A Comparison of Sonication and Microwave-assisted Extraction Method for Speciation of Arsenic in Fish Tissue, DORM-2 (어류중 비소의 종분화 분석을 위한 초음파 추출법과 마이크로파 추출법의 비교)

  • Comparison of a microwave-assisted extraction with sonication extraction was performed for arsenic speciation in fish tissue with chromatographic separation and inductively coupled plasma mass spectrometry detection. The detection limits of arsenicals with ultrasonic nebulizerand cross-flow nebulizer were shown to be similar. The arsenicals investigated were arsenobetaine (AsB), arsenite [As(III)], dimethylarsine acid (DMA), monomethylarsonic acid (MMA), arsenate [As(v)], and phenylarsonic acid (PAA). Quantitative extraction of arsenicals from dogfish muscle, DORM-2, standard reference material of NRCC (National Research Council of Canada) was achieved using 50% (v/v) methanol-water in both extraction methods. Extraction efficiency of arsenobetaine in both methods is greater than 82% with RSDs on replicates of less than 5%. The concentrations of AsB determined in extract of microwave assisted extraction and sonication methods were $14.18{\pm}0.42mg\;kg^{-1}$ and $13.54 {\pm}0.84mg\;kg^{-1}$, respectively. And the concentrations of DMA were $0.45{\pm}0.06mg\;kg^{-1}$ and $0.44{\pm}0.06mg\;kg^{-1}$, respectively.

  • This study was conducted to confirm the change in the contents of the ingredients according to the extraction method of nipa palm. The contents were analyzed by extraction according to the ethanol ratio, extraction time, and extraction temperature. The contents of polyphenols and flavonoids according to the ratio of the extraction solvent were the highest at 36.91 and 27.62 mg/g, respectively, when extracted with 50% ethanol. Polyphenols and flavonoids according to extraction temperature and extraction time showed the highest content of 40.83 and 37.63 mg/g, respectively, when extracted for 6 hours at 60℃. The contents of the major component of nipa palm according to the ethanol ratio were 2.08 mg/g in 70% ethanol for 5-O-caffeoylshikimic acid, 0.10 mg/g for 4-hydroxybenzoic acid in 30% ethanol, and 0.12 mg/g for 3,4- hydroxybenzoic acid in 50% ethanol. It is expected that it can be used as basic research data when developing natural materials such as food and cosmetics through the change in the contents of the ingredients contained in nipa palm according to the extraction methods.

A STUDY ON THE ANALYSIS OF VOLATILE ORGANIC ACIDS IN TOBACCO LEAVES (I) (잎담배중의 휘발성 유기산 분석에 관한 연구(I))

  • Extraction efficienties of volatile organic acids in tobacco leaves according to several extraction methods were investigated and contents of volatile organic acids in 16 kinds of tobacco leaf varieties were compared. Extraction efficiency according to simultaneous distillation and extraction was 5 to 10 times higher than that according to solvent extraction, steam distillation, essential oil extraction or continuous extraction and distillation. Total contents of volatile organic acids were 1.62 to 12.94mg per l00g of sample in aromatic tobacco varieties,0.12 to 2.08mg in flue-cured tobacco leaves, and trace in burleys. Among the Korean aromatic tobacco varieties, total contents of volatile organic acids in ST374-3 were the highest, 4.66mg per l00g of sample, and those in Sohyang and Hyangcho were low, 1.69mg and 1.62mg, respectively. Among flue-cured tobacco varieties, those in NC2326 were the highest, 2.08mg per 100g of sample, but those in the other varieties were not more than 0.80mg per 1 00g of sample. Total contents of volatile organic acids in acidic hydrolysis with 0.1 M tartaric acid were higher than in non-acidic condition in all kinds of tobacco varieties.

Optimization of Extraction Conditions for Total Phenolics from Sapium japonicum Using a Pressurized Liquid Extractor

  • Sapium japonicum was extracted by a pressurized liquid. Operating parameters such as the type and the ratio of solvent to water, temperature, pressure, and number of extractions were investigated as the main variables that influence the extraction efficiencies of total phenolics (TP). MeOH extracted the highest level of TP as 50.4 mg GAE/g compared to 48.8 and 27.2 mg GAE/g with $H_2O$ and EtOH, respectively. $EtOH:H_2O$ (40:60, v/v) was found to be the best solvent for TP extraction as 90.3 mg GAE/g compared to 85.0 and 84.3 mg GAE/g in 40:60 and 60:40 of $MeOH:H_2O$, respectively. TP were increased with the increase of the number of extraction steps. TP content was increased by 11% as the extraction temperature was increased from 40 (97.4) to $50{\circ}C$ (108.3 mg GAE/g). The optimum extraction conditions of TP were; extraction solvent, $EtOH:H_2O$ (40:60, v/v); temperature, $50{\circ}C$; pressure, 10.2 MPa; 2 extraction steps.

Mineral Carbonation of Serpentinite: Extraction, pH swing, and Carbonation (사문암(Serpentinite)을 이용한 광물탄산화: Mg 추출과 pH swing 및 탄산화)

  • Mineral carbonation by indirect method has been studied by serpentinite as cation source. Through the carbonation of $CO_2$ and alkaline earth ions (calcium and magnesium) from serpentinite, the pure carbonates including $MgCO_3$ and $CaCO_3$ were synthesized. The extraction solvent used to extract magnesium (Mg) was ammonium sulfate ($(NH_4)_2SO_4$), and the investigated experimental factors were the concentration of $(NH_4)_2SO_4$, reaction temperature, and ratio of serpentinite to the extraction solvent. From this study, the Mg extraction efficiency of approximately 80 wt% was obtained under the conditions of 2 M $(NH_4)_2SO_4$, $300^{\circ}C$, and a ratio of 5 g of serpentinite/75 mL of extraction solvent. The Mg extraction efficiency was proportional to the concentration and reaction temperature. $NH_3$ produced from the Mg extraction of serpentinite was used as a pH swing agent for carbonation to increase the pH value. About 1.78 M of $NH_3$ as the form of $NH_4{^+}$ was recovered after Mg extraction from serpentinite. And, the main step in Mg extraction process of serpentinite was estimated by geochemical modeling.

Lutein, β-Carotene, and Polyphenol Contents of Sweet Potato Leaves under Different Extraction Conditions (추출조건에 따른 고구마 잎의 Lutein, β-Carotene 및 Polyphenol 함량)

  • This study was carried out to determine the simultaneous extraction conditions of functional components (lutein, ${\beta}$-carotene, total polyphenol, flavonoids, and phenolic compounds) from sweet potato leaves and to evaluate the antioxidant activities. Extraction conditions included different ethanol concentrations (1st extraction: 99.9% ethanol; 2nd extraction: 50~90% ethanol) and times (30, 60, and 90 min). The highest values of lutein and ${\beta}$-carotene content were obtained by the 2nd extraction at an ethanol concentration of 90%. The extraction yields of lutein and ${\beta}$-carotene decreased with increasing extraction time. The maximum polyphenol, flavonoid, and total phenolic acid contents and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) and 1,1-diphenyl-2-picrylhydrazyl radical scavenging activities were 32.3 mg gallic acid equivalent/g, 17.0 mg catechin equivalent/g, 2,842.6 mg/100 g, 17.0 mg ascorbic acid equivalent/g, and 1.94 mg/mL ($IC_{50}$) at the 2nd extraction with an ethanol concentration of 60%. The optimum extraction conditions were as follows; ethanol concentrations of the extraction solvent were 99.9% (1st extraction) and 60% (2nd extraction), and extraction time was 30 min.

Changes in Component of Five-fruit Tea, with Various Extraction Time (오과차(five-fruit tea)의 추출시간에 따른 성분 변화)

  • This thesis is to analyze the ingredients of five-fruit tea depending on extraction time as a basic experiment for quality improvement and industrialization of traditional Korean beverage. According to extraction time, the acidity of the tea is 2.4% at 30 minutes, 2.3% at 50 minutes and 1.7% at 70 minutes, and soluble solid is 0.22 g, 0.31 g and 0.41 g. The pH of five-fruit tea, which is 4.70, 4.85 and 4.98 at each time, and total sugar is 54.01 m9, 108.82 m9 and 142.92 mg as extraction time increases. The total amount of free sugars is 1.14 mg, 1.36 mg and 2.17 mg, and glucose, surcose and fructose which are 98.2%, 92.6% and 92.6% of the whole percentage occupy the most part of free sugars. The total free amino acids content increases to 84.94 mg, 99.67 mg and 120.40 mg. The five kinds of amino acids like serine, glutamic acid, threonine, proline and alanine amount to 94.5%, 93.6% and 94.7% at all. she variation in the percentage of free amino acids content in accordance with extraction time is as follows : threonine, extracted for 50 minutes, is reduced considerably and glutamic acid decreases gradually as extraction time increases. When proline is extracted for 50 minutes, it's rate of increase Is most high.

Optimization of Supercritical Fluid Extraction of Tocotrienol from Grape Seed (초임계유체 추출을 이용한 포도씨 tocotrienol 추출조건 최적화)

  • In this study, supercritical carbon dioxide extraction (SFE) was utilized for the extraction of tocotrienol from grape seeds. The optimal conditions for vitamin E and tocotrienol extraction were determined via response surface methodology (RSM). Central composite design was utilized to assess the effects of oven temperature (30-$50^{\circ}C$, X1), operating pressure (17-25 MPa, X2), and extraction time (1-5 hr, X3) of supercritical fluid extraction. Vitamin E and tocotrienol contents were 8.65 mg/100 g and 7.88 mg/100 g at $40^{\circ}C$, 20MPa and 5 hr, respectively. The predicted extraction condition was validated via actual experimentation. The predicted extraction conditions were $40^{\circ}C$, 3.8 hr, and 20.7MPa. The vitamin E and tocotrienol contents under these conditions were 8.20 mg/100 g and 7.42 mg/100 g, respectively. The vitamin E and tocotrienol contents of solvent extraction with hexane were 8.18 mg/100 g and 7.24 mg/100 g, respectively.

[Retracted] Optimization of Jirisan Mountain Cudrania tricuspidata leaf substance extraction across solvents and temperatures

  • Objective: The aim of this study is to optimize the extraction of beneficial substance from Cudrania tricuspidata leaves grown at Jirisan Mountain in South Korea by three different solvents depending on extraction time and at different temperature. Methods: The total phenolic contents were determined by the method reported by $S{\acute{a}}nchez$-Moreno et al. The total flavonoid contents were analyzed by Slinkard and Singleton. The DPPH radical scavenging activity was determined according to the method reported by Blois Results: The extraction yield for each solvent is 9.05-14.1%, 2.17-5.67%, and 2.3-3.9% for D.W., ethanol, and hexane, respectively. The overall results were maximized for the extract obtained with D.W. for 5 min at $100^{\circ}C$. The average phenol contents were 77.11, 45.64, and 0.343 mg/g at $100^{\circ}C$ in water, $78^{\circ}C$ in ethanol, and $68^{\circ}C$ in hexane, respectively. The flavonoid contents were the highest in the materials extracted with D.W., and were increased with increasing temperature, regardless of the extraction solvents, whether water (green), polar organic ethanol, or nonpolar organic hexane. In the ethanol extract, the flavonoid contents are increased gradually from 5.66 mg/g to 7.73 mg/g. The total flavonoid contents were proportional to the concentrations of the water extracts, ranging from 4.14 mg/g to 48.89 mg/g. The antioxidative activities of the water-extracted compounds are generally increased with increasing temperature from 42.5% to 85.5%. Those of the hexane extracts are increased slowly from 3.79% to 8.8%, while those of ethanol extracts are increased from 29.8% to 47.4%. Conclusion: The extraction yields were dependent upon solvents for extraction as well as extraction time and the temperature. The optimal extraction time was 5 min and the extraction yields were increased with increasing temperature excepted hexane. Of the three tested extraction solvents, the greenest solvent of water shows excellent results, suggesting that water is among the most effective solvents for natural sample extractions for general medicinal, pharmaceutical, and food applications.

Optimization of Extraction Conditions of Salidroside, Hepatoprotective Compound from Acer Tegmentosum (산겨릅나무 간 보호 화합물 salidroside의 추출조건 최적화)

  • Salidroside is a major phenolic glycoside of Acer tegmentosum (Aceraceae) and known to be a hepatoprotective compound. Extraction conditions are one of the most important factors to save time and cost from an economic point of view. For this reason, this study was conducted to optimize the extraction condition for maximum yield of salidroside. For optimization, three extraction factors such as ethanol concentration (%), extraction temperature (℃), and solvent to material ratio (mg/mL) were tested and optimized for maximum yield of salidroside using response surface methodology (RSM). The optimal condition was obtained as an ethanol concentration of 53.4%, an extraction temperature at 67.11℃ and a solvent to material ratio (mg/mL), 195.55 mg/ml. The salidroside yield under optimal conditions was found to be 1.59 mg/g dried samples, which were well-matched with the predicted value of 1.56 mg/g dried samples. These results will provide useful information about optimized extraction conditions for the development of salidroside as hepatoprotective therapeutics.
