  • We have shown that zeolite microcrystals can be readily organized in the form of uniformly oriented monoand multilayers on various substrates by well-defined chemical linkages based on covalent, ionic, and hydrogen bondings between the microcrystals and the substrates. This finding establishes the fact that micrometer-scale building blocks can be readily organized into organized entities through interconnection of the surface-tethered large number of functional groups. Since zeolite crystals have highly regular and uniform nanochannels and nanopores within them, the resulting mono and multilayers of zeolite microcrystals bear great potential to be utilized in various novel applications.

  • Cryo-electron microscopy (cryo-EM) is arguably the most powerful tool used in structural biology. It is an important analytical technique that is used for gaining insight into the functional and molecular mechanisms of biomolecules involved in several physiological processes. Cryo-EM can be separated into the following three groups according to the analytical purposes and the features of the biological samples: cryo-electron tomography (cryo-ET), cryo-single-particle reconstruction, and cryo-electron crystallography. Cryo-tomography is a unique EM technique that is used to study intact biomolecular complexes within their original environments; it can provide mechanistic insights that are challenging for other EM-methods. However, the resolution of reconstructed three-dimensional (3D) models generated by cryo-ET is relatively low, while single-particle reconstruction can reproduce biomolecular structures having near-atomic resolution without the need for crystallization unless the samples are large (>200 kDa) and highly symmetrical. Cryo-electron crystallography is subdivided into the following two categories according to the types of samples: one category that deals with two-dimensional (2D) crystalline arrays and the other category that uses 3D crystals. These two categories of electron-crystallographic techniques use different diffraction data obtained from still diffraction and continuous-rotation diffraction. In this paper, we review crystal-based cryo-EM techniques and focus on the recently developed 3D electron-crystallographic technique called microcrystal-electron diffraction.

  • Well-defined solid-state microcrystalline structures from bispyridyl Zn-porphyrin have been successfully synthesized. The coordinative interactions between pyridine and Zn are main responsible for this translation of porphyrin molecular building blocks to crystalline microscopic objects. The hexagonal plates are obtained from acetonitrile and rhombus plates are grown from toluene solution. With a simple manipulation during the microcrystal growth, such as growth temperature and time, the morphologies can be controlled by adopting different molecular packing. Consequently, morphologies of microcrystals have been diversified.

  • The optical properties of individual 3-to 14-micron diameter CdS crystalline spheres embedded in poly(methyl methacrylate) were studied using elastic scattering. The presence of well defined sharp peaks in the 550 to 600 nm elastic scattering spectra confirmed that each microcrystal acts as an optical cavity with cavity quality factors exceeding 104. Such natural resonator microcrystals should lead to greatly enhanced local field effects near the surface of CdS, quantum electrodynamic modification of optical transition rates of nearby species and altered photochemistry. Absorptive heating following high intensity laser irradiation was found to induce a transient washout of the high Q modes.

  • Perovskite structured barium titanate particles ($BaTiO_3$) platelets were synthesized by molten salt synthesis and topochemical microcrystal conversion. As the precursors of $BaTiO_3$, plate-like $BaBi_4Ti_4O_{15}$ particles were first synthesized by the reaction of $Bi_4Ti_3O_{12}$, $BaCO_3$, and $TiO_2$ at $1080^{\circ}C$ for 3 h in $BaCl_2$-KCl molten salt. After the topochemical reactions, layer-structured $BaBi_4Ti_4O_{15}$ particles transformed to the perovskite $BaTiO_3$ platelets. $BaTiO_3$ particles with thickness of approximately $0.5{\mu}m$ and a length of $10-15{\mu}m$ retained the morphology feature of the $BaBi_4Ti_4O_{15}$ precursor. For <001> $Pb(Mg_{1/3}Nb_{2/3})O_3-32.5PbTiO_3$ (PMNT)-5 wt % PbO piezoelectric ceramics textured with 5 vol % of $BaTiO_3$ templates, the Lotgering factor reached 0.82, and $d_{33}$ was 870 pC/N.

  • Silver chlorobromoiodide cubic seed emulsion was prepared'in the presence of ammonia and phenylcarbamyl gelatin solution. When benzenethiosulfonate sodium salt (BTS) and 4-hydroxy6-methyl-1,3,3a,7-tetraazaindene (TAI), as growth modifiers, were added in the seed emulsion, the silver halide microcrystals were obtained in the forms of cubo-octahedrons, octahedrons, rhombic dodecahedron by variation of pAg value through controlled double jet. The crystal habits and shapes were determined by means of X-ray diffraction (D/MAX BIB, Rigaku) and electron microscopy (JSM-840, JEOL), respectively.

  • We have developed a simple method for tuning the morphologies of $Cu_2O$ microcrystals. $Cu_2O$ microcrystals were prepared by precipitation at room temperature from a mixture of $Cu(CH_3COO)_2{\cdot}H_2O$, N,N,N',N'-tetramethyl ethylenediamine (TMEDA), ascorbic acid, and polyethylene glycol (PEG). TMEDA was used to promote the formation of copper-TMEDA complexes. A variety of $Cu_2O$ microcrystal morphologies were obtained simply by varying the concentrations of TMEDA and ascorbic acid. Aggregated $Cu_2O$ microspheres are formed at higher concentrations of ascorbic acid in the absence of TMEDA. Aggregated $Cu_2O$ microcubes are formed at lower concentrations of ascorbic acid and higher concentrations of TMEDA. The crystal growth mechanism of these $Cu_2O$ morphologies is explained.

  • Amphiphilic (porphyrin)Sn$(OH)_2$ molecular building block can directly translate into well-defined solid-state microcrystalline structures. The crystalline diamond plates are obtained from ethanol and crystalline square plates are grown from methanol solution. With a simple synthetic manipulation during the microcrystal growth, the morphologies can be controlled by adopting different molecular packing. Consequently, morphologies of microcrystals have been diversified. Furthermore, the macroscopic crystals were obtained in the presence of cetyltrimethylammonium bromide (CTAB).

  • This study was designed to investigate the stability of a lipase fused with a cellulosebinding domain (CBD) to cellulase. The fusion protein was derived from a gene cluster of a CBD fragment of a cellulase gene in Trichoderma hazianum and a lipase gene in Bacillus stearothermophilus L1. Due to the CBD, this lipase can be immobilized to a cellulose material. Factors affecting the lipase stability were divided into the reaction-independent factors (RIF), and the reaction-dependent factors (RDF). RIF includes the reaction conditions such as pH and temperature, whereas substrate limitation and product inhibition are examples of RDF. As pH 10 and $50^{\circ}C$ were found to be optimum reaction conditions for oil hydrolysis by this lipase, the stability of the free and the immobilized lipase was studied under these conditions. Avicel (microcrystalline cellulose) was used as a support for lipase immobilization. The effects of both RIF and RDF on the enzyme activity were less for the immobilized lipase than for the free lipase. Due to the irreversible binding of CBD to Avicel and the high stability of the immobilized lipase, the enzyme activity after five times of use was over $70\%$ of the initial activity.

  • In this study, the yeast Pichia pastoris was genetically modified to assemble minicellulosomes on its cell surface by the heterologous expression of a truncated scaffoldin CipA from Clostridium acetobutylicum. Fluorescence microscopy and western blot analysis confirmed that CipA was targeted to the yeast cell surface and that NtEGD, the Nasutitermes takasagoensis endoglucanase that was fused with dockerin, interacted with CipA on the yeast cell surface, suggesting that the cohesin and dockerin domains and cellulose-binding module of C. acetobutylicum were functional in the yeasts. The enzymatic activities of the cellulases in the minicellulosomes that were displayed on the yeast cell surfaces increased dramatically following interaction with the cohesin-dockerin domains. Additionally, the hydrolysis efficiencies of NtEGD for carboxymethyl cellulose, microcrystal cellulose, and filter paper increased up to 1.4-fold, 2.0-fold, and 3.2-fold, respectively. To the best of our knowledge, this is the first report describing the expression of C. acetobutylicum minicellulosomes in yeast and the incorporation of animal cellulases into cellulosomes. This strategy of heterologous cellulase incorporation lends novel insight into the process of cellulosome assembly. Potentially, the surface display of cellulosomes, such as that reported in this study, may be utilized in the engineering of S. cerevisiae for ethanol production from cellulose and additional future applications.
