Studies on Constituents and Culture of the Higher Fungi of Korea (한국산(韓國産) 고등균류(高等菌類)의 성분(成分)및 배양(培養)에 관한 연구(硏究))

  • The objectives of this investigation were to produce artificially an antitumor constituent by submerged culture of the mycelium of Coriolus versicolor (Fr.) Quel., to characterize the influence of various modifications of the nutrient and culture conditions with respect to the pro­duction, to determine chemical composition of the antitumor constituent, and to examine effects of the constituent on the immune response of mice. Submerged agitation of the mycelium in flasks containing a nutrient solution showed its adequate growth. Especially the mycelial growth in the medium containing glucose and yeast extract was abundant. The addition of cotton seed flour or ginseng waste to the medium increased the yield of mycelial growth and the production of the antitumor constituent. The replacement of glucose with starch also yielded the adequate growth. The antitumor constituent extracted from the mycelium and isolated from the culture filtrate was a protein-bound polysaccharide. The analyses of this constituent by GLC and amino acid autoanalysis showed that it contained four monosaccharides and fifteen amino acids. The protein-free polysaccharide of the constituent was also found to exert greater antitumor activity against sarcoma-180 in mice than the entire constituent. The antitumor constituent was found to potentiate the immune response of mice against sheep red blood cell. The protein-bound polysaccharide exerted more favorable influence on the immunity than the protein-free moiety.

Gellan-type Microbial Polysaccharide Production in Continuous Fermentation (Gellan형 미생물 다당류의 연속생산)

  • The Gellan-type polysaccharide produced by Pseudomonas elodea(ATCC 31461) is one of the new heteropolysaccharides, having useful properties as gelling, suspending, stabilizing, emulsifying and binding agents in aqueous systems. Medium compositions for growth stage and production stage are improved. The problems of low cell concentration and poor productivity in highly viscous fermentation were attributed to inadequate mixing accompanied by insufficient oxygen transfer. During continuous culture, cell growth and polysaccharide production were greatly affected by the apparent viscosity, and they showed oscillation behavior, i.e. as the product concentration increases, cell concentration decreases. With improved culture conditions, the productivity of continuous culture increased up to 0.6g/$\ell$/hr(6-fold that of batch culture ) at dilution rate, D=$0.14hr^{-1}$.

Optimum Conditions for the Biological Production of Lactic Acid by a Newly Isolated Lactic Acid Bacterium, Lactobacillus sp. RKY2

  • Lactic acid is a green chemical that can be used as a raw material for biodegradable polymer. To produce lactic acid through microbial fermentation, we previously screened a novel lactic acid bacterium. In this work, we optimized lactic acid fermentation using a newly isolated and homofermentative lactic acid bacterium. The optimum medium components were found to be glucose, yeast extract, $(NH_4)_{2}HPO_4,\;and\;MnSO_4$. The optimum pH and temperature for a batch culture of Lactobacillus sp. RKY2 was found to be 6.0 and $36^{\circ}C$, respectively. Under the optimized culture conditions, the maximum lactic acid concentration (153.9 g/L) was obtained from 200 g/L of glucose and 15 g/L of yeast extract, and maximum lactic acid productivity ($6.21\;gL^{-1}h^{-1}$) was obtained from 100 g/L of glucose and 20 g/L of yeast extract. In all cases, the lactic acid yields were found to be above 0.91 g/g. This article provides the optimized conditions for a batch culture of Lactobacillus sp. RKY2, which resulted in highest productivity of lactic acid.

Biopolyrner Production of Zoogloea ramigera in Batch, Fed-Batch and Continuous Culture Processes (Zoogloea ramigera의 회분식, 유가배양, 연속배양에 의한 생물고분자 생산)

  • Zoogloea ramigera 115 was selected for the production of viscous microbial polysaccharide for bioflocculants usage. Batch, fed-batch, and continuous culture processes were examined with regard to the high biopolymer production. Several carbon sources were tested, including glucose, lactose, molasses, and cheese whey. The C/N ratio of 90 was most effective for biopolymer production from glucose, while the C/N ratios of 30 for lactose and 60 for both molasses and cheese whey substrate gave a maximum production. Fed batch culture proved more effective to increase final biopolymer concentration than batch culture. Continuous fermentation with two stages modifying C/N ratio increased the productivity. The production rates were a maximum at dilution rate of 0.048 $hr^{-1}$ for molasses and at 0.096 $hr^{-1}$for cheese whey.

The Case Study of Lactobacillus mixture culture fluid on Atopic dermatitis (아토피피부염에 대한 Lactobacillus 혼합배양액의 임상 증례)

  • Objectives: Recently lactic acid formulation was known as the adjuvant therapy on atopic dermatitis(AD) symptoms because of little side effects. In this study, it was studied that Lactobacillus mixed culture appliment was effective on not on AD symptoms. Methods: The case-photos 30-40 times of AD symptoms from case were observed with literatures. The case-photos were acquired with environmental AD dermatitis experience program which is conducted in SUNCHANG RESERCH INSTITUTE OF HEALTH AND LONGEVITY from 2014.01 to 2014.08. Experience applicants were limited the oral administration and chemical external administration. Results: Lactobacillus mixed culture appliment was effective because of the mitigation or disappearance of AD primary symptoms like pruritus, erythema, edema, effusion, dry skin, scaly, scab etc. AD symptom degree was dependent on lactobacillus mixed culture appliment times, and classified as Reaction Period (RAP), Reduction Period (RDP), Efection Period (EP), Reproduction Period (RPP) on a single mixed culture appliment time. And AD symptoms which were appeared in RPP were classified as Rebound Period (RBP), Effection Period (EP), Subclinical Period (SCP). Conclusions: Lactobacillus mixed culture appliment is considered to be useful for AD symptoms and is estimated to be effective by different mechanism with oral administration or steroid ointment application. Lactobacillus mixed culture appliment is expected to induce a synergistic effects on AD symptom reliefs with the other oral administration.

Biological Control of Aphid Using Fungal Culture and Culture Filtrates of Beauveria bassiana

  • Aphids are one of the most destructive pests in crop production such as pepper, cucumber, and eggplants. The importance of entomopathogenic fungi as alternative pest control agents is increasing. Conidia of entomopathogenic fungi are influenced by environmental conditions, such as temperature and relative humidity, and cause slow and fluctuating mortality. These factors have prevented wider application and use of biocontrol agents. For investigation of means of mitigation of such problems, we conducted bioassays with 47 fungal culture filtrates in order to evaluate the potential of secondary metabolites produced by entomopathogenic fungi for use in aphid control. Among 47 culture filtrates cultured potato dextrose broth, filtrate of Beauveria bassiana Bb08 showed the highest mortality (78%) against green peach aphid three days after treatments. Filtrate of Bb08 cultured in Adamek's medium showed higher toxicity as 100% to third instar nymphs of the aphid compared with seven other filtrates cultured in different broths amended with colloidal chitin or oil. The culture filtrates and fungal cultures from media amended with colloidal chitin or oil had lower control efficacies than filtrates without these additives in three different media. These results indicate that the fungal culture fluid or culture filtrate of B. bassiana Bb08 cultured in Adamek's medium has potential for development as a mycopesticide for aphid control.

Environmental toxicity and biodegradation of Pseudomonas sp. EL-G527 producing biosurfactant

  • A biosurfactant-producing microorganism, .Pseudomouos sp. EL-G527 was isolated from activated sludge by enrichment culture when grown on mineral salt medium containing n-hexadecane as a carbon source. The biosurfactant from .Pseudomonar sp. EL-G527 exhibited lesser toxicity to bacterial population than synthetic surfactants and in the biodegradation test, biosurfactant was rapidly degraded and lost its activity as surface active material after 1 day incubation. In this study, the biosurfactant from Pseudomonas sp. EL-G527 was effective surface-active compound, more biodegradable and less toxic to microbial ecosystem than various synthetic surfactants.

  • ${\cdot}$ The LAB as an integrated part of kimchi were well adapted to the new habitat of fermenting sausage and exhibited good souring properties that are comparable to those commercial starter cultures. ${\cdot}$ With the added kimchi (5-15%) and kimchi-powder (2-5%), the necessary microbial stability of real fermented sausages was achieved. ${\cdot}$ In particular, kimchi-powder contributed to improving the safety of the fermented sausages as compared to the conventional one treated with starter culture.

Studies on the Microbial Pigment(II) (미생물의 색소에 관한 연구 2)

  • Glucose and galactose were the inhibitors of pigmentation of Serrratia marcescens. Other sugars, however, even the fructose which is the structural isomer of glucose and galactose did not affect to pigmentatioin. The yield of pigmentation was descreased when the glucose was added to culture medium. And it was known to that the antibiotics was roled as the inhibitors of pigmentation. The limit concentration of the inhibitors were as followings :rifampicin, $1{\mu}g/ml$. Addition of rifampicin$(1{\mu}g/ml)$ at 6 hrs cultures inhibited the formation of pigment completely.

  • Capillary electrophoretic direct chiral separation method is described for the determination of the absolute configuration of chiral aromatic acids, The enantiomeric separation was achieved by capillary electrophoresis using HP $\beta$-cyclodextrin (CD) as the chiral selector. The effect of CD concentration was investigated to optimize the chiral separation and resolution. When applied to microbial culture fluid. the present method allowed positive identification of chiral aromatic acids and their chirality as well.
