Evaluation of the Quality of Yogurt Using Ginseng Extract Powder and Probiotic Lactobacillus plantarum NK181

  • The objective of this study was to evaluate the composition, pH, titratable activity, microbial properties, and antioxidant effect of yogurt using ginseng extract powder (GEP), Lactobacillus plantarum NK181, and Streptococcus thermophilus as the starter culture. Different concentration of GEP (0%, 0.5%, 1%, 1.5%, and 2% (w/v)) were used in the yogurt. During yogurt fermentation, pH was decreased; however, titratable acidity and viable cell counts were increased. The addition of GEP to yogurt led to a decrease in moisture content and an increase in the fat, ash, and total solids content. The antioxidant effect using 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging, ${\beta}$-carotene bleaching, and ferric reducing antioxidant power (FRAP) assay gradually increased with added GEP. Overall, yogurt fermented with 1% GEP was acceptable in terms of cell viability and antioxidant effect. These results might provide information regarding development of ginseng dairy products with enhanced antioxidant activities and probiotic properties.

  • Septic arthritis of the temporomandibular joint (TMJ) is an uncommon disease caused by microbial pathogens through hematogenous infection, local spread, or iatrogenic infection. As the symptoms have an insidious onset, the early stage of septic arthritis is often confused with other diseases. A 49-year-old man was referred for increasing preauricular pain, swelling, and restricted mouth opening. He had been initially diagnosed as having a conventional temporomandibular joint disorder and trigeminal neuralgia and had been treated for the same. Imaging studies including panoramic view, lateral tomography, computed tomography with contrast enhancement, and magnetic resonance imaging were performed. Erosive bone change with displacement of the involved condyle, diffuse swelling of adjacent soft tissue, and fluid collection in the joint space were noted. Needle aspiration of the joint space and bacterial culture confirmed the diagnosis of septic arthritis of the TMJ and he was treated with antibiotic therapy and surgical drainage. Clinicians should always consider the diagnosis of septic arthritis of the TMJ in patients with preauricular pain or swelling.

Synthetic Biology Tools for Novel Secondary Metabolite Discovery in Streptomyces

  • Streptomyces are attractive microbial cell factories that have industrial capability to produce a wide array of bioactive secondary metabolites. However, the genetic potential of the Streptomyces species has not been fully utilized because most of their secondary metabolite biosynthetic gene clusters (SM-BGCs) are silent under laboratory culture conditions. In an effort to activate SM-BGCs encoded in Streptomyces genomes, synthetic biology has emerged as a robust strategy to understand, design, and engineer the biosynthetic capability of Streptomyces secondary metabolites. In this regard, diverse synthetic biology tools have been developed for Streptomyces species with technical advances in DNA synthesis, sequencing, and editing. Here, we review recent progress in the development of synthetic biology tools for the production of novel secondary metabolites in Streptomyces, including genomic elements and genome engineering tools for Streptomyces, the heterologous gene expression strategy of designed biosynthetic gene clusters in the Streptomyces chassis strain, and future directions to expand diversity of novel secondary metabolites.

Antimicrobial Activity of Essential Oil against Oral Strain

  • Objective: In this study, we tried to investigate the antimicrobial activity of natural herbaceous plant-derived essential oils against oral disease-causing bacterial strains and establish the scientific evidences to apply them to the dentifrice, mouthwash, and other uses. Methods: We used the disk diffusion method with 23 types of 100% natural essential oils to verify the antimicrobial effect against Streptococcus mutans, Porphyromonas gingivalis, and Lactobacillus rhamnosus. After adhering to the surface of the plate culture medium, $20{\mu}l$ of the essential oil was prepared, and the diameter of the transparent ring was measured after incubation for 24 hours. All the experiments were repeated three times. Results: Among 23 types of oils, 13 were effective against all three strains: myrrh, ginger, basil, carrot seed, tea tree, patchouli, ylang ylang, cypress, lemongrass, cinnamon, peppermint, lavender, and eucalryptus. Seventeen oils were effective against S. mutans, and myrrh, basil, and carrot seed showed high antimicrobial activity. Eighteen oils were effective against P. gingivalis, and tea tree, carrot seed, and cinnamons showed high antimicrobial activity. Sixteen oils were effective against L. rhamnosus, and carrot seed and peppermint cinnamon showed high antimicrobial activity. Conclusion: Consequently, 13 essential oils showed the antimicrobial activity against three bacterial strains, which indicates these essential oils might be used as the effective materials to suppress the growth of oral-disease inducing microorganisms. Moreover, essential oils that have been analyzed activities in this study will be mixed up within the constant range and analyzed their antimicrobial effects to examine the synergistic activity among them.

An Efficient Micropropagation to Obtain the Disease-free Bulbs from Scales for Cryopreservation in Lilium

  • Cryopreservation is one of the ideal and suitable methods for long-term storage of plant germplasm. The plant contaminated with diseases and pathogens are decreased the multiplication rate, survival rate and high quality of plants after cryopreservation. The aim of this work was to improve a micropropagation method for lily in Korea, which are cultivars and indigenous plant. In the last process of rinsing scales after surface-sterilization, we tried to control the diseases and pathogens lived within the tissue by rinsing in 0.03% sodium hypochlorite (NaClO) instead of sterile distilled water. Bulb scales of Lilium were cultured in vitro on MS medium supplemented with Plant Preservative Mixture (PPM). The newly small bulb formed from bulb-scales was transferred to MS medium. We checked the non-contamination and survival rate after 2 weeks in culture. Non-contamination was shown to be 70 to 90% in formed small bulbs. This study will help to mitigate microbial contamination in Lilium species micropropagation for cryopreservation.

The Interkingdom Interaction with Staphylococcus Influences the Antifungal Susceptibility of the Cutaneous Fungus Malassezia

  • The skin is a dynamic ecosystem on which diverse microbes reside. The interkingdom interaction between microbial species in the skin microbiota is thought to influence the health and disease of the skin although the roles of the intra- and interkingdom interactions remain to be elucidated. In this context, the interactions between Malassezia and Staphylococcus, the most dominant microorganisms in the skin microbiota, have gained attention. This study investigated how the interaction between Malassezia and Staphylococcus affected the antifungal susceptibility of the fungus to the azole antifungal drug ketoconazole. The susceptibility was significantly decreased when Malassezia was co-cultured with Staphylococcus. We found that acidification of the environment by organic acids produced by Staphylococcus influenced the decrease of the ketoconazole susceptibility of M. restricta in the co-culturing condition. Furthermore, our data demonstrated that the significant increased ergosterol content and cell membrane and wall thickness of the M. restricta cells grown in the acidic environment may be the main cause of the altered azole susceptibility of the fungus. Overall, our study suggests that the interaction between Malassezia and Staphylococcus influences the antifungal susceptibility of the fungus and that pH has a critical role in the polymicrobial interaction in the skin environment.

The Antibacterial Properties of Filtrates from Chinese Cabbage Kimchi

  • Lactobacillus plantarum and Leuconostoc mesenteroides are crucial functional starters and predominant isolates in a wide range of fermented foods, particularly kimchi, whose constituents exhibit bioactive properties. We previously developed a methodology using anion exchange resins to purify peptidyl compounds from Lb. plantarum LBP-K10. Antibacterial cultures of Lb. plantarum LBP-K10 were obtained from the respective cultures' supernatants and filtrates. However, conclusive evidence of the efficacy of kimchi filtrates in eradicating pathogenic bacteria is lacking. We aimed to simulate the potential effects of antibacterial filtrates that contained antibacterial compounds which were derived from cultures of Lb. plantarum LBP-K10. We acquired the kimchi filtrates using a combination of centrifugation and filtration methodologies, without the requirement for inoculation. The filtered liquid from Chinese cabbage kimchi, inoculated with Lb. plantarum LBP-K10 as a starter culture, and the non-inoculated liquid from Chinese cabbage kimchi (referred to as CCK and CCKRef, respectively) were were examined. CCK demonstrated greater inhibitory activity and a more significant bactericidal effect against the bacterial indicator strains. The minimum inhibitory concentration demonstrated comparable outcomes in tests against both Gram-positive and Gram-negative bacteria. This research offers a groundbreaking examination that displays the effectiveness of profiling peptidyl compounds within kimchi filtrates for curing bacterial infections.

닭 우모 분해세균 Elizabethkingia meningoseptica CS2-1이 생산하는 단백질분해효소의 특성 및 아미노산 생산을 위한 배양조건 (Characterization of Protease Produced by Elizabethkingia meningoseptica CS2-1 and Optimization of Cultural Conditions for Amino Acid Production)

  • 우모분으로부터 높은 함량의 아미노산을 생성하는 Elizabethkingia meningoseptica CS2-1 (729.7 ${\mu}mol/mL$)를 선발하였다. 선발 균주의 protease 활성과 우모 분해능을 위한 최적배지 및 배양조건을 확립하고 우모 분해산물 아미노산의 특성을 조사하였다. E. meningoseptica CS2-1의 최적 배양조건은 $25^{\circ}C$, pH 7.5, 180 rpm이었으며, 효소는 pH 8.0, $40^{\circ}C$에서 가장 높은 활성을 나타내었다. E. meningoseptica CS2-1의 아미노산 생산을 위한 최적 배지조건은 $NH_4Cl$ 0.05%, NaCl 0.05%, $K_2HPO_4$ 0.03%, $KH_2PO_4$ 0.03%, $MgCl_2{\cdot}6H_2O$ 0.01%, urea 0.1%, 닭 우모분 2%이었다. E. meningoseptica CS2-1를 최적 배지 및 배양조건에서 배양한 결과, 아미노산의 총 함량은 1063 ${\mu}mol/mL$로 기본조건 (729.7 ${\mu}mol/mL$)보다 46%로 향상되었고 필수 아미노산 함유량은 315.9 ${\mu}mol/mL$로 기본조건 (219.3 ${\mu}mol/mL$)보다 44%로 향상되었다. 닭 우모분으로부터 추출되 는 17 종류의 아미노산 중 proline (14%), aspartic acid (12%), serine (10%), alanine (10%), glycine (9%), tyrosine (7%)이 주요 아미노산으로 추출되는 특징을 나타내었다. 따라서, E. meningoseptica CS2-1은 닭 우모로부터 아미노산 생산을 위한 미생물유전자원 소재로서 잠재적 가치가 클 것으로 판단된다.

라이보좀 RNA 염기서열 분석을 이용한 집식배양된 식물추출물발효음료의 미생물 다양성 (Microbial Diversity in the Enrichment Cultures from the Fermented Beverage of Plant Extract Using Ribosomal RNA Sequence Analysis)

  • 식물추출발효음료(fermented beverage of plant extract, FBPE)는 과일류, 야채류, 약초류, 및 해조류를 설탕에 의한 당장법으로 발효시킨 액상발효음료이다. 본 연구에서는 FBPE의 이화학적 특성과 16S 및 26S rRNA 염기서열을 이용하여 집식배양된 FBPE의 미생물 다양성을 조사하였다. FBPE의 pH는 3.48, 산도는 1.68%, 당도는 $70^{\circ}$, 환원당은 1,026 g/L 및 알코올은 3.5%이었다. 유리당과 유기산은 glucose (567.83 g/L)와 tartaric acid (936.8 mg/L)로 나타났다. Lactobacillus homohiochii는 모든 집식배양 시료에서 우점종이었고 L. fructivorans는 20B 라이브러리에서만 나타났다. 세 가지의 다른 당 농도에서 집식배양된 FPEB의 우점종은 0Y 라이브러리(설탕농도 0%)에서는 Candida zeylanides (45.2%), 20Y 라이브러리(설탕농도 20%)에서는 C. lactis-condensi (35.7%)와 C. zeylanoides (35.7%) 및 40Y 라이브러리(설탕농도 40%)에서는 C. lactis-condensi (38.1%)이었다. 이외에 0Y 라이브러리에서는 C. lactis-condensi (40.5%), Pichia farinosa (7.1%), C. parapsilosis(4.8%) 및 Zygosaccharomyces bailii (2.4%)가 나타났으며, 20Y 라이브러리에서는 P. guilliermondii (14.3%), Pichia farinosa (7.1%), C. parapsilosis (4.8%) 및 Kazachstania exigua (2.4%)로 나타났고 40Y 라이브러리에서는 C. zeylanoides (30.9%), C. parapsilosis (11.9%), Z. bailii (11.9%), Pichia farinosa (4.8%), P. guilliermondii (2.4%)이 확인되었다. 이 결과는 앞으로 FBPE의 미생물 변화 연구에 아주 유용한 자료로 제공할 수 있다.

천연항균제를 사용한 냉장떡류의 유통 안정성 평가연구 (A Study on the Evaluation of Distribution Stability of Refrigerated Rice Cake Using Natural Antibiotics)

  • 냉장유통을 목적으로 하는 떡류의 저장안전성을 높이기 위하여, 1차로 공정의 변화, 2차로 천연항균성 물질을 사용하여 미생물을 억제하고자 하였다. 사용한 떡류로는 녹두깨찰떡과 통팥찰떡을 사용하였고, 천연항균제로서는 자몽종자추출물, 포도씨추출물, 복합황금추출물, polylysine, chitosan, ethylpyrubate 및 glycine을 사용하였다. 액상의 천연추출물은 0, 0.25, 0.5, 1, 2, 4% 농도까지 조정하여 미생물 변화를 확인하였고 냉장온도를 위해 4, 8, $12^{\circ}C$ 측정하였다. 자몽추출물의 경우 $4^{\circ}C$ 저장에서 모든 농도에서 미생물 안전성이 충분히 유지되는 것을 확인하였으나 냉장온도 중상온에 해당하는 $8^{\circ}C$, 21일 저장에서 이미 $10^5cfu/g$를 넘어 기준에 적합하지 않은 것을 확인하였고, $12^{\circ}C$ 저장에서는 자몽추출물의 농도가 2%에서 급격히 세균수가 안정화를 이루는 것으로 보여 이 농도 또한 미생물 항균활성이 유지되는 것을 확인하였다. 포도씨추출물의 경우 $8^{\circ}C$ 배양에서 4%에서만 최종 21일 배양에서 식품유통 안전기한의 요구조건을 충족하였고, $12^{\circ}C$에서는 4% 농도에서 18일까지만 안전기준을 만족하였다. 복합황금추출물의 농도별 처리에 따른 일반 총균수를 보여주고 있는데 저온($4^{\circ}C$)에서 안정적으로 미생물의 유통안전이 유지되는 것을 확인할 수 있었으나, $8^{\circ}C$, $12^{\circ}C$에서는 다른 처리 시료와 동일하게 유통한 계치를 넘었다. Polylysine을 녹두깨 찰떡에 적용한 경우 $4^{\circ}C$에서 무첨가군을 제외한 모든 군에서 활성을 나타내는 경향을 보여주고 있으나 $8^{\circ}C$$12^{\circ}C$에서는 그러한 특성들이 보이지 않았다. Cchitosan의 농도 0.5% 이상에서는 매우 안정적으로 미생물의 생육이 억제되는 것으로 보여지고, 8, $12^{\circ}C$에서도 유사한 경향성이 보이고 있어 미생물을 억제하는 항균성 소재로서 가능성이 있을 것으로 보이나, $12^{\circ}C$에서는 모두 일반세균수에 대한 유통기준을 상회하였다. Ethyl-pyruvate는 $4^{\circ}C$, 1% 농도에서 미생물 안전이 유지되는 것을 보이고 있고 이후의 2, 4%에서도 식품으로서의 안전이 안정적으로 유지되는 것을 알 수 있다. Glycine을 0.25~4%까지 농도를 달리하여 4, 8, $12^{\circ}C$의 냉장조건에서 성장억제 현상을 관찰하였는데 저온인 $4^{\circ}C$에서는 매우 우수하게 효과적으로 유통안전성이 유지되는 것을 확인하였다. 그러나 $8^{\circ}C$, $12^{\circ}C$에서는 다른 항균활성물질의 첨가와 마찬가지로 14일의 유통기한을 유지하지 못하였다.
