돈 분뇨 액비에 미생물 첨가가 배추의 발아지수에 미치는 영향 (Effect of Microbial Inoculant to Pig Liquid Fertilizer on Germination Index of Chinese Cabbage)

  • 본 연구는 돈 분뇨 액비에 미생물 첨가가 배추의 발아지수에 미치는 효과를 구명하기 위해서 수행하였다. 돈 분뇨 액비는 107일 동안 폭기와 무폭기 상태로 액상비료를 제조하여 공시하였고 미생물 첨가는 $0.05\%,\;0.1\%,\;0.2\%,\;0.3\%$ 수준으로 하였다. $0.05\%$ 미생물 접종 처리구에서 식물 독성이 낮고 발아지수가 높게 나타났다. 첨가수준별 돈분 액비 이용시험에서 초기 돈 슬러리의 액비성상은 T-N 2,873 ppm, T-P 753 ppm, $NH_4-N$ 1,441.6 ppm, $NO_3-N$ 16.48ppm으로 50일이 되면서 폭기조건에서 T-N 3,672 ppm, T-P 164 ppm, $NH_4-N$ 183.87 ppm, $NO_3-N$ 21.97 ppm로 변화하였으며, 무폭기 조건에서 T-N 1,261 ppm, T-P 68 ppm, $NH_4-N$ 161 ppm, $NO_3-N$ 16.87 ppm로 나타났다. 미생물을 처리하지 않고 107일동안 폭기조건과 무폭기 조건으로 액비화한 시료의 발아지수는 각각 83과 40.4이였다. 돈 분뇨 액비를 무산소 제조시 $40\%$ 이상, 호기제조시 $50\%$ 이상의 발아지수가 개선되었다. 이와 같은 결과는 액비제조 처리가 무산소 제조보다는 호기제조 처리가 작물에 양호한 것으로 판단되어지며 이는 호기처리시 총질소의 함량을 높게 유지할 수 있도록 하는 결과인 것으로 사료된다.

수확시 숙기 및 젖산균 제제가 호밀 라운드베일 사일리지의 품질에 미치는 영향 (Effect of Maturity at Harvest and Inoculants on the Quality of Round Baled Rye Silage)

  • 본 시험은 1998년 축산기술연구소 초지사료과 시험포장에서 수확시 숙기 및 젖산균 제제가 호밀 라운드 베일 사일리지의 품징에 미치는 영향을 구명하기 위하여 수행되었다. 시험설계는 분할구 배치법으로 주구로는 수잉기, 출수기 및 개화기에 수확하는 수확시 숙기를 두고 세구로는 무처리(control), Inoculant A 및 Inoculant B를 두고 3 반복으로 수행하였으며 라운드베일 사일리지 제조시 호밀은 수잉기에는 1일 그리고 출수기 및 개화기에는 0.5일간 예건 하였다. 저장 2개월 후 호밀 라운드베일 사일리지의 섬유소 함량(ADF 및 NDF)은 수확시 숙기가 지연됨에 따라 증가하였고(p<0.01), 첨가제 처리에 따른 차이는 없었다. 한편 IVDMD에 있어서도 수확이 지연됨에 따라 감소되었다. pH의 변화는 개화기에서 평균 4.35로 가장 낮았고 출수기에서 평균 4.91로 가장 높았다. 또한 젖산균 처리구는 대조구에 비해 유의적으로 낮게 나타났다. 무처리구의 건물 함량이 젖산균 처리구보다 높게 나타났다. 암모니아태 질소의 함량은 전체적으로 10% 미만으로 고품질 사일리지의 적정 수준에 도달하였으며, 젖산균 제제 이용은 암모니아태 질소 함량을 줄였다. 젖산 함량은 첨가제 처리로 증가한 반면 초산 및 낙산 함량은 감소하였다. 젖산 생성균의 수는 젖산균 처리구에서 대조구에 비해 2~3배 높게 나타났으며 개화기의 Inoculant B구에서 가장 높았다. 이상의 결과를 종합할 때 고품질의 호밀 라운드베일 사일리지 제조시에는 출수기 이후에 수확하여 젖산균 제제를 처리하는 것이 바람직한 것으로 나타났다.

Effects of Microbial Additives on Chemical Composition and Fermentation Characteristics of Barley Silage

  • This study examined the effects of bacterial inoculants on chemical composition and fermentation indices of barley silage. Barley forage (Youngyang) was harvested at 24% dry matter (DM) and wilted to 47.9% DM. The wilted barley forage was chopped to 3-5 cm length and applied with no inoculant (CON), L. plantarum ($1{\times}10^{10}cfu/g$, LP) or Effective Microorganisms ($0.5{\times}10^9cfu/g$, EM). Then the forages were ensiled in four replications for each treatment in 20 L mini silos and stored for 100 days. The contents of crude protein and ether extract were higher in CON silage ensiled for 100-d, while the contents of DM and crude ash were higher in EM silage (p<0.05). The contents of ADF, NDF and hemicellulose as well as the in vitro DM digestibility were not affected by microbial inoculation (p>0.05). The pH, ammonia-N concentration and lactate to acetate ratio were higher (p<0.05) in CON silage, while lactate concentrations were higher (p<0.05) in CON and LP silage. Acetate concentration and lactic acid bacteria was increased (p<0.05) by both inoculants (LP and EM), but propionate concentration and yeast was increased (p<0.05) by EM and LP, respectively. These results indicated that the fermentation quality of barley silage was improved by the application of bacterial inoculants.

Streptomyces sp. AC-3을 이용한 배추 무사마귀병의 생물학적 방제 (Biocontrol of Cabbage Clubroot by the Organic Fertilizer Using Streptomyces sp. AC-3.)

  • 본 연구는 배추 무사마귀병의 생물학적 방제를 위해 수행되었다. 배추 무사마귀병균 Plasmodiophora sp외 길항미생물은 낙엽부식토양에서 분리한 350여 종의 토양미생물 중에서 가장 길항력이 높은 방선균 AC-3균주를 이용하였고, AC-3 균주는 16S rDNA 염기서열분석방법으로 Streptomyces sp.로 동정되었다. Streptomyces sp. AC-3은 배양액 $m\ell$당 9.3 units의 chitinase를 생산하였다. 그 결과 배추 무사마귀병균 Plasmodiophora sp.의 배양액에 Streptomyces sp. AC-3을 접종하고 배양하면 Plasmodiophora sp.의 균체가 팽윤되거나 세포벽이 용해된 모습이 관찰되었다. Streptomyces sp. AC-3은 전딘도 배추 무사마귀병이 만연했던 포장에서 재배 시험하기 위해 제형화시키고 유기질비료에 첨가하여 이용하였다. 미생물제제를 첨가하지 많고 제조한 유기질비료(H-1)(대조구)와 미생물제제 첨가하여 제조한 유기질비료(H-2)(처리구)에 의한 포장시험에서 배추의 생육은 재배 40일 까지는 거의 차이를 나타나지 않았으나 재배 60일에는 다소 차이를 나타내었다. 배추의 병 발생율을 조사한 결과 무사마귀병은 H-1 비료 처리구에서는 36.17% 병이 발생되었으나 H-2 비료 처리구에서는 18.28% 발병되어 약 50%의 방제효과를 나타내었으며, 균핵병, 잎마름병, 그루썩음병 등에서도 길항효과를 나타내어 생물학적 방제가 가능한 균주로 확인되었다.

Influence of microbial additive on microbial populations, ensiling characteristics, and spoilage loss of delayed sealing silage of Napier grass

  • Objective: To measure whether a microbial additive could effectively improve the fermentation quality of delayed-sealing (DS) silage, we studied the effects of inoculants of lactic acid bacteria (LAB) and cellulase enzyme on microbial populations, ensiling characteristics, and spoilage loss of DS silage of Napier grass in Africa. Methods: Quick-sealing (QS) and DS silages were prepared with and without LAB (Lactobacillus plantarum) inoculant, cellulase enzymes, and their combination. The QS material was directly chopped and packed into a bunker silo. The DS material was packed into the silo with a delay of 24 h from harvest. Results: In the QS silage, LAB was dominant in the microbial population and produced large amounts of lactic acid. When the silage was treated with LAB and cellulase, the fermentation quality was improved. In the DS silage, aerobic bacteria and yeasts were the dominant microbes and all the silages were of poor quality. The yeast and mold counts in the DS silage were high, and they increased rapidly during aerobic exposure. As a result, the DS silages spoiled faster than the QS silages upon aerobic exposure. Conclusion: DS results in poor silage fermentation and aerobic deterioration. The microbial additive improved QS silage fermentation but was not effective for DS silage.

Development of a new lactic acid bacterial inoculant for fresh rice straw silage

  • Objective: Effects of newly isolated Lactobacillus plantarum on the fermentation and chemical composition of fresh rice straw silage was evaluated in this study. Methods: Lactic acid bacteria (LAB) from good crop silage were screened by growing them in MRS broth and a minimal medium with low carbohydrate content. Selected LAB (LAB 1821) were Gram-positive, rods, catalase negative, and were identified to be Lactobacillus plantarum based on their biochemical characteristics and a 16S rRNA analysis. Fresh rice straw was ensiled with two isolated LAB (1821 and 1841), two commercial inoculants (HM/F and P1132) and no additive as a control. Results: After 2 months of storage at ambient temperature, rice straw silages treated with additives were well-preserved, the pH values and butyric and acetic acid contents were lower, and the lactic acid content and lactic/acetic acid ratio were higher than those in the control (p<0.05). Acidity (pH) was lowest, and lactic acid highest, in 1821-treated silage (p<0.05). The $NH_3-N$ content decreased significantly in inoculant-treated silage (p<0.05) and the $NH_3-N$ content in 1821-treated silage was lowest among the treatments. The dry matter (DM) content of the control silage was lower than that of fresh rice straw (p<0.05), while that of the 1841- and p1174-inoculant-treated silages was significantly higher than that of HM/F-treated silage. Microbial additives did not have any significant (p>0.05) effect on acid detergent fiber or neutral detergent fiber contents. Crude protein (CP) content and in vitro DM digestibility (IVDMD) increased after inoculation of LAB 1821 (p<0.05). Conclusion: LAB 1821 increased the CP, IVDMD, lactic acid content and ratio of lactic acid to acetic acid in rice straw silage and decreased the pH, acetic acid, $NH_3-N$, and butyric acid contents. Therefore, adding LAB 1821 improved the fermentation quality and feed value of rice straw silage.

Changes in fermentation pattern and quality of Italian ryegrass (Lolium multiflorum Lam.) silage by wilting and inoculant treatments

  • Objective: This study was conducted to investigate the effects of wilting and microbial inoculant treatment on the fermentation pattern and quality of Italian ryegrass silage. Methods: Italian ryegrass was harvested at heading stage and ensiled into vinyl bags (20 cm×30 cm) for 60d. Italian ryegrass was ensiled with 4 treatments (NWNA, no-wilting noadditive; NWA, no-wilting with additive; WNA, wilting no-additive; WA, wilting with additive) in 3 replications, wilting time was 5 hours and additives were treated with 106 cfu/g of Lactobacillus plantarum. The silages samples were collected at 1, 2, 3, 5, 10, 20, 30, 45, and 60 days after ensiling and analyzed for the ensiling quality and characteristics of fermentation patterns. Results: Wilting treatment resulted in lower crude protein and in vitro dry matter digestibility and there were no significant differences in acid detergent fiber (ADF), total digestible nutrient (TDN), water-soluble carbohydrate (WSC), ammonia content, and pH (p>0.05). However, wilting treatment resulted in higher ADF and neutral detergent fiber content of Italian ryegrass silage (p<0.05), and the WNA treatment showed the lowest TDN and in vitro dry matter digestibility. The pH of the silage was higher in the wilting group (WNA and WA) and lower in the additive treatment group. Meanwhile, the decrease in pH occurred sharply between the 3-5th day of storage. The ammonia nitrogen content was significantly lower in the additive treatment (p<0.05), and wilting had no effect. As fermentation progressed, the lactic and acetic acid contents were increased and showed the highest content at 30 days of storage. Conclusion: The wilting treatment did not significantly improve the silage fermentation, but the inoculant treatment improved the fermentation patterns and quality of the silage. So, inoculation before ensiling is recommended when preparing high quality of Italian ryegrass silage, and when wilting, it is recommended to combine inoculation for making high quality silage.

  • 전국(全國) 223개(個) 지역(地域)에서 수집(蒐集)한 두과작물(豆科作物)의 근류(根瘤)에서 총(總) 590주(株)의 근류균(根瘤菌)을 분리(分離)하여 숙주특이성(宿主特異性)에 따라, R. japonicum 218종(種), R. phaseoli 101종(種), R. trifolii 97종(種), R. meliloti 4종(種), R. leguminosarum 1종(種), Rhizobium species 101종(種), 미분류(未分類)된 균주(菌株) 159종(種)으로 분류(分類)하였다. 분리(分離)된 R. japonicum 218종(種)의 근류형성능(根瘤形成能)과 질소고정능(窒素固定能)을 비교(比較)하여. R. japonicum R-138, R-168, R-214 균주(菌株)를 우수균주(優秀菌株)로 하였다. 분리(分離) 선발(選拔)된 R. japonicum 중(中) 질소고정능(窒素固定能)이 우수(優秀)한 균주(菌株)의 plasmid를 확인(確認)한 결과(結果), fast-growing group 균주(菌株)들은 $1{\sim}4$개(個)의 거대(巨大) plasmid를 함유(含有)하고 있었으며 그 분자량(分子量)은 약(約) $35{\sim}300{\;}Md$ 정도(程度)이었다. 반면 대부분(大部分)의 slow-growing group 균주(菌株)들은 plasmid가 검출(檢出)되지 않았다.

Effects of sodium diacetate and microbial inoculants on fermentation of forage rye

  • Rye (Secale cereale L.) is a valuable annual forage crop in Korea but there is limited information about the impact of chemical and biological additives on fermentation characteristics of the crop. This experiment was conducted to investigate fermentation dynamics of wilted forage rye treated with the following six additives; control (no additive), sodium diacetate applied at 3 g/kg wilted forage weight (SDA3), 6 g/kg wilted forage weight (SDA6), inoculations (106 CFU/g wilted forage) of Lactobacillus plantarum (LP), L. buchneri (LB), or LP+LB. The ensiled rye sampled at 1, 2, 3, 5, 10, 20, 30, and 45 days indicated that the acidification occurred fast within five days of storage than the rest of the storage period. The microbial inoculants decline the pH of ensiled forage, more rapidly than the control or SDA treated, which accompanied by the decrease of water-soluble carbohydrates and increase of lactic acid. Compared with the control silage, all treatments suppressed ammonia-nitrogen formation below to 35 g/kg DM throughout the sampling period. Suppression of total microbial counting occurred in SDA6, LP, and LP + LB. The lactic acid production rates were generally higher in microbial inoculation treatments. Acetic acid concentration was lowest in the LP-treated silage and highest in the SDA- and LB-treated silages. The in vitro dry matter (DM) digestibility and total digestible nutrients were the highest in the silage treated with SDA (6 g/kg) at day 45 of ensiling. Based on lower ammonia-nitrogen concentrations and higher feed value, ensiling forage rye treated with SDA at 6 g/kg is promising through enhanced silage quality.

Effect of microbial inoculants on fermentation quality and aerobic stability of sweet potato vine silage

  • Objective: This study was conducted to evaluate the effect of homo or hetero fermentative inoculants on fermentation quality and aerobic stability of sweet potato vine (SPV) silage containing Italian ryegrass hay as moisture absorbent. Methods: The SPV was harvested at 15% dry matter, mixed with Italian ryegrass hay at 1:1 ratio on a fresh weight basis, and chopped to 3 to 5 cm length. After then, the chopped forage mixture was ensiled into 20-L mini silos in quadruplicate for 7, 48, and 100 days after application of microbial inoculants at $1.2{\times}10^5$ colony forming units (cfu)/g of forage following: no inoculant (CON), Lactobacillus plantarum as a homo fermentative (LP), Lactobacillus buchneri as a hetero fermentative (LB), and mixture of LP and LB at 1:1 ratio as a combo fermentative (MIX). Results: The LP and MIX silages had lowest pH (p<0.001) on 7 and 48 days, while MIX and CON silages had greatest lactate concentrations (p<0.05) on 7 and 48 days, respectively. Acetate concentrations were highest (p<0.01) in LB and MIX silages on 7 days, and in LB silage on 48 days, while lactate to acetate ratios were lowest (p<0.001) in LB silages. The chemical compositions and nutrient digestibility of silage ensiled for 100 days was not affected by inoculants. On 100 days of ensiling, LB silage had lowest (p<0.01) lactate concentration and lactate to acetate ratio, but highest acetate concentration. Aerobic stability was highest (p<0.001) in LB silage followed in MIX silage. On contrast, LB silage had lowest (p<0.05) lactic acid bacteria and mold. Conclusion: The results indicated that application of LB solely had a better effect on aerobic stability than not only LP, but also MIX. However, LP application did not show beneficial effects from the viewpoints of fermentation quality and aerobic stability compared to CON.
