갯벌 토양 내에 존재하는 방선균, 세균, 곰팡이의 다양성 및 개체군 분석을 위해 토양 깊이(지표, 지하 10 cm, 지하 20 cm, 지하 30 cm)와 염농도 (균분리 배지상에 바닷물, 1.5% NaCl, 5% NaCl을 첨가함)에 따른 생태학적 특성을 분석하였다. 방선균으로서는 총 175 균주를 분리할 수 있었고 이를 대상으로 RAPD-PCR 방법을 이용하여 군집분석을 하였다. 방선균의 경우 깊이 10 cm에서 가장 많은 개체수를 보였으며 NaCl이나 바닷물이 포함되지 않은 HVagar 배지에서 가장 많은 수의 균주들이 나타났고, 토양깊이에 따른 다양성의 경우 지표에서 29균주, 지하 10 cm에서 74 균주, 지하 20 cm에서 39 균주, 지하 30 cm에서 37 균주가 분리됨으로써 지하 10 cm 이내에서 다양한 방선균이 존재함을 알 수 있었다. 방선균 군집분석을 한 결과 15개의 cluster로 나눌 수 있었으며, Promicromonospora citrea, Microtetraspora angiospora, Kineosporia aurantiaca, Sebekia benithana, Kibdelosporangium aridum과 동일한 cluster에 속하는 균주들이 많았고, 그 외에 다양한 균주들이 존재함을 알 수 있었다. 곰팡이의 경우 지표면의 토양을 바닷물이 함유된 배지에서 배양했을 때 가장 많은 수가 나타났고, 세균의 경우도 지표면의 토양을 염이 첨가되지 않은 nutrient agar 배지로 배양하였을 때 가장 많은 세균을 분리할 수 있었다. 이러한 결과는 갯벌토양에서는 표층 및 지하 10 cm 이내에서 가장 다양하고 많은 수의 미생물이 존재하고 있음을 나타낸다고 하겠다.
본 논문은 제주연안에서 채집한 소라(Turbo cornutus)의 장내세균을 분리하고 군집의 다양성을 조사하였다. 표준배지를 사용하여 순수 정체 배양 결과 MA agar에서 가장 많은 군집을 나타났다. 일반 배양 CFU값은 평균 1.8×105, 혐기 배양 CFU값은 평균 0.4×10으로 보다 적게 나타났다. 기존 표준균주와 16S rDNA sequence 유사도 비교 분석 결과 크게 4문 12과 26속 67종으로 분류되었다. 표준균주와의 상동성 범위는 93-100%로 나타났다. 소라장내세균 군집은 크게 Proteobacteria 39% (α-proteobacteria; Phyllobacteriaceae (1), Rhodobacteraceae (8) / γ-proteobacteria; Alteromonadaecae (1), Shewanellaceae (4), Vibrionaceae(12))로 가장 우점하였고, Firmicutes 34% (Bacillaceae (21), Paenibacillaceae (2)), Actinobacteria 21% (Cellulomonadaceae (1), Mycobacteriaceae (6), Nocardiaceae (4), Streptomycetacea (3)), Bacteroidetes 6% (Flavobacteriaceae (4))로 각각 나타났다. Bacillus sp., Vibrio sp.이 가장 우점 하였으며, 그 외 대부분의 분리 균주는 해양 유래 관련 균주로 나타났다. 이전 보고된 제주 연안에 서식하는 해양동물 장내세균군과 비슷한 양상을 보였다. 분리된 일부 균주가 단당류(Cellulose), 다당류(alginate, agar)분해능을 갖고 있는 것으로 나타났는데, 대부분이 해조류 유래 세균으로 소라의 섭이가 장내세균군과 관련됨을 알 수 있었다. 동시에 probiotics 기능을 갖고 있는 일부 균주도 분리되었다.
인삼부산물 및 인삼밭 토양에서 분리한 9균주 중 𝛽-Glucosidase 생성 균주 GYP-1과 GYP-3-3 균주를 선발하였다. 선발된 𝛽-Glucosidase 생성 균주에 대하여 16S rRNA 유전자 염기서열과 ITS 염기서열을 기반으로 계통 분석을 실시한 결과, GYP-1 균주는 Rhodotorula 속에 속하며, GYP-3-3은 Brachybacterium 속에 속하는 것으로 확인되었다. 특히, Rhodotorula sp. GYP-1 균주는 호기성 효모종으로 biomass 생산량이 높아 최종 우수 균주로 선발하였다. Rhodotorula sp. GYP-1가 생성하는 𝛽-Glucosidase의 온도 및 pH에 따른 효소 활성 및 안정성을 검정한 결과, 30 ℃에서 6.7 unit/ml로 가장 높은 활성을 나타내었고, 20 ℃ ~ 40 ℃에서 효소 활성의 약 70 % 이상을 유지하는 것으로 확인하였다. pH에 따른 효소의 활성 및 안정의 경우, pH 5에서 6.8 unit/ml으로 가장 높은 활성을 나타내었고 pH 5~pH 8까지 93.3 % 이상의 효소 활성을 유지하는 것으로 확인하였다. Rhodotorula sp. GYP-1가 생성하는 𝛽-Glucosidase는 ginsenoside Rb1 minor 진세노사이드로 분해하는 것으로 확인되었다. 또한, 인삼 뿌리 병원균(Botrytis cinerea)에 대해 항진균능을 갖는 것으로 확인되었다.
본 연구는 부패미생물의 종류와 차이를 알아보기 위해 유기농산물과 일반농산물(깻잎, 양배추, 상추)을 구입하여 $4^{\circ}C$에 14일 저장한 후 일반목적배지를 사용하여 미생물을 분리 및 16S rRNA sequencing방법을 이용하여 동정하였다. 유기농산물 깻잎, 양배추, 상추의 총균수는 $5-7{\log}_{10}CFU/g$ 수준이었고, 일반농산물의 총균수는 $5-8{\log}_{10}CFU/g$ 수준이었다. 유기농산물보다는 일반농산물이 총균수를 더 많이 내재하고 있으나 유의적인 차이는 나타나지 않았다(p > 0.05). 유기농산물 깻잎에서 발견된 주요 균은 Microbacterium sp.이었으며, 일반농산물 깻잎에서 발견된 주요균은 Stenotrophomonas maltophilia이었다. 또한, 유기농산물에서 부패를 일으킬 수 있는 균인 Klebsiella pneumonia가 발견되었으며, 일반농산물보다 유기농산물에서 발견된 균의 종류가 더 많은 것을 확인할 수 있었다. 이에 본 연구 결과는 부패미생물에 대한 이해를 높이고 농산물의 신선도 및 유통기한을 늘리기 위한 새로운 살균소독제 및 부패 억제제를 개발하는데 좋은 기초자료로 사용될 수 있을 것으로 사료된다.
식물 근권 토양으로부터 색소생성 균주 106균주를 수집하여 색소 생성능이 우수한 균주로부터 노란색(33개), 주황색(12개), 분홍색, 빨간색, 갈색 그리고 보라색 총 49개의 세균색소를 추출하였다. 식물병원균에 대한 항균활성능이 우수한 색소를 선발하기 위하여 고추점무늬병원균(Xanthomonas axonopodis), 흑마병원균(Xanthomonas campestris)과 딸기잿빛곰팡이병원균(Botrytis cinerea), 고추탄저병원균(Colletotrichum acutatum), 그리고 시들음병원균(Fusarium oxysporum)을 대상으로 항균활성 검정을 수행하였다. 색소생성 Chryseobacterium sp. RBR9 균주가 생산하는 노란색 색소와 Methylobacterium sp. RI13 균주가 생산하는 빨간색 색소는 X. axonopodis와 X. campestris에 항세균 활성을 나타내었다. 차나무 토양으로부터 분리된 Collimonas sp. DEC-B5가 생산하는 보라색 색소는 항세균 활성과 더불어 B. cinerea와 Colletotrichum acutatum에 항진균활성을 나타내었다. 특히, 보라색 색소는 최소저해 농도 $20{\mu}M$에서 B. cinerea를 65% 이상 생육 저해하였다. 항진균활성 보라색 색소를 HPLC 분석한 결과, violacein (91.6%)와 deoxyviolacein(8.4%)으로 동정되었다. 보라색 색소 violacein의 생산량은 SCB 배지에서 $43.2{\mu}M$이었고 D-mannitol 1.5%, yeast extract 0.2%를 첨가한 경우 $431.6{\mu}M$로 약 10배 높은 색소 생성량을 나타내었다. 본 연구에서 분리된 Collimonas sp. DEC-B5가 생산하는 violacein 색소는 딸기잿빛곰팡이병원균 방제제로 활용 가능성이 확인되었다.
Background: The study was conducted to investigate the distributions of faecal bacteria in commercial oriental medicine herb products. Methods and Results: A survey was conducted on the microbial contamination levels and antimicrobial specificity of Bacillus cereus and other microbes using 106 oriental medicine herb products on sale in Seoul. Pouring and isolation methods such as standard plate counts were used to identify the bacteria. The isolated bacterias included coliforms, Bacillus spp., Enterococcus spp., Staphylococcus spp., Listeria spp.were identified by using gram staining and an API (analytical profile index) kit. Antimicrobial drugs discs were determined by CLSI (clinical and laboratory standards institute). Conclusions: The bacterial isolates present in the herbal medicines included 98 coliforms, 45 Bacillus spp., 29 Enterococcus spp., and 2 Listeria spp. Among these, there were nine Bacillus cereus strains, one Enterococcus faecium strain, and one Enterococcus faecalis strain present. The 9 Bacillus cereus strains were tested for susceptibility to 36 types of antibiotics products by the disc diffusion method. The strains showed resistance to 13 of these antibiotic products and semi-resistance to 5 antibiotic products. On the basis of these results, any oriental medicine herb product can be assumed to be contain resistant or semi-resistant bacterial strains. Therefore, we suggest prescribing guidelines and special management for the use of antibiotics in farms producing oriental medicine herb products.

Skin sooty dapple disease, a fungal disease that lowers Asian pear fruit quality, has emerged recently in Korea but has not yet been thoroughly characterized. This disease affects the surface of fruit, leaves, and young shoots of the Asian pear, typically appearing as a dark or pale black dapple on the fruit surface. The disease initiates on the fruit with small circular lesions that become bigger, eventually spreading to form large circular or indefinite lesions. Sparse dark or flourishing white-greyish aerial mycelia and appearance of a dark or pale black dapple on the fruit surface are typical signs of this disease. The disease was severe during cold storage of the Niitaka and Chuhwangbae varieties, but more limited on the Gamcheonbae and Hwangkeumbae varieties. To identify causal pathogens, 123 fungal isolates were obtained from lesions. The fungi that caused typical skin sooty dapple disease symptoms in our bioassay were identified. Based on their morphological characteristics, 74% of the isolates were Cladosporium sp. and 5-7 % of the isolates were Leptosphaerulina sp., Tripospermum sp., or Tilletiopsis sp. None of the isolates caused severe soft rot by injection to a wound plug, but some of the Cladosporium sp. isolates caused mild maceration. Therefore this microbiol complex cannot account for the soft rot also observed in stored fruits. The high frequency of isolation of Cladosporium sp. from disease tissues and bioassay on pear fruit surface suggest that Cladosporium sp. could be a major pathogen in the microbial complex associated with skin sooty dapple disease of the Asian pear in Korea.
섬유소계 바이오매스로부터 바이오 연료 등과 같은 유용한 물질을 생산하기 위해서는 바이오매스로부터 유래하는 혼합당을 효과적으로 대사할 수 있는 균주의 개발이 필수적이다. 본 연구에서는 xylose를 대사가 가능한 효모인 P. stipitis를 적응진화하여 cellobiose 대사효율이 향상되고 cellobiose와 xylose를 동시에 대사할 수 있는 균주를 개발하고자 하였다. 총 10회의 계대배양을 통해 얻어진 진화된 P. stipitis 돌연변이 균주는 모균주에 비해 6배 이상 증가된 cellobiose 대사속도를 나타내었으며 ethanol 생산수율을 0에서 0.4 (g ethanol/g cellobiose)로 향상시켰다. 아울러 본 실험에서 개발한 돌연변이 균주는 cellobiose와 xylose 혼합당 조건에서 모균주에 비해 2배 가까이 향상된 ethanol 생산 및 생산속도를 나타내었다.

The diesel-degrading activities of biofilms sampled from petroleum-contaminated groundwaters in urban subway drainage systems were examined in liquid cultures, and the microbial populations of the biofilms were characterized by denaturing gel gradient electrophoresis (DGGE) and 16S rDNA sequence analysis. Biofilm samples derived from two sites (19 K and 20 K) at subway Station N and Station I could degrade around $80\%$ of applied diesel within 20 and 40 days, respectively, at $15^{\circ}C$, and these results were strongly correlated with the growth patterns of the biofilms. The closest phylogenetic neighbor of a dominant component in the 19 K biofilm was Thiothrix fructosivorans strain Q ($100\%$ similarity). Four dominant strains in the 20 K biofilm were closely related to Thiothrix fructosivorans strain Q ($100\%$ similarity), Thiothrix sp. CC-5 ($100\%$ similarity), Sphaerotilus sp. IF14 ($99\%$ similarity), and Cytophaga-Flexibacter-Bacterioides (CFB) group bacterium RW262 ($98\%$ similarity). Three dominant members in the Station I biofilms were very similar to uncultured Cytophagales clone CRE-PA82 ($91\%$ similarity), Pseudomonas sp. WDL5 ($97\%$ similarity), and uncultured CFB group bacterium LCK-64 ($94\%$ similarity). The microbial components of the biofilms differed depending on the sampling site. This is the first report on the isolation of clones highly similar to Thiothrix fructosivorans and Thiothrix sp. from biofilms in petroleum-polluted groundwaters, and the first evidence that these organisms may play major roles in petroleum degradation and/or biofilm-development.
Crack remediation on the surface of cement mortar using microbiological calcium carbonate ($CaCO_3$) precipitation (MICP) has been investigated as a microbial sealing agent on construction materials. However, MICP research has never acknowledged the antifungal properties of calcite-forming bacteria (CFB). Since fungal colonization on concrete surfaces can trigger biodeterioration processes, fungi on concrete buildings have to be prevented. Therefore, to develop a microbial sealing agent that has antifungal properties to remediate cement cracks without deteriorative fungal colonization, we introduced an antifungal CFB isolated from oceanic islands (Dokdo islands, territory of South Korea, located at the edge of the East Sea in Korea.). The isolation of CFB was done using B4 or urea-$CaCl_2$ media. Furthermore, antifungal assays were done using the pairing culture and disk diffusion methods. Five isolated CFB showed $CaCO_3$ precipitation and antifungal activities against deteriorative fungal strains. Subsequently, five candidate bacteria were identified using 16S rDNA sequence analysis. Crack remediation, fungi growth inhibition, and water permeability reduction of antifungal CFB-treated cement surfaces were tested. All antifungal CFB showed crack remediation abilities, but only three strains (KNUC2100, 2103, and 2106) reduced the water permeability. Furthermore, these three strains showed fungi growth inhibition. This paper is the first application research of CFB that have antifungal activity, for an eco-friendly improvement of construction materials.
