Pitting Characteristics and Electrochemical Polarization Behaviors in Al-Cu-Si-Mg-Ag-Zr Alloys with Ageing (Al-Cu-Li-Mg-Ag-Zr합금의 시효에 따른 전기화학적 분극 거동과 공식특성)

  • In this paper, we studied on both electrochemical polarization behaviors and pitting characteristics of ultra high strength Al-Cu-Li-Mg-Ag-Zr alloys(named C1 and C2) and 2090 alloy according to their treatments in the deaerated 3.5% NaCl, using by the potentiodynamic and the potentiostatic method, SEM micrograph and surface roughness including depth of pitting attack. With the cyclic polarization curves, the hysteresis of the C1 and C2 alloys appeared more remarkably than that of the 2090 alloy, because of precipitation microstructural difference between C1, C2 alloys and 2090 alloy. In the pitting experiments, the correlations between pitting growth and aging conditions were analyzed with the SEM micrograph and measurement of the pit depth.

The Properties of Diffraction Efficiency in Polarization Holography using the Ag and MgF2/AsGeSeS Multi-layer (Ag 및 MgF2/AsGeSeS 다층박막에서의 편광 홀로그래피 회절효율 특성)

  • We have carried out two-beam interference experiments to form holographic gratings on As$_{40}$ Se$_{15}$ S$_{35}$Ge$_{10}$ single layer, Ag/As$_{40}$ Se$_{15}$ S$_{35}$Ge$_{10}$ multi-layer. In this study, holographic gratings have been formed using He-Ne laser(632.8nm) under different polarization combinations(intensity polarization holography, phase polarization holography). The diffraction efficiency was obtained by the +lst order intensity. The maximum diffraction efficiency of As$_{40}$ Se$_{15}$ S$_{35}$Ge$_{10}$ single layer, As$_{40}$ Se$_{15}$ S$_{35}$Ge$_{10}$ and MgF$_2$/As$_{40}$ Se$_{15}$ S$_{35}$Ge$_{10}$ multi-layer were 0.8%, 1.4% and 3.1% under intensity polarization holography, respectively.

Selective leaching of valuable metals (Au, Ag etc.) from waste printed circuit boards (PCB)

  • This study was carried out to recover gold, silver and other valuable metals from the printed circuit boards (PCB) of waste computers. PCB samples were crushed to under 1mm by a shredder and initially separated into 30% conducting and 70% non-conducting materials by an electrostatic separator. The conducting materials, which contained the valuable metals, were then used as the feed material for magnetic separation where it was found that 42% was magnetic and 58% non- magnetic. The non-magnetic materials contained 0.227mg/g Au and 0.697mg/g Ag. Further leaching of the non-magnetic component using 2.0M sulfuric acid and 0.2M hydrogen peroxide at 85$^{\circ}C$ extracted more than 95% copper, iron, zinc, nickel and aluminium. Au and Ag were not extracted in this solution, however, more than 95% of Au and 100% of Ag were selectively leached with a mixed solvent (0.2M ammonium thiosulfate, 0.02M copper sulfate, 0.4M ammonium hydroxide). Finally, the residues were reacted with a NaCl solution to leach out Pb while sulfuric acid was used to leach out Sn. Recoveries reached 95% and 98% in solution, respectively.

Ultrastructural Alterations in the Gastric Mucous Epithelial Cells of Mouse Inoculated with Ehrlich Carcinoma Cells, Induced by 5-Fluorouracil, Mitomycin C or Acriflavine-Guanosine Compound (AG60) (5-Fluorouracil, Mitomycin C 및 Acriflavine-Guanosine 복합제가 Ehrlich 암세포를 이식한 생쥐 위점막 점액상피세포의 미세구조에 미치는 영향)

  • This experiment was performed to evaluate the morphological responses of the gastric epithelial cells of the mouse, inoculated with Ehrlich carcinoma cells in the inguinal area, following administration of 5-fluorouracil, mitomycin C or Acriflavine-Guanosine compound (AG60). In this study, each mouse was inoculated with $1{\times}10^7$ Ehrlich carcinoma cells subcutaneously in the inguinal area. From next day after inoculations, 0.2 mL of saline, 5-fluorouracil (30 mg/kg), mitomycin C ($400{\mu}g/kg$) or AG60 (30 mg/kg) were injected to the animals every other day, respectively. Each animals were sacrificed after 7th injection and tissue were taken from the gastric mucosa. Thereafter, the ultrathin sections were stained with uranyl acetate and lead citrate. In the 5-fluorouracil-, mitomycin C- or AG60-treated mice, myelin figures and multivesicular bodies within the gastric mucous epithelial cells were observed more frequently than those of the normal control. In the 5-fluorouracil-treated mice, membrane structures containing a few mucous granules in the luminal space were observed. Indeed, bulging cytoplasmic process containing mucous granules protruding into the gastric lumen were observed in the mitomycin Ctreated mice. Therefore, this study suggested that AG60 as compared with 5-fluorourail and mitomycin C may effective medicine without damage to the secretion ability of gastric mucous epithelial cells.

Physicochemical Properties of Recominant Hepatitis B Surface Antigen Expressed in Mammalian Cell(C127)

  • The physicochmical properties of recombinant hepatitis B surface antigen (r-HBsAg), which was expressed in C127 mammalian cell were studied. Using roller bottle culture in DMEM supplemented with fetal bovine serum, 10-15 mg/L of r-HBsAg was produced with about 31% of purification yield. The purity of r-HBsAg by HPLC was 99.8% and electron microscopic examination showed homogeneous spherical particle with 22 nm in diameter, a morphological characteristic of HBsAg. The density of r-HBsAg by CsCI density gradient method was 1.19g/ml and the isoelectric point by Mono $P^{TM}$ HR 5/20 column was 4.6. The analysis of subunit protein pattern using SDS-PAGE followed by scanning densitometry gave 81.3% of S protein and 18.7% of pre-S protein. fluorophore-assisted-carbohydrate-electrophoresis analysis showed the relative amount of carbohydrate to protein was 1.7% and it smajr component was N-acetyl glucosamine, which was about 39% of total carbohydrate. The relative amount of lipid to protein determined by vanillin phosphoric acid method was 32.5% and its major component was phospholipid, which was about 70% of total lipid. The physicochemical properties of C127 mammalian cell-derved r-HBsAg are similar to those of p-HBsAg, suggesting that the r-HBsAg can be used in developing a new preventive vaccine against hepatitis B.

Screening of Antifungal Medicinal Plants for Turfgrass Fungal Disease Control (잔디 병해 방제를 위한 항균성 약용식물의 탐색)

  • Brown patch (Rhizoctonia solani AG1-1), dollar spot (Sclerotinia homoeocarpa), pythium blight (Pythium spp.), anthracnose (Colletotrichum graminicola), yellow patch (Rhizoctonia cerealis) and Zoysia patch (Rhizoctonia solani AG2-2) are the major turfgrasses diseases in Korea. In this study, 23 medicinal plant extracts were tested for antifungal activities against turfgrass pathogenic fungi. In paper disk diffusion method, 12 medicinal plant extracts, including Sophora flavescens, showed antifungal activity. Also, in the test of antifungal activity on media contained the extracts of S. flavescens, Curcuma longa, Rheum undulatum, Coptis chinensis and Asiasarum sieboldi showed above 80% inhibitory effects on the mycelial growth in 110 mg/10 ml concentration of the extracts. S. flavescens, in particularly, showed antifungal activity against the six turfgrass pathogenic fungi. The inhibition rate of S. homoeocarpa was 100% in 10 mg/10 ml, 5 mg/10 ml and 2 mg/10 ml concentrations of C. longa extract. In case of Pythium spp., the extracts of S. flavescens, R. undulatum and C. chinensis showed 100 % inhibition rate on the test media.

Soil Mineral Nutrients and Microbes Are Responsible for Large Patch Disease Caused by Rhizoctonia solani AG2-2 in Zoysiagrass Turf (골프장 한국잔디의 Rhizoctonia solani AG2-2에 의한 Large Patch 발생 토양에서 근권 미생물과 무기영양 평가)

  • Mineral nutrients and population dynamics of soil microbes in the root zones of zoysiagrass infected by Rhizoctonia solani AG2-2 and that of healthy plants were sampled from ten golf courses using a cup cutter(diameter $10\;cm\;{\times}\;8\;cm$ deep). Analysis of variance(ANOVA) showed significant differences in content of $NO_3$-N(P = 0.05), $NH_4$-N(P = 0.1), and K(P = 0.1) between infected and healthy samples. The content of $NO_3$-N in the soils of large patch was 9.49 mg/kg and that in soil of healthγ plants was 7.02 mg/kg. However, the content of $NH_4$-N in the soil of large patch was 12.02 mg/kg whereas 14.40 mg/kg for the soil under the healthy plants. The content of K in the soil of large patch was lower than that of soil of healthy plants. There was few numbers of Pseudomonas colonies In the soils of large patch compared to that of healthy plants. These results indicated that the content of $NO_3$-N, NH4-N, and K and the microbial population dynamics in root zones correlated to occurrence of large patch.

In Vitro Plant Regeneration for Mass Propagation of Epimedium koreanum Nakai (삼지구엽초의 다량번식을 위한 기내 식물체 분화)

  • As an endeavor to establish a micropropagation system for Epimedium koreanum Nakai., this study was carried out to define methods to disinfect its explants and media for callus induction, proliferation and plant regeneration. The lowest infection rates by fungi or bacteria on apical and axillary bud explants of rhizome were observed when they were immerged in 0.3% NaOCl solution for 20 min after soaked in 0.1% $AgNO_3$ solution for 30 min, but leaf explants were seldom infected with fungi or bacteria by this disinfectant method. The highest rate of plantlet formation was obtained from the explants disinfected in 0.3% NaOCl solution for 20 min after soaked in 0.1% $AgNO_3$ solution for 60 min for tip buds and in 0.1 % $AgNO_3$ solution for 30 min for axillary buds of rhizome. Induction rate of callus was the highest from the explants disinfectd in 0.3% NaOCl solution for 20 min after soaked in 0.2% $AgNO_3$ solution for 15 min. Callus growth was proper in a modified 1/2 MS medium including half strength of $NH_4NO_3$ with $0.02-0.2mg{\cdot}L^{-1}$ BA and $2.0mg{\cdot}L^{-1}$ NAA. Low rate of plantlet regeneration was obtained in 1/2 UM or 1/2 White medium with $2.0mg{\cdot}L^{-1}$ BA and $0.2mg{\cdot}L^{-1}$ AA.
