Quinone profile과 PCR-DGGE를 이용한 정수장 침전지에서의 부착조류 및 미생물군집의 계절적 변화해석 (Analysis of the Seasonal Change in Attached Algae and Microbial Community Structure in Sediment Basin Trough of Water Treatment Plant By Using Quinone Profile and PCR-DGGE)

  • The seasonal change in attached algae and microbial community structure at sedimentation basin of water treatment plant was investigated by using quinone profiles and denaturing gel gradient electrophoresis (DGGE). The photosynthetic bacteria and algae contains PQ-9 and VK-1 as major quinone are major component of the total quinone fraction in attached algae and microorganisms on sedimentation basin trough. The microorganisms containing menaquinones appear to be sensitivity to the change in temperature than those containing ubiquinones. The plot of the mole fraction of dominant quinone species ($f_d$) to the DQ values showed higher sensitivity to the seasonal change in the microbial community structure. The results indicated that quinone and DGGE are useful tool for the evaluation of the changes in the microbial community structure.

  • The differences in microbial community structures between planktonic microorganism and biofilm in rivers were investigated using respiratory quinone profiles. The compositions of microbial quinone for 4 tributaries of the Kyongan Stream located in/flowing through Yongin City, Gyeonggi-Do were analyzed. Ubiquinone(UQ)-8, UQ-9, menaquinone(MK)-6 and Plastoquinone(PQ)-9 were observed in all samples of planktonic microorganism and biofilm for the sites investigated, Most planktonic microorganism and biofilm had UQ-8(15 to 30%) and PQ-9(over 30%) as the dominant quinone type. These results indicated that oxygenic phototrophic microbes(cyanobacteria and/or eukaryotic phytoplankton) and UQ-8 containing proteobacteria constituted major microbial populations in the river. The quinone concentration in the river waters tested, which reflects the concentration of planktonic microorganisms, increases with increasing DOC. Further research into this is required. The microbial diversities of planktonic microorganism and biofilm calculated based on the composition of all quinones were in the range from 4.2 to 7.5, which was lower than those for activated sludge(ranging from 11 to 14.8) and soils(ranging from 13.4 to 16.8). The use of quinone profile appears to be a useful tool for the analysis of microbial community structure in river.

  • The respiratory quinone profile was used as a tool for the study on microbial community structure in the mine tailings of abandoned mines over the depth. For the study, the area of Jingok mine located in Bongwha, Korea has been selected. The distributions of Cd, Cu, Pb, Al, Fe and Mn showed the following common patterns; the highest values in the upper part of mine failings (0-20 cm), rapid decrease with increasing depth. The dominant quinone species of the mine tailings were UQ-9 followed by UQ-10, suggesting that microbes had contributed to heavy metal degradation. The quinone contents in mine tailings ranged from 5.0 to 24.9 nmol/kg. The microbial diversity in the upper part of mine tailings (0-40 cm) was higher than that of lower part of mine tailings (100-120 cm).

도시 공원의 토양에서 중금속이 미생물의 생체량과 활성에 미치는 영향 (The effects of heavy metals on microbial biomass and activity in contaminated urban park soils)

  • The relationship between pb, zn, cd, and the microbial biomass and activity were investigated in three public park soils of central and outer london. Variability with distance from the roadside and profile were studied. The heavey metal concentrations were the highest in hampstead heath and hyde park with high trafic density and the lowest in hainault. The highest concentrations of heavy metals were found adjacent to the roadside in the upper parts of the soil profile. Dehydrogenase activity, adenosine tri-phosphate and ergosterol contents used as indices of micrbial biomass and activity, were generally higher in hainadult, and also higher in the upper pats of the soil profile. Simple regression analysis indicated that the microbial biomass and activity were affected significantly by moisture content, water holding capacity, total organic carbon, total nitrogen, and organic mater rather than heavy metal concentration. Highest inputs of nitrogen and carbon were associated with high inputs of heavey metals, all three being derived from vehicle emissions adjacent to the road. The hyde park and hampstead heath microbial populations were able to respond to the c and n input positively by increase in biomass and activity, whereas the hainault populations could not. This rsult suggrsts adaptation in he former to heavy matals, but not in the latter.

Insulin Resistance Does Not Influence Gene Expression in Skeletal Muscle

  • Insulin resistance is commonly observed in patients prior to the development of type 2 diabetes and may predict the onset of the disease. We tested the hypothesis that impairment in insulin stimulated glucose-disposal in insulin resistant patients would be reflected in the gene expression profile of skeletal muscle. We performed gene expression profiling on skeletal muscle of insulin resistant and insulin sensitive subjects using microarrays. Microarray analysis of 19,000 genes in skeletal muscle did not display a significant difference between insulin resistant and insulin sensitive muscle. This was confirmed with real-time PCR. Our results suggest that insulin resistance is not reflected by changes in the gene expression profile in skeletal muscle.

Effect of Feeding Ficus infectoria Leaves on Rumen Microbial Profile and Nutrient Utilization in Goats

  • A feeding trial was conducted to study the effect of tannin rich Pakar (Ficus infectoria) leaves on microbial profile, rumen fermentation and nutrient utilization in goats. Eight goats divided in two groups were fed pakar leaves (experimental group) and green oats (control group) as sole roughage source along with a fixed quantity of concentrate mixture for a period of 3 months. Two metabolic trials of six days duration were conducted after 30 and 90 days of experimental feeding. The dry matter intake was significantly higher (p<0.05) and digestibility's of DM, OM, CP, EE, NDF and ADF were reduced in experimental as compared with the control group. The TDN intake was similar (236.52 vs. 240.39 g/d) in both the groups. All the animals were in positive nitrogen balance. The concentration of ammonia nitrogen, TVFA, lactic acid and activities of xylanase and protease were reduced in pakar leaves fed goats. The rumen microbial profile as obtained by MPN technique showed no change in total bacterial population but total fungi and cellulolytic bacteria were reduced (p<0.05), whereas, tannin degrading/tolerant bacteria increased with the feeding of pakar leaves. Real time PCR data revealed a decrease in Ruminococcus flavefaciens, an increase in methanogens and no change in the Fibrobacter succinogenes population by feeding of pakar leaves.

Characterization of Microbial Community in Biological Wastewater Treatment System Using Respiratory Quinone Profiles

  • The dynamics of microbial community structure of the various domestic wastewater treatment processes were examined using a novel approach of quinone profiles. The compositions of microbial quinone of 5 sites fer plant and lab-scale activated sludge were analyzed. More than 14 kinds of quinones were observed in the activated sludges tested in this study. The microbial community structure of the plant activated sludge processes a little differed from that of the lab-scale submerged MBR systems. The dominant quinones were UQ-8, UQ-10 followed $MK-8(H_4)$, MK-7 and MK-6. The molar ratio of ubiquinones to menaquinones (UQ/MK) changed from 0.81 to 1.9, indicating that aerobic bacteria dominated the microbial community of the activated sludge examined. The microbial diversity of the activated sludges calculated from the all quinone compositions was 9.5-11.9 and the microbial equability of the activated sludges was 0.64-0.79.

논과 밭 토양에서 토층간 미생물 군집의 차이 (Variation of Microbial Community Along Depth in Paddy and Upland Field)

  • 인지질 지방산을 분석하여 특정 미생물군의 수직적 분포와 토층간 미생물 군집 패턴을 조사하였다. 경북 농업기술원에 위치하고, 질소, 인산, 가리의 화학비료만 장기 연용한 논과 밭 포장에서 15 cm 깊이까지 토양을 채취하였다. 인지질 지표 지방산을 주요인 분석으로 분석하여 토양 미생물 군집을 분석한 결과 논과 밭 토양의 미생물 군집은 뚜렷하게 구분되었으며, 토층간 차이보다 논과 밭의 차이가 더 컸다. 논보다 밭은 토층이 깊어짐에 따라 미생물 군집이 급격하게 변하였는데, 미생물 군집 측면에서 밭보다 논의 표층이 더 두껍다고 볼 수 있다. cyclopropyl/monoenoic precursor 비율과 전체 포화지방산/전체 불포화 지방산 비율은 토심이 깊어짐에 따라 증가하였는데, 이는 토심이 깊어질수록 탄소원과 통기가 부족하기 때문에 일어나는 현상으로 보인다. 대체로 표토는 그램음성균, 곰팡이 등의 상대적 비율이 높고 토심이 깊어질수록 세균과 방선균의 상대적 비율이 높아졌다.

  • The objective of this study was to investigate community structure of attached algae and microbes in sedimentation basin of water and wastewater treatment plants by using respiratory quinone profile. There was an evident difference, in microbial community structure and attached algae species, between inclination plate settler and drainage canal in the sedimentation basin. The algae was composed of species in following order; Chlorophyceae>Bascillariophyceae>Cyanophyceae. The dominant quinone types of attached microorganisms in the wastewater treatment plant were plastoquinone (PQ)-9, vitamin(V)K-1 followed by UQ-8, but those for water treatment plant were VK-1, PQ-9 followed by UQ-8. These results suggests that nutrients, seasons and material of sedimentation basin have notable influence on composition of attached algae and microbial community structure in water and wastewater treatment plants.

PCR-DGGE를 이용한 누룩에서의 미생물 다양성 분석 (Analysis of Microbial Diversity in Nuruk Using PCR-DGGE)

  • 누룩은 탁주와 약주의 제조를 위한 당화효소와 알코올발효를 위한 미생물의 공급원으로서 제품의 맛과 품질을 결정하는 중요한 역할을 한다. 본 연구에서는 산성누룩의 세균과 진균의 다양성을 조사하기 위해 순수분리 종과 16S 및 28S rRNA gene를 대상으로 한 PCR-DGGE를 이용한 분석을 수행하였다. 누룩 내 세균의 수는 $2.7{\times}10^9$ CFU/g이었으며 순수분리와 PCR-DGGE 분석에서 우점종은 Kocuria spp., Pantoea spp., Lactobacillus spp., Pediococcous spp., Weissella spp., Staphylococcus spp. 그 외 endophytic bacterium, uncultured gamma-proteobacteria, uncultured Cyanobacteria와 Actinobacteria였다. PCR-DGGE profile에서 주된 우점종은 Pediococcous pentosaceus와 uncultured Cyanobacteria 이었다. 누룩 내 진균의 수는 $3.5{\times}10^8$ CFU/g이었으며 순수분리와 PCR-DGGE 분석에서 우점종은 Trichomonascus spp., Pichia spp., Torulaspora spp., Wickerhamomyces spp., Sacharomycopsis spp., Lichtheimia spp., Mucor spp., Rhizopus spp., Aspergillus spp., Cladosporium spp.였다. PCR-DGGE profile에서 주된 우점종은 Pichia kudriavzevii와 Aspergillus oryzae이었다. PCR-DGGE 기술은 본 연구에서 누룩의 미생물군집을 평가하기 위해 처음으로 사용되었으며 미생물 다양성을 설명하는 데 효과적임을 입증하였다.
