Yak-kong and Soybean Induced Expression of Osteoprotegerin in MG-63 Human Osteoblastic Cells Requires Estrogen Receptor-$\beta$

  • Phytoestrogens, especially Yak-kong or soybean-derived isoflavones have been traditionally used as a supplement of estrogen for preventing postmemopausal osteoporosis in oriental folk medicine. In our previous study, the treatment of Yak-kong and soybean increased estrogen receptor-a (ERa) expression and proliferation of MG-63 osteoblastic cells. In contrast, the increase of estrogen receptor-$\beta$ (ER$\beta$) expression in proliferating MG-63 cells with Yak-kong and soybean treatment was less pronounced, which suggested that ER$\beta$ may play a role rather in the regulation of bone cell differentiation To determine the role of ER$\beta$ in Yak-kong or soybean mediated regulation of bone cell differentiation, we established MG-63 cell lines stably expressing either ER$\beta$ or antisense ER$\beta$ RNAs. Increased expression of ER$\beta$ did not affect ERa expression and proliferation of MG-63 cells. However, increased expression of ER$\beta$ in MG-63 cells (ER$\beta$-MG63 cells) selectively enhanced Yak-kong or soybean induced expression of osteoprotegerin (OPG), a novel soluble glycoprotein which is secreted from osteoblasts and mediates the signal for osteoclast differentiation. Inhibition of ER$\beta$ expression by antisense ER$\beta$ RNAs (As-ER$\beta$-MG63) caused these cells to insensitize Yak-kong or soybean induced expression of OPG but increased MG-63 cell proliferation. Furthermore, the comparable effects between Yak-kong and the combined treatment of genistein and daidzein at $0.5{\times}l0^{-8}$ M, which is a concentration of these two isoflavones similar to Yak-kong at 0.001 mg/mL, on OPG expression in ER$\beta$-MG63 cell demonstrate that the enhanced expression of OPG with Yak-kong treatment is mediated by the synergistic effect of low leveled isoflavones in the extracts. Together, coupled with low level of ER expression in osteoclasts, our data demonstrate that ER$\beta$ in osteoblasts plays an important role in Yak-kong and soybean mediated inhibition of osteoclast differentiation indirectly by enhancing the expression of OPG.

  • Aspalatone, a new antithrombotic agent, was administered orally to pregnant Sprague-Dawley rats during the organogenetic period at dose levels of 0, 20, 100 and 500 mg/kg/day. All dams were subjected to caesarean section on day 20 of pregnancy. Effects of test substance on dams and embryonic development of F1 fetuses were examined There were treatment-related decreases in body weight and food consumption in the 500 mg/kg group. There was a increase in the spleen weight in the 100 and 500 mg/kg groups. Develo-pmental toxicity was evident as decreased fetal body weights and increased fetal malformations in the 500 mg/ kg group. External and skeletal malformations of fetuses occurred at an incidence of 1 and 8.2%, respectively. In addition, there was a delay in ossification of sternebrae and sacrocaudal vertebrae in the 500 mg/kg group. The results show that the no observed adverse effect dose level (NOAEL) for maternal toxicity was 20 mg/kg/ day and for developmental toxicity was 100 mg/kg/day.

Trace Metals In Several Edible Marine Algae Of Korea

  • The present paper deals with nine kinds of trace metal contents among 79 samples, more than 20 species of edible marine algae collected during 1973∼1975 along the coasts of Korea. Among the metals detected by the atomic absorption spectrometer, Mg content is variable in 2.09∼13.84mg/g-dry on an average and 5.31mg/g-dry in a mean of green, brown and red algae, whereas Fe is 0.148∼2.343 and 0.747mg/g-dry; Mn 0.015∼0.191 and 0.063mg/g-dry; Zn 0.007∼0.082 and 0.030 mg/g-dry; Cu 0.005∼0.054 and 0.017mg/g-dry; Pb 0.0017∼0.082 and 0.0043mg/g-dry Co 0.0009∼0.0024 and 0.0016mg/g-dry; Cd 0.0002∼0.0034 and 0.0011mg/g-dry, and Cr is 0.0007∼0.0074 and 0.0021mg/g-dry, both on an average and in a mean respectively. The trace metal contents in the seaweeds investigated are variable according to species, collecting seasons, places and kinds of metal It is interesting that in Sargassum thunbergii some metal contents such as Fe, Mn, and Zn are specially high in September1974 through the year investigated.

  • A conventional oxide method was used to fabricate $Ba_{0.5}Sr_{0.5}TiO_3$(BST) ceramic plates doped by MgO from 10 to 60 wt%. The structural and dielectric properties of BST were investigated as a fraction of MgO dopant concentration. The dielectric properties of the MgO doped BST were strongly dependent on the MgO contents. The dielectric constant and dielectric loss of MgO doped BST decreased with increasing MgO content.

Purification and Characterization of a Thermophilic Cellulase from a Novel Cellulolytic Strain, Paenibacillus barcinonensis

  • A novel bacterial strain, MG7, with high cellulase activity was isolated and identified by morphological characteristics and molecular phylogeny analysis as Paenibacillus barcinonensis. Maximum production of cellulase by MG7 was observed at pH 7.0 and $35^{\circ}C$. The enzyme was purified with a specific activity of 16.88 U/mg, the cellulase activity was observed in a zymogram, and its molecular mass (58.6 kDa) was confirmed by SDS-PAGE. The purified enzyme showed maximum activity at pH 6.0 and $65^{\circ}C$ and degraded cellulosic substrates such as carboxy methyl cellulose (CMC), Avicel, filter paper, and ${\beta}$-glucan. The enzyme showed stability with 0.5% concentration of various surfactants. The $K_m$ and $V_{max}$ of cellulase for CMC and Avicel were found to be 0.459mg/ml and 10.46mg/ml/h, and 1.01 mg/ml and 10.0 mg/ml/h, respectively. The high catalytic activity and its stability to temperature, pH, surfactants, and metal ions indicated that the cellulase enzyme by MG7 is a good candidate for biotechnological applications.

Effect of Polygonatum Odoratum on Lowering Lipid and Antioxidation (옥죽(玉竹)의 지질강하 및 항산화효과)

  • Objectives : This study was designed to examine the effects of Polygonatum odoratum EtOH ext. on lowering lipid and anti-oxidation using hyperlipidemic rat. Methods : Male rats weighting $195.21{\pm}4.93g$ were divided into 4 groups and fed high fat diet for 8 weeks. Each of 7 rats was divided into a control and sample group. We fed a control group of rats a basal diet and administered normal saline(100 mg/kg, 1 time/1 day) for 4 weeks. And we fed each experimental group of rats basal diet and administered an extract of Polygonatum odoratum(100 mg/kg, 200 mg/kg, 300 mg/kg, 1 time/1 day) for 4 weeks. At the end of the experiment, the rats were sacrificed to determine their chemical composition. We measured lipid of plasma and liver, concentration of anti-oxidative activity and tumor necrosis factor-$\alpha$($TNF-{\alpha}$). Results : 1. Concentration of plasma free fatty acid, low density lipoprotein-cholesterol showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. Concentration of plasma total cholesterol, triglyceride showed a significant decrement in all Polygonatum odoratum EtOH ext. groups than that of control group. However, concentration of plasma high density lipoprotein-cholesterol was not significantly different in all the treatment groups. 2. Concentration of liver total cholesterol showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. Concentration of liver triglyceride(TG) showed a significant decrement in all Polygonatum odoratum EtOH ext. groups than that of control group. 3. Concentration of plasma thiobarbituric acid reactive substance, and liver thiobarbituric acid reactive substance showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. 4. The values of glutathione peroxidase activity showed a significant increment in all Polygonatum odoratum EtOH ext. groups than that of control group. The values of superoxide dismutase activity and catalase activity showed a significant increment in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. 5. The values of plasma aspartate aminotransferase and alanine aminotransferase activities were not significantly different in all treatment groups. 6. Concentration of liver $TNF-{\alpha}$ showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. Conclusions : Based on the results in this study, the Polygonatum odoratum EtOH ext. showed a positive effect in lowering lipid and anti-oxidation.

Comparative Analysis of the Amino Acids and Effective Components from Viola mandshurica (자화지정(Viola mandshurica)의 아미노산과 유용성분 비교 분석)

  • This study compared amino acids and effective components of Viola mandshurica produced in Korea with those of Chinese plants, as part of a study on nutritional andfunctional materials, to aid in the development of a valuable food. The contents of reducing sugar, free sugar, polyphenols, and soluble protein in the flowers of V. mandshurica were higher than in other fractions, being 10,485.11 mg%, 118.85 mg%, 1,259.26 mg%, and 23.67 mg%, respectively. Total flavonoid content of Chinese leaves was the highest at 2,370.75 mg%, followed by 1,844.43 mg% in Korean leaves. Free amino acids were highest in Korean leaves (5,345.21 mg%), followed by roots (4,692.24 mg%), flowers (3,807.87 mg%), and leaves of China plants (3,613.81 mg%). Arginine accounted for 55% of free amino acids in the roots, and all plant fractions contained more valine, aspartic acid, glutamic acid, alanine, and arginine, than other amino acids. The total content of amino acid derivatives was the highest (9,058.80 mg%) in Korean leaves. Especially, $\alpha$-aminoadipic acid constituted over 50% of total amino acid derivatives in all plant parts of V. mandshurica. When mineral contents were analyzed, leaves grown in China showed the highest content of 5,917.57 mg%, and the contents of K, Ca, and Mg were much greater than those of other minerals, together comprising over 95% of total mineral content in leaves and flowers from Korea.

Chemical Composition of Rose Petals (Rosa hybrida L.) As A Food Material (식품 소재로서 장미꽃의 화학성분 조성)

  • To evaluate a flower of rose, Rosa hybrids L. as a new food material, its chemical composition was analysed. The contents of crude protein, crude fat, crude ash and crude fiber in rose petals were 16.3, 2.9, 5.4, 16.1% on dry basis, respectively. Free sugars showed 74.3 mg/g of fructose, 49.6 mg/g of glucose and 16.6 mg/g of xylose. The contents of ${\beta}$-carotene and ascorbic acid were 205.2 ug/100 g and 129.5 mg/100 g, respectively. The major minerals of rose petals were K, P, Mg, Ca, Na and Fe, and among them K was the most abundant as 1,981.7 mg/100 g. The major amino acids were aspartic acid as 4,007.3 mg/100 g, glutamic acid as 1,114.8 mg/100 g, lysine as 672.6 mg/100 g and leucine as 661.0 mg/100 g. Fatty acids were mainly unsaturated fatty acids as 76.3%.

Screening of Antiviral Medicinal Plants against Avian Influenza Virus H1N1 for Food Safety

  • Various extracts from 30 medicinal plants were evaluated for their antiviral activity against influenza virus A/Puerto Rico/8/34 (H1N1) and cytotoxicity in MDCK cell culture. The plant material (30 g) was extracted with methanol (300 mL) at room temperature for 24 h, after which the methanolic extracts were filtered, evaporated, and subsequently lyophilized. Evaluation of the potential antiviral activity was conducted by a viral replication inhibition test. Among these medicinal plants, Tussilago farfara, Brassica juncea, Prunus armeniaca, Astragalus membranaceus, Patrinia villosa, and Citrus unshiu showed marked antiviral activity against influenza virus A/H1N1 at concentrations ranging from 0.15625 mg/mL to 1.25 mg/mL, 0.3125 mg/mL to 10 mg/mL, 5 mg/mL to 10 mg/mL, 0.625 mg/mL to 10 mg/mL, 0.625 mg/mL to 10 mg/mL, and 0.3125 mg/mL to 5 mg/mL, respectively. The extracts of Tussilago farfara showed cytotoxicity at concentrations greater than 2.5 mg/mL, whereas the other five main extracts showed no cytotoxicity at concentrations of 10 mg/mL. Taken together, the present results indicated that methanolic extracts of the six main plants might be useful for the treatment of influenza virus H1N1.

Preparation and thermodynamics consideration of MgO-Al spinel by self-propagation high- temperature synthesis (자전고온연소합성법에 의한 MgO-Al 스피넬 제조 및 열역학적 고찰)

  • Self-propagating high temperature synthesis (SHS) technique was used to synthesize the spinel phase of $MgAl_2O_4$ from MgO and Al powder. Thermit reaction products of MgO and Al, The reaction products were heat treated at the temperature $800^{\circ}C$ preheating. Processing factors such as DTA/TG, combustium product and maxium temperature, synthesis of MgO and Al from "$MgO+2Al+3/2O_2$\rightarrow$MgAl_2O_4$". An activation energy (${\Delta}H^{\circ}$)-264.8 kcal/mol and reaction of maxium temperature 5634 K was calculated to form a $MgAl_2O_4$ spinel from unreacted materials. Pellet were increased volume 6% after thermit reaction. reaction.
