저주파대 음향 현미경의 구성과 그 응용에 관한 연구 (A Study on the construction of Low-Frequency Acoustic Microscope and Its Application)

  • In this paper, a low frequency acoustic microscope system has been built for the purpose of detecting subsurface defects in materials. The lateral resolution of acoustic microscope is studied and analyzed in order to evaluate that system performance. And a NDE technique is demonstrated by using this system. In the focused and defocused mode of operation, the acoustic microscope system showed in experiment that its lateral resolution was about 0.5 mm at a frequency of operation of 3MHz on a fused-quartz sample with seeded circular cracks ranged in size from 0.2 mm to 1.0 mm in diameter wihtin 1 mm of the surface. It also showed the acoustic image of 100 won coin with fine contrast.

  • We demonstrate a high-speed recording based on field-induced manipulation in combination with an optical reading of recorded bits on Au cluster films using the atomic force microscope (AFM) and the near-field scanning optical microscope (NSOM). We reproduced 50 nm-sized mounds by applying short electrical pulses to conducting tips in a non-contact mode as a writing process. The recorded marks were then optically read using bent fiber probes in a transmission mode. A strong enhancement of light transmission is attributed to the local surface plasmon excitation on the protruded dots.

혐기성 조건에서 담체에 부착된 미생물의 관찰 (Microscope Examination of Attached Biofilm under Anaerobic Conditions)

  • Microstructural examinations were performed on the anaerobic biofilm from reactor filled with PE support media. Optical microscope, SEM and fluorescent microscope were used for qualitative and morphological studies on the attached microorganism under anaerobic condition. Microorganisms were attached in crevices where protection from shear forces of surfaces where easy to contact with support media surface. A hypothesis for biofilm accumulation occurs on a surface such as polymer support media is presented schematically : 1st step ; cell-support media attachment, 2nd step ; cell-support media attachment and cell-cell attachment, 3rd step ; attached biofilm from neighboring crevices joins together and growing, 4th step ; mature and irregualar biofilm was formed. In SEM photographs, shape and structures of biofilm were observed, but microorganism species and methanogens were not identified. A large number of methanogenic bacteria were identified on the surface of PE substratum by fluorescence under 480nm of radiation and it was estimated that methanogenic bacteria was related to initial attachment of bacteria under anaerobic condition.

  • The most powerful analytical equipment usually comes at the cost of having the highest demand for space. Where electron microscopes has traditionally required a room to themselves, not just for reasons of their size but because of ancillary demands for pipes and service. The simple optical microscopes, of course, can occupy the desk-top, but because their performance is limited by the wavelength of light, their powers of magnification and resolution are inferior to that of the electron microscope. Mini SEM will sit comfortably on a desk-top but offers magnification and resolution performances much closer to that of a standard SEM. This new technique extends the scope of SEM as a high-resolution microscope, relatively cheap and widely available imaging tool, for a wider variety of samples.

대두(조사)의 발효에 의한 미세구조 변화에 관한 연구 (Studies on the Microstructure of Soybean (Irradiated) During Fermentation)

  • It was observed by electron microscope (transmission electron microscope, Scanning electron microscope) as a study on microstructure of soybean after r-ray irradiation with the intensity of 5KGY, 7KGY, 10KGY and 15KGY, fermented with the named Bacillus subtilis SCF, which newly separated and identified. According to the progress fermentation, changes of soybean microstructure have been increased, especially irradiated soybeans more increased than non-irradiated them. Observation of microstructure by electron microscope showed that each protein body became more. expanded in the dimension and decomposed, spherosome around the protein body in unit area dispersed and dwindled in the numbers of it. As the fermentation on progress, changes of soybean microstructure were suitable on fermentation period of 7KGY soybean, 48-72hrs fermentation.

근접장 주사 광학 현미경을 이용한 표면 플라즈몬의 측정 (Measurement of surface plasmon using near-field scanning optical microscope)

  • 표면 플라즈몬은 금속 표면에 존재하는 자유전자의 밀도파이며, 광자와 결합하여 존재한다. 본 논문에서는 근접장 주사 광학 현미경을 구성하고 이를 이용하여 표면에서 지수함수적으로 감소하는 표면 플라즈몬과 두 표면 플라즈몬에 의한 간섭무늬를 측정하였다. 표면 플라즈몬은 표면의 작은 광학적 분포에도 민감하게 변화하여, 광학 탐침이 금속 표면에 접근함에 따라서 표면 플라즈몬의 공명 조건에 영향을 준다. 이로 인해 광학 탐침이 금속 표면으로부터 약 250nm 떨어진 곳에서 광의 세기가 최대가 됨을 확인하였다.

PC카메라와 일반광학현미경을 이용한 빵효모 배양액의 효모세포 자동계수 (Automatic Counting of Yeast Cells in Baker's Yeast Culture Using PC Camera and Conventional Light Microscope)

  • Automatic counting of yeast cells in baker's yeast culture was tried using a conventional light microscope equipped with a pc camera. Relatively good binary image was obtained by using white LED as microscope light source, but uneven brightness distribution in original image hindered counting accuracy. A block binarization method using local thresholds proportional to local brightnesses was used to get improved binary images. The brightnesses of the blocks were expressed as the value component in HSV color model. Good quality binary images were obtained by binarization on $8{\times}6$ blocks of original images and connected-component labelling of the binarized images produced reliable counting results in the concentration range $1.4{\times}10^5/mL{\sim}1.4{\times}10^7\;cells/mL$.

유기 발광소자내 dark spot의 마이크로파 근접장 현미경(near-field scanning microwave microscope)을 이용한 연구 (Investigation of dark spots in OLEDs by using a near-field scanning microwave microscope)

  • 유기발광소자 안에 존재하는 비발광영역(dark spot)의 전압에 대한 영향을 근접장 마이크로파 현미경(near-field scanning microwave microscope)을 이용하여 관찰하였다. 유기발광소자는 glass/indiumtin oxide(ITO)/Cu-Pc/tris-(8-hydroquinoline)aluminum(Alq3)/aluminum(Al) 의 기본구조로 제작하였다. Dark spot은 ITO 기판을 부분적으로 에칭하여서 형성시켰다. Dark spot에 $0{\sim}l5 V$ 까지 전압을 인가시키면서 인가 전압에 따른 전기적 특성을 근접장 마이크로파 현미경 image의 변화와 반사계수인 $S_{11}$ 측정을 통하여 연구하였다.

  • In this study, we used Brewster-Angle Microscope(BAM) to study on the molecular orientation of monolayer on the water surface. The behaviors of molecules on three different subphase have been observed. Reproducible $\pi$-A isotherm have been obtained only on information about phase transition by molecular area. BAM facilitates the observation of morphology by optical anisotropy and thickness in monolayer and multilayers as BAM is shown to be sensitive to anisotropy of film. Every transition was found by BAM technique, either as a dramatic change in degree of contrast or as a sudden alteration of molecular action and $\pi$-A isotherm.

  • 열 증착 방법을 이용하여 copper(II)-phthalocyanine(CuPc) 박막을 glass 기판 위에 제작하였다. 열처리 조건은 $150^{\circ}C$에서 후열(annealing) 처리 하는 방식과 예열하는 두 가지 방식으로 달리하였다. 제작된 박막의 전기전도도를 평가하기 위해 마이크로웨이브 근접장 효과를 이용한 근접장 현미경(near-field scanning microwave microscope)을 이용하여 비파괴적인 방식으로 CuPc 박막의 반사계수(reflection coefficient)를 측정하였다. CuPc 박막의 전기전도도 특성을 UV 흡수도를 통한 에너지 밴드갭의 shift 현상과 관련지어 설명하고 또한 x-ray diffraction(XRD) data를 통해 박막의 결정 특성과 비교하였다. 박막 표면 특성은 SEM(scanning microscope microscopy)을 통해 관측하였다. 열처리 조건에 따른 CuPc 박막의 전기전도도 특성은 후열 처리한 박막의 경우 예열 처리한 박막보다 전기 전도 특성이 향상되었음을 관측할 수 있었다.
