In this experiment forty-eight Sprague Dawley male rats were chosen and used in order to measure the growth rates and to see the effects of lead acumulation in their organs resulting from variously controlled lead protein diet. Protein sources were casein and isolated soyprotein (ISP), and each source was divided into three groups : 7% low protein [LP], 20% standard protein (SP) and 40% high protein (HP) groups. The six experimental groups were given lead acetate(25 mg/kg B.W.) and six control groups were given sodium chloride by oral administration 6 times a week for weeks. The results from this experiment were summeraized as following ; 1) Food consumption, weight gain, organ weight and food efficiency ; Lead acetate administration with protein source had no effects on food consumption, weight gain and organ weight . By their different levels of protein, food consumption of LP group was less the that of SP and HP groups after 3 weeks, weight gain of LP group was less than that of SP and HO groups after 1 weeks. The organ weight in LP group was significantly lower than SP and HP groups except teeth and adrenal s. Effect of lead acetate administration on food efficiency have significantly lower in LP-ISP diet and HP -casein diet than other groups only first week. By their different levels LP group showed significantly lower than SP group until 3 weeks. 2) Hematopoietic effect ; The hematopoieteic effect was not influencec by lead acdtate administration and protein source. Objective: Interleukin-6 (IL-6) is a T cell-derived B cell stimulating factor which plays an important role in inflammatory diseases. In this study, the pharmacokinetic properties of LMT-28 including physicochemical property, in vitro liver microsomal stability and an in vivo pharmacokinetic study using BALB/c mice were characterized. Methods: LMT-28 has been synthesized and is being developed as a novel therapeutic IL-6 inhibitor. The physicochemical properties and in vitro pharmacokinetic profiles such as liver microsomal stability and Madin-Darby canine kidney (MDCK) cell permeability assay were examined. For in vivo pharmacokinetic studies, pharmacokinetic parameters using BALB/c mice were calculated. Results: The logarithm of the partition coefficient value (LogP; 3.65) and the apparent permeability coefficient values (Papp; 9.7×10-6 cm/s) showed that LMT-28 possesses a moderate-high cell permeability property across MDCK cell monolayers. The plasma protein binding rate of LMT-28 was 92.4% and mostly bound to serum albumin. The metabolic half-life (t1/2) values of LMT-28 were 15.3 min for rat and 21.9 min for human at the concentration 1 μM. The area under the plasma drug concentration-time curve and Cmax after oral administration (5 mg/kg) of LMT-28 were 302±209 h·ng/mL and 137±100 ng/mL, respectively. Conclusion: These data suggest that LMT-28 may have good physicochemical and pharmacokinetic properties and may be a novel oral drug candidate as the first synthetic IL-6 inhibitor to ameliorate mammalian inflammation.
The present study was designed to evaluate the protective effects of dietary vitamin A or E. and of combination of vitamins A and E on peroxidative deterioration of heart in adriamycin-treated rats. Male Sprague-Dawley rats were assigned to 5 groups according to the dietary supplementation of vitamin A or E Except control rats a dose of 2mg ADR/kg of B. W was injected to these animals intraperitoneally on the same day every week. Adriamycin treatment significantly decreased the weight gain of experimental rats compared with that of control rats, But this decrement was not modified by dietary supplementation of vitamin A or E. Lipid peroxide values of plasma were elevated by ADR treatment. The combined use of ADR and dietary vitamin A or E significantly reduced these values, The interaction between vitamins A and E seemed to be present in the lipid peroxide value of plasma. Catalase and superoxide dismutase(SOD) activities in rat heart were decrased by ADR treatment but glutathione peroxidase(GSH-Px) activity was elevated. Dietary supplmen-tation of vitamin A or E enhanced the heart catalase and SOD activities. except only vitamin A-supplemented group. GSH-Px activity of rat heart tended to be decreased by dietary supple-mentation of vitamin A or E. With ADR treatment polyunsaturated fatty acids such as archido-nic acid(20:4) and docosahexaenoic acid(22:6) were decreased in rat heart. However dietary supplementation of vitamins A and E reduced this decrease. The retinol and tocopherol contents of rat plasma were decreased by ADR treatment. Dietary vitamin A or E influence vitamin A or E content of plasma. The interaction between dietary vitamins A and E was observed in vitamin A or E level of rat plasma.
This study examined the antimicrobial activity of Myagropsis yendoi (MY). The antimicrobial activity of ethanol and aqueous MY extracts were measured using a paper disc assay. The MY ethanol extract had the best antimicrobial activity. It inhibited the growth of Candida tropicalis and Gram-positive bacteria such as Bacillus subtilis, Listeria innocua, L. monocytogenes, and Staphylococcus aureus at 4 mg/mL. The minimum inhibitory concentration (MIC) of the MY ethanol extract ranged from 0.1 to 0.5%. The MY ethanol extract inhibited the growth of B. subtilis and L. innocuaat concentrations of 0.15% and 0.1%, respectively. The antimicrobial activity of the MY ethanol extract was not affected by heating at $121^{\circ}C$ for 15 min or pH 2.10. Therefore, the main substances responsible for the antimicrobial activity of the MY ethanol extract are believed to be stable with changing heat and pH.
Development of the strain and the fermentation process of Hansenula polymorpha was implemented for the production of ${\gamma}$-linolenic acid ($GLA,\;C18:3{\Delta}^{6,9,12}$), an n-6 polyunsaturated fatty acid (PUFA) that has been reported to possess a number of health benefits. The mutated ${\Delta}^6$-desaturase (S213A) gene of Mucor rouxii was expressed in H. polymorpha under the control of the methanol oxidase (MOX) promoter. Without the utilization of methanol, a high-cell-density culture of the yeast recombinant carrying the ${\Delta}^6$-desaturase gene was then achieved by fed-batch fermentation under glycerol-limited conditions. As a result, high levels of the ${\Delta}^6$-desaturated products, octadecadienoic acid ($C18:2{\Delta}^{6,9}$), GLA, and stearidonic acid ($C18:4{\Delta}^{6,9,12,15}$), were accumulated under the derepression conditions. The GLA production was also optimized by adjusting the specific growth rate. The results show that the specific growth rate affected both the lipid content and the fatty acid composition of the GLA-producing recombinant. Among the various specific growth rates tested, the highest GLA concentration of 697 mg/l was obtained in the culture with a specific growth rate of 0.08 /h. Interestingly, the fatty acid profile of the yeast recombinant bearing the Mucor ${\Delta}^6$-desaturase gene was similar to that of blackcurrant oil, with both containing similar proportions of n-3 and n-6 essential fatty acids.
Silicon is commercially prepared by the reaction of high-purity silica with wood, charcoal, and coal, in an electric arc furnace using carbon electrodes, so called the metallurgical refining process, which produces ~98% pure Si (MG-Si). This can be further purified to solar grade silicon (SoG-Si) by various techniques. The most problematic impurity elements are B and P because of their high segregation coefficients. In this study, we explored the possibility of the using Cat-CVD for Si purification. The existing hot-wire CVD was modified to accommodate the catalyzer and the heating source. Mo boat (1.5 cm ${\times}$ 1 cm ${\times}$ 0.2 cm) was used as a heating source. Commercially available Si was purchased from Nilaco corporation (~99% pure). This powder was kept in the Mo-boat and heated to the purification temperature. In addition to the purification by cat-CVD technique, other methods such as thermal CVD, plasma enhanced CVD, vacuum annealing was also tried. It is found that the impurities are reduced to a great extent when treated with cat-CVD method.
The present study is designed to explore an anti-tumor activity on crude extracts of Oplopanax elatus. Water extractions of Oplopanax elatus were performed at $100^{\circ}C$(OeE-100). OeE-100 doses up to $62.5{\mu}g/m{\ell}$ had no cytotoxicity on the tumor cell lines in vitro. In experimental lung metastasis of colon26-M3.1 carcinoma or B16-BL6 melanoma, the prophylactic intravenous ($4{\sim}100{\mu}g/mouse$) or oral (2 mg/mouse) administration of OeE-100 significantly inhibited tumor metastasis as compared with tumor controls. Peritoneal macrophages stimulated with OeE-100 produced various cytokines such as TNF-${\alpha}$, IL-6 and IL-12. In an analysis of NK-cell activities, i.v. administration of OeE-100 ($10{\sim}100{\mu}g/mouse$) significantly augmented the cytotoxicity to YAC-1 tumor cells. Vaccination of mice with boiling-treated tumor cells (BT-vaccine) in combination with OeE-100 ($100{\mu}g/mouse$) showed higher inhibitions in tumor metastasis when compared with the mice of BT-vaccine treatment. In addition, the splenocytes from OeE-100 admixed BT-vaccine immunized mice secreted a higher concentration of Th1 type cytokine such as IFN-${\gamma}$. These results suggested that the OeE-100 stimulated immune system and was a good candidate adjuvant of anti-tumor immune responses.
The purpose of this study was to measure the antioxidant activity of yacon root and to manufacture yacon pickles for commercialization as a functional food. The total phenol content in yacon roots were the highest, 29.96 mg GAE/ 100 g, in the ethyl acetate fraction and the DPPH radical scavenging activity and ABTS radical scavenging activity was 37.07%, and 91.60%, respectively, in the ethyl acetate fraction. The optimum recipe for the pickling solution was determined to be vinegar, water and high fructose corn syrup, at 34%, 22% and 44%, respectively. The quality characteristics of yacon pickles using this solution were monitored during storage at $25^{\circ}C$ for 43 days. The pH value decreased to 2.98 (42 days) during storage and $^{\circ}Brix$ gradually increased during storage. The L-value decreased significantly during storage but there were no significant changes detected in the a and b values. No significant changes were detected in the hardness of the yacon pickles. According to the sensory evaluation, no significant changes in taste, texture and overall acceptance of yacon pickle were observed during storage. These results indicated that yacon was an excellent functional food in terms of its antioxidant activity and that pickling might be a promising methods to maintain the taste and quality of yacon during storage.
Purpose: This study aimed to investigate the quality characteristics of traditional and commercial soybean pastes (Doenjang) for development of traditional soybean pastes. Methods: Proximate compositions, amino acid nitrogen contents, pH, acidity, salt contents, chromaticity, viable bacteria and inorganic substance contents of nineteen traditional and three commercial Doenjang were investigated. Results: Analysis of proximate compositions indicated a significant difference between samples. In moisture content, D10 did not meet the standard of food codex. The contents of moisture, fat, protein and ash were slightly higher in traditional than commercial Doenjang. However, the carbohydrate content was 2-fold higher in commercial than traditional, and thus, the calories were also higher. The amino nitrogen content was lower in commercial than traditional Doenjang. pH, acidity and salt contents of Doenjang were pH 4.67-6.15, 1.53-3.29%, and 9.01-18.78%, respectively. pH and acidity showed no significant differences between traditional and commercial Doenjang, however, salt contents were significantly higher in traditional than commercial Doenjang. In case of chromaticity, the averages of L, a, b values were higher in traditional Doenjang. In microbiological analysis, total bacteria counts were higher in traditional than commercial Doenjang. The number of Bacillus cereus in D12 and D19 exceeded the standard of food codex. Staphylococcus aureust was undetected in all samples. The number of fungi varied widely between samples, with no detection in five traditional and all commercial Doenjang. Results of inorganic substance analysis indicated that contents of inorganic substances in Doenjang were in order of Na, K, Mg and Ca. Overall, Na content was higher in traditional Doenjang. Contents of inorganic substances showed wide variations in traditional Doenjang. Conclusion: For the development of traditional Doenjang, quality standardization and ensuring safety are considered necessary.
