Protein Profiles Associated with Anoikis Resistance of Metastatic MDA-MB-231 Breast Cancer Cells

  • Resistance to anoikis, a cell-detachment induced apoptosis, is one of the malignant phenotypes which support tumor metastasis. Molecular mechanisms underlying the establishment of this phenotype require further investigation. This study aimed at exploring protein expression profiles associated with anoikis resistance of a metastatic breast cancer cell. Cell survival of suspension cultures of non-metastatic MCF-7 and metastatic MDA-MB-231 cells were compared with their adherent cultures. Trypan blue exclusion assays demonstrated a significantly higher percentage of viable cells in MDA-MB-231 than MCF-7 cell cultures, consistent with analysis of annexin V-7-AAD stained cells indicating that MDA-MB-231 possess anti-apoptotic ability 1.7 fold higher than MCF-7 cells. GeLC-MS/MS analysis of protein lysates of MDA-MB-231 and MCF-7 cells grown under both culture conditions identified 925 proteins which are differentially expressed, 54 of which were expressed only in suspended and adherent MDA-MB-231 but not in MCF-7 cells. These proteins have been implicated in various cellular processes, including DNA replication and repair, transcription, translation, protein modification, cytoskeleton, transport and cell signaling. Analysis based on the STITCH database predicted the interaction of phospholipases, PLC and PLD, and 14-3-3 beta/alpha, YWHAB, with the intrinsic and extrinsic apoptotic signaling network, suggesting putative roles in controlling anti-anoikis ability. MDA-MB-231 cells grown in the presence of inhibitors of phospholipase C, U73122, and phospholipase D, FIPI, demonstrated reduced ability to survive in suspension culture, indicating functional roles of PLC and PLD in the process of anti-anoikis. Our study identified intracellular mediators potentially associated with establishment of anoikis resistance of metastatic cells. These proteins require further clarification as prognostic and therapeutic targets for advanced breast cancer.

  • It is frequently said that mortgage-backed securities (MBS) of different security designs are issued in an attempt to meet the varying needs and expectations of investors. If this is true indeed, MBS of complicated risk-return characteristics are likely to be priced higher than MBS of simple risk-return characteristics we. We test this implication by establishing a recombining binomial interest rate prepayment model with a burnout effect embedded. More specifically, we compare the relative values (utilities) of a pass-through and a PAC- Support collateralized mortgage obligation(CMO), and theoretically show why and how the CMO is more highly valued than is the pass-through. The model is established such that mortgage prepayment is a function of the current value of, and the past path of, the mort-gage market rate. Since we work on not the total value of the two MBS but the value of each tranche of either MBS, the test results could be robust to slightly different versions of similar tests, which may be done in the future.

The Sterilization Effect of Methylene Blue, Formalin, and Iodine on Egg and Adult Stage of Marine Medaka, Oryzias dancena

  • The sterilization effects of methylene blue (MB), formalin, and iodine on the egg of marine medaka, Oryzias dancena, were investigated for disinfecting naididae worm, Chaetogaster diastrophus through sterilization. To determine harmfulness of MB, formalin, and iodine, lethal concentrations 50 ($LC_{50}$) of three chemicals were analyzed in the eggs of marine medaka. The sterilized periods of each chemical were set at 1 hr. Sterilized rates of naididae worm in each chemical were significantly affected and increased drastically as the concentration of each chemical increased (p<0.05). Sterilization abilities of naididae worm were most effective for formalin, but survival rates of egg and hatched rates for formalin were lowest among each chemical. The $LC_{50}$ of MB over 96 hrs were 185.26, 103.84, and 127.15 ppm for adults, juveniles, and eggs respectively. The toxic effects of MB were clearly dose dependent for each life stage (p<0.05). The toxicity sensitivity of juveniles to MB was dramatically higher than that of other groups. In 48 hrs after sterilization, cortisol and glucose concentrations of the adult group with MB treatment were significantly higher than those of the adult group with no treatment (p<0.05). This research provides useful data on sterilization effect of MB, formalin, and iodine, acute toxicity in marine medaka egg and toxicity, sensitivity of life stage of MB in marine medaka.

Effect of Grape Skin Extract on Apoptosis in MDA-MB-231 Human Breast Cancer Cells (포도껍질 추출물이 인체유방암세포 사멸에 미치는 영향)

  • We investigated the apoptotic effects of grape skin extracts (GSE) and related gene expressions in human breast cancer MDA-MB-231 cells cultured in the presence of 0, 0.5, 1 and 1.5 mg/mL of GSE for 72 hours. MTT assay, trypan blue and nuclei staining showed lower cellular mitochondrial activities and increased cell deaths with a higher concentration of GSE (p<0.05). Increased cell number with fragmentated DNA of sub-G1 phase was calculated as a measure of apoptotic cell death by FACS analysis (p<0.05). In particular, apoptotic cell death caused markedly increased in the 1 and 1.5 mg/mL of GSE groups, as revealed by flow cytometry (Annexin V-FITC). RT-PCR analysis was performed on apoptotic and preapoptotic genes. Expression of the apoptosis suppressor gene bcl-2 significantly decreased, proapoptotic gene bax was significantly increased and procaspase-3 showing the presence of caspase-3 significantly decreased (p<0.05). Furthermore, bcl-2/bax ratio which is considered to be an important indicator of apoptosis, significantly decreased in a concentration-dependent manner (p<0.05). These results indicated that GSE induces apoptosis in MDA-MB-231 human breast cancer cells.

  • The efficienty of ATM is based on the statical multiplexing of fixed-length packets, which are called cells. The most important technical point for realizing ATM switching network is an arrangement of the buffers and switches. Current most ATM switching networks are being achieved by using the switching modules based on the unit switch of $8{\times}8$ 150Mb/s or $16{\times}16$ 150Mb/s, the unit switch of $32{\times}32$150Mb/s for a large scale system is under study in many countries. In this paper, we proposed a new $32{\times}32$(4.9Gb/s throughput) ATM switch using Shared buffer memory switch which provides superior traffic characteristics in the cell loss, delay and throughput performance and easy LSI(Large Scale Integrated circuit). We analytically estimated and simulated by computer the buffer size into it. We also proposed the configuration of the large capacity ATM switching network($M{\times}M$.M>1,000) consisting of multistage to improve the link speed by non-blocking.

MB-OFDM UWB Technology for Increasing Transmission Reach of Wireless Speaker Systems (차세대 무선 스피커 시스템의 전송거리 증대를 위한 MB-OFDM UWB 기술)

  • We present the Multi-band orthogonal frequency division multiplexing ultra-wideband (MB-OFDM UWB) technology for increasing the transmission reach of wireless speaker systems. The proposed scheme adopts the Reed-Solomon coding for preventing the random error perfectly and shows the SNR gain in low bit error rate (BER) especially. So, we can increase the maximum reach of MB-OFDM UWB technology since the receiver sensitivity is improved. The simulation environment includes most effects of realistic channel environments such as Additive White Gaussian Noise (AWGN), CM1 channel model, Sampling frequency offset (SFO), Carrier frequency offset (CFO) to improve the simulation accuracy. The simulation results show that the proposed scheme can give a maximum 2 dB SNR gain and increase the transmission reach up to 12.6m.

G0/G1 Switch 2 Induces Cell Survival and Metastasis through Integrin-Mediated Signal Transduction in Human Invasive Breast Cancer Cells

  • Human breast cancer cell line, MDA-MB-231, is highly invasive and aggressive, compared to less invasive cell line, MCF-7. To explore the genes that might influence the malignancy of MDA-MB-231, DNA microarray analysis was performed. The results showed that G0/G1 switch 2 (G0S2) was one of the most highly expressed genes among the genes upregulated in MDA-MB-231. Although G0S2 acts as a direct inhibitor of adipose triglyceride lipase, action of G0S2 in cancer progression is not yet understood. To investigate whether G0S2 affects invasiveness of MDA-MB-231 cells, G0S2 expression was inhibited using siRNA, which led to decreased cell proliferation, migration, and invasion of MDA-MB-231 cells. Consequently, G0S2 inhibition inactivated integrin-regulated FAK-Src signaling, which promoted Hippo signaling and inactivated ERK1/2 signaling. In addition, G0S2 downregulation decreased ${\beta}$-catenin expression, while E-cadherin expression was increased. It was demonstrated for the first time that G0S2 mediates the Hippo pathway and induces epithelial to mesenchymal transition (EMT). Taken together, our results suggest that G0S2 is a major factor contributing to cell survival and metastasis of MDA-MB-231 cells.

Design of a CMOS LNA for MB-OFDM UWB Systems (MB-OFDM 방식의 UWB 시스템을 위한 CMOS LNA 설계)

  • A CMOS LNA based on a single-stage cascode configuration is designed for MB-OFDM ultra-wide band(UWB) systems. Wideband($3.1GHz\~4.9GHz$) input matching is performed using a simple bandpass filter to minimize the chip size and the noise figure degradation. The simulation results using $0.18{\mu}m$ CMOS process parameters show a power gain of 9.7dB, a 3dB band width of $2.1GHz\~7.1GHz$, a minimum NF of 2dB, an IIP3 of -2dBm. better than -11.8dB of input matching while occupying only $0.74mm^2$ of chip area. It consumes 25.8mW from a 1.8V supply.

  • A new line code design technique based on the BUDA(Binary Unit DSV and ASV) concept is introduced. The new line code called MB34 and designed by this new technique is of the minimum bandwidth, dc-free, and runlength limited. To confirm the performance of the new code, its power spectrum and eye pattern are obtained, wherein spectral nulls at dc(f=0) and Nyguist frequency (f=1/2Ts) are clearly identified. It is also discussed how the transmission errors can be detected by monitoring the DSV, the ASV, and the runlength.
