Ethyl linoleate inhibits α-MSH-induced melanogenesis through Akt/GSK3β/β-catenin signal pathway

  • Ethyl linoleate is an unsaturated fatty acid used in many cosmetics for its various attributes, such as antibacterial and anti-inflammatory properties and clinically proven to be an effective anti-acne agent. In this study, we investigated the effect of ethyl linoleate on the melanogenesis and the mechanism underlying its action on melanogenesis in B16F10 murine melanoma cells. Our results revealed that ethyl linoleate significantly inhibited melanin content and intracellular tyrosinase activity in ${\alpha}$-MSH-induced B16F10 cells, but it did not directly inhibit activity of mushroom tyrosinase. Ethyl linoleate inhibited the expression of microphthalmia-associated transcription factor (MITF), tyrosinase, and tyrosinase related protein 1 (TRP1) in governing melanin pigment synthesis. We observed that ethyl linoleate inhibited phosphorylation of Akt and glycogen synthase kinase $3{\beta}$ ($GSK3{\beta}$) and reduced the level of ${\beta}-catenin$, suggesting that ethyl linoleate inhibits melanogenesis through $Akt/GSK3{\beta}/{\beta}-catenin$ signal pathway. Therefore, we propose that ethyl linoleate may be useful as a safe whitening agent in cosmetic and a potential therapeutic agent for reducing skin hyperpigmentation in clinics.

당귀와 구기자 에탄올 추출물의 피부 미백작용 (Skin Whitening Effects of Extracts from Angelicae Gigantis Radix and Lycii fructus Ethanol Extracts)

  • Objective : The purpose of this study was to investigate the synergistic effect of Angelicae gigantis Radix (AG) and Lycii fructus (LF) ethanol extracts on skin-whitening effects. Method : LFAG extracts were prepared by extracting with 80% ethanol. The efficacy of LFAG was judged by measurement of cell viability, tyrosinase activity, melanin production, tyrosinase and microphthalmia-associated transcription factor (MITF) expression in B16F10 murine melanoma cells by lipopolysaccharides (LPS) treatment. Results : Each extract (LF or AG) inhibited the tyrosinase activity in a dose-dependent manner. The co-treatment of LFAG extracts ($25{\mu}g/mL$ LF plus $25{\mu}g/mL$ AG) markedly suppressed the LPS-induced cellular tyrosine activity, melanin production, tyrosinase and MMP-1 expression in B16F10 murine melanoma cells. These suppressive effects were synergistically increased by their combination. Conclusions : With these observations, we suggest that the extracts from Lycii fructus and Angelica gigantis Radix could be potent natural materials for whitening skin.

  • The purpose of this study was to investigate the anti-melanogenic effects of the extracts from Decaschistia intermedia craib (EDI). In this study, we examined the effects of EDI on mushroom tyrosinase activity in in vitro, melanin contents, and expression levels of mRNA and proteins of melanogenesis-related genes in B16F10 melanoma cells. The treatment of EDI significantly decreased both tyrosinase activity and melanin contents in B16F10 cells with dose-dependent manner. In addition, we found that the expression of mRNA or proteins of melanogenic proteins, such as, a-melanocyte-stimulating hormone (a-MSH)-induced microphthalmia associated transcription factor (MITF), tyrosinase, tyrosinase related protein-1 (TRP-1), and TRP-2 was significantly downregulated with dose-dependent manner in the EDI-treated B16F10 cells compared to controls. Our results suggest that the EDI inhibits cellular melanogenesis through downregulation of a-MSH-stimulated melanin synthesis. Thus EDI may potentially be an effective whitening agent.

ERK Activation by Fucoidan Leads to Inhibition of Melanogenesis in Mel-Ab Cells

  • Fucoidan, a fucose-rich sulfated polysaccharide derived from brown seaweed in the class Phaeophyceae, has been widely studied for its possible health benefits. However, the potential of fucoidan as a possible treatment for hyperpigmentation is not fully understood. This study investigated the effects of fucoidan on melanogenesis and related signaling pathways using Mel-Ab cells. Fucoidan significantly decreased melanin content. While fucoidan treatment decreased tyrosinase activity, it did not do so directly. Western blot analysis indicated that fucoidan downregulated microphthalmia-associated transcription factor and reduced tyrosinase protein expression. Further investigation showed that fucoidan activated the extracellular signal-regulated kinase (ERK) pathway, suggesting a possible mechanism for the inhibition of melanin synthesis. Treatment with PD98059, a specific ERK inhibitor, resulted in the recovery of melanin production. Taken together, these findings suggest that fucoidan inhibits melanogenesis via ERK phosphorylation.

Melanogenesis Inhibitory Activity of Epicatechin-3-O-Gallate Isolated from Polygonum amphibium L.

  • This study aimed to investigate the melanogenesis inhibitory activity of epicatechin-3-O-gallate (ECG) isolated from Polygonum amphibium L. ECG was isolated from the ethanol extract of P. amphibium L, and its chemical structure was determined using spectroscopic methods such as LC-ESI-MS, 1D-NMR, and UV spectroscopy. ECG inhibited the melanogenesis of B16F10 cells in a dose-dependent manner. Particularly, it decreased the melanin content by 27.4% at 200 µM concentration, compared with the control, in B16F10 cells, without causing cytotoxicity. It is noteworthy that the expression of three key proteins, including tyrosinase, tyrosinase-related protein-1 (TRP-1), TRP-2, and microphthalmia-associated transcription factor (MITF), involved in melanogenesis, is significantly inhibited by ECG. The ECG isolated in this study caused the inhibition of body pigmentation and tyrosinase activity in vivo in the zebrafish model. These results suggest that the ECG isolated from P. amphibium L. is an effective anti-melanogenesis agent.

흑미의 주정 추출물이 melanin 생성과 관련된 tyrosinase 및 항산화 활성에 미치는 영향 (Effect of Black Rice (Oryza sativa L. indica) Ethanolic Extract on Tyrosinase Activity and Antioxidant Activity Related to Melanin Production)

  • 백모 발생은 연령이 증가함에 따라 모낭에 있는 melanocyte 수가 감소됨과 동시에 세포활성이 저하되고 동시에 과산화수소($H_2O_2$)가 축적되어 melanin 합성이 감소되어 진행된다. 본 연구의 목적은 흑미 주정추출물인 Black oryzasativa ethanolic extract (BLEE)가 백모에서 흑모로 전환되는데 관련되어 있는 melanin 합성에 어떠한 영향을 미치는지 알아보고자 하였다. 본 연구에서 사용된 BLEE는 DPPH radical scavenging assay 및 환원력 실험에서 $64{\mu}g/ml$에서 항산화 효과가 있는 것으로 나타났으며, tyrosinase 활성 및 melanin 생성 촉진효과는 $16{\mu}g/ml$ 이상에서 효과가 나타났다. DCF fluorescence assay에서 세포내 $H_2O_2$를 소거하는 효과가 있는 것으로 나타났고, $H_2O_2$를 처리한 세포에서 melanin 생성 촉진효과가 있는 것으로 나타났다. 그러나 Western blot 분석에서는 catalase의 발현을 감소시키는 것으로 나타났으며 melanin 생성에 관여하는 tyrosinase, TRP-1, TRP-2, 효소 및 MITF 전사인자의 발현촉진에는 영향이 없는 것으로 나타났다. 그러므로 BLEE의 melanin 생성 촉진효과는 tyrosinase 활성 증가와 세포내 $H_2O_2$의 생성을 감소시켜 melanin 생성을 촉진시키는 것으로 나타났다. 결론적으로 BLEE는 melanin 생성을 촉진시키는데 있어서 중요한 역할을 하고 있는 것으로 확인되었으며, 본 연구에서 개발된 BLEE는 흑모성장과 연관이 있는 melanin생성 촉진과 관련된 모발화장품 개발분야에서 하나의 잠재적인 기능성 소재로 응용 될 수 있다는 것을 시사하고 있다.

MSH에 의해 자극된 B16F10세포에서 사간(射干)의 멜라닌 합성 억제 효과 (Inhibitory Effect of Belamcandae Rhizoma on the Melanogenesis in MSH-stimulated B16F10 cells)

  • Objective : The present study was designed to assess the potential inhibitory activity of an ethanol extract of Belamcandae Rhizoma (EBR) on the alpha-melanocyte stimulating hormone (${\alpha}$-MSH)-induced melanogenesis signal pathway in B16F10 melanoma cells. Methods : Several experiments were performed in B16F10 melanoma cells. We studied tyrosinase activity, melanin content, cell-free tyrosinase activity and DOPA stain, and performed Western blots and RT-PCR for proteins and mRNA involved in melanogenesis. Results : ${\alpha}$-MSH-induced tyrosinase activity and melanin content were inhibited significantly by EBR. EBR markedly suppressed the protein expression level of tyrosinase in B16F10 melanoma cells. On the other hand, the expression of tyrosinase-related protein-1 (TRP-1) and -2 (TRP-2; DCT) were not affected by EBR. To elucidate the mechanism of the depigmenting property of EBR, we examined the involvement EBR in cAMP response element binding (CREB) protein phosphorylation and microphthalmia-associated transcription factor (MITF) signalling induced by ${\alpha}$-MSH. EBR did not regulate CREB phosphorylation and MITF expression by ${\alpha}$-MSH. Nevertheless, the mRNA expression of tyrosinase was significantly attenuated by EBR treatment without changes in the expression of TRP-1 and -2 mRNA. Conclusion : Our study suggested that EBR inhibits ${\alpha}$-MSH-induced melanogenesis by suppressing tyrosinase mRNA.

은행 열매 추출물의 멜라닌 생성 저해효과 (Inhibitory Effect of Ginkgo biloba Extracts on Melanin Biosynthesis)

  • 본 연구는 은행 열매 오일의 멜라닌 생성 억제 효과를 확인한 것이다. 은행나무 열매 오일은 DPPH assay와 FRAP assay를 사용하여 라디컬 소거능을 시험하였다. 결과적으로 은행나무 열매 오일은 DMSO를 용매로 0.06% 녹였을 때, DPPH assay에서 9.96% 소거활성을 나타내었고 FRAP는 1.33 mM의 ferric sulfate ($FeSO_4$)를 생성하였다. 은행 열매 오일은 tyrosinase inhibition assay에서 37.72%의 억제력을 가졌고 B16/F10 세포에 멜라닌 생합성 실험을 통해 확인하였다. 은행 오일 0.06%에서 ${\alpha}$-MSH 처리 구에 비해 48.02%의 멜라닌 생성을 억제하였다. Tyrosinase, tyrosinase related protein-1 (TRP-1), tyrosinase related protein-2 (TRP-2), microphthalmia-associated transcription factor (MITF)의 유전자 발현 수준은 control군에 비해 0.04%와 0.06% 농도 군이 크게 감소하였다. 결과적으로 은행 열매 오일 추출물이 멜라닌 생성을 억제하는 효과가 있는 것으로 확인되었다.

Inhibitory Effect of Pinus rigida × Pinus taeda on Melanogenesis in B16 F10 Cells

  • The cone of Pinus rigida × Pinus taeda (PRT), a plant in the Pinaceae family, has long been used in traditional medicine to treat hemostasis, bruises, and burns. Previous research has shown that regulating oxidation-reduction reactions in reactive oxygen species can help inhibit melanogenesis, the process of melanin synthesis, which is a common target for addressing hyperpigmentation. Inhibiting tyrosinase is also known to be effective in this regard. Based on these findings, we conducted an investigation into the inhibitory effect of the ethyl acetate fraction of PRT (ERT) on melanogenesis in B16 F10 cells. We know that the expression levels of melanin biosynthesis-related proteins, including tyrosinase, TRP-1, and TRP-2, are regulated by MITF (microphthalmia-associated transcription factor) and cAMP, with cAMP affecting the activity of protein kinase A (PKA). PKA can reduce melanogenesis, and CREB reduces the phosphorylation of melanin-producing enzymes. In addition, the MAPK signaling pathway, composed of ERK, JNK, p38, and other factors, is also known to play a role in the inhibition of melanogenesis in melanocytes. Our immunoblotting results showed that ERT inhibited the expression of melanin production-related proteins (tyrosinase, TRP-1, TRP-2, and MITF) that were significantly increased by a-MSH treatment to promote melanin production. Furthermore, the phosphorylation levels of factors related to cAMP/PKA/CREB and MAPK signaling pathways were significantly reduced without affecting the total form. In conclusion, we believe that treatment with ERT can inhibit melanin synthesis by modulating the phosphorylation of cAMP/PKA/CREB and MAPK signaling pathways at the cellular level. These findings suggest the potential of ERT as a raw material for functional cosmetics and pharmaceuticals, thanks to its antioxidant activity and ability to inhibit melanogenesis. We thought that these findings of ERT as a natural plant resource will inspire further research and development in this area.

Dendrobium speciosum 에탄올 추출물의 melanin 생성 억제 효능 및 주름개선 효과 (Anti-melanogenesis and Anti-wrinkle Properties of Korean Native Dendrobium speciosum Ethanol Extract)

  • 본 연구에서는 Dendrobium speciosum 에탄올추출물의 티로시나아제 억제를 통한 멜라닌형성 저해 효과 및 주름개선효과에 대해 연구하였다. Dendrobium speciosum추출물은 α-MSH만 처리한 군에 비해 티로시나아제 활성을 농도 의존적으로 저해시킴으로서 멜라닌 함량을 감소시켰다. 더 자세한 메카니즘을 알아보기 위하여 멜라닌 합성 관련 유전자인 Tyrosinase-related protein-1, tyrosinase-related protein-2, Microphthalmia-associated transcription factor 발현을 확인하였을 때 Dendrobium speciosum처리군에서 유전자 발현이 농도 의존적으로 감소하였다. 또한 Dendrobium speciosum처리 시 type I procollagen 합성이 증가함으로써 Dendrobium speciosum처리가 collagen 생합성을 증대시킬 것으로 사료된다. 이 결과를 바탕으로 Dendrobium speciosum에탄올 추출물 처리 시 멜라닌합성 관련 유전자의 발현 및 티로시나아제 활성을 저해시켜 멜라닌 합성이 감소됨에 따라 효과적인 미백활성을 가지며 collagen합성을 증대시킴으로서 주름개선에 탁월한 효과를 가지는 것으로 사료되며 이는 기능성 화장품 소재로 사용가능할 것으로 사료된다.
