This study was performed to evaluate the inactivation and microbial regrowth of heterotrophic and nitrifying bacteria using chloramine as a secondary disinfectant for drinding water distribution system. Three sets of the three reactors filled with the $Cl_2/NH_3-N$ ratio of 3:1, 4:1, and 5:1 were used in these experiments. Chloramine concentration were applied to each set of the reactors with $1mg/\ell$,\;2mg/\ell\;and\;3mg/\ell$, respectively. For the set with elapsed time and reached to zero level after 7 days. Heterotrophic bacteria remarkably increased and nitrification through the experimenatal period (21 day). Furthermore the regrowth of heterotrophic bacteria and nitrification were not found. More than $2mg/\ell$ of chloramine with $Cl_2/NH_3-N$ ratio of 3:1, the nitrification could be inhibited by 2 days of contact time.
A new phenol and hydrazide derivatives were obtained for the first time from the C. giganteawhite by silica gel column chromatography. The structure of the isolated compounds was identified by UV, IR NMR and MS. C. gigantea was scientifically reported for several medicinal properties viz. analgesic, antimicrobial and cytotoxic. In this screening work, anti-microbial activity of test compounds was found to be active against all organisms. Additionally, anti-inflammatory activity of the test groups has reduced the thickness of edema of the hind paw compared to the control group.
This study was performed to investigate the physical characteristics like compressive strength, permeability, porosity and the water quality removal characteristics of permeable concrete pavement filled with microbial-soil sheet to remove SS, organic matter and nutrients in artificial rainfall. As a result, it can show the removal efficiency is SS 90~95%, COD 85~93%, BOD 80~83%, T-N 61~75%, T-P 71~78% on WAPS I(W1) and WAPS II(W2). Therefore, permeable concrete pavement filled with microbial-soil sheet shows higher removal efficiency(SS 10%, organic matter and nutrients 30%) than a conventional porous concrete(W3). By filling microbial-soil sheet to permeable concrete pavement, we confirm that the function and efficiency are improved significantly and that a naturally-friendly facility can be developed and applied to treat non-point sources.
The xynA gene encoding an alikali-tolerant endo-1,4-${\beta}$-xylanase (XYN) was cloned from the alkalophilic Bacillus pumilus A-30. The nucleotide sequence of a 974-bp DNA fragment containing the xynA was determined. An ORF of 684 nucleotides that encoded a protein of 228 amino aicds was detected. Asparagine-71 of XYN from B. Pumilus A-30 showed to be highly conservative in alkaline xylanases of family G/11, upon comparing the amino acid sequences of 17 family G/11 xylanases. Site-directed mutation of N71D of the xynA gene resulted in a decrease of 12.4% in the specific acitivity and a significant decline in the enzyme activity in the alkaline pH range.
On a urea-molasses-straw (3:15:82; UMS) based diet effect of graded levels of cottonseed cake (CSC) supplementation on the performance of native (Bos indicus) bulls has been studied for 167 days. Eighteen growing bulls of $129{\pm}13.4kg$ weight and about 14 months old were randomly allocated to three dietary treatments designed in a completely randomized design, having six animals in each treatment. Three dietary treatments were 0, 0.5 and 1.0 kg CSC per head/d. In addition, each animal also received ad lib. UMS, 4 kg Napier (Pennisetum purpureum) grass, 500 g of each of rice and wheat bran and 60 g mineral mix daily. For unit increase in CSC, total DM intake was increased by $1g/kg\;W^{0.75}/d$ but the straw DM intake decreased by $0.54g/kg\;W^{0.75}/d$. Whole gut digestibility of DM and OM was not effected but N and ADF digestibility increased with incremental increase in dietary CSC. For unit (1kg) increase in dietary CSC intake N and ADF digestibility increased by 10 (${\pm}1.155$) and 3 (${\pm}1.732$) unit respectively. Microbial N yield for the 0, 0.5 and 1.0 kg CSC were 5.63, 5.28 and 5.16 g/kg OM apparently fermented in the rumen respectively. For each gram increase in CSC, N intake and N balance increased by 0.626 (${\pm}0.015$) and 0.625 (${\pm}0.0814$) mg/kg $W^{0.75}/d$. High apparent N balance was contrasted with low live weight gain, e.g., for 1 kg increase in CSC supplementation, live weight gain increased by only 0.077 (${\pm}0.00288$) kg/d ($r^{2}=0.99$; p<0.01). The conversion efficiency was 12.98 kg CSC per kg of live weight gain. It was concluded that unless the protein is being protected from the rumen degradation, addition of CSC to UMS diet would have little nutritional or economic advantages.
To investigate the function of N-glycosylation of Cel5A (endoglucanase II) from Hypocrea jecorina, two N-glycosylation site deletion Cel5A mutants (rN124D and rN124H) were expressed in Saccharomyces cerevisiae. The weights of these recombinant mutants were 54 kDa, which were lower than that of rCel5A. This result was expected to be attributed to deglycosylation. The enzyme activity of rN124H was greatly reduced to 60.6% compared with rCel5A, whereas rN124D showed slightly lower activity (10%) than that of rCel5A. rN124D and rN124H showed different thermal stabilities compared with the glycosylated rCel5A, especially at lower pH value. Thermal stabilities were reduced and improved for rN124D and rN124H, respectively. Circular dichroism spectroscopy showed that the modification of secondary structure by mutation may be the reason for the change in enzymatic activity and thermal stability.

The objective of this study was to determine the effect of rain tree pod meal with palm oil supplementation on feed intake, digestibility, microbial protein synthesis and microbial populations in dairy cows. Four, multiparous early-lactation Holstein-Friesian crossbred (75%) lactating dairy cows with an initial body weight (BW) of $405{\pm}40$ kg and $36{\pm}8$ DIM were randomly assigned to receive dietary treatments according to a $4{\times}4$ Latin square design. The four dietary treatments were un-supplementation (control), supplementation with rain tree pod meal (RPM) at 60 g/kg, supplementation with palm oil (PO) at 20 g/kg, and supplementation with RPM at 60 g/kg and PO at 20 g/kg (RPO), of total dry matter intake. The cows were offered concentrates, at a ratio of concentrate to milk production of 1:2, and chopped 30 g/kg of urea treated rice straw was fed ad libitum. The RPM contained condensed tannins and crude saponins at 88 and 141 g/kg of DM, respectively. It was found that supplementation with RPM and/or PO to dairy cows diets did not show negative effects on feed intake and ruminal pH and BUN at any times of sampling (p>0.05). However, RPM supplementation resulted in lower crude protein digestibility, $NH_3$-N concentration and number of proteolytic bacteria. It resulted in greater allantoin absorption and microbial crude protein (p<0.05). In addition, dairy cows showed a higher efficiency of microbial N supply (EMNS) in both RPM and RPO treatments. Moreover, NDF digestibility and cellulolytic bacteria numbers were highest in RPO supplementation (p<0.05) while, supplementation with RPM and/or PO decreased the protozoa population in dairy cows. Based on this study, supplementation with RPM and/or PO in diets could improve fiber digestibility, microbial protein synthesis in terms of quantity and efficiency and microbial populations in dairy cows.

In this paper, a counting algorithm hybridized with an adaptive automatic thresholding method based on Otsu's method and the algorithm that elongates markers obtained by the well-known watershed algorithm is proposed to enhance the exactness of the microcell counting in microscopic images. The proposed counting algorithm can be stated as follows. The transformed full image captured by CCD camera set up at microscope is divided into cropped images of m$\times$n blocks with an appropriate size. The thresholding value of the cropped image is obtained by Otsu's method and the image is transformed into binary image. The microbial cell images below prespecified pixels are regarded as noise and are removed in tile binary image. The smoothing procedure is done by the area opening and the morphological filter. Watershed algorithm and the elongating marker algorithm are applied. By repeating the above stated procedure for m$\times$n blocks, the m$\times$n segmented images are obtained. A superposed image with the size of 640$\times$480 pixels as same as original image is obtained from the m$\times$n segmented block images. By labeling the superposed image, the counting result on the image of microbial cells is achieved. To prove the effectiveness of the proposed mettled in counting the microbial cell on the image, we used Acinetobacter sp., a kind of ammonia-oxidizing bacteria, and compared the proposed method with the global Otsu's method the traditional watershed algorithm based on global thresholding value and human visual method. The result counted by the proposed method shows more approximated result to the human visual counting method than the result counted by any other method.
Peptides are formed in the rumen as the result of microbial proteinase activity. The predominant type of activity is cysteine ptoteinase, but others, such as serine proteinases, are also present. Many species of protozoa, bacteria and fungi are involved in ptoteolysis; large animal-to-animal variability is found when proteinase activities in different animals are compared. The peptides formed from proteolysis are broken down to amino acids by peptidases. Different peptides are broken down at different rates, depending on their chemical composition and particularly their N-terminal structure. Indeed, chemical addition to the N-terminus of small peptides, such as by acetylation, causes the peptides to become stable to breakdown by the rumen microbial population; the microorganisms do not appear to adapt to hydrolyse acetylated peptides even after several weeks exposure to dietary acetylated peptides, and the amino acids present in acetylated peptides are absorbed from the small intestine. The amino acids present in some acetylated peptides remain available in nutritional trials with rats, but the nutritive value of the whole amino acid mixture is decreased by acetylation. The genus Prevotella is responsible for most of the catabolic peptidase activity in the rumen, via its dipeptidyl peptidase activities, which release dipeptides rather than free amino acids from the N-terminus of oligopeptides. Studies with dipeptidyl peptidase mutants of Prevotella suggest that it may be possible to slow the rate of peptide hydrolysis by the mixed rumen microbial population by inhibiting dipeptidyl peptidase activity of Prevotella or the rate of peptide uptake by this genus. Peptides and amino acids also stimulate the growth of rumen microorganisms, and are necessary for optimal growth rates of many species growing on tapidly fermented substrates; in rich medium, most bacteria use pre-formed amino acids for more than 90% of their amino acid requirements. Cellulolytic species are exceptional in this respect, but they still incorporate about half of their cell N from pre-formed amino acids in rich medium. However, the extent to which bacteria use ammonia vs. peptides and amino acids for protein synthesis also depends on the concentrations of each, such that preformed amino acids and peptides are probably used to a much lesser extent in vivo than many in vitro experiments might suggest.
In this study, characteristics of dissolved organic matter (DOM) from Lake Paldang and seven other DOM sources (lake plankton, plants, soil, composite, treated sewage) were studied using XAD resin fractionation and 28-day microbial incubation experiment. Distribution patterns of DOM-fractions, which include hydrophilic acids (HiA), hydrophilic bases (HiB), hydrophilic neutrals (HiN), hydrophobic acids (HoA), hydrophobic neutrals (HoN) and the extent of DOM biodegradation (i.e., biodegradability) were different depending on the origins of the DOM samples. The DOM distribution pattern and the biodegradability were found to be effective for distinguishing the different DOM sources. The biodegradability (%) had negative correlations with the content (%) of hydrophobic fractions (Ho) and specific UV absorbance of DOM, which indicate that the Ho fractions contain more aromatic carbon structures and relatively stable during biodegradation, irrespective of the sources. To gain additional insight into the microbial transformation of the DOM, we also investigated the changes in the fraction's distribution for plankton, leaf litter and composite samples after the incubation. The results showed that biodegradation of hydrophilic fraction (Hi) causes an increase in the proportion of Ho (HoA, HoN), while biodegradation of HoA increases the HoN production.
