Change of Sludge Consortium in Response to Sequential Adaptation to Benzene, Toluene, and o-Xylene

  • Activated sludge was sequentially adapted to benzene, toluene, and o-xylene (BTX) to study the effects on the change of microbial community. Sludge adapted to BTX separately degraded each by various rates in the following order; toluene>o-xylene>benzene. Degradation rates were increased after exposure to repeated spikes of substrates. Eleven different kinds of sludge were prepared by the combination of BTX sequential adaptations. Clustering analyses (Jaccard, Dice, Pearson, and cosine product coefficient and dimensional analysis of MDS and PCA for DGGE patterns) revealed that acclimated sludge had different features from nonacclimated sludge and could be grouped together according to their prior treatment. Benzene- and xylene-adapted sludge communities showed similar profiles. The sludge profile was affected from the point of the final adaptation substrate regardless of the adaptation sequence followed. In the sludge adapted to 50 ppm toluene, Nitrosomonas sp. and bacterium were dominant, but these bands were not dominant in benzene and benzene after toluene adaptations. Instead, Flexibacter sp. was dominant in these cultures. Dechloromonas sp. was dominant in the culture adapted to 50 ppm benzene. Thauera sp. was the main band in the sludge adapted to 50 ppm xylene, but became vaguer as the xylene concentration was increased. Rather, Flexibacter sp. dominated in the sludge adapted to 100 ppm xylene, although not in the culture adapted to 250 ppm xylene. Two bacterial species dominated in the sludge adapted to 250 ppm xylene, and they also existed in the sludge adapted to 250 ppm xylene after toluene and benzene.

  • 생물학적 복구법의 효율은 미생물 균수의 계수, 호흡율, 분해율 등으로 측정할 수 있으며 부작용 시험은 Daphnia,굴의 유충, 무지개 송어 를 사용하여 측정된다. On-Site처리에 있어서는 산소 전달이 문제가 되는데 이에는 과산화 수소가 사용될수 있으며(벤젠계통의 용매), 유류오염 방제시에는 친유화적 질소 및 인을 첨가할 수 있다. 접종균주는 혼합균주 또는 순수 균주가 사용되는데 후자에는 Pseudomonas와 Phaneochate 등이 있다. 때로는 효울을 높이기 위해 효소를 첨가 하거나 광분해법과 병용되기도 한다. 토양의 유류오염물질, 예컨대 폐윤활유, 기계유, 오일 슬러지들을 처리할때는 토양상부 15∼20cm를 갈고 유류오염물질을 5%의 농도로 주입한다. 이 때 적정 pH는 7∼8, 탄화수소 질소 = 100 : 1, 탄화수소 : 인 = 800 : 1이며 적절한 배수가 필요하다. 지하 유류오염물질의 처리에 있어서는 특히 다량의 산소가 필요하며 해양유류오염물질의 처리에 있어서는 친유화성 비료를 첨가하면 효과가 있다 생물학적 복구법에 의해 대기오염물질 특히, 악취 물질을 처리할 수 있는데 반응기로는 생물여과법과 생물세정기가 쓰인다.

Effects of Pumpkin (Cucurbita moschata Duch.) Leaf Ethanolic Extracts on Lipid Oxidation and Microbial Activity in Refrigerated Raw Ground Pork

  • This study was conducted to evaluate the antioxidant effects of pumpkin leaf extracted using a 50% ethanol on ground pork during storage. The pumpkin leaf extracts were added at concentrations of 0.05 (PE-0.05), 0.1 (PE-0.1), and 0.2% (PE-0.2) to ground pork, and 0.05% of ascorbic acid (As-0.05) was added as a control. Each sample was collected after 1, 4, 7, and 10 d of storage and the pH, total viable counts (TVC), conjugated dienes (CD), free fatty acids (FFA), and thiobarbituric reaction substance (TBARS) values were measured. The pH of the pork samples decreased until day 7, and then increased thereafter, except for the control and PE-0.05 sample. Lower CIE $a^*$ values were observed for pork samples containing PE relative to As-0.05 at increasing storage time (p<0.05). The addition of PE decreased the TVC, CD, FFA and TBARS values levels in the ground pork when compared to the control during 10 d of storage. These results indicate that PE can produce notable effects on meat products, such as inhibiting lipid oxidation and discoloration.

Effects of two litter amendments on air NH3 levels in broiler closed-houses

  • Objective: High $NH_3$ emissions from poultry houses are reported to have negative impacts on health, welfare and safety of birds and humans, and on the environment. Objective of the present study was to determine the effects of two litter amendments on the $NH_3$ levels in broiler closed houses under hot-humid conditions. Methods: Giving a completely randomize design, nine closed houses, each housed 32,500 birds on paddy husk litter, were randomly allocated into two treatment (Mizuho; a bacterial culture mix and Rydall OE; an enzymatic biocatalyst) and control groups. $NH_3$ levels were determined thrice a day (0600, 1200, and 1800 h), at three heights from the litter surface (30, 90, and 150 cm), at 20 predetermined locations of a house, from day 1 to 41. Results: Rydall significantly reduced the $NH_3$ level compared to control and Mizuho. $NH_3$ levels at 30 cm were significantly higher than that of 90 and 150 cm. The $NH_3$ levels at 30 cm height were higher than 25 ppm level from day 9, 11, and 13 in Mizuho, control, and Rydall groups, respectively to day 41. $NH_3$ levels at 150 cm height were higher than maximum threshold limit of 50 ppm for human exposure from day 12, 14, and 15 in Mizuho, control, and Rydall groups, respectively to day 33. Being significantly different among each other, the $NH_3$ level was highest and lowest at 0600 and 1800 h. Litter amendments had no significant effects on growth performance. Rydall significantly increased the litter N content on day 24. Conclusion: It was concluded that the $NH_3$ levels of closed house broiler production facilities under tropical condition are so high that both birds and workers are exposed to above recommended levels during many days of the growing period. Compared to microbial culture, the enzymatic biocatalyst was found to be more effective in reducing $NH_3$ level.

Culture-Independent Analysis of Microbial Succession During Composting of Swine Slurry and Mushroom Cultural Wastes

  • Bacterial diversity and the composition of individual communities during the composting process of swine and mushroom cultural wastes in a field-scale composter (Hazaka system) were examined using a PCR-based approach. The composting process was divided into six stages based on recorded temperature changes. Phylogenetic analysis of eighty 16S rRNA sequences from uncultured composting bacterial groups revealed the presence of representatives from three divisions, including plant pathogenic bacteria, high-molecule-degrading bacteria and spore-forming bacteria. The plant pathogen A. tumefaciens gradually decreased in abundance during the composting process and eventually disappeared during the thermophilic and cooling stage. A bacterium homologous to Bacillus humi first appeared at the early thermophilic stage and was established at the intermediate thermophilic, post-thermophilic, and cooling stages. It was not possible to isolate the B. humi during any of the stages using general culture techniques.

Enhanced Production of Human Serum Albumin by Fed-Batch Culture of Hansenula polymorpha with High-Purity Oxygen

  • Fed-batch cultures of Hansenula polymorpha were studied to develop an efficient biosystem to produce recombinant human serum albumin (HSA). To comply with this purpose, we used a high-purity oxygen-supplying strategy to increase the viable cell density in a bioreactor and enhance the production of target protein. A mutant strain, H. polymorpha GOT7, was utilized in this study as a host strain in both 5-l and 30-l scale fermentors. To supply high-purity oxygen into a bioreactor, nearly 100% high-purity oxygen from a commercial bomb or higher than 93% oxygen available in situ from a pressure swing adsorption (PSA) oxygen generator was employed. Under the optimal fermentation of H. polymorpha with highpurity oxygen, the final cell densities and produced HSA concentrations were 24.6 g/l and 5.1 g/l in the 5-l fermentor, and 24.8 g/l and 4.5 g/l in the 30-l fermentor, respectively. These were about 2-10 times higher than those obtained in air-based fed-batch fermentations. The discrepancies between the 5-l and 30-l fermentors with air supply were presumably due to the higher contribution of surface aeration over submerged aeration in the 5-l fermentor. This study, therefore, proved the positive effect of high-purity oxygen in enhancing viable cell density as well as target recombinant protein production in microbial fermentations.

Purification and Characterization of the Fibrinolytic Enzyme Produced by Bacillus subtilis KCK-7 from Chungkookjang

  • A fibrinolytic enzyme has been found in several bacteria isolated from fermented food. This study was carried out to investigate the purification and characteristics of the fibrinolytic enzyme produced by Bacillus subtilis KCK-7 originated from Chungkookjang. The fibrinolytic enzyme was purified to homogeneity from the culture supernatant using ammonium sulfate fractionation and chromatographies on DEAE-cellulose and on Sephadex G-100. The final specific activity of the purified enzyme increased 11.0-fold, and the protein amount in the purified enzyme was about 16% of that in the culture supernatant. The molecular weight of the purified enzyme was estimated to be about 45,000 by SDS-PAGE. The optimum pH and temperature for the enzyme activity were pH 7.0 and $60^{\circ}C$, respectively. The enzyme activity was relatively stable up to $60^{\circ}C$ over the pH range of 7.0-10.0. The fibrinolytic enzyme activity increased by $Ca^{2+}$ and $Cu^{2+}$, whereas it was inhibited by $Hg^{2+}$ and $Ba^{2+}$. In addition, it was severely inhibited by PMSF and DFT. It is suggested that the purified enzyme was a serine protease for the fibrinolysis. The purified enzyme could completely hydrolyze fibrin in vitro within 8 h. Hence, it is suggested that the purified enzyme can be put into practice as an effective thrombolytic agent.

Characterization of the Biodiversity of the Spoilage Microbiota in Chicken Meat Using Next Generation Sequencing and Culture Dependent Approach

  • This study investigated the psychrotrophic bacteria isolated from chicken meat to characterize their microbial composition during refrigerated storage. The bacterial community was identified by the Illumina MiSeq method based on bacterial DNA extracted from spoiled chicken meat. Molecular identification of the isolated psychrotrophic bacteria was carried out using 16S rDNA sequencing and their putrefactive potential was investigated by the growth at low temperature as well as their proteolytic activities in chicken meat. From the Illumina sequencing, a total of 187,671 reads were obtained from 12 chicken samples. Regardless of the type of chicken meat (i.e., whole meat and chicken breast) and storage temperatures ($4^{\circ}C$ and $10^{\circ}C$), Pseudomonas weihenstephanensis and Pseudomonas congelans were the most prominent bacterial species. Serratia spp. and Acinetobacter spp. were prominent in chicken breast and whole chicken meat, respectively. The 118 isolated strains of psychrotrophic bacteria comprised Pseudomonas spp. (58.48%), Serratia spp. (10.17%), and Morganella spp. (6.78%). All isolates grew well at $10^{\circ}C$ and they induced different proteolytic activities depending on the species and strains. Parallel analysis of the next generation sequencing and culture dependent approach provides in-depth information on the biodiversity of the spoilage microbiota in chicken meat. Further study is needed to develop better preservation methods against these spoilage bacteria.

마늘첨가가 김치의 숙성 및 품질에 미치는 영향 (Effect of the Garlic on the Fermentation and Quality of Kimchi)

  • 마늘이 김치의 숙성과 품질에 미치는 영향을 조사하고 아울러 cyclodextrin과 sorbitol의 마늘냄새 masking 효과를 실험하였다. 김치 숙성 초기의 산도는 마늘 첨가량이 많을수록 대조구 비해서 높았고 환원당 함량은 마늘 첨가량이 많을수록 높았으나 숙성말기에는 거의 비슷한 수준을 유지하였다. 미생물수의 경우 숙성초기에는 마늘 첨가에 의해 성장이 촉진되었으나 이후에 그 수준을 유지한 후에는 대조구에 비해서 오히려 억제되었다. 관능검사 결과는 온도에 관계없이 마늘을 첨가하지 않은 김치가 모든 관능적인 면에서 유의적으로 나쁘게 평가되었으며 마늘을 $2{\sim}3%$ 첨가한 김치가 유의적으로 가장 좋게 평가되었다. Cyclodextrin 또는 sorbitol을 1%씩 첨가한 결과 마늘맛을 masking하는데는 효과가 없었고 관능적으로도 나쁘게 평가되었다.

Effect of Chlorine Dioxide and Commercial Chlorine Sanitizer on Inhibiting Foodborne Pathogens and on Preventing the Formation of Chemically Injured Cells on Radish Sprouts

  • This study assessed the efficacy of aqueous chlorine dioxide ($ClO_2$) and commercial chlorine sanitizer in terms of its ability to eliminate Listeria monocytogenes, Salmonella Typhimurium, and Escherichia coli O157:H7 on radish sprouts (Raphanus sativus L.). Radish sprouts were inoculated with a cocktail containing one each of three strains of three different foodborne pathogens, then treated with distilled water (control) or chemical sanitizers (100 ppm commercial chlorine, and 50, 100, 200 ppm $C1O_2$) for 1, 5, and 10 min at room temperature ($22{\pm}2^{\circ}C$). Populations of S. Typhimurium, E. coli O157:H7 and L. monocytogenes were counted at 4.64, 6.05, and 4.29 log CFU/g, respectively, after inoculation. Treatment with water did not significantly reduce the levels of any of the three foodborne pathogens. The levels of all three pathogens were reduced by treatment with chemical sanitizers; however, the observed levels of reduction of E. coli O157:H7 and L. monocytogenes were not significant as compared with the controls. The levels of the three pathogens were reduced most profoundly when treated for 10 min with 200 ppm of $C1O_2$, and the reduction levels of S. Typhimurium, E. coli O157:H7, and L. monocytogenes were 1.17, 1.63, and 0.96 log CFU/g, respectively. When chemically injured cells were investigated using SPRAB for E. coli O157 :H7 and by selective overlay methods for S. Typhimurium and L. monocytogenes, respectively, it was noted that commercial chlorine sanitizer generated more numbers of injured pathogens than did $C1O_2$. These data indicate that $C1O_2$ treatment may prove useful in reducing the numbers of pathogenic bacteria in radish sprouts.
