Ninety samples showing mosaic symptoms on soybean (Glycine max) cv. Sukryangputkong were collected from the Cheongsongkun area, Kyungbuk province in Korea. Initially, DAS-ELISA was conducted far detection of Soybean mosaic virus (SMV). Negative samples were chosen at random and mechanically inoculated on soybean cv. Buffalo, which reported not to produce mosaic symptoms when mechanically inoculated with SMV. An isolate of SMV, designated as B-1, from Buffalo showing mosaic and mottle symptoms was used for identification and biological characterization of the causal vim. The purified B-1 isolate had spherical particles of approximately 24nm. It positively reacted with the antiserum against Cowpea mosaic virus (CPMV) but not with Cucumber mosaic virus (CMV) and SMV antisera. CPMV was newly isolated from soybean and had been characterized by host range and by serological and electron microscopic methods. Results of this study suggest that CPMV is the possible cause of mosaic disease in vegetable soybean and that based on sympto-matology, a difference between the typical mosaic and rugose symptoms caused by SMV and CPMV was observed. This is first report of CPMV from soybean in Korea.
Three wood coffins of Chosun Dynasty period buried about 450 years ago were excavated in the sound condition in Andong area in the early 1998. The proprietors of wood coffins were grandparents, Mr. Myoung Jong Lee and Mrs. Mun, and their grandson, Mr. Eung Tae Lee, and the social standing of their family was known to belong to the nobility in those days by the clan genealogy. All the wood coffins investigated through light microscopy had same anatomical characteristics as follows: abrupt to somewhat abrupt tracheid transition from earlywood to latewood; normal longitudinal and horizontal resin canals with thin-walled epithelium; tylosoids in resin canals; bordered pits frequently in 1 row on radial walls of tracheids; 1 or 2 window-like pits per cross-field; uniseriate and fusiform rays; heterogeneous rays composed of body ray parenchyma cells and marginal ray tracheids or homgeneous rays composed of only ray tracheids; dentate ray tracheids; occasional trabeculae traversing tracheids in radial direction. Based on theses microscopic characteristics, all the wood coffins were identified to be Korean red pine (Pinus densiflora) or Korean black pine (Pinus thunbergii). Korean black pine growing naturally in coastal area might not be probable because the site of excavation, Andong area, was mountainous and inland area of Korea Thus, Korean red pine was thought to be the possible species for the wood coffins because of its natural distribution through the Korean Peninsula and the easy availability.
Wood identification is regularly performed by observing the wood anatomy, such as colour, texture, fibre direction, and other characteristics. The manual process, however, could be time consuming, especially when identification work is required at high quantity. Considering this condition, a convolutional neural networks (CNN)-based program is applied to improve the image classification results. The research focuses on the algorithm accuracy and efficiency in dealing with the dataset limitations. For this, it is proposed to do the sample selection process or only take a small portion of the existing image. Still, it can be expected to represent the overall picture to maintain and improve the generalisation capabilities of the CNN method in the classification stages. The experiments yielded an incredible F1 score average up to 93.4% for medium sample area sizes (200 × 200 pixels) on each CNN architecture (VGG16, ResNet50, MobileNet, DenseNet121, and Xception based). Whereas DenseNet121-based architecture was found to be the best architecture in maintaining the generalisation of its model for each sample area size (100, 200, and 300 pixels). The experimental results showed that the proposed algorithm can be an accurate and reliable solution.
In order to identify an unknown wood sample native to Korea. the softwood databases(KSWCHUN; Korean SoftWood CHUN) and the hardwood databases(KHWCHUN; Korean HardWood CHUN) had been built. and the new computer searching programs(IDINEX; IDentification INformation EXpress) has been written in Turbo Pascal(V.5.0) and in Macro Assembly(V.5.0). The characters of the data were based on the 74 features of softwood and on the 148 features of hardwood which are a part of new "IAWA list of microscopic features for hardwood identification" published in 1989. For the purpose of this investigation the wood anatomical nature of 25 species of softwood(13 genera of 5 families) and of 112 species of hardwood(57 genera of 31 families) were observed under a scanning electron microscope and light microscope. and a lot of literature used. The IDINEX programs are based on edge-punched card keys. with several improvements. The maximum number of features in the IDINEX is 229. but that is fixed for a given database. Large numbers of taxa are handled efficiently and new taxa easily added. A search may be based on sequence numbers of features. Comparisons are made sequentially by feature and taxon using the entire suite of features specified to produce the list of possible matching taxa. The results are followings. (1) The databases of Korean wood and the searching programs(IDINEX) had been built. (2) The databases of Korean wood could be an information to search an unknown wood. (3) The databases would be valuable. for the new features, which were not mentioned in Korean wood up to the present. were observed in details. (4) The ultrastructures of the cell walls(warty layer) and crystals observed under a scanning electron microscope will be helpful to search an unknown wood in particular. (5) The searching process is more quick and accurate than the others. 6) We can obtain the information on the differences of a species from the other and search an unknown wood using probability. in IDINEX, (7) The IDINEX will be utilized to identify and classify an animal life, vegetable world, mineral kingdom, and so on.
Objectives : Southern, northern, and wild types of Osterici Radix, and Notopterigii Rhizoma et Radix imported from China are circulated in Korea. Morphological identification was attempted by comparing external and internal forms of these materials. Methods : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by olymphus-BHT. Results : 1. Southern type of Osterici Radix has short rhizome and many small and long roots, while northern type of it is mostly composed of rhizomes with little small reds and sometimes looks like bamboo because of the extention of the parts between joints. 2. Parenchymal cell of southern type of Osterici Radix is filled with starch grain, while that of nothern type includes less starch grain with lamination crossed on its cell wall. 3. Surface of root and rhizome of wild Osterici Radix has some marks of fibrous root raised in the form of nodule, but cultivated Osterici Radix does not have this kind of marks. 4. Notopterigii Rhizoma et Radix is mostly composed of rhizomes, and its form is similar to that of a silkworm or joints of bamboo. Parenchymal cell is filled with starch grains, and lamination is crossed on its cell wall but not salient. Conclusion : Southern, northern, and wild types of Osterici Radix, and Notopterigii Rhizoma et Radix circulated in Korea can be easily identified because of their differences in their external and internal forms.

Objectives : The consumption of licorice is large in Korea, but domestic production is insufficient due to the lack of adaptability. This study aimed to provide a morphological basis for adding interspecific hybrid licorice with improved adaptability to pharmacopoeia. Methods : This study was to establish identification criteria for the original plants, external and internal morphology of the authentic herbal medicines (Glycyrrhiza uralensis, G. glabra and G. inflata), market products and artificially interspecific hybrid forms of licorice. For this purpose, previous studies were investigated and visual and histological observations were carried out. We focused on the internal morphology by microscopic observation for securing objectivity. Finally, we proposed the identification keys for precise classification of each part. Results : 1) Original plant : Licorice species in the compendium were distinguished by the number of leaflets, presence of hair on the fruit, curvature and swelling of the fruit. 2) External morphology : Licorice species were distinguished by degree of powderiness, tearing gap, radial structure in the cross section and existence of protrusion of outer epidermis. 3) Internal morphology : Licorice species were distinguished by the degree of development of phloem fiber bundle, development of obliterated sieve, whether the secondary medullary ray are branched. In the case of interspecific hybrids, the characteristics of both species used for hybridization were mixed in all observation methods. Conclusions : These results suggest that the interspecific crossbred licorice is suitable for the pharmacopoeial standard. Therefore, it can be applied as a herbal medicine through additional supplementary study.
Fungal isolates from infected Chinese quince trees were found to cause black rot in Yeongcheon, Gyeongsangbuk Province, Korea. The quince leaves withered and turned reddish-brown and fruits underwent black mummification. To elucidate the cause of these symptoms, the pathogen was isolated from infected leaf and fruit tissues on potato dextrose agar and Levan media. Several fungal colonies forming a fluffy white or dark gray mycelium and two types of fungi forming an aerial white mycelium, growing widely at the edges, were isolated. Microscopic observations, investigation of fungal growth characteristics on various media, and molecular identification using an internal transcribed spacer, β-tubulin, and translation elongation factor 1-α genes were performed. The fungal pathogens were identified as Diplodia parva and Diplodia crataegicola. Pathogenicity tests revealed that the pathogen-inoculated fruits exhibited a layered pattern, turning brown rotting; leaves showed circular brown necrotic lesions. The developed symptoms were similar to those observed in the field. Fungal pathogens were reisolated to fulfill Koch's postulates. Apples were inoculated with fungal pathogens to investigate the host range. Strong pathogenicity was evident in the fruits, with browning and rotting symptoms 3 days after inoculation. To determine pathogen control, a fungicidal sensitivity test was conducted using four registered fungicides. Thiophanate-methyl, propineb, and tebuconazole inhibited the mycelial growth of pathogens. To the best of our knowledge, this is the first report on the isolation and identification of the fungal pathogens D. parva and D. crataegicola from infected fruits and leaves of Chinese quince, causing black rot disease in Korea.
Natural products used for oriental medicine often come from various geographical sources, after several different distribution channels. Therefore some form of quality control procedure is required to safeguard naturl products for prescriptions purposes. To achieve this, systematic apprroaches such as morphological examination, microscopic analysis of powdered herbs and chemical analysis can be carried out. However, to ensure absolute criteria for quality assurance of natural products, DNA fingerprinting method such as RAPD(Random amplified polymorphism DNA) analysis can be used for authentication of natural products for authenticatin of natural products. In this study, warious oligonucleotide primers will be synthesized for the detection of RAPD markers and also parameters of affecting PCR(Polymerase Chain Reaction) in the detection of RAPD markers of rare and high-priced natural products will be studied with genomic DNA of chosen samples.

For isolating and identifying Bifidobacterium spp. originating from Korea, feces were sampled from healthy Korean infants nursery school and postpartum care center. Through the use of gram staining and microscopic examination for cell morphology, 87 bacterial strains presumed to be the Bifidobacterium strains were isolated from 59 Koreans. To identify the Bifidobacterium strains at the genus level, these bacteria were then analyzed using the TLC method. As a result, 29 of the isolated strains were confirmed as members of the genus Bifidobacterium. 29 Bifidobacterium strains were tested acid, bile salts and oxygen tolerance and investigated antioxidative effect specially. And determined the superiority of 5 strains out of 29 Bifidobacterium strains. Finally, the selected bifidobacterium was identified with using designed 16S-ITS rDNA primer.

Potato mop-top virus(PMTV) was identified from Solanum tuberosum cv. Gawon showing bright chlorotic mottle symptom in Namwon, Korea. Samples were collected green-house in February, 2003. Electron microscopic examination of negatively stained preparation revealed that PMTV were rigid-rod shaped particles about 100-150, 250-300 nm x 18-20 nm in length. In ultrathin sections of leaf tissue from diseased potato plants, cluster of viruses particles were observed in the cytoplasm. TAS-ELISA determined that the virus was serologically related to PMTV. PMTV produced double ring necrotic local lesion in inoculated leaf of Chenopodium amaranticolor in incubated at 15$^{\circ}C$. The PMTV could be detected with RT-PCR using PMTV detectable primer set designed to amplify about 540 bp of the partial CP gene of PMTV
