Ganoderma Lucidum Polysaccharides Target a Fas/Caspase Dependent Pathway to Induce Apoptosis in Human Colon Cancer Cells

  • Ganoderma lucidum polysaccharides (GLP) extracted from Ganoderma lucidum have been shown to induce cell death in some kinds of cancer cells. This study investigated the cytotoxic and apoptotic effect of GLP on HCT-116 human colon cancer cells and the molecular mechanisms involved. Cell proliferation, cell migration, lactate dehydrogenase (LDH) levels and intracellular free calcium levels ($[Ca^{2+}]i$) were determined by MTT, wound-healing, LDH release and fluorescence assays, respectively. Cell apoptosis was observed by scanning and transmission electron microscopy. For the mechanism studies, caspase-8 activation, and Fas and caspase-3 expression were evaluated. Treatment of HCT-116 cells with various concentrations of GLP (0.625-5 mg/mL) resulted in a significant decrease in cell viability (P< 0.01). This study showed that the antitumor activity of GLP was related to cell migration inhibition, cell morphology changes, intracellular $Ca^{2+}$ elevation and LDH release. Also, increase in the levels of caspase-8 activity was involved in GLP-induced apoptosis. Western blotting indicated that Fas and caspase-3 protein expression was up-regulated after exposure to GLP. This investigation demonstrated for the first time that GLP shows prominent anticancer activities against the HCT-116 human colon cancer cell line through triggering intracellular calcium release and the death receptor pathway.

Macro- and Micro-nutrient Utilization and Milk Production in Crossbred Dairy Cows Fed Finger Millet (Eleucine coracana) and Rice (Oryza sativa) Straw as Dry Roughage Source

  • Finger millet straw and rice straw are the major source of dry roughage in southern India. They distinctly vary in their morphological and nutritional characters. Hence an effort was made to study the nutrient utilization, milk yield and composition in crossbred dairy cows fed either finger millet (group 1) or rice straw (group 2) as a source of dry roughage. The cows in both the groups were fed as per requirement with concentrate, green fodder and straw in the ratio of 30:45:25 parts (DM). At the end of 50 days of preliminary feeding a digestibility trial was conducted for 7 days and pooled samples of feed, fodder, feces, urine and milk were analysed for macro and micro nutrient content. Finger millet straw contained more CP, Ca, P, Mg, Cu, Zn and Co than rice straw and rice straw contained higher ADF, ash and silica. The intake of DM, CP, EE, NDF, ADF and most micronutrients (Ca, P, Mg, Cu, Zn, Fe, Mn and Co) was significantly higher in cows fed finger millet straw. The digestibility of DM, CP, NDF and ADF was significantly higher in cows fed finger millet straw and the gut absorption of Ca, Cu, Mn and Co was significantly higher in cows fed finger millet straw. The dietary requirement of all micronutrients in both the group of cows could be met irrespective of the type of roughage fed except that of Ca, which was low (0.61 and 0.40%) in rice straw fed cows. The average daily milk yield (L/cow) was also higher (7.0 L) in cows fed finger millet straw as compared to cows fed rice straw (6.3 L). The average milk composition also did not differ except that of milk fat which was significantly (4.7 and 4.5%) low in cows fed rice straw. The overall results of this study have indicated that finger millet straw is a better source of dry fodder than rice straw and while feeding rice straw as the sole roughage to dairy cows there is need to supplement additional calcium as this could be one of the limiting nutrients for milk production.

  • The utilization methods of algae biomass have been studied constantly in whole world. These are $\circled1$the wastewater treatment if waste stabilization pond and oxidation ditch etc. and $\circled2$the biosorption of heavy metals and recovery of strategic' precious metals and $\circled3$the single-celled protein production and the production of chemicals like coloring agent and $\circled4$the production of electric energy through methane gasification. The culture system also has been developed constantly in relation with such utilization method developments. In the result of experimental operation under continuous and cyclic irradiation of light, using high rate algae biomass culture pond(HRABCP), which had been made so as to be an association system with the various items which had been managed to have high efficiency for algae culture, the algae production of the 12 hours-irradiance pond was 41.48 Chlorophyll-a ${\mu}g/L$ only in spite of having the more chance of $CO_2$ synthesis to algae cell than the 24 hours-irradiance pond. This means that the energy supply required for dark-reaction of photosynthesis is very important like this. The difference of algae production between continuous and cyclc irradiation explains that the dark-reaction of photosynthesis acts on algae production as the biggest primary factor. The continuous irradiance on HRABCP made the good algae-production($1403.97{\;}{\mu}g$ Chlorophyll-a/mg) and the good oxygen-production(5.8 mg $O_2/L$) and the good solid-liquid seperation. especially, DO concentration through the oxygen-production was enough to fishes' survival.

  • For the production of microbial cells from cellulosie materials by cellulore-assimilating bacteria, Cellulomonas flavigena GFB 24-1, isolated by authors, utilization of this organism on some microbiological properties was investigated. The results of these studies were summarized at follows; 1. When the organism was incubated in the growth medium at pH 7.0 for 50 hours, its growth was the most effective and the level of excreted total protein in the menstruum increased continuously during the stationary phase of cell growth. 2. The optimal enzyme activity was observed in the pH region of 5 to 7 and culture period of 40 to 50 hours. 3. The microbial degestibility of cellulosic wastes such as sawdust, rice hull, rice straw, peanut hull and used newspaper was less than 30%, whereas that of cellulose powder was 47.1% and rice straw was digested 77% by NaOH treatment. 4. Bacterial cells incubated in the growth medium were increased up to 8% of sustrate concentration and showed a decrease on further concentration. 5. The production of microbial cells from NaOH treated rice straw was obtained 10.6mg/ml of culture medium.

Lysolecithins improved growth performance, nutrient digestibility, immunity, and antioxidant ability in broiler chickens

  • Objective: This study investigated the effects of dietary supplementation with lysolecithins (LPC) on growth performance, nutrient digestibility, blood profiles, immunity, and liver health in broiler chickens. Methods: A cohort of 240 one-day-old male Arbor Acres broilers of comparable weight was divided into four treatment groups, each comprising six replicates of 10 birds. The groups were defined as follows: positive control with recommended metabolizable energy (PC+ME), negative control with 90 kcal/kg reduced ME (NC+ME), PC supplemented with 300 mg/kg LPC (PC+LPC), and NC supplemented with 300 mg/kg LPC (NC+LPC). Results: LPC supplementation led to a statistically significant reduction in the feed conversion ratio (p = 0.05) and a decrease in the proportion of abdominal fat and the liver (p<0.05). Digestibility of dry matter was also enhanced (p<0.05). Malondialdehyde concentrations in the liver were significantly reduced by LPC (p<0.01), with a noteworthy interaction between energy levels and LPC affecting this reduction (p<0.05). Serum levels of interleukin-6 were reduced on day 21, and both endotoxin and interleukin-6 levels were lower on day 42. Notably, a significant interaction was observed between the energy levels and LPC on relative liver weight and endotoxin concentrations in the serum (p<0.05). Conclusion: The study concluded that LPC positively affects growth performance, nutrient digestibility, immune response, and antioxidative capacity in broiler chickens, affirming its value as a beneficial feed additive in poultry nutrition.

Nutrition Components in Different Parts of Korean Loquat(Eriobotrya japonica Lindl.) (한국산 비파의 부위별 영양성분)

  • To furnish basic data about the utilization of Korean loquat as flood, this experiment was conducted. Nutritional components in leaves, fruit excluded seed, flesh and seed of Korean loquat were analyzed as follows : moisture 48.7%, 87.8%, 88.3% and 59.5% ; total sugar 1.57%, 7.21%, 7.36% and 2.41% ; crude protein 5.23%, 1.61%, 1.44% and 4.31% in each portion, respectively. The highest mineral contents of loquat leaves, fruit excluded seed, flesh and seed were Ca 2,458ppm, K 661ppm, 654ppm and 1,528ppm, and water soluble vitamins such as ascorbic acid, thiamin and pyridoxine were confirmed in different pares of Korean loquat, the contents of those were high thiamin 5.86mg% in leaves and ascorbic acid 1.l0mg%, 1.26mg% and 4.90mg% in fruit excluded seed flesh and seed, respectively. The contents of free sugars were high sucrose 0.87%, glucose 0.62%, 0.6475 and rhamnose 0.20%, and major organic acid were detected oxalic acid 1,693.70mg%, malic acid 201.70mg%, 207.60mg% and citric acid 55.70mg% in each portion, respectively. Free amino acid were identified 21, 14, 14 and 16 kinds of leaves, fruit excluded seed, flesh and seed, respectively and their contents in each portion were highest glutamic acid 280.22mg%, proline 35.l0mg%, glutamic acid 56.96mg% and sarcosine 230.24mg%, respectively. Volatile components were identified 25 and 11 kinds of leaves and flesh and their contents were highest d-nerolidol 28.70ppm, hexadecanoic acid 16.67ppm, respectively.

Interaction of Dietary Wheat Bran and Dietary Calcium Levels ell Calcium Utilization and Bone Mass in Post-breeding Female Rats

  • This study was conducted to determine the interaction of dietary wheat bran and dietary calcium levels n)n calcium utilization in post-breeding female rats. It was designed to compare the effects of four different levels (2.5, 5, 10 and 20%) of wheat bran and two different levels (0.5 and 1%) of calcium on bone and calcium balance in post-breeding female rats over a ten-week period. The effects of diet on animal weight gain, serum calcium, femur weight, femur calcium concentration, bone mass and calcium balance were determined and statistically analyzed. The addition of 20% wheat bran significantly (p$\leq$0.05) decreased the weight gain of rats. Serum calcium and bone calcium contents were more affected by dietary calcium level than by dietary wheat bran level. There was no significant difference in fat-free solid, ash, percentage of ash to fat-free solid and percentage of calcium to ash among groups. Groups fed the 1% calcium diet had a higher percentage of calcium to fat-free solid. All rats were in positive calcium balance during the three-4ay experimental period. The average calcium balance of the rats fed 1% calcium diet ranged from 25.34 to 53.90mg and the average calcium balance of the rats fed the 0.5% calcium diet ranged from 26.71 to 32.90mg. In rats receiving 2.5% wheat bran, the difference in calcium balance between the group fed the 1% calcium diet and the group fed the 0.5% calcium diet was only 1.37mg, which was not significantly (p$\leq$0.05) different. In rats receiving 20% wheat bran, the difference in calcium balance between the group fed the 1% calcium diet and the group fed the 0.5% calcium diet was 19.S7mg, which was significantly (p$\leq$0.05) different. The addition of wheat bran caused an increase in the calcium balance of the rats adminslesed the 1% calcium diet. On the other hand, the addition of wheat bran had no effect on the calcium balance of the rats adminislesed the 0.5% calcium diet. In conclusion, calcium utilization of rats wire more positively affected by the interaction of both dietary wheat bran levels and dietary calcium levels than either dietary wheat bran levels or dietary calcium levels alone. Moderate wheat bran consumption did not interfere with the calcium metabolism of rats when calcium intake was high enough.

  • The ferrokinetics and red cell life spans of the patients with chronic glomerulonephritis were investigated by the double tracing method using radioactive iron ($^{59}Fe$) and chromium ($^{51}Cr$). According to the serum NPN levels, the patients were subdivided into 3 groups: Group 1. 6 patients, had the levels below 40 mg/dl Group 2. 6 patients, had the levels between 41 mg/dl to 80 mg/dl Group 3. 10 patients, had the levels above 80 mg/dl The results were as follows: 1) Red blood cell-, hematocrit- and hemoglobin values were moderately reduced in patients with normal serum NPN levels, while markedly reduced in patients with elevated serum NPN levels. 2) The plasma volume was increased, while the red cell volume was decreased in patients with elevated serum NPN levels, hence, total blood volume was unchanged. 3) The serum iron level was slightly reduced h patients of groups 1 and 2, while was within the normal ranges in patients of group 3. 4) i) In patients with normal serum NPN levels, the plasma iron disappearance rate, red cell iron utilization rate, red cell iron turnover rate, daily red cell iron renewal rate, circulating red cell iron and red cell iron concentration were within the normal ranges, while the plasma iron turnover rate was slightly reduced. ii) In patients with elevated serum NPN levels, the plasma iron disappearance rate was delayed, while the plasma iron turnover rate was within the normal ranges. The red cell iron utilization rate, red cell iron turnover rate and circulating red cell iron were decreased and the period in which the red cell iron utilization rate reachd its peak was delayed in Group 3 patients. The daily red cell iron renewal rate and the red cell iron concentration were unchanged. iii) The mean red cell life span was within the normal ranges in patients with normal serum NPN levels, while was shortened in patients with elevated serum NPN levels.

  • Magnetization studies have been carried out on $MgB_2$ polycrystalline samples in the temperature range of 5 - 44 K and in the magnetic field up to 7 Tesla. The critical current density was calculated from hysteresis loops using the Bean's critical state model, and the highest value of $J_c$ at 20 K was $2.7{\times}10^5\;A/cm^2$ at 2 Tesla. The hysteresis loops were carefully examined to determine the temperature and magnetic field range where flux jumps appeared. The first jump occurred typically at 1 Tesla. Due to the strong pinning, we observed the presence of flux jump below H = 1 Tesla at temperature below 30 K.

  • Studies on protein and antinutritional factors of rapeseeds are ncessary for effective utiliza-tion of defatted rapeseed meal. Proteins were extracted from seeds of several species of rape-seeds and analyzed by SDS-PAGE and the contents of glucosinolate and phytate were determi-ned. One percent solution extraction and extraction yield was relatively higherfor B. campestris and B. juncea than for other species. SDS-PAGE revealed that rapeseeds of most species were rich in low molecular weight proteins and that in particular the roteins of B. napus var. Halla and B. juncea were composed of simpler subunits as compared with other species. The content of glucosinolate was around 10mg/g of defatted meal for B. juncea however for var. Halla it was 7.26mg/g of defatted meal and 0.46mg/g of protein concentrate which were the lowest values. The level of phytate was between 2.7 and 4.6% for all species tested. Our results indicate thT B, napus var. Halla is the desired species for the utilization of rapeseed proteins.
