Effects of a specific blend of essential oils on apparent nutrient digestion, rumen fermentation and rumen microbial populations in sheep fed a 50:50 alfalfa hay:concentrate diet

  • Objective: An experiment was conducted to investigate the effects of a specific mixture of essential oils (MEO), containing thyme, clove and cinnamon EO, on rumen microbial fermentation, nutrient apparent digestibility and blood metabolites in fistulated sheep. Methods: Six sheep fitted with ruminal fistulas were used in a repeated measurement design with two 24-d periods to investigate the effect of adding MEO at 0 (control), 0.8, and 1.6 mL/d on apparent nutrient digestibility, rumen fermentation characteristics, rumen microbial population and blood chemical metabolites. Animals were fed with a 50:50 alfalfa hay:concentrate diet. Results: Ruminal pH, total volatile fatty acids (VFA) concentration, molar proportion of individual VFA, acetate: propionate ratio and methane production were not affected with MEO. Relative to the control, Small peptides plus amino acid nitrogen and large peptides nitrogen concentration in rumen fluid were not affected with MEO supplementation; while, rumen fluid ammonia nitrogen concentration at 0 and 6 h after morning feeding in sheep fed with 1.6 mL/d of MEO was lower (p<0.05) compared to the control and 0.8 mL/d of MEO. At 0 h after morning feeding, ammonia nitrogen concentration was higher (p<0.05) in sheep fed 0.8 mL/d of MEO relative to 1.6 mL/d and control diet. Ruminal protozoa and hyper ammonia producing (HAP) bacteria counts were not affected by addition of MEO in the diet. Relative to the control, no changes were observed in the red and white blood cells, hemoglobin, hematocrit, glucose, beta-hydroxybutyric acid, cholesterol, total protein, albumin, blood urea nitrogen and aspartate aminotransferase and alanine aminotransferase concentration. Apparent total tract digestibility of dry matter, crude proten, organic matter, and neutral detergent fiber were not influenced by MEO supplementation. Conclusion:The results of the present study suggested that supplementation of MEO may have limited effects on apparent nutrient digestibility, ruminal fermentation and protozoa and HAP bacteria count, blood cells and metabolites.

Microbiological Quality Evaluation for Implementation of a HACCP System in Day-Care Center Foodservice Operations I. Focus on Heating Process and After-Heating Process (보육시설급식소의 HACCP시스템 적용을 위한 미생물적 품질평가 I. 가열조리 및 가열조리후 처리 공정을 중심으로)

  • The objective of this study was to evaluate the microbiological quality of heating and after-heating processed foods for implementation of a HACCP system in day-care center foodservice operations. The evaluating points were microbial assessment and temperature of foods during receiving, cooking, and serving in heating process. In non-heating process, in addition to monitoring microbial assessment of food during preparation, cooking, and serving steps, the microbial populations of employees' hands and utensils and serving temperature were also evaluated. Microbiological quality was assessed using 3M Petrifilm$^{TM}$ to measure total plate count and coliforms for foods and utensils and Staphylococcus aureus for hands in five Gumi day-care centers. Microbiological quality assessment for foods and utensils is summarized as follows. Microbiological quality of the heating processed foods was satisfactory for cooking and serving steps. The internal temperature of food was above 74$^{\circ}C$. However, temperature control before the serving step was not achieved due to inappropriate time management between the cooking and serving steps. In the after-heating process, the total plate counts of boiled mungbean sprouts salad, blanched spinach salad, com vegetable salad were below the standard at the serving step. The majority of samples showed that coliforms exceeded the norm, which is thought to be the result of the cross-contamination from utensils. These results suggest that it is essential to educate employees on the importance of hand washing and of avoiding cross-contamination by using clean, sanitized equipment to serve food in the after-heating process. Establishing Sanitation Standard Operating Procedures (SSOPs) is an essential part of any HACCP system in day-care center foodservice operations.

Effects of Electron Beam Radiation on the Microflora and Sensory Characteristics of Pork Loin (전자선 조사에 의한 국내산 신선 돈육의 미생물학적 및 관능적 특성 변화)

  • The effects of electron-beam irradiation on the microbial and sensory qualities of pork loins were studied. The pork loins were aerobically packed in polyethylene films and exposed to absorbed doses of electron-beam at 0, 0.5, 1 and 2 kGy in ambient temperature. All samples were stored at $4{\pm}1^{\circ}C$. As an irradiation dose increased, decontaminating effects increased in terms of total microbial count, lactic acid bacteria and psychrotrophs during storage. All difference degrees in the triangle test were very low levels. In the descriptive analysis, Low dose irradiation did not affect aroma, off-flavor and acceptability in fresh and cooked meats.

Improvement of HACCP Verification Checklist in School Foodservices - A Case Study on Cooked Squid with Seasoned Fresh Vegetable - (학교급식 HACCP 검증 체크리스트 개선 - 오징어채소무침을 중심으로 -)

  • The purpose of this study was the improvement and modification of the HACCP verification checklist in school foodservices. For this, the HACCP verification checklist was modified on the basis of an existing school foodservice format. The modified checklist was composed of 28 items, including CCPs (critical control points), microbial test, and other components of the HACCP system than CCPs. To confirm the suitability of the modified checklist, comparisons were made based on the microbiological quality of cooked foods, utensils, and number of aerial microbes in the working area. In this study, the applicability of the modified checklist was determined by focusing on cooked squid with seasoned fresh vegetables (Ojingeochaesomoochim). The following results were obtained from 14 schools in Changwon. The checklist scores for maintaining hot foods over $60^{\circ}C$ or serving within 2 hours, microbial tests of drinking water, food contact surfaces and cooking utensils, monitoring tools, and usage of suitable sanitizers were 2 points each (The possible highest score is 2 points). On the contrary, the checklist score for microbial test of cooked foods was the lowest of all the items. The correlation coefficient (r) between the improved checklist and microbiological quality of cooked foods was 0.699 (P<0.01), whereas that between the improved checklist and microbiological quality of cooking utensils was 0.612 (P<0.05). The correlation coefficient between the improved checklist and aerial plate count in the working area was -0.556 (P<0.05). Our results indicate the potential possibility of using the HACCP verification checklist in school foodservices.

Changes in Quality Characteristics of Cherry Tomato Packaged with Different Films (포장 조건에 따른 방울토마토의 저장 중 품질 특성 변화)

  • Cherry tomato was packed with several plastic films, and investigated the quality characteristics such as gas composition, weight loss, microbial load, soluble solid content, total acidity during storage at 12$\^{C}$. Cast polypropylene(CPP), low density polyethylene(LDPE), polyolefin(MPD, PD961) and perforated polyolefin(PY85) were used as packaging film. Oxygen and carbon dioxide concentration inside packages generally showed a stabilized levels after 10 days, but those of CPP and PD961 changed throughout the storage. Weight loss of PY85 was 25.6 % after 20 days, while others maintained around 1.0%. Total microbial count showed a increase pattern similar to yeast and mold, and CPP maintained the highest microbial load after 10 days. MPD retarded the decay of cherry tomato to 5.3%, while others showed above 20% after 20 days.

Effect of Electron Beam Irradiation on the Microbial Safety and Qualities of Sliced Dried Squid (전자선 조사가 건조 오징어의 미생물학적 안정성 및 품질에 미치는 영향)

  • Electron beam irradiation was applied to examine the microbial safety and qualities of sliced dried squid. Sliced dried squid was irradiated at dose of 2, 4, 8, 12, and 16 kGy. Microorganisms contaminated in sliced dried squid were significantly decreased by irradiation. Decimal reduction dose (D/sub 10/ value) of total bacteria count, yeast and mold, coliforms in sliced dried squid were 8.57, 4.60, and 8.10 kGy, respectively. Electron beam irradiation caused negligible changes in Hunter color L, a, and b value. Sensory evaluations of irradiated sliced dried squid showed that there were no significant changes among the samples. These results indicate that electron beam irradiation improves the microbial safety and qualities of sliced dried squid.

  • This study was performed to investigate the physical, chemical and microbial characteristics of Gwamegi to provide basic data for the standardization of marine processed foods and for the improvement of the quality of commercial Gwamegi. The acid values of commercial Gwamegi were 5.8, 5.3 and 5.2 mg KOH/g for fillet type (F-type), "two divide" type (T-type) and whole type (W-type), respectively, and the peroxide values were 51.6, 51.5 and 53.2 meq/kg for each. There was a positive correlation between the acid value and the peroxide value (r=0.555) at confidence intervals (CI) of 99%. Trimethylamine (TMA) content of F-type, T-type and W-type products were 2.9, 2.6 and 3.6 mg%, respectively, while volatile basic nitrogen (VBN) contents were 22.4, 21.5 and 21.8 mg%. There was a strong positive correlation between TMA and VBN (r=0.961) at a CI of 99%. The histamine content was detected to be as much as 122 mg/kg, with about 36 % of the samples exceeding the CODEX criteria for histamine of 100 mg/kg. The total microbial count of 4 products exceeded 5 Log CFU/g and coliform group of 11 products exceeded the criteria of less than 1 Log CFU/g. Staphylococcus aureus in 27% of the samples exceeded the criteria of less than 2 Log CFU/g.

Microbial Quality of Street Foods Sold by Season (계절에 따른 길거리 제조 식품의 미생물 오염 특성)

  • This study examined microbiological contamination of street foods(kimbab, fish cake, Korean sausage) by microbiological analysis. A collection of 360 samples of street foods was obtained seasonally in four major cities(Seoul, Daejeon, Busan, Gwangju) in Korea. Aerobic mesophilic counts ranged between 1.0 and 9.9 log CFU/g, with the highest count recorded from Kimbab. Counts of psychrotrophic microorganisms were as high as those of mesophilic microorganisms. Total coliform populations between <1.0 and 7.5 log CFU/g were found in 53.6% of samples. Escherichia coli 4.4%, Staphylococcus aureus 7.8% and Clostridium perfringens 3.3%. Salmonella spp. and Listeria monocytogenes were not detected in any of the samples. Kimbab purchased in spring and summer showed higher S. aureus and Cl. perfringens contamination rates. Microbial contamination levels determined in the present study may be used as the primary data to execute microbial risk assessment of street foods.

Changes in Quality Characteristics of Bell Pepper Packaged with Different Films (포장 조건에 따른 피망의 저장 중 품질 특성 변화)

  • Bell pepper was packed with several plastic films, and investigated the quality Characteristics such as gas composition, weight loss, microbial load, soluble solid content, ascorbic acid and color during storage at 10$^{\circ}C$. Cast polypropylene (CPP), low density Polyethylene (LDPE), Polyolefin (MPD, PD961) and linear low density polyethylene (LLDPE) were used for this purpose. Oxygen concentration inside packages generally showed a 10∼14% after 1∼2 weeks, but package with CPP maintained above 15% throughout the storage. Carbon dioxide concentration of CPP was above 2%, but other treatments showed below 2%. Weight loss of all treatments was below 3.0% after 5 weeks. Total microbial count showed a similar pattern compared to yeast and mold, and CPP maintained the highest microbial load after 3 weeks. Yellowness (b value) of bell pepper was generally increased throughout the storage, and MPD and PD961 were higher than other treatments.

Effect of Electron Beam Irradiation on Microbial Qualities of Whole Black Pepper Powder and Commercial Sunsik (전자선 조사가 통후추 분말과 시판 선식의 미생물학적 안전성 및 품질에 미치는 영향)

Whole black pepper powder and commercial Sunsik were irradiated at 2, 4, 8, 12, and 16 kGy. Microorganism contamination in black pepper powder and commercial Sunsik were significantly decreased by irradiation. Decimal reduction doses ($D_{10}\;value$) of total bacteria count in black pepper powder and commercial Sunsik were 5.32 and 1.56 kGy, respectively. $D_{10}\;value$ of yeast and mold were 2.54 and 2.14 kGy for black pepper powder and commercial Sunsik, respectively. Electron beam irradiation caused negligible changes in Hunter color L, a, and b values. Sensory evaluations of black pepper powder and commercial Sunsik showed no significant changes among samples. These results indicate electron beam irradiation improves microbial safety and qualities of black pepper powder and commercial Sunsik.
