림프구의 미소핵을 지표로 영광 원자력발전소 주변 사육 돼지의 방사선 생물학적 평가 (Radiobiological Evaluation in Pig Bred in the Vicinity of Yeonggwang Nuclear Power Station Using Micronuclei in Cytokinesis-blocked Lymphocyte)

  • Cytogenetic and hematological analysis was performed in peripheral blood of pig in the vicinity of Yeonggwang nuclear power station and control area. The frequency of micronuclei (MN) in peripheral blood lymphocytes from pig was used as a biomarker of radiobiological effects resulting from exposure to environmental radiation. An estimated dose of radiation was calculated by a best fitting linear-quadratic model based on the radiation-induced MN formation from the swine lymphocytes exposed in vitro to radiation over the range from 0 Gy to 4 Gy. MN rates in lymphocytes of pig from Yeonggwang nuclear power station and control area were 10.60/1,000 and 11.10/1,000, respectively. There were no significant differences in MN frequencies and hematological values in pig between Yeonggwang and control area. The study indicates that the MN assay in lymphocyte of pig is a rapid, sensitive and accurate method that can be used to monitor a large population exposed to radiation.

  • The frequencies of gamma-ray-induced micronuclei (MN) in cytokinesis-blocked (CB) lymphocytes at several doses were measured in three donors of Korean native cattle and Korean native goat. Measurements performed after irradiation showed a dose-related increases in MN frequency in each of the donors studied. When analysed by linear-quadratic model the line of best fit was cattle : y : 0.1016D +$0.0118D^2$+0.0147, goat : y = 0.1353D +$0.0043D^2$+0.0087 (y : number of MN/CB cells and D = irradiation dose in Gy). The relative sensitivity of goat lymphocytes compared with cattle lymphocytes was estimated by best fitting linear-quadratic model based on the radiation-induced MN data over the range from 0 Gy to 4 Gy. In the case of MN frequency with 0.05, 0.1, 0.2, 0.4 and 0.8, the relative sensitivities of goat lymphocytes were 1.106. 1.166. 1.140, 1.069 and 0.976 respectively. Our in vitro radiobiological study confirmed that the cytogenetic response obtained in blood from cattle and goat can be utilized for application in environmental studies.

  • The interaction of extremely low frequency magnetic field (ELF-MF) on the frequency of micronuclei (MN) induced by benzo(a)pyrene (BP) in human lymphocytes was examined. A 60 Hz ELF-MF of 0.8 mT field strength was applied for 24 hours either alone or with the tumour initiator, BP. The frequency of MN induced by BP increased in a dose-dependent manner.(omitted)

  • Endocrine disruptors have been implicated in carcinogenesis in animal studies, but carcinogenetic effects on human remain controversial. In order to examine the genotoxicity of two common endocrine disruptors, Bisphenol A and Diethylstilbestrol, cytogenetic endpoints including chromosome aberration (CA), sister chromatid exchange (SCE), micronuclei (MN) analyses and DNA damage by single cell gel electrophoresis (SCGE) were assessed. The effects of Bisphenol A and Diethylstilbestrol on the frequencies of CA and MN were increased in a dose-dependent manner and that of Bispheol A was more significant by Kendall'$\tau$test. Bisphenol A and Diethylstilbestrol also increased the frequency of SCE. Bisphenol A and Diethylstilbestrol induced DNA damage in a dose-dependent manner and the DNA damage induced by Diethylstilbestrol in human blood lymphocytes was more significant.

마우스 소핵(Micronuclei, MN) 시험방법을 이용한 감잎차 추출물의 돌연변이 억제효과 (Antimutagenic Effects of Persimmon Leaf Tea Extract (PLTE) in Mice Using Micronucleus Induction (MN) Test)

  • The antimutagenic effects of persimmon leaf tea extract (PLTE) at concentration levels consumed by human were examined in mice using micronuleus induction with MMC(mitomycin C) or 4-NQO (4-nitroquinoline-1-oxide). When mice received oral gavage of 10 equivalent to PLTE 24 hr and 6 hr before, and 5 equivalent to PLTE 6 hr before and 3 hr after intraperitoneal injection of MMC, a significant decrease in the frequency of micronuclei were observed. The induction of micronuclei by 4-NQO was suppressed by oral dosage of PLTE at 5 equivalent to PLTE 6 hr before and 3 hr after, 10 equivalent to PLTE 3 hr before and 3 hr after intraperitoneal injection of MMC. Though the components of PLTE have not been analyzed so far, our present results suggest the existence of several bio-antimutagens and/or desmutagens in PLTE, beside catechin, well-known antimutagen.

An estimated dose of radiation was calculated by a best fitting linear-quadratic model based on the radiation-induced MN formation from the bovine lymphocytes exposed in vitro to radiation over the range from 0 Gy to 4 Gy. MN ratio in lymphocytes of cattle from Uljin nuclear power station and control area were 8.90/1,000 and 9.60/1,000, respectively. There were no significant differences in MN frequencies and hematological values in cattle between Uljin and control area.

Increased Frequency of Micronuclei in Binucleated Lymphocytes among Occupationally Pesticide-exposed Populations: A Meta-analysis

  • Background: The cytokinesis-block micronucleus (CBMN) assay is a standard cytogenetic tool employed to evaluate chromosomal damage subsequent to pesticide exposure. Objectives: To evaluate the pooled levels of total micronuclei (MN) and binucleated cells with micronuclei (MNC) in 1000 binucleated lymphocytes among population occupationally exposed to pesticides and further determine the more sensitive biomarker of CBMN. Materials and Methods: A meta-analysis on the pooled levels of MN and MNC in binucleated lymphocytes among occupationally pesticide-exposed populations was conducted using STATA 10.0 software and Review Manager 5.0.24 in this study. Results: We found significant differences in frequencies of MN and MNC in 1000 binucleated lymphocytes between pesticide-exposed groups and controls, and the summary estimates of weighted mean difference were 6.82 [95% confidence interval (95% CI): 4.86-8.78] and 5.08 (95% CI: 2.93-7.23), respectively. However, when we conducted sensitivity analyses further, only the MN remained statistically different, but not the MNC, the summary estimates of weight mean difference were 2.86 (95% CI: 2.51-3.21) and 0.50 (95% CI: -0.16-1.17), respectively. We also observed pesticide-exposed subjects had significantly higher MN frequencies than controls among smokers and nonsmokers, male and female populations, and American, Asian and European countries in stratified analyses. Conclusions: The frequency of MN in peripheral blood lymphocytes might be a more sensitive indicator of early genetic effects than MNC using the CBMN assay for occupationally pesticide-exposed populations.

Relationships between Malignant Melanoma and Chromosome Damage in Human Peripheral Blood Lymphocytes

  • The incidence of malignant melanoma increases with age. One significiant effect of aging processes is an accumulation of oxidative damage in the genetical material. In this study, the relationship between malignant melanoma and damage in chromosomes and proliferative effectiveness of human peripheral lymphocytes were investigated by the micronucleus (MN) technique. A total of 15 malignant melanoma patients and appropriately matching 15 healthy controls were involved in the study. MN frequencies and proliferative indexes (PI) after non toxic levels of hydrogen peroxide treatment were also measured to determine damaging effect of oxidative stress in genome in addition to measuring the spontenous levels of micronuclei and PI. The patient group had a significantly higher rate of spontaneous MN than the control group (p<0.01). After treatment with $H_2O_2$, MN frequencies in the patient group was significantly decreased (p<0.01) although there was no difference between the treated and untreated results of control group (p=0.29). There was also difference (p<0.01) between the MN frequencies of the patient and the control group either in the spontaneous levels or in the $H_2O_2$ treated groups. The same significant difference persisted when the PI values were compared between patient and control groups. Increase in the MN frequency in patients could mean the alterations in the chromosomal structure which may lead to the chromosome instability and therefore genetic susceptibility to cancer. This increased number of micronuclei can also be used for cytological marker in identifying high risk cases for malignant melanoma.

  • A technique to improve the analysis of micronuclei(MN) in lymphocytes as a cytogenetic indicator is reported. For the purpose of diminishing the variation of the result from individual reader and making it easier to distinguish accurately a cytokinesis blicked(CB) lymphocyte and micronuclei, we tried a modified one-step silver staining technique as a method for detection of the argyrophilic nucleolar organizer region(AgNOR) with or without conventional Giemsa stain in the slide from CB method. Compared with the conventional Giemsa stain, the preparation processed with this method are especially useful for the accurate analysis of MN of cultured lymphocyte with cytochalasin B. This method will be a useful technique for automated calculation of MN.

An estimated dare of radiation was calculated by a best fitting linear-quadratic model based on the radiation-induced MN formation from the bovine lymphocytes exposed in vitro to radiation over the range from 0 Gy to 4 Gy. MN rates in lymphocytes of cattle from Wolsong nuclear power plant and control area were 9.87/1,000 and 9.60/1,000, respectively. There were no significant differences in MN frequencies and hematological values in cattle between Wolsong and control area. The study indicates that the MN assay is a rapid, sensitive and accurate method that can be used to monitor a large population exposed to radiation.
