Effects of Chitosanoligosaccharide on the Mouse Hepatotoxicity Induced by Cadmium (카드뮴으로 유발된 생쥐 간독성에 대한 키토산올리고당의 효과)

  • This research was conducted to determine the effects of chitosanoligosaccharide on liver poisoning induced by cadmium (Cd). Three groups of mice were used in this research. The group was only injected with cadmium (5.0 mg/kg; i.p.) (group Cd) and the other group was injected with cadmium and chitosanoligosaccharide (0.5% solution) at the same time (group Cd+Chi). In order to investigate the inhibitory action of chitosanoligosaccharide on liver damage, cadmium concentration in liver tissues and metallothionein (MT) concentration were relatively measured. In addition, histological observations were made to determine the morphologic injury of liver tissues. Cadmium concentration in liver tissues was drastically lower in groups Cd+Chi than in group Cd. MT concentration in liver tissues was lower in group Cd than in groups Cd+Chi. As the result of electron microscopic observation, mitochondria in group Cd showed a severe swelling phenomenon, RER fragment and ribosome dropout. However, in groups Cd+Chi, mitochondria with high electron density were distributed and RER forming a typical lamellae with ribosome was observed. From these results, cadmium toxicity on rat liver tissues could be lessened by chitosanoligosaccharide.

Histochemical and Electron Microscopic Study on the Zinc-containing Neurons in Rat Spinal Cord (흰쥐의 척수에서 아연이 함유된 신경원에 대한 조직화학 및 전자현미경적 연구)

  • The somata and boutons of the zinc-containing neurons in the spinal cord of the rats were labeled by intraperitoneal injection of sodium selenite and silver amplification. The labeled somata of the neurons were located in laminae V, VI, VII and X of the gray matter. The zinc selenium reaction products were retrogradely transported and precipitated into somata of the neuron with survival 8 hours. This observation suggest that all or part of the spinal cord zinc-containing neurons are interneurons. At survival 1 hour, the loaded axonal boutons with zinc precipitates of zinc-containing neurons were distributed in the gray matter and in the processes of lateral and ventral funiculi of the white matter. In particular, AMG stained boutons with huge form were appeared in the lamina IX. Ultrastructurally, the zinc precipitates were located in the cytoplasmic lysosomes or the vesicle within boutons of the zinc-containing neuron in accordance with survival times.

VirtualDub as a Useful Program for Video Recording in Real-time TEM Analysis (실시간 TEM 분석에 유용한 영상 기록 프로그램, VirtualDub)

  • The capability of real-time observation in TEM is quite useful to study dynamic phenomena of materials in a certain variable ambience. In performing the experiment, the choice of video recording program is an important factor to obtain high quality of movie streaming. Window Movie Maker (WMM) is generally recommended as a default video recording program if one uses "DV Capture" function in DigitalMicrograph$^{TM}$ (DM) software. However, the image quality does not often satisfy the condition for high-resolution microscopic analysis since the severe information loss in the final result occurs during the conversion process. As a good candidate to overcome this problem, Virtual-Dub is highly recommended since the information loss can be minimized through the streaming process. In this report, we demonstrated how useful VirtualDub works in a high-resolution movie recording. Quantitative comparison of the information quality between the images recorded by each software, WMM and VirtualDub, was carried out based on histogram analysis. As a result, the image recorded by VirtualDub was improved ~13% in brightness and ~122% in contrast compared with the image obtained by WMM at the same imaging condition. Remarkably, the gray gradation (meaning an amount of information) becomes wider up to ~115% than that of the WMM result.

Differential Expression Patterns of Crystallin Genes during Ocular Development of Olive Flounder (Paralichthys olivaceus)

  • Olive flounder Paralichthys olivaceus is one of the most widely cultured fish species in Korea. Although olive flounder receive attention from aquaculture and fisheries and extensive research has been conducted eye morphological change in metamorphosis, but little information was known to molecular mechanism and gene expression of eye development- related genes during the early part of eye formation period. For the reason of eyesight is the most important sense in flounder larvae to search prey, the screening and identification of expressed genes in the eye will provide useful insight into the molecular regulation mechanism of eye development in olive flounder. Through the search of an olive flounder DNA database of expressed sequence tags (EST), we found a partial sequence that was similar to crystallin beta A1 and gamma S. Microscopic observation of retinal formation correspond with the time of expression of the crystallin beta A1 and gamma S gene in the developmental stage, these result suggesting that beta A1 and gamma S play a vital role in the remodeling of the retina during eye development. The expression of crystallin beta A1 and gamma S were obviously strong in eye at all tested developing stage, it is also hypothesized that crystallin acts as a molecular chaperone to prevent protein aggregation during maturation and aging in the eye.

Expression Analysis of Visual Arrestin gene during Ocular Development of Olive Flounder (Paralichthys olivaceus)

  • Olive flounder (Paralichthys olivaceus) is one of the commercial important flatfish species in Korea. The ocular signal transduction pathway is important in newly hatched flounders because it is closely involved in the initial feeding phase thus essential for survival during the juvenile period. However, the study of gene expression during ocular development is incomplete in olive flounder. Therefore we examined the expression analysis of specifically induced genes during the development of the visual system in newly hatched flounders. We searched ocular development-involved gene in the database of expressed sequence tags (ESTs) from olive flounder eye and this gene similar to arrestin with a partial sequence homology. Microscopic observation of retinal formation corresponded with the time of expression of the arrestin gene in the developmental stage. These results suggest that arrestin plays a vital role in the visual signal transduction pathway of the retina during ocular development. The expression of arrestin was strong in the ocular system during the entirety of the development stages. Our findings regarding arrestin have important implications with respect to its biological role and evolution of G-protein coupled receptor (GPCR) signaling in olive flounder. Further studies are required on the GPCR-mediated signaling pathway and to decipher the functional role of arrestin.

Petrochemistry of granitic rocks from the Eonyang and the Ulsan area (언양(彦陽) 및 울산지역(蔚山地域) 화강암질암류(花崗岩質岩類)의 암석화학적(岩石化學的) 연구(硏究))

  • Granitic rocks, from the Eonyang and the northwestern part of Ulsan area, were mainly studied from the petrochemical point of view. From field work, microscopic observation and the result of K-Ar ages, these granitic rocks are divided into six rock types of a, b, c, d, e and e', of which modal composition are given. Type a, c, e and e' are mainly granodiorite, type b is adamellite, and type d is granodiorite, adamellite and others (porphyritic rock, porphyry and felsitic rocks). Bulk chemical analyses of 22 samples of the granitic rocks are given. The petrographical and petrochemical characteristics of these rocks are discussed briefly. In the petrochemical compositions with their characteristic variation trends of several oxides and norm Or-Ab-An triangular diagram, type a, b and c show some similarity to that of San-yo granite of Japan and younger granite of Ogcheon geosynclinal zone, and then, type e and e', to San-in granite of Japan. But, some of granitic rock samples of type d are similar to San-yo granite and the others of type d to San-in granite because this type is composed of porphyritic rock, porphyry and felsitic rocks. According to the result of K-Ar ages (1976, Lee et al.) of rock samples from type a, b and care Cretaceous, on the otherwise, type d, e and e', are Tertiary granitic rocks. Judging from these evidences, granitic rocks in the area are correlated to the Cretaceous and Tertiary granitic rocks in the, southwestern Japan, that is, type a, band c are correlated to San-yo granite, and type d, e and e' to San-in granite.

The Influence of Hen Aging on Eggshell Ultrastructure and Shell Mineral Components

  • The eggshell, which is a complex and highly ordered structure, is very important factor for food safety and egg marketing. This study investigated the changes in eggshell structure and shell components in relationship to hen age. For this study, we examined the histological change of the endometrium of the 30-, 60-, and 72-wk-old commercial layers, and analyzed the ultrastructure and ionic composition of their eggshells. The results showed that histological deformation, fibrosis, atrophy and elimination of micro-villi in the uterus endometrium were found through microscopic observation that was associated with increasing hen age. Concentration of blood-ion components such as $Ca^{2+}$, $Na^+$, $K^+$, and $Cl^-$ ions did not change with age. Along with the results from the ultrastructure analysis of the eggshell, the palisade layer ratio and the density of mammillary knobs were significantly decreased in older hens. In addition, the type B mammillary knobs were frequently observed with increasing hen age. In the mineral element assay from the eggshell, $Ca^{2+}$, $S^{2-}$, and $Co^{2+}$ significantly decreased with increasing hen age, whereas $Na^+$, $K^+$, and $V^{2+}$ significantly increased. Therefore, the damages of endometrial tissue inhibit the processes of ion transmission and the crystallization of eggshell formation, resulting in a large and non-uniform mammillary knob formation. This means the conditions of endometrial cells affect the formation of the eggshell structure. In conclusion, hen aging causes the weakness of the eggshell and degrades the eggshell quality.

The study on the Fluorescence Characteristics of Several Freshwater Bloom Forming Algal Species and Its Application (수종 담수적조 원인종들의 형광특성과 적용연구)

  • The freshwater blooms mainly blue-green algal blooms occur frequently in the lower Naktong River in summer, which provoke many socio-economical problems; therefore, the early detection of bloom events are demanding through the quantitative and qualitative analyses of blue green algal species. The in vivo fluorescence properties of cultured strains of Microcystis aeruginosa, M. viridis, M. wesenbergii, M. ichthyoblabe, Anabaena cylindrica, A. flos-aquae, and Synedra sp. were investigated. Wild phytoplankton communities of the lower Naktong River were also monitored at four stations in terms of their standing stocks, biomass and fluorescence properties compared with its absorption spectram. The 77K fluorescence emission spectra of each cultured strains normalized at 620 nm was very specific and enabled to detect of blue green algal biomass qualitatively and quantitatively. The relative chlorophyll a concentration determined by chlorophyll fluorescence analysis method showed significant relationship with chlorophyll a concentration determined by solvent extraction method ($R^2$ = 0.906), and the blue-green algal cell number determined by microscopic observation ($R^2$ = 0.588), which gives insight into applications to early detection of blue green algal bloom.

The Effects of Triamcinolone Acetonide by Iontiophoretic Transdermal Delivery on Inflammation Induced Rat (백서에서 유발된 염증에 대한 Triamcinolone Acetonide 이온도입 경피투과 효과)

  • This study investigated the effects of triamcinolone acetonide by iontophoretic transdermal drug delivery on anti-inflammatory action into the rats and which had carrageenan-induced hyperalgesia and edema in the feet, trauma-induced tissue damage in the thigh. Each group was treated under the fellowing conditions. 1. Group I : Control group 2. Group II : Application of direct current 3. Group III : Application of 0.1$\%$ triamcinolone acetonide solution 4. Group IV : Iontophoresis of 0.1$\%$ triamcinolone acetonide solution The degree of anti-inflammation was evaluated by the paw withdrawal latency, the change in volume of foot the change of paw edema, histological change in rats. 1. In paw withdrawal latency, group IV showed the most significant therapeutic effect than the other groups at 0, 3, 6 and 9 hours(p < 0.001). 2. In paw edema experiment in the foot, group IV showed the most significant effect than group I at 0, 3, 6 and 9 hours. It meant that there was effective anti-inflammatory reaction in group I (p < 0.001). 3. In the light microscopic observation, group IV showed the most significant reduction of haemorrhage, hyperemia and infiltrative inflammation. From the results, the iontophoresis with triamcinolone acetonide is more effective than using each groups. It is one of the effective physical agent which delivered large molecular weight drug into the body. The continuous study is needed for many interesting issues of iontophoretic transdermal drug delivery in new future.

Growth retardation and differential regulation of expansin genes in chilling-stressed sweetpotato

  • We report here a first evaluation of chilling-responsive gene regulation in the sweetpotato. The growth of sweetpotato plants was severely retarded at $12^{\circ}C$; the lengths of the leaf, petiole, and root were markedly reduced and microscopic observation revealed that the elongation growth of the epidermal cells in each of these organs was significantly reduced. We examined the transcriptional regulation of three sweetpotato expansin genes (IbEXP1, IbEXP2 and IbEXPL1) in response to various chilling temperatures (12, 16, 22, and $28^{\circ}C$). In the leaf and petiole, the highest transcript levels were those of IbEXP1 at $28^{\circ}C$, whereas IbEXPL1 transcript levels were highest in the root. IbEXP1 mRNA levels in the $12^{\circ}C-treated$ petiole showed a fluctuating pattern (transient decrease-recovery-stable decrease) for 48 h. In the leaf and petiole, IbEXP1 and IbEXPL1 exhibited a similar response to chilling in that their mRNA levels decreased at $22^{\circ}C$, increased at $16^{\circ}C$, and decreased dramatically at $12^{\circ}C$. In contrast, mRNA levels of IbEXP2 in the leaf fell gradually as the temperature fell from 28 to $12^{\circ}C$, while they remained unaltered in the petiole. In the root, mRNA levels of IbEXPL1 and IbEXP1 reached maximum levels at $16^{\circ}C$, and decreased significantly at $12^{\circ}C$. These data demonstrated that expression of these three expansin genes was ultimately down-regulated at $12^{\circ}C$; however, transcriptional regulation of each expansin gene exhibited its own distinctive pattern in response to various chilling temperatures.
