Production of Saponin by Hairy Root Cultures of Ginseng (Panax ginseng C.A. Meyer) Transformed with Agrobacterium rhizogenes (Agrobacterium rhizogenes에 의하여 형질전환된 인삼(Panax ginseng C.A. Meyer)의 모상근 배양에 의한 Saponin 생산)

  • Cultures of hairy root induced from ginseng(Panax C.A. Meyer) transformed with Agrobacterium rhizogenes (strain A4, ATCC 15834) were established and morphologically two different hairy root strains (HB1, HB2) were obtained. To determine the optimum growth rate, the hairy root (HB2) was cultured in various liquid medium supplemented with or without plant growth hormone. The growth rate of hairy root cultured on MS medium was 1.3-3.1 times higher than those cultured on other media, and the optimum sucrose concentration and pH were 3-6%, 5.5-6.5, respectively. Also, the growth rate of hairy root was increased when 0.02 M ammonium nitrate, 1.2 mM potassium phosphate (monobasic) and 0.5 mg/l IBA were supplied to liquid medium. The saponin patterns and contents of hairy root (HB2) were determined by TLC and HPLC. The crude saponin contents were 4.67% and the total saponin contents were 1.0%, on dry weight basis.

  • Induction and culture of hairy roots from ginseng(Panax ginseng C. A. Meyer) roots discs by A. rhizogenes strain $A_4$ were studied. After 6-12 weeks infected with A. rhizogenes tumor and hairy roots emerged from the root discs. The ratio of hairy root induction on root discs was higher in 5-year old than in 3, 4, and 6-year old ginseng. On treatment with IAA, IBA, 2, 4-D and tryptophan, hairy roots formation showed a significant increase at 15-30mg/1 tryptophan treated. Subsequently, hairy roots were cultured on hormone-free RCM medium(pH 4.5, sucrose 30g/1).

  • This study was conducted to investigate the characteristics of nonstarchy polysaccharides in Korean ginseng, (Panax ginseng C.A. Meyer). The results obtained are as follows. 1. The total sugar content of ginseng roots were decreased with increasing the cultural period. On the other hand, the crude fiber content was increased with that of the ginseng leaves or stems. But the crude fiber in root was much less than that of leaves and stems. 2. The dietary fiber content of ginseng root on 5 years old was 14.20% as neutral detergent fiber, 9.08% as acid detergent fiber, hemicellulose 5.12%, cellulose 7.98% and lignin of 1.10%, respectively. 3. Much more pectin was found in ginseng roots which was cultivated for shooter Period. And it was contained much more in the root than in the leaves and stems. 4. ginseng hemicellulose content in root was 5% to 10%. It was decreased with increasing: cultivated period. Hemicellulose was constituted of xylose, arabinose, glucose, rhamnose and xylose of these sugars was the predominant. 5. X-ray diffraction Pattern of ginseng cellulose showed maximum intensity at tile interplanar angle of 4.1$^{\circ}$.

  • Kastle-Meyer (KM) test is one of presumptive tests used to detect latent blood in crime scenes. While this method is more sensitive than others, false positive reaction can be shown where blood does not exist. In this study, we tried to suggest a new application method to solve this problem. Reaction time and aspect of reaction of this new method was compared with two conventional methods in order to identify the effectiveness and identifiability. As a result, there was no statistically significant difference in sensitivity between the methods. In addition, in case of blood with a dilution ratio of 5000:1 or less, positive reaction showed immediately, making it easy to distinguish the reaction from false positive reaction. However, it became difficult to distinguish them with the reaction time as the dilution ratio increased, and this phenomenon could be supplemented observing aspect of the reaction when using the new method. Therefore, this study suggested a new method for KM test that can be used more accurately in the field.

Effects of virtual reality training on upper extremity function and activities of daily living in patients with sub-acute stroke (가상현실 훈련이 아급성 뇌졸중 환자의 상지기능과 일상생활활동에 미치는 효과)

  • The aim of this study was to investigate the effects of virtual reality training on upper extremity function and activities of daily living in patients with sub-acute stroke. The present study enrolled 18 patients with sub-acute stroke. All subjects were assigned into either the experimental group (n=9) or control group (n=9). Both groups received conventional occupational therapy for 30 minutes/day, 5 times a week, for 4 weeks. Additionally, the experimental group performed virtual reality training in each session for 30 minutes/day, and the control group conducted conventional occupational therapy in each session for 30 minutes/day. The outcome measures were performed through the Fugl-Meyer Assessment (FMA) and the Korean-modified Barthel Index (K-MBI) before and after intervention. In results, the experimental group showed significant improvements in the scores of FMA and K-MBI after intervention (p<.05). The control group showed significant improvements in the shoulder/elbow/forearm, wrist, and hand sub-domains of the FMA and K-MBI (p<.05). After intervention, the experimental group showed significantly greater improvements in the total score and in the wrist and hand sub-domains of the FMA than control group (p<.05). These findings suggest that virtual reality training may have positive effects on the improvements of upper extremity function in patients with sub-acute stroke.
