Histomorphometric Analysis of the Spine and Femur in Ovariectomized Rats Using Micro-Computed Tomographic Scan

  • Objective : The purpose of this study was to evaluate the different patterns of bone loss between the lumbar spine and the femur after ovariectomy in rats. Methods : Twenty-four female Sprague-Dawley rats underwent a sham operation (the sham group) or bilateral ovariectomy (the ovariectomized group). Four and eight weeks after operation, six rats from each of the two groups were euthanized. Serum biochemical markers of bone turnover including osteocalcin and alkaline phosphatase (ALP), which are sensitive biochemical markers of bone formation, and the telopeptide fragment of type I collagen C-terminus (CTX), which is a sensitive biochemical marker of bone resorption, were analyzed. Bone histomorphometric parameters of the 4th lumbar vertebrae and femur were determined by micro-computed tomography. Results : Ovariectomized rats were found to have higher osteocalcin, ALP and CTX levels than sham controls. Additionally, 8 weeks after ovariectomy in the OVX group, serum levels of osteocalcin, ALP and CTX were significantly higher than those of 4 weeks after ovariectomy. Bone loss after ovariectomy was more extensive in the 4th lumbar spine compared to the femur. Bone loss in the 4th lumbar spine was mainly caused by trabecular thinning, but in the femur, it was mainly caused by trabecular elimination. Conclusion : The present study demonstrates different patterns of bone loss between the 4th lumbar spine and the femur in ovariectomized rats. Therefore, when considering animal models of osteoporosis, it is important that bone sites should be taken into account.

Micro-CT Analysis of Cranial Bone and Tooth Density in Mice Deficient for GDF11 or Myostatin

  • Purpose: Growth differentiation factor 11 (GDF11) and myostatin (MSTN) are closely-related transforming growth factor β family members reported to play crucial roles in bone formation. We previously reported that, in contrast to MSTN, GDF11 promotes osteogenesis of vertebrae and limbs. GDF11 has been also reported as an important regulator in tooth development by inducing differentiation of pulp stem cells into odontoblasts for reparative dentin formation. The goal of this study was to investigate the differential roles of GDF11 and MSTN in dental and cranial bone formation. Methods: Micro-computed tomography analysis was performed on cranial bones, including frontal, parietal, and interparietal bones, and lower incisors of wild-type, Gdf11 knockout (Gdf11-/-), and Mstn knockout (Mstn-/-) mice. Tissue volume, thickness, and mineral density were evaluated for both cranial bone and lower incisors. Lower incisor lengths were also measured. Because Gdf11-/- mice die shortly after birth, analysis was performed on newborn (P0) mice. Results: Compared to those of Mstn-/- mice, cranial bone volume, thickness, and mineral density levels were all significantly diminished in Gdf11-/- mice. Tissue mineral density of Gdf11-/- mice were also significantly decreased compared to wild-type mice. Likewise, lower incisor length, tissue volume, thickness, and mineral density levels were all significantly reduced in Gdf11-/- mice compared to Mstn-/- mice. Incisor length was also significantly decreased in Gdf11-/- mice compared to wild-type mice. Mstn-/- mice exhibited mildly increased levels of tissue volume, thickness, and density in cranial bone and lower incisor compared to wild-type mice although statistically not significant. Conclusions: Our findings suggest that GDF11, unlike MSTN, endogenously promotes cranial bone and tooth development.

Morphological characteristics of the mesiobuccal root in the presence of a second mesiobuccal canal: a micro-CT study

  • Objectives: This study investigated the internal morphology of mesiobuccal (MB) roots of maxillary molars with a second mesiobuccal (MB2) canal. Materials and Methods: Forty-seven maxillary first or second molars from Brazilians were scanned using micro-computed tomography. The following measurements were obtained from the MB roots: root thickness, root width, and dentin thickness of the buccal aspect of the first mesiobuccal (MB1) canal, between the MB1 and MB2 canals, and the palatal aspect of the MB2 and MB1 canals at 3 mm from the root apex and in the furcation region. For statistical analysis, the Student's t-test and analysis of variance with the post-hoc Tukey test were used (α = 0.05). Results: In maxillary molars with an MB2 canal, MB roots were significantly thicker (p = 0.0014) and narrower (p = 0.0016) than in maxillary molars without an MB2 canal. The dentin thickness of the palatal aspect of the MB1 canal was also significantly greater than that of MB roots without an MB2 canal at 3 mm from the root apex (p = 0.0007) and in the furcation region (p < 0.0001). In the furcation region of maxillary molars with an MB2 canal, the dentin thickness between the MB1 and MB2 canals was significantly smaller than that in the buccal and palatal aspects (p < 0.0001). Conclusions: The internal morphology of MB roots of maxillary molars with an MB2 canal revealed differences in dentin thickness, root diameter, and distance between the canals when compared with maxillary molars without an MB2 canal.

성장 중인 쥐에서 음식물의 경도가 하악 과두의 해면골에 미치는 영향 : 미세전산화 단층촬영을 이용한 연구 (THE EFFECTS OF DIETARY CONSISTENCY ON THE TRABECULAR BONE ARCHITECTURE IN GROWING MOUSE MANDIBULAR CONDYLE : A STUDY USING MICRO-CONFUTED TOMOGRAPHY)

  • 하악 과두의 발달과 증식은 측두하악관절 부위의 생역학적 환경의 변화에 따라 변경될 수 있다. 이 부위에 전달되는 생역학적 하중은 섭취하는 음식물의 경도를 다르게 함으로써 변화시킬 수 있다. 이번 연구의 목적은 성장 중인 쥐에서 부드러운 음식물의 섭취에 의해 저작력을 변화시키는 것이 하악 과두의 해면골의 형태를 변화시킬 수 있는지 미세전산화 단층촬영을 이용하여 분석하는 것이었다. 생후 21일 된 C57BL/6 쥐 36마리를 무작위로 두 그룹으로 나누었다. 8주 동안 대조군의 쥐들은 일반적인 딱딱한 덩어리의 사료를, 실험군의 쥐들은 덩어리의 사료를 잘게 갈은 후 물과 섞어 부드럽게 만들어 먹였다. 또한 실험군의 쥐들의 하악 절치를 일주일에 두 번씩 잘라서 짧게 만들었다. 8주 후 모든 쥐들을 희생하여 우측 하악 과두를 준비하였다. 미세전산화 단층촬영과 삼차원 영상 분석프로그램을 이용하여 하악 과두 해면골의 bone volume(BV), bone surface(BS), total volume(TV) bone volume fraction(BV/TV), surface to volume ratio(BS/BV), trabecular thickness(Tb. Th.), structure model index(SMI)와 degree of anisotropy(DA)를 측정하고 이들 값으로부터 trabecular number(Tb. N.)와 trabecular separation (Tb. Sp.)을 계산하였다 미세전산화 단층영상을 얻은 후 하악 과두의 조직 표본을 만들었다. 연구 결과는 다음과 같았다. 1. Bone volume fraction(BV/TV), trabecular thickness(Tb. Th.)와 trabecular number(Tb. N.)가 대조군에 비해 부드러운 음식을 먹인 실험군에서 유의하게 감소되었다(p<0.05). 2. Trabecular separation(Tb. Sp.)은 부드러운 음식을 먹인 실험군에서 유의하게 증가하였다(p<0.05). 3. Surface to volume ratio(BS/BV), structure model index(SMI), degree of anisotropy (DA)는 두 군 사이에 유의한 차이를 보이지 않았다(p>0.05). 4. 조직 절편을 관찰한 결과, 부드러운 사료를 먹인 실험군에서 하악 과두의 연골층의 증식 층과 전체 두께가 상당히 감소하였다.

Real-Time PCR과 Internal Standard Addition법을 이용한 돼지고기 소시지에 혼합된 닭고기의 정량 (Use of Real-Time PCR and Internal Standard Addition Method for Identifying Mixed Ratio of Chicken Meat in Sausages)

  • 본 연구는 돼지고기로 제조된 소시지에 저가원료인 닭고기 혼합비율을 정량하기 위해 real-time PCR 법과 소량 함유된 닭의 정량의 정확도를 높이기 위해 닭의 내부 표준물질을 첨가하는 방법을 적용함으로써 소시지의 진위판별 가능성을 제시하였다. DNA 회수율과 PCR 증폭효율을 높이기 위해 QIAamp DNA Micro Kit을 사용하였고, 최종 PCR 반응액 $20{\mu}L$에 template DNA($5{\sim}10ng/{\mu}L$)는 $3.0{\sim}5.0{\mu}L$, primer 농도는 $0.5{\mu}L(10pmol)$, $2{\times}$ Cybrgreen buffer는 $10{\mu}L$로 조정하였을 때 가장 적합하였고, PCR 증폭조건은 annealing 온도를 $62^{\circ}C$, extension 온도를 $68^{\circ}C$, final extension 시간은 33초, 최종 PCR cycle은 40 cycle로 했을 때 가장 PCR 증폭효율이 좋은 것으로 평가되었다. 돼지와 닭의 DNA를 50 ng에서 0.05 ng으로 순차적으로 10배씩 희석한 template DNA를 이용해 민감도(최소 검출한계)를 확인한 결과 돼지와 닭 모두 0.05 ng 이상으로 각각 확인되었다. 표준곡선의 결정계수($R^2$)도 모두 0.995 이상으로 표준곡선의 linearity가 정량에 적합한 것으로 확인되었다. 돼지고기와 닭고기를 각각 70:30 비율로 혼합한 3점의 시료를 $70^{\circ}C$에서 10분간 열처리한 후 정량분석을 실시하여 기대치에 의한 측정치를 비교한 결과, 64.3:35.7의 비율로써 평균 5.7%의 차이를 나타내 생육(raw meat) 또는 가열처리된 시료의 상태에 따라 DNA에 영향을 주어 PCR 증폭효율 및 DNA 정량 값에 일부 영향을 주는 것으로 판단되었다. 소시지에 함유된 돼지고기와 닭고기의 함량을 분석한 결과 돼지고기에 비해 닭고기 함량이 적은 소시지에서는 닭고기(6%, 12%, 25%, 38%)의 검출이 어려웠고, Ct(threshold cycle) 값에 따른 DNA 정량값이 매우 낮아 배합비율을 환산이 어려웠다. 그러나 소시지에 닭의 표준물질 DNA(50 ng)를 첨가함으로써 배합비율이 증가할수록 Ct값도 점차 낮아져서 배합비율을 반영하고 있음에 따라 Ct값의 평균치${\pm}$오차범위 값으로 간접적으로 배합비율을 추정할 수 있을 것으로 생각되었다.

Deterioration of tensile behavior of concrete exposed to artificial acid rain environment

  • This study is focused on evaluation of the tensile properties of concrete exposed to acid rain environment. Acid rain environment was simulated by the mixture of sulfate and nitric acid in the laboratory. The dumbell-shaped concrete specimens were submerged in pure water and acid solution for accelerated conditioning. Weighing, tensile test, CT, SEM/EDS test and microanalysis were performed on the specimens. Tensile characteristics of the damaged concrete are obtained quantitatively. Evolution characteristics of the voids, micro cracks, chemical compounds, elemental distribution and contents in the concrete are examined. The deterioration mechanisms of concrete exposed to acid rain are well elucidated.

Electrospinning으로 제조된 PMMA/PVA Multilayer bone plate의 생체적합성에 관한 연구

  • Bone plate는 골절된 뼈의 골 유합을 지지하기 위해서 정형외과, 신경외과, 성형외과 및 치과 등에서 널리 사용되고 있다. 하지만 기존의 bone plate는 대부분 금속으로 제작되어 있어 장기간 이식에 따른 부식 및 천연골 강도저하 등으로 인해 1~2년 후 재수술을 해야 하는 문제점을 갖고 있다. 본 연구에서는이런 금속 bone plate의 단점을 개선하고자 생체적합성이 우수한 생분해성 고분자 bone plate를 제작하였다. 사용된 고분자는 생체적합성과 생분해성이우수한 PVA(polyvinly alcohol)와 강도를 유지하기 위한 PMMA(poly methyl methacrylate)를 사용였다. Electrospinning 법으로 PVA와 PMMA fibrous mat를 제작하여 각 mat를 적층시킨 후 열압착을 하여 강도를 증가시킨 PMMA/PVA Mutlilayer bone plate을 제작하였다. 제작된 PMMA/PVA Mutlilayer bone plate의 생체적합성 평가를 위해 MTT assay, 생분해 특성을 관찰하기 위해 Micro-CT와 SBF(simulated body fluid) 내에서의 용해도를 관찰하였다. 또한조골세포의 부착과 분화에 미치는 영향을 SEM(scanning electron microscope)을통해 관찰하였고, 조골세포의 유전자 발현에 미치는 영향을 RT-PCR을통해 확인하였다.

척추 골절의 예방적 치료법에 관한 미세 구조해석 (A Microstructural Analysis for Preventive Treatments of Vertebral Fracture)

  • It is reported that the mechanical properties of vertebral trabecular bone depend on the density and the mass of bones. Osteoporosis is a systemic skeletal disease caused by low bone mass and microstructure deterioration of trabecular bone. Silva and Gibson (1997) studied the treatment of age-related bone loss using drug therapy. Vertebroplasty is a minimally invasive surgery for the treatment of osteoporosis vertebrae. This procedure includes puncturing vertebrae and filling with Polymethylmethacrylate (PMMA). However, the relative effect of drug therapy and bone cement for osteoporosis treatment is not reported yet. In this study, several 2D models of human vertebral trabecular bone are analyzed by finite element method. The mechanical behaviors of the vertebral trabecular bone treated by the drug therapy and the bone cement are compared. This study shows that bone cement treatment is more effective strategy than drug therapy to prevent the degradation of bone strength.

유한요소해석을 이용한 하중조건에 따른 대퇴골의 거동예측 (Predicted the behavior of the femur according to the loading condition using FEM)

  • Falling related injuries are categorized as the most serious and common medical problems experienced by the elderly. Hip joint fracture, one of the most serious consequences of falling in the elderly, occurs in only about 1% of falling. In this study, according to the loading conditions, the analysis is the behavior of the femur. The CT images using the commercial program "Mimics" the bones of three-dimensional CAD data generated, and we will analyze the results of finite element analysis. The boundary conditions on the basis of existing research has been simplified. In this paper, the whole femur was assumed to be isotropic linear elastic material. Predicted the behavior of the femur according to the loading condition, it can be help the development of high-precision artificial bones and joints can be treated with surgery and will be able to perform efficiently.

쾌속조형시스템을 이용한 생체 조직 재생용 지지체 제작과 특성분석 (Bio-degradable 3D-scaffold fabrication using rapid-prototyping system)

  • The purpose of tissue engineering is to repair or replace damaged tissues or organs by a combination of cells, scaffold, suitable biochemical and physio-chemical factors. Among the three components, the biodegradable scaffold plays an important role in cell attachment and migration. In this study, we designed 3D porous scaffold by Rapid Prototyping (RP) system and fabricated layer-by-layer 3D structure using Polycarprolactone (PCL) - one of the most flexible biodegradable polymer. Furthermore, the physical and mechanical properties of the scaffolds were evaluated by changing the pore size and the strand diameter of the scaffold. We changed nozzle diameter (strand diameter) and strand to strand distance (pore size) to find the effect on the mechanical property of the scaffold. And the surface morphology, inner structure and storage modulus of PCL scaffold were analyzed with SEM, Micro-CT and DMA.
