Synthesis and Characterization of Magnetic Nanoparticles and Its Application in Lipase Immobilization

  • We demonstrate herein the synthesis and modification of magnetic nanoparticles and its use in the immobilization of the lipase. Magnetic $Fe_3O_4$ nanoparticles (MNPs) were prepared by simple co-precipitation method in aqueous medium and then subsequently modified with tetraethyl orthosilicate (TEOS) and 3-aminopropyl triethylenesilane (APTES). Silanization magnetic nanoparticles (SMNP) and amino magnetic nanomicrosphere (AMNP) were synthesized successfully. The morphology, structure, magnetic property and chemical composition of the synthetic MNP and its derivatives were characterized using transmission electron microscopy (TEM), Fourier transform infrared spectroscopy (FT-IR) analysis, X-ray diffraction, superconducting quantum interference device (SQUID) and thermogravimetric analyses (TGA). All of these three nanoparticles exhibited good crystallization performance, apparent superparamagnetism, and the saturation magnetization of MNP, SMNP, AMNP were 47.9 emu/g, 33.0 emu/g and 19.5 emu/g, respectively. The amino content was 5.66%. The AMNP was used to immobilize lipase, and the maximum adsorption capacity of the protein was 26.3 mg/g. The maximum maintained activity (88 percent) was achieved while the amount of immobilized lipase was 23.7 mg $g^{-1}$. Immobilization of enzyme on the magnetic nanoparticles can facilitate the isolation of reaction products from reaction mixture and thus lowers the cost of enzyme application.

Fe-Nanoparticle Amalgamation Using Lagenaria siceraria Leaf Aqueous Extract with Focus on Dye Removal and Antibacterial Efficacy

  • Iron nanoparticles (Fe-NPs) were synthesized employing Lagenaria siceraria (LS) leaf aqueous extract as a reducing and capping medium to remove methylene blue (MB) dye and have antibacterial properties against G-negative (Escherichia coli) and G-positive bacteria (Staphylococcus aureus). The formation of LS-Fe-NPs (Lagenaria-siceraria-iron-nanoparticles) was confirmed by a change in color from pale yellow to dark brown. Characterization techniques, such as particle size analysis (PSA), transmission electron microscopy (TEM) and scanning electron microscopy (SEM), were employed to prove nano spherical particles of size range between 80-100 nm. Phytochemicals and the presence of iron in LS-Fe-NPs nanoparticles were proved by UV-visible spectrophotometry. Further, Fourier transform infrared spectroscopy (FTIR) analysis results confirmed the existence of bioactive molecules in the plants. The magnetic property was analyzed using a vibrating sample magnetometer (VSM), which displayed that the synthesized nanoparticles were superparamagnetic and exhibiting a saturation magnetization of 12.5 emu/g. Synthesized magnetic nanoparticles were used in methylene blue (MB) dye removal through adsorption. About 83% of 100 mg/L MB dye was removed within 120 min at pH 6 with a maximum adsorption capacity of 246.8 mg/g. Antibacterial efficacy of LS-Fe-NPs was screened against G-negative (Escherichia coli) and G-positive bacteria (Staphylococcus aureus), respectively, and found that LS-Fe-NPs were effective against Staphylococcus aureus.

Acute Toxicity and Tissue Distribution of Cerium Oxide Nanoparticles by a Single Oral Administration in Rats

  • Cerium oxide nanoparticles (size: 30 nm) were prepared by the supercritical synthesis method, Acute oral toxicity and tissue distribution of the nanoparticles were evaluated by a single administration in rats. Oral administration of the nanoparticles to the rats did not lead to death when the animals were treated by a dose of 5 g/kg (high dose) as well as 100 mg/kg (low dose). Abnormal clinical signs, changes in serum biochemistry and hematology were not observed in high-dose treated group compared to the vehicle control group. Lesions in liver, lung and kidney were not observed in high-dose treated group by histopathological examination. Tissue distribution analysis in liver, kidney, spleen, lung, testis and brain was performed on day 1, day 7 and day 14 after treatment. The average values of the accumulated cerium oxide nanoparticles were elevated in all tissues but statistical significance was only shown in lung. Low levels of tissue distributions after a single oral administration seem to be the low bioavailability of the nanoparticles.

  • 본 연구는 레시틴으로 나노입자화 시킨 티아민 디라우릴 설페이트의 향장활성 증진에 관한 것이다. TDS를 포집시킨 나노입자는 150 ~ 200 nm의 크기를 나타내는 구형이며, 또한 제타포텐셜을 측정하여 여러 pH 범위에서 안정한 것을 확인하였다. TDS 나노입자는 인간 섬유아세포(CCD-986sk)에 높은 농도를 처리하여도 85%의 세포생존률을 보였다. 자유라디칼소거활성 실험을 진행한 결과 나노입자화하지 않은 TDS 희석액(1.0 mg/mL)은 81.6%의 활성을 나타내었고, 나노입자화한 TDS 용액은 이보다 더 높은 88.1%의 높은 라디칼 소거활성을 보였다. TDS 나노입자는 자외선을 조사시킨 CCD-986sk에서 MMP-1의 발현을 41.4% 감소시켰다. TDS 용액과 TDS 나노입자를 가지고 salmonella typhimurium, listeria monocytogenes에 대하여 항균활성을 측정하였다. TDS 나노입자의 경우 양성대조군의 항균활성과 비슷한 결과를 나타내었다. 이러한 결과들로 TDS 나노입자가 항산화, 미백, 주름개선 효능같은 향장 소재로서의 적용이 가능할 것이라 생각된다.

Green Synthesis to Develop Iron-Nano Formulations and Its Toxicity Assays

  • Objectives: In the past few years, herbal medicines have gained popularity over synthetic drugs because of their natural source and minimal side effects which has led to a tremendous growth of phytopharmaceuticals usage. With the development of nanotechnology, it provides alternative approaches to overcome several limitations using nano-formulations. In spite of considerable quantity of antianemic preparations with different iron forms available, currently additives are used and represented in modern pharmaceutical market. Iron deficiency anemia is a major global public health problem which particularly affects pregnant women, children and elderly persons. The situation is complicated because of disadvantages and drug side effects from existing antianemic medicines. There is a great demand for the development of new antianemic preparations. Green synthesis of iron oxide nanoparticles, possess high potential in this field. Methods: Our study focuses on developing green synthesis of iron oxide nanoparticles (IONPs) of 10-50 nm with spherical shape where different dosages were used -1 mg/kg, 10 mg/kg and 100 mg/kg for exposure in Wistar albino female rats for 28 days. The toxicity was assessed using various parameters such as measurements of the rat body and organ mass, hematology, biochemical evaluation and histopathological examinations. Results: No significant differences were observed in body and organ weights. Hematological indices also indicated no significant differences whereas biochemical factors showed increase in levels of direct bilirubin and globulin of medium as well as high dose and SGPT levels were increased only in high dose. The major organs (heart, kidney and liver) showed histopathological alterations in 10 and 100 mg/kg whereas brain showed only in 100 mg/kg. Conclusion: The toxicity of IONPs was found to be more significant when the concentration was increased; however, low doses can be used for further investigation as an antianemic preparation.

Retinoic acid loaded with chitosan nanoparticles improves spermatogenesis in scrotal hyperthermia in mice

  • Objective: High temperatures can trigger cellular oxidative stress and disrupt spermatogenesis, potentially leading to male infertility. We investigated the effects of retinoic acid (RA), chitosan nanoparticles (CHNPs), and retinoic acid loaded with chitosan nanoparticles (RACHNPs) on spermatogenesis in mice induced by scrotal hyperthermia (Hyp). Methods: Thirty mice (weighing 25 to 30 g) were divided into five experimental groups of six mice each. The groups were as follows: control, Hyp induced by a water bath (43 ℃C for 30 minutes/day for 5 weeks), Hyp+RA (2 mg/kg/day), Hyp+CHNPs (2 mg/kg/72 hours), and Hyp+RACHNPs (4 mg/kg/72 hours). The mice were treated for 35 days. After the experimental treatments, the animals were euthanized. Sperm samples were collected for analysis of sperm parameters, and blood serum was isolated for testosterone measurement. Testis samples were also collected for histopathology assessment, reactive oxygen species (ROS) evaluation, and RNA extraction, which was done to compare the expression levels of the bax, bcl2, p53, Fas, and FasL genes among groups. Additionally, immunohistochemical staining was performed. Results: Treatment with RACHNPs significantly increased stereological parameters such as testicular volume, seminiferous tubule length, and testicular cell count. Additionally, it increased testosterone concentration and improved sperm parameters. We observed significant decreases in ROS production and caspase-3 immunostaining in the RACHNP group. Moreover, the expression levels of bax, p53, Fas, and FasL significantly decreased in the groups treated with RACHNPs and RA. Conclusion: RACHNPs can be considered a potent antioxidative and antiapoptotic agent for therapeutic strategies in reproductive and regenerative medicine.

솔잎 추출물이 삽입된 마그네슘-층상규산염 샌드위치 나노입자의 합성과 피부 상재균에 대한 항균 특성에 관한 연구 (Study on Synthesis of Pine Leaf Extract Intercalated Mg-Phyllosilicate Sandwich Nanoparticles and Antimicrobial Activity against Cutaneous Microorganisms)

  • 본 연구에서는 마그네슘-층상규산염(MgP) 층간에 솔잎 추출물(PLE)이 삽입되어 겹겹이 쌓여있는 샌드위치 구조의 나노입자(PLE/MgP)를 one-pot으로 합성하였다. XRD 분석을 통해 팔면체와 사면체 구조로 MgP가 성공적으로 합성되었으며, PLE가 층간에 삽입되어 MgP 간의 층간거리(d-spacing)가 벌어진 것을 확인하였다. HR-TEM을 통해 모폴로지 변화를 관찰하였으며, 열중량분석법을 통해 PLE가 MgP 대비 10%까지 함유된 것을 확인하였다. 최소저해농도(MIC) 및 최소 사멸농도(MBC) 분석을 통해 피부 상재균에 대한 항균 활성을 검토한 결과 MgP 또는 PLE보다 PLE/MgP의 항균 활성이 더욱 향상된 것을 확인하였다. 이러한 결과는 본 연구에서 개발한 PLE/MgP 유/무기 복합 소재가 화장품 소재, 의료용 소재 등의 다양한 분야에 적용 가능함을 확인하였다.

Low temperature wet-chemical synthesis of spherical hydroxyapatite nanoparticles and their in situ cytotoxicity study

  • The present research work reports a low temperature ($40^{\circ}C$) chemical precipitation technique for synthesizing hydroxyapatite (HAp) nanoparticles of spherical morphology through a simple reaction of calcium nitrate tetrahydrate and di-ammonium hydrogen phosphate at pH 11. The crystallinity of the single-phase nanoparticles could be improved by calcinating at $600^{\circ}C$ in air. Thermogravimetric and differential thermal analysis (TG-DTA) revealed the synthesized HAp is stable up to $1200^{\circ}C$. Scanning electron microscopy (SEM) and energy dispersive spectroscopy (EDS) studies confirmed the formation of spherical nanoparticles with average size of $23.15{\pm}2.56nm$ and Ca/P ratio of 1.70. Brunauer-Emmett-Teller (BET) isotherm of the nanoparticles revealed their porous structure with average pore size of about 24.47 nm and average surface area of $78.4m2g^{-1}$. Fourier transform infrared spectroscopy (FTIR) was used to confirm the formation of P-O, OH, C-O chemical bonds. Cytotoxicity and MTT assay on MG63 osteogenic cell lines revealed nontoxic bioactive nature of the synthesized HAp nanoparticles.

Production of High-purity Magnetite Nanoparticles from a Low-grade Iron Ore via Solvent Extraction

  • We produced magnetite nanoparticles (MNPs) and a Mg-rich solution as a nano-adsorbent and a coagulant for water treatment, respectively, using a low-grade iron ore. The ore was leached with aqueous hydrochloric acid and its impurities were removed by solvent extraction of the leachate using tri-n-butyl phosphate as an extractant. The content of Si and Mg, which inhibit the formation of MNPs, was reduced from 10.3 wt% and 15.5 wt% to 28.1 mg/L and < 1.4 mg/L, respectively. Consequently, the Fe content increased from 68.6 wt% to 99.8 wt%. The high-purity $Fe^{3+}$ solution recovered was used to prepare 5-15-nm MNPs by coprecipitation. The wastewater produced contained a large amount of $Mg^{2+}$ and can be used to precipitate struvite in sewage treatment. This process helps reduce the cost of both sewage and iron-orewastewater treatments, as well as in the economic production of the nano-adsorbent.

Antiproliferative Effects of Free and Encapsulated Hypericum Perforatum L. Extract and Its Potential Interaction with Doxorubicin for Esophageal Squamous Cell Carcinoma

  • Objectives: Esophageal squamous cell carcinoma (ESCC) is considered as a deadly medical condition that affects a growing number of people worldwide. Targeted therapy of ESCC has been suggested recently and required extensive research. With cyclin D1 as a therapeutic target, the present study aimed at evaluating the anticancer effects of doxorubicin (Dox) or Hypericum perforatum L. (HP) extract encapsulated in poly(lactic-co-glycolic acid) (PLGA) nanoparticles on the ESCC cell line KYSE30. Methods: Nanoparticles were prepared using double emulsion method. Cytotoxicity assay was carried out to measure the anti-proliferation activity of Dox-loaded (Dox NPs) and HP-loaded nanoparticles (HP NPs) against both cancer and normal cell lines. The mRNA gene expression of cyclin D1 was evaluated to validate the cytotoxicity studies at molecular level. Results: Free drugs and nanoparticles significantly inhibited KYSE30 cells by 55-73% and slightly affected normal cells up to 29%. The IC50 of Dox NPs and HP NPs was ~ 0.04-0.06 mg/mL and ~ 0.6-0.7 mg/mL, respectively. Significant decrease occurred in cyclin D1 expression by Dox NPs and HP NPs (P < 0.05). Exposure of KYSE-30 cells to combined treatments including both Dox and HP extract significantly increased the level of cyclin D1 expression as compared to those with individual treatments (P < 0.05). Conclusion: Dox NPs and HP NPs can successfully and specifically target ESCC cells through downregulation of cyclin D1. The simultaneous use of Dox and HP extract should be avoided for the treatment of ESCC.
