Purification and Characterization of an Insect Antibacterial Peptide, Defensin, Expressed in Saccharomyces cerevisiae (Saccharomyces cerevisiae에서 발현한 곤충 항균펩티드, defensin의 정제 및 특성 조사)

  • We investigated the biochemical properties of insect defensin expressed and secreted from Saccharomyces corevisiae. The defensin showed extremely high resistance to boiling for up to 30 min and to pH values tested from 2.0 to 12.0. The treatment of defensin with various proteases abolished antibacterial activity. However, amylases, cellulase, lipase and catalase had no effect on the activity. The defensin was purified to homogeneity through ammonium sulfate concentration of culture supernatant, SP-Sepharose column chromatography and RP-HPLC. Tricin-SDS-PAGE analysis revealed that the molecular weight of the defensin was about 4.0 kDa. The antibacterial activity of the purified defensin was verified by renaturation of stained gel and gel pouring assay using Micrococcus luteus as a test organism.

Isolation and Identification of Bacteria from Air Conditioners and its Hygiene (에어컨내 세균의 분리, 동정 및 위생)

  • This study was performed to identify the microorganisms from air conditioners and to investigate hygiene management on air conditioners. Eight species of bacteria were isolated and identified from twenty samples of coolers in air conditioners; Pantoea sp., Bacillus circulans, Bacillus pumilus, Corynebacterium, Flavimonas oryzihabitans, Ochrobacterum anthropi, Micrococcus sp., and non fermented bacilli (NFB). One thousand and three hundreds twenty-two persons who used air conditioners in their houses were investigated about the state of hygiene management in their air conditioners. One thousand and one hundred thirty eight persons (86%) of the total investigated persons ventilated their air conditioners and 1,128 persons (85%) of them cleaned their air conditioners. However, 864 persons (66%) of them did not clean filters and 1,089 persons (82%) did not clean the heat exchangers, both of which air conditioners could be easily contaminated by microorganisms. From these results, we could conclude that the contaminants, bacteria as mentioned the above, in air conditioners could cause human disease such as respiratory infections if the number of bacteria increase in air conditioners. Thus, the removal of contaminants and the improved hygiene of the air conditioners can prevent human diseases caused by the released bacteria during the use of air conditioners.

Characterization of Bunker Oil-Related Compounds Degrading Bacteria Isolated from Pusan Coastal Waters (부산근해에서 분리한 Bunker Oil 관련화합물 분해세균의 특성)

  • Microorganisms utilizing petroleum as substrate were screened from the seawater in Pusan coastal area. Among them, fifty strains utilized bunker-A oil as a sole carbon and energy source. Five of these fifty strains were selected to experiment this study. According to the taxonomic characteristics of its morphological, cultural and biochemical properties, the selected stains were named Pseudomonas sp. EL-12, Flavobacterium sp. EL-15, Acinetobacter sp. EL-18, Enterobacter sp. EL-27 and Micrococcus sp. EL-43, respectively. The optimal medium compositions and cultural conditions for assimilation of bunker-A oil by the selected strains were 1.5-2% bunker-A oil, 0.1% $NH_4NO_3$, 1-1.5% $MgSO_4$.$7H_2O$, 0.05-0.15% KCl, 0.1-0.15% $CaCl_2$.$2H_2O$, 2.5-3.5% NaCl, initial pH 8-9, temperature 3$0^{\circ}C$ and aeration, respectively. The utilization and degradation characteristics on the various hydrocarbons by the selected stains were showed that bunker oil, n-alkane and branched alkane compounds were highly activity than cyclic alkane and aromatic hydrocarbon compounds.

Red-Colored Phenomena of Ginseng(Panax ginseng C. A. Meyer) Root and Soil Environment (인삼근 적변현상과 근권 토양환경)

  • In order to elucidate the mechanism of red-colored phenomena(RCP) in ginseng(Panax ginseng C.A. Meyer), distribution of inorganic elements of ginseng root and its surrounding soil, and microflora in the soil were investigated. Red brown colored-substances were accumulated in the cell wall of epidermis at early stage of red-colored ginseng (RCG). Cell wall of the late stage of RCG was disordered and microorganisms were shown in the disordered cell wall. Al, Si and Fe contents among inorpanic elements in the epidermis of RCG were higher at two or three times than that of healthy ginseng. On the other hand, K content was higher at three times in healthy ginseng than that of RCG. Especially, Fe content was higher at three times in lateral roots of RCG than that of healthy ginseng. Total 21 strains of microorganisms were isolated on the 523 medium from surface soil, surrounding soil of both healthy and RCG, and RCG. Six strains of microorganisms among them were resistant to 2 mM Fe. Two species in Bacillus app. and Lactobacillus app. , and one species in Micrococcus sp. and Npisseria sp. respectively were identified. It seemed that RCP was closely related with the distribution and uptake of inorganic elements, was also correlated Fe-independent metabolism of microorganisms.

Effects of Organic Nutrients on Chitinase Production in Minimal Media (배양액중의 유기영양물이 Chitinase 생산에 미치는 영향)

  • Four chitinase producing bacteria, Arthrobacter nicotinae CH4, Arthrobacter nicotinae CHI3, Arthrobacter sp. CH5 and Micrococcus sp. CH3, were isolated from small crabs and shrimps. We investigated the optimum medium condition for the production of enzyme and high cell mass. The preferable medium composition was as follows: colchitin 0.1 %(w/v), glycerol 0.25%(w/v) and yeast extract 0.05%(w/v) in minimal midium ($K_{2}HPO_{4}$ 0.7 g/l, $KH_{2}PO_{4}$ 0.3 g/l, $MgSO_{4}{\cdot}5H_{2}O$ 0.5 g/l, $FeSO_{4}}{\cdot}7H_{2}O$ 0.01 g/l, $ZnSO_{4}$ 0.001 g/l, $MnCI_2$ 0.001 g/l, pH 7.0). This cell culture medium could be used directly as sample for measuring chitinase activity. Because it hardly conreducing sugar such as glucose (blank value=0), the detected reducing sugar can be considered as a chitinase reaction product. The results can be used for easy preparation method for determination of enzyme activity and analysis of enzyme-substrate reaction in step of screening of chitinase producing bacteria.

Characterization of the Extracellular Autolysins from Moraxella sp. CK-1. (Moraxella sp. CK-1의 세포외 Autolysins 특성 연구)

  • We studied the characteristics of extracellular autolysins from Moraxella sp. CK-1 which has been known to lyse the cyanobacterial cell walls. This bacterium excreted autolysins from the early exponential growth phase. These enzymes showed optimal action condition of 60-$70^{\circ}C$ and pH 9.0. Whereas $Na^{+}$, $K^{+}$ and $Li^{+}$ ions exhibited positive effect on the enzyme activity, $Ba^{2+}$, $Mg^{2+}$, $Ca^{2+}$ and $Mn^{2+}$ ions exhibited negative effect. Especially, $Fe^{2+}$ and $Cu^{2+}$ ions almost completely suppressed the activity. Four extracellular autolysins of 30, 32, 38 and 41 kDa were detected in renaturing SOS-PAGE gel containing 0.2% heat-killed Micrococcus luteus cells as substrate. Among these 4 autolysins, 2 enzymes of 32 and 41 kDa distributed in the culture medium throughout the experimental time, but the 38 kOa enzyme diminished and 30 kOa began to appear at mid-exponential growth phase. When SOS-insoluble peptidoglycan of M. luteus was treated with the autolysins of Moraxella sp. CK-l, the concentration of free amino groups in reaction mixture increased. This indicates that the autolysins are N-acetylmuramyl-L-alanine amidase or endopeptidase.

Purification and Characterization of the Bacteriocin Produced by Lactococcus sp. KD 28 Isolated from Kimchi (김치에서 분리한 Lactococcus lactis가 생산하는 박테리오신의 정제 및 특성)

  • The bacterial strain isolated from Kimchi showed antibacterial activity against Micrococcus luteus IAM 1056. The selected strain was identified as Lactococcus lactis by 16S rRNA nucleotide sequence analysis and named as Lactococcus sp. KD 28. The treatment of culture supernatant with proteinase K removed antibacterial activity, indicating its proteinaceous nature, a bacteriocin. This bacteriocin was sensitive to hydrolytic enzymes such as ${\alpha}$-chymotrypsion, trypsin, proteinase K, lipase, ${\alpha}$-amylase and subtilisin A. The bacteriocin was highly thermostable and resistant to heating at $80^{\circ}C$ for up to an hour but 50 % of the total activity was remained at $100^{\circ}C$ for 30 min. The pH range from 2.0 to 8.0 had no effect on bacteriocin activity and it was not affected by solvents such as acetonitrile, isopropanol, methanol, chloroform and acetone up to 50% concentration. The bacteriocin showed antibacterial activity against M. luteus IAM 1056, Lactobacillus delbrueckii subsp. lactis KCTC 1058, Enterococcus faecium KCTC 3095, Bacillus cereus KCTC 1013, B. subtilis KCTC 1023, Listeria ivanovii subsp. ivanovii KCTC 3444, Staphylococcus aureus subsp. aureus KCTC 1916, B. megaterium KCTC 1098 and B. sphaericus KCTC 1184. The bacteriocin was purified through ammonium sulfate concentration, SP-Sepharose chromatography and RP-HPLC. The molecular weight was estimated to be about 3.4 kDa by tricine-SDS-PAGE analysis.

Diagnostic test for bovine mastitis by the determination of ATP based on firefly bioluminescence (Bioluminescence 반응에 의한 ATP 측정을 이용한 젖소 유방염 진단에 관한 연구)

  • This study was carried out to diagnostic test for bovine mastitis by the determination of adenosine triphosphate (ATP) based on firefly bioluminescence. The results obtained are follow; 1. The infection rate of bovine mastitis investigated with 521 cows in 47 dairy farms were found to be 3.6% of clinical form and 44.1% of subclinical form according to the degree of infection. 2. The light yield produced in firefly bioluminescence system was proportional to the concentration of ATP giving stright line within the range of 100PM~1uM. 3. When the number of somatic cell in milk was determined by the ATP assay and compared with three conventional methods such Fossomatic, California mastatic test (CMT), and rolling ball viscometer (RBV), it was shown that r=0.92 for Fossomatic, 0.63 for CMT and 0.7 for RBV. 4. The microorganisms causing mastitis were isolated Staphylococcus sp. (53.3%), Streptococcus sp. (17.9%), Micrococcus sp. (13.5%), Gram negative bacilli (6.3%), Gram positive bacilli (5.5%) and Yeast-like fungi (5.4%). 5. The endogeneous ATP levels of bacteria in a raw milk determined by the firefly bioluminescence system and compared with the results of the conventional methods. The correlation was 0.88 for raw milk.

  • The rational approach to antimicrobial therapy of infected root canals is based on accurate identification of the infecting organism and on the organism's susceptibility to antimicrobial agents as measured by standardized techniques. In establishing criteria for the selection of antibiotics, a susceptibility test should be performed. The purpose of this study was to investigate the susceptibility of 224 aerobic and anaerobic microbial strains isolated from infected root canals to various antibiotics. This was performed by using 7 antibiotic sensi-disc: Penicillin (10 units), Ampicillin (10 mcg), Tetracycline (30 mcg), Streptomycin (10 mcg), Kanamycin (30 mcg), Lincomycin (2 mcg), and Clindamycin (2 mcg). The results were as follows; 1. Strains isolated from infected root canals was shown to be most susceptible to Clindamycin, while Streptomycin exhibited least antibacterial properties. 2. Anaerobes were found to be susceptible to Penicillin, Ampicillin, and Clindamycin. 3. Streptomycin and Kanamycin were shown to be effective against Staphylococcus aureus and Staphylococcus epidermis, however, unidentified G(+) cocci organisms were found to be resistant to these agents. 4. Bifidobacterium sp. was susceptible to Lincomycin while G(+) rods were resistant to it. 5. Staphylococcus aureus, Micrococcus sp., and anaerobes were highly susceptible to Clindamycin. 6. All of the antibiotics tested were shown to be very effective against Eubacterium sp.
