAE and UAE, it was very rough and destroyed by strong impact.
A simple and efficient microwave-assisted extraction procedure was developed and optimized for the extraction of paclitaxel from the plant cell cultures of Taxus chinensis. The biomass, immersed in a methanol-water mixture, was irradiated with microwaves in a closed-vessel system. The microwave-assisted extraction was compared with the existing conventional solvent extraction in terms of yield, extraction time, and solvent consumption. The use of microwave energy allows rapid recovery of paclitaxel from biomass and dramatically reduces extraction time and solvent usage compared to conventional solvent extraction. The paclitaxel was completely extracted from biomass by microwave-assisted extraction for 3 min at $50^{\circ}C$, for 6 min at $30^{\circ}C$ and $40^{\circ}C$, respectively.
Comparison of a microwave-assisted extraction with sonication extraction was performed for arsenic speciation in fish tissue with chromatographic separation and inductively coupled plasma mass spectrometry detection. The detection limits of arsenicals with ultrasonic nebulizerand cross-flow nebulizer were shown to be similar. The arsenicals investigated were arsenobetaine (AsB), arsenite [As(III)], dimethylarsine acid (DMA), monomethylarsonic acid (MMA), arsenate [As(v)], and phenylarsonic acid (PAA). Quantitative extraction of arsenicals from dogfish muscle, DORM-2, standard reference material of NRCC (National Research Council of Canada) was achieved using 50% (v/v) methanol-water in both extraction methods. Extraction efficiency of arsenobetaine in both methods is greater than 82% with RSDs on replicates of less than 5%. The concentrations of AsB determined in extract of microwave assisted extraction and sonication methods were $14.18{\pm}0.42mg\;kg^{-1}$ and $13.54 {\pm}0.84mg\;kg^{-1}$, respectively. And the concentrations of DMA were $0.45{\pm}0.06mg\;kg^{-1}$ and $0.44{\pm}0.06mg\;kg^{-1}$, respectively.
This study was carried out to develop an efficient process far the elimination of phenol pollutant from soils. An microwave-assisted process (MAP) and a conventional Soxhlet extraction method (SEM) were employed to extract phenol from two types of soils. The effects of extraction methods, aged time of the spiked soil samples, extraction solvent and extraction time on the extraction performance were compared. Our results demonstrate that the recoveries from standard soil spiked were at least 10% higher fer MAP than these f3r the conventional Soxhlet. The extraction time by MAP requires significantly shelter time (1 min) than 15 h of the conventional Soxhlet. The recoveries from non-contaminated soil spiked with phenol were also almost identical f3r above results. The reduction of the extraction times with efficiency higher than that afforded by the conventional Soxhlet technique supports the suitability of the MAP method.
Physiologically active material ${\beta}$-glucan extracted from mushroom has an antitumor effect. Agaricus blazei murill(A blazel) fruiting bodies were reported to contain large amounts of ${\beta}$-glucan. The purpose of this study was to maximize ${\beta}$-glucn extraction from A. blazei by microwave assisted extraction method. Optimal extraction conditions were obtained under various extraction time and applied power which was examined as 200, 500 and 650 watt. The structure and concentrations of the raffinates were analyzed by nuclear magnetic resonance spectroscopy(NMR) and infrared spectroscopy(IR) It was proved that the extraction yield of microwave assisted extraction method was higher than that of conventional solvent extraction method. It was also observed that microwave assisted extraction method requires less amounts of solvents and extraction time compared to the conventional solvent extraction method.
The use of the microwave-assisted process for the extraction of effective constituents from ginseng was investigated at various operating conditions. The influence of solvent (ethanol-water, 50% v/v) to ginseng ratio, particle size and applied microwave power on the efficiency of extraction was examined. The microwave extraction system used was custom manufactured so that the intensity of microwave may be varied by using anode-voltage controller. It was found that the ratio of 6 : 1 (vol/mass) gave a good extraction efficiency. Small particle size gave high yield but it caused difficulties in the separation of solvent from the sludge. The higher power was no guarantee of the efficient extraction yield. The more important factor than the employed power was the adequate temperature under sufficient contact time. Using deionized-water as swelling agent, the degree of swelling of ginseng by microwave heating and conventional heating in water-bath was also studied. It was observed that the microwave heating enhanced the swelling much more than the conventional heating. It is believed that this enhanced swelling was responsible for the rapid microwave-assisted extraction rate.
Considering the thermal unstableness of ginseng components, microwave-assisted extraction(MAE) was performed under the atmospheric pressure condition. The monitoring of extraction characteristics and the optimization of extraction conditions were made by response surface methodology. The extraction efficiency of soluble ginseng components was high at lower ethanol concentration and at higher microwave power, while crude saponin content was easily extracted at higher ethanol concentration. Estimated conditions for the maximized extraction of soluble components including crude saponin, total phenolics and electron donating ability were $54{\sim}60%$ in ethanol concentration, $41{\sim}90$ W in microwave power, and within 4 min in extraction time. Predicted values at the optimum condition(60% ethanol, 80 W microwave power and 4 min extraction time) were in good agreement with observed values.
Microwave-assisted process (MAP) was applied to extract antioxidant-related components from Thymus quinquecostatus var. japonica Hara. Microwave power(2,450 MHz, $0{\sim}160$ W) and extraction time($1{\sim}5\;min$) were used as independent variables($X_i$) for central composite design to yield 10 different extraction conditions. Response surface methodology was applied to predict optimum extraction conditions for dependent variables of extracts, such as total yield, total phenolics, flavonoid, and electron donation ability depending on different powers and extraction times of MAP. Determination coefficients($R^2$) of regression equations for dependent variables were higher than 0.93 excluding that of total phenolics, and microwave power was predicted more influential than extraction time in MAP (p<0.05). The optimal extraction time for each dependent variable was ranged from 3.36 to 4.97 min, but microwave power showed wide ranges depending on variables. The superimposed contour maps for maximized dependent variables illustrated extraction conditions of 64 to 100 W in microwave power and 2.9 to 4.0 min in extraction time.
