Development of a Meta-Information System for Microbial Resources

  • Microbes are one of the most important bioresources in bioindustry and provide high economic values. Although there are currently about 6,000 bacterial species with validly published names, microbiologists generally assume that the number may account for less than 1% of the bacterial species present on Earth. To discover the remaining species, studies of metagenomes, metabolomes, and proteomes related to microbes have recently been carried out in various fields. We have constructed an information system that integrates various data on microbial resources and manages bioinformation to support efficient research of microorganisms. We have designated this system 'Bio-Meta Information System (Bio-MIS).' Bio-MIS consists of an integrated microbial resource database, a microbial resource input system, an integrated microbial resource search engine, a microbial resource online distribution system, a portal service, and management via the Internet. In the future, this system is expected to be connected with various public databases. We plan to implement useful bioinformatics software for analyzing microbial genome resources. The Web site is accessible at http://biomis.probionic.com.

  • There are currently about 6000 bacterial species with validly published names, but scientists assume that these may be less than 1% of bacterial species present on the earth. Microbial resource is one of the most important bioresources in bioinderstry and provides us with high economic values. To find missing ones, the studies of metagenome, metabolome, and proteome about microbes have started recently in developed countries. We construct the information system that integrates information on microbial genome resources and manages the information to support efficient research of microbial genome application, and name this system 'Bio-Meta Information System (Bio-MIS)'. Bio-MIS consists of integrated microbial genome resources database, microbial genome resources input system, integrated microbial genome resources search engine, microbial resources on-line distribution system, portal service and management via internet. In the future, we will include public database connection and implement useful bioinformatics software for analyzing microbial genome resources. Experimental study on solidification of uranium tailings by microbial grouting combined with electroosmosis

  • The present microbial reinforcement of rock and soil exhibits limitations, such as uneven reinforcement effectiveness and low calcium carbonate generation rate, resulting in limited solidification strength. This study introduces electroosmosis as a standard microbial grouting reinforcement technique and investigates its solidification effects on microbial-reinforced uranium tailings. The most effective electroosmosis effect on uranium tailings occurs under a potential gradient of 1.25 V/cm. The findings indicate that a weak electric field can effectively promote microbial growth and biological activity and accelerate bacterial metabolism. The largest calcium carbonate production occurred under the gradient of 0.5 V/cm, featuring a good crystal combination and the best cementation effect. Staged electroosmosis and electrode conversion efficiently drive the migration of anions and cations. Under electroosmosis, the cohesion of uranium tailings reinforced by microorganisms increased by 37.3% and 64.8% compared to those reinforced by common microorganisms and undisturbed uranium tailings, respectively. The internal friction angle is also improved, significantly enhancing the uniformity of reinforcement and a denser and stronger microscopic structure. This research demonstrates that MICP technology enhances the solidification effects and uniformity of uranium tailings, providing a novel approach to maintaining the safety and stability of uranium tailings dams.

Stable Microbial Community and Specific Beneficial Taxa Associated with Natural Healthy Banana Rhizosphere

  • Banana planting altered microbial communities and induced the enrichment of Fusarium oxysporum in rhizosphere compared with that of forest soil. Diseased plant rhizosphere soil (WR) harbored increased pathogen abundance and showed distinct microbial structures from healthy plant rhizosphere soil (HR). The enriched taxon of Bordetella and key taxon of Chaetomium together with some other taxa showed negative associations with pathogen in HR, indicating their importance in pathogen inhibition. Furthermore, a more stable microbiota was observed in HR than in WR. Taken together, the lower pathogen abundance, specific beneficial microbial taxa and stable microbiota contributed to disease suppression.

산란계에 Microbial Phytase 첨가시 산란율, 계란 품질, 인 이용율 및 영양소 대사율에 미치는 영향 (Effect of Dietary Microbial Phytase on Laying Performance, Egg Quality, Phosphorus Utilization and Nutrient Metabolizability in Laying Hens)

  • 본 시험은 microbial phytase을 첨가하여 산란계에 급여시 산란율, 계란 품질, 인 이용율 및 영양소 소화율에 미치는 영향을 조사하기 위하여 수행하였다. 32주령 Hy-Line Brown 갈색계 120수를 공시하여, 4주간 사양시험을 실시하였다. 시험 설계는 CON(control), LP(low phosphorus diet), NP(low phosphorus diet + 0.03% normal microbial phytase) 및 CP(low phosphorus diet + 0.03% coated microbial phytase)로 4개 처리를 하여 처리당 5반복, 반복당 6수씩 완전 임의 배치하였다. 전체 기간 동안 산란율은 CON과 CP 처리구가 LP 처리구보다 높게 나타났다(P<0.05). 난각 강도와 난각 두께는 LP 처리구에 비해 CON, CP 및 NP 처리구가 높게 나타났다(P<0.05). 난황색과 haugh unit에서는 CON, CP 및 NP 처리구가 LP 처리구보다 높게 나타났다(P<0.05). 혈액내 칼슘과 인 농도는 CP 처리구가 LP 처리구에 비해 높게 나타났다(P<0.05). 건물 소화율과 질소 소화율에서는 CP 처리구가 CON, LP 및 NP 처리구에 비하여 유의적으로 증가하였다(P<0.05). 회분소화율은 CON, CP 및 NP 처리구가 LP 처리구에 비해 증가하였다(P<0.05). 칼슘 소화율과 인 소화율에서는 CP와 NP 처리구가 CON과 LP 처리구에 비해 높게 나타났다(P<0.05). 칼슘 흡수량은 CON, CP 및 NP 처리구가 LP 처리구보다 유의적으로 높게 나타났다(P<0.05). 인 흡수량은 CP와 NP 처리구가 CON과 LP 처리구와 비교하여 유의적으로 높게 나타내었다(P<0.05). 질소와 칼슘 배출량은 CON, NP 및 CP 처리구가 LP 처리구에 비해 유의적으로 낮게 나타냈다(P<0.05). 인 배출량은 LP, NP 및 CP 처리구가 CON 처리구에 비해 유의적으로 낮게 나타내었다(P<0.05). 결론적으로, 산란계에 있어 코팅된 microbial phytase의 첨가는 산란율, 계란 품질, 혈액 내 칼슘과 무기태 인, 영양소 소화율과 칼슘 및 인 흡수량을 증진시키며, 질소와 칼슘 배출량을 감소시키는 것으로 나타났다.

Effect of dietary Achyranthes japonica extract on growth performance of growing pigs and absorption rate of quercetin in blood

  • This study was done to investigate the effects of the incorporation of Achyranthes japonica extracts (AJE) in diet on the production parameters of growing pigs. Exp 1: Total, 105 crossbred pigs (average body weight: 24.47 ± 2.46 kg) were used in a 6-week feeding trial. Pigs (seven replicates, five pigs per pen) were allotted randomly to three treatments. Dietary treatments: CON (basal diet); basal diet with 0.025% AJE, and basal diet + 0.050% AJE). Growth performance, nutrient digestibility, fecal microbial count, and fecal noxious gas were assessed in this study. Average daily gain (ADG), average daily feed intake (ADFI), and gain to feed ratio (G:F) were not affected by the addition of up to 0.05% AJE. In the case of apparent total tract digestibility (ATTD), dry matter (DM), nitrogen (N), and digestible energy (DE) were not changed in 3rd and 6th weeks of the feeding trial through the addition of AJE up to 0.05% in the growing pig diet. In microbial count, Lactobacillus and Escherichia coli count at 3rd and 6th week was similar in all the treatment diets. The inclusion of AJE at levels up to 0.05% in growing pig diet had no effect on the production of NH3, H2S, acetic acid, and CO2 in the feces. After ending the Exp 1, a total of nine pigs were divided into three treatment groups. Treatment diets were included, TRT1, basal diet + powder quercetin 30 g; TRT2, basal diet + powder quercetin 150 g; TRT3, basal diet + powder quercetin 300g. Rate of absorption in blood was increased with the higher dose of quercetin. The results suggested incorporation of AJE up to 0.05% has no significant effect on ADG, ADFI, and G:F, as well as DM, N, and DE digestibility, fecal microbial count, and fecal noxious gas emission in growing pigs, even though no negative effect was found.

Optimization Studies for the Production of Microbial Transglutaminase from a Newly Isolated Strain of Streptomyces sp.

  • Covalent cross-links between a number of proteins and peptides explain why transglutaminase may be widely used by food processing industries. The objective of this work was optimization of the fermentation process to produce transglutaminase from a new microbial source, the Streptomyces sp. P20. The strategy adopted to modify the usual literature media was: (1) fractional factorial design (FFD) to elucidate the key medium ingredients, (2) central composite design (CCD) to optimise the concentration of the key components. Optimization of the medium resulted in not only an 86% increase in microbial transglutaminase activity as compared to the media cited in the literature, but also a reduction in the production cost. Optimal fermentation conditions - namely temperature and agitation rate - were also studied, using CCD methodology. Usual conditions of $30^{\circ}C$ and 100 rpm were within the optimal area. All other parameters for enzyme production were experimentally proven to be optimum fermentation conditions.

살균${\cdot}$숙성된 남은 음식물의 공정별 미생물 분포 및 Pepsin과 In vitro 소화율 평가 (Changes of the Microbial Population and Determination of Pepsin and In vitro Digestibilities of Pasteurized and Cured Food Wastes)

  • 본 연구는 남은 음식물에 수분흡수제 또는 영양 보충제로 유기성 부산물인 제과부산물, 밀기울, 맥강, 육계분 등을 혼합한 후 rotary drum 방식의 반건식 살균처리 및 호기적 숙성 과정 별 혼합사료 내의 미생물 분포 및 사료단백질 pepsin 소화율과 in vitro 영양소 소화율을 측정하여 실제 동물 급여 시의 효능을 사전 예측하고자 실시하였다. 살균 처리 공정($80^{\circ}C$에서 30분간)은 전반적으로 균수를 감소시키는 경향과 곰팡이를 사멸시키는 효과가 있었으며(P<0.05), 연이은 숙성 과정은 살균과정에서 감소된 lactic acid bacteria를 증식시키는 효과가 있었다(P<0.05). 열처리 살균 공정은 혼합사료 단백질의 pepsin 소화율을 감소시켰다(P<0.05). 사료원료 건물 및 유기물의 in vitro 소화율은 제과부산물>밀기울>남은 음식물=맥강 순으로 높았다. 상기한 연구 결과는 반건식 남은 음식물 혼합사료의 동물 급여시 중요한 기초 자료로 활용될 것이다.

Butyric Acid Fermentation of Sodium Hydroxide Pretreated Rice Straw with Undefined Mixed Culture

  • This study describes an alternative mixed culture fermentation technology to anaerobically convert lignocellulosic biomass into butyric acid, a valuable product with wide application, without supplementary cellulolytic enzymes. Rice straw was soaked in 1% NaOH solution to increase digestibility. Among the tested pretreatment conditions, soaking rice straw at $50^{\circ}C$ for 72 h removed ~66% of the lignin, but retained ~84% of the cellulose and ~71% of the hemicellulose. By using an undefined cellulose-degrading butyrate-producing microbial community as butyric acid producer in batch fermentation, about 6 g/l of butyric acid was produced from the pretreated rice straw, which accounted for ~76% of the total volatile fatty acids. In the repeated-batch operation, the butyric acid production declined batch by batch, which was most possibly caused by the shift of microbial community structure monitored by denaturing gradient gel electrophoresis. In this study, batch operation was observed to be more suitable for butyric acid production.
