Amino acid digestibility in plant protein sources fed to growing pigs

  • Objective: The objective was to determine standardized ileal digestibility (SID) of amino acids (AA) in 11 plant protein sources fed to growing pigs. Methods: Eleven feed ingredients used were sesame meal, two sources of soybean meal (SBM) produced in the Republic of Korea, a source of SBM produced in India, high-protein distillers dried grains (HPDDG), perilla meal, canola meal, copra meal, corn germ meal, palm kernel expeller, and tapioca distillers dried grains (TDDG). Experimental diets were prepared to contain each test ingredient as a sole source of AA, and a nitrogen-free diet was also prepared to estimate the basal ileal endogenous losses of AA. Twelve barrows surgically fitted with T-cannulas at the distal ileum with an initial body weight of 29.0 kg (standard deviation = 3.0) were individually housed in metabolism crates equipped with a feeder and a nipple drinker. A $12{\times}9$ incomplete Latin square design was employed with 12 experimental diets, 12 animals, and 9 periods. After a 5-d adaptation period, ileal digesta were collected on d 6 and 7 in each experimental period. Results: Values for apparent ileal digestibility of most indispensable AA in three sources of SBM were greater compared with other test ingredients except HPDDG and canola meal (p<0.05). Pigs fed diets containing SBM sources had also greater SID of most indispensable AA compared with those fed diets containing other test ingredients (p<0.05) except for HPDDG and canola meal. There was no difference in the apparent ileal digestibility and SID of AA among sources of SBM. The TDDG had the least value for the SID of methionine among test ingredients (p<0.05). Conclusion: The SID of most AA in SBM, HPDDG, and canola meal were greater than those in sesame meal, perilla meal, copra meal, and TDDG.

  • This study was conducted to examine a mixture of plant protein sources as a fish meal (FM) substitute. Two feeding trials were carried out using similar dietary formulations but different FM levels. In Experiments 1 and 2, the basal diets were formulated to contain 65% and 60% of FM, respectively. The other five diets were formulated replacing FM by 10, 15, 20, 25 and 30% with a mixture of soybean meal, wheat gluten and soy protein concentrate. Three synthetic amino acids (lysine, threonine and methionine) were added to the test diet. Groups of fish in experiment 1 ($6.76{\pm}0.03g$) and experiment 2 ($32.5{\pm}0.1g$) were fed one of the experimental diets for 7 and 9 weeks, respectively. Each experiment was carried out in triplicate. There were no significant differences among groups in terms of growth performance, feed utilization, survival or hematological parameters in either experiment. The results indicated that a mixture of soybean meal, wheat gluten and soy protein concentrate, supplemented with three synthetic amino acid, can replace fish meal by up to 30% in diets for juvenile olive flounder.

자돈에서 대체 단백질 원료사료의 외관상 회장 아미노산 소화율 및 표준 회장 아미노산 소화율 평가 (Evaluation of Apparent Ileal and Standardized Ileal Digestibility of Amino Acid in Feed Ingredients to Alterate the Protein Sources for Weaned Pigs)

  • 본 시험은 돼지 사료 내 단백질원으로 사용되는 원료사료의 외관상 회장 아미노산 소화율과 표준 회장 아미노산 소화율을 평가하기 위해 수행하였다. 본 시험에서 T-cannula를 회장에 설치한 자돈(Landrace×Yorkshire×Duroc, 14.4±0.35kg) 6두를 공시동물로 사용하였으며, 6×6 라틴스퀘어 설계법(Latin square design)을 이용하였다. 평가 원료사료로서 옥수수 배아, 옥수수 글루텐 밀, 옥수수 글루텐 피드, 클로렐라 및 라이신 부산물이 사용하였으며, 각 원료사료별 5일의 적응기간과 2일의 소화물 채취기간을 두었다. 라이신의 외관상 회장 소화율에서 클로렐라와 라이신 부산물이 유의적으로 높았으며(p<0.05), 각각 79.1% 및 78.7%이였다. 메치오닌의 외관상 회장 소화율은 원료사료들 간의 유의적인 차이가 없었다. 트레오닌의 외관상 회장 소화율은 클로렐라가 유의적으로 높게 나타났으며(p<0.05), 81.8%이였다. 라이신의 표준 회장 소화율에서 클로렐라, 라이신 부산물 및 옥수수 글루텐밀이 유의적으로 높게 나타났으며(p<0.05), 각각 80.4%, 79.5% 및 77.0%이였다. 메치오닌의 표준 회장 소화율은 원료사료들 간의 유의적인 차이가 없었다. 트레오닌의 표준 회장 소화율은 클로렐라가 라이신 부산물, 옥수수 글루텐 피드에 비해 유의적으로 높게 나타났으며(p<0.05), 84.2%이였다. 본 연구결과, 클로렐라와 라이신 부산물의 외관상 및 표준 회장 아미노산 소화율이 가장 우수하였으며, 이 원료들은 돼지 사료 내 단백질 대체 원료사료로서 가치가 있음을 시사한다.

벼 잎집무늬마름병균의 균사생장 및 균핵형성에 미치는 질소원의 효과 (Effect of Nitrogen Sources on Mycelial Growth and Sclerotial Formation of Rhizoctonia solani Causing Rice Sheath Blight)

  • 벼 잎집무늬마름병균(Rhizoctonia solani Kuhn)의 생리적 특성을 조사하기 위하여 충북지역을 중심으로 채집 분리한 60여 Rhizoctonia 균중 균사생장 및 균핵형성에 특징이 있는 3개 균주를 선발하여 그들의 배양적 특성, 병원성 및 균핵형성에 미치는 질소원의 효과를 시험하였다. 이들 분리균주에서 PSA 배지상의 균사생장 및 균핵형성 등의 배양적 특성에 따라 선발한 3개 분리균주의 병원성은 균주간에 달랐다. 질소원으로서 Arginine, Alanine, Urea, Ammonium sulfate 및 Sodium nitrate 등은 균사생장 및 균사형성에 효과적이었으나 Cystine, Methionine, Lysine, Histidine, Tryptophan 및 Tyrosine 등은 뚜렷한 효과가 없었다. Urea, Sodium nitrate, Agrinine, Ammonium sulfate 및 Lysine 등은 질소원의 농도가 낮게 첨가된 배지에서 균사생장이 양호하였으나 Proline, Histidine, Alanine은 높은 농도 수준에서 양호하였으며 균핵형성은 모든 공시 질소원의 높은 농도 수준에서 양호하였다. 균사생장 및 균핵형성에 양호한 질소원인 Arginine, Alanine, Urea, Ammonium sulfate는 병원성도 증가시켰다. Proline 첨가 배지에서는 각 균주 공히 큰 균핵이 형성되었으며 Tryptophan을 함유한 배지에서 각 균주 모두 생육이 저조하였고, 배지의 갈변화가 가장 심하였다.

Effects of Different Sources of Dietary Chromium on Growth, Blood Profiles and Carcass Traits in Growing-finishing Pigs

  • This study was carried out to evaluate the effects of dietary supplementation of different sources of chromium on growth performance, blood profile and carcass trait in growing-finishing pigs. A total of 200 growing pigs (Landrace${\times}$Yorkshire)${\times}$Duroc, average initial weight 8.5 kg) were allotted to 5 treatments with 4 replicates per treatment and 10 pigs per replicate. Five treatments were designated as follows according to the source of chromium. i) Control (No chromium): corn-soybean meal based basal diet, ii) $CrCl_{3}$: control diet+200 ppb Cr as $CrCl_{3}$, iii) CrPic: control diet+200 ppb Cr as Cr picolinate, iv) CrMet-1: control diet+100 ppb Cr as Cr methionine, and v) CrMet-2: control diet+200 ppb Cr as Cr methionine. After the feeding trial, three pigs per replicate (12 pigs per treatment) were slaughtered for the evaluation of carcass traits. Average daily gain (ADG), average daily feed intake (ADFI), and feed: gain ratio (F/G) were not different (p>0.05) among dietary Cr sources. However, whole-period ADG of pigs fed CrPic, CrMet-1 and CrMet-2 diets was higher (p<0.05) than for the control diet. Nutrient digestibility was not different (p>0.05) among dietary Cr sources, but the nutrient digestibility of pigs fed CrPic, CrMet-1 and CrMet-2 diets was higher (p<0.05) than for the control diet. BUN level decreased with more magnitude (p<0.05) in pigs fed Cr during the 20 to 50 kg period. Although both serum cholesterol and triglyceride were different (p<0.05) among treatments, there was no consistent response that could be related to the dietary Cr sources regardless of growth phase. However, the overall data suggested that serum cholesterol level increased as BW of pigs increased. Blood total protein (TP) increased (p<0.05) in pigs fed Cr only during the 90-110 kg phase, and blood creatinine (Creat) level was higher in $CrCl_{3}$ and CrPic treatments than in the control only during the 90-110 kg phase. Backfat thickness was thinner (p<0.05) in pigs fed CrMet-2 than in the control treatment. Therefore, lean percentage was higher (p<0.05) in CrMet-2 than in control pigs. However, dressing percentage and Longissimus muscle area (LMA) were not different (p>0.05) among treatments. In conclusion, dietary supplementation of 200 ppb Cr, via either CrPic or CrMet, improved pig growth performance and nutrient digestibility. Moreover, dietary CrMet supplementation for the growing-finishing pig is evidently remarkable for improving both lean percentage of the carcass and backfat thickness.

Serratia marcens Arylsulfatase의 정제와 성질 (Purification and Properties of Arylsulfatase of Serratia marcescens)

  • Arylsulfatase는 간단한 Phenols류의 황에스테르 화합물로부터 S $O_{4}$$^{[-992]}$ - 이온의 유리를 촉매한다. 이 효소는 토양센균을 포함한 많은 미생물과 동식물의 조직등에 널리 분포하여 있으며 이와 같은 넓은 분포는 이 효소의 기본적 기능이 환경학적으로 매우 중요한 의미를 갖는다고 하겠다. Arylsulfatase에 대한보고는 Klebsiella sp를 사용하여 몇몇 보고가 있다. 본 연구는 6종의 Serratia sp를 사용하여 arylsulfatase 합성조건을 검토하고 효소의 정제조건과 성질에 대하여 조사하여 Serratia marcescens를 선정하였다. Serratia mrcescens는 탄소원으로서 xylose rhamnose, glucosamine 그리고 arabinose등과 같은 몇몇 당을 이용하지 못했으며 glucose와 mannitol을 잘 이용하였으나 glucose methioniue의 경우 효소 합성을 억제시키었다. 유황원으로서는 무기유황염과 methionine의 첨가는 억제되었으며 tyramine의 첨가에 의해서 효소 함성의 억제효과는 해제되었다. 효소의 정제는 황산암모늄 포화용액의 분획과 DEAE-Cellulose, CM-Cellulose 그리고 DEAE-Sephadex A-25로 연결되는 구분 분획에 의해서 행하여졌다. 효소의 분자량은 SDS-gelelectrophoresis와 Sephadex G-100 column chromatography에 의하여 각각 46,000과 49,000으로 측정되었고 최적 PH는 6.8이었다. P-Nitrophenyl sulfate를 사용한 Km과 Vmak치는 각각 2.5$\times$$10^{4-}$M과 20 nmoles/min/mg protein이었다. 기질에 대한 특성은 phenylsulfte와 ο-, p-nit-rophenyl sulfate 그리고 p-nitro catechol sulfate에 대해서 높은 활성을 보였다. Hydroxylamine, inorganic fluoride, sulfide 그리고 Phosphate등은 강한 효소 저해작용을 나타내었고 무기유산염은 저해작용을 보여주지 않았다. Tyramine, octopamine그리고 dopamine과 같은 amino acid 또한 강한 저해 작용을 보였다.

민산초나무 종실(種實)의 유지(油脂) 및 단백질조성(蛋白質組成)에 관한 연구(硏究) (Analytical Studies on the Composition of Oil and Protein in the Seed of Thornless Chinese Pepper Zanthoxylum schinifoilum var. inermis Nakai)

  • 야생식용유자원(野生食用油資源)의 개발(開發)을 위하여 민산초나무 종실(種實)의 일반성분(一般成分)과 종실유(種實油)의 이화학적(理化學的) 성상(性狀)을 A. O.AC. 법(法)으로, 종실유(種實油)의 지방산(脂肪酸) 조성(組成) 및 종실단백질(種實蛋白質)의 amino 산조성(酸組成)을 GLC법(法) 및 amino acid auto analyzer 로 각각 분석(分析) 측정(測定)한 결과(結果)를 다음과 같이 적요(摘要)한다 민산초나무의 종실(種實)은 비교적 다량(多量)의 유지(油脂)(47.05%), 단백질(蛋白質)(20.14%) 및 회분(灰分)(3.1%)을 함유(含有)하고 있는 영양가(榮養價)가 높은 반식물(飯食物) 또는 식용유(食用油)로 인정(認定)되었다. 민산초나무 종실(種實)에는 극미량(極微量)의 유황(硫黃)(0.02%)과 cyan 화합물(化合物)(0.00005%)이 함유(含有)되였으나 모두 한계량이하(限界量以下)로서 식용유(食用油) 및 생과(生菓)로서 이용이 안전(安全)하다. 종실유(種實油)는 옥도가(沃度價) 145.2로서 건성유(乾性油)에 속(屬)하며 산가(酸價), 감화가, 굴절율(屈折率)등 이화학적(理化學的) 성질(性質)도 불포화고급(不飽和高級) 지방산조성(脂肪酸組成)에서 특성(特性)을 나타냈다. 종실유(種實油)의 지방산(脂肪酸) 조성(組成)은 oleic acid(34.8%), linoleic acid(25.1%), linolenie acid(26.1%)등 필수지방산(必須脂肪酸)의 함량(含量)이 80%이상으로서 식용유(食用油)에 적합(適合)한 것으로 인정(認定)되였다. 종실단백질(種實蛋白質)에서 17종(種)의 amino산(酸)을 분석정량(分析定量)하였으며, lysine, threonine, valine, methionine, isoleucine, leucine, phenylalanine등 필수(必須) amino산(酸)의 함량(含量)이 비교적 높았고, glutamic acid의 함량(含量)(4.985%)이 특히 높아서 단백질원(蛋白質源)으로서 유용(有用)하다고 인정(認定)되었다.

변형번데기 농축단백질 아미노산 조성 및 주사전자현미경에 의한 관찰 (Amino Acid Composition and Scanning Electron Micrographs of Modified Silkworm Larvae Protein Concentrates)

  • 제사공장에서 부산물로 나오는 번데기는 단백질의 함량이 높을 뿐 아니라 필수 아미노산 조성도 우수하여 영양적으로 좋은 단백질원으로 알려져 있다. 이에 번데기로 농축단백질을 조제해서 무수초산과 호박산으로 아실화시켜 아미노산조성 및 주사전자현미경에 의한 관찰을 대두농축단백질과 비교 검토하여 다음과 같은 결과를 얻었다. 1. 번데기 농축단백질의 단백질 함량은 84.1%로서 대두농축단백질의 70.3%보다 높았다. 2. 번데기 농축단백질은 아미노산 조성에 있어서 대두농축단백질보다 필수 아미노산 함량이 높았으며 특히 대두에 부족한 methionine은 4.21(g/100g)로 매우 높았고, F.A.O.의 scoring pattern과 비교시 전체적으로 필수 아미노산이 함량이 높았다. 또한 번데기 농축단백질은 alanine, valine, phenylanine, isoleucine 등 지방족 아미노산의 함량이 높게 나타났다. 한편 변형번데기단백질에서는 아미노산의 함량에 약간의 손실을 가져왔으며 숙시닐화에서 더 큰 경향을 나타내었고 특히 lysine의 함량이 현저하게 감소되었다. 3. 주사전자현미경에 의한 관찰에서 탈지번데기분은 작고 입자가 많이 모여 하나의 agglomerate의 형태로 거칠고 세포성이 그대로 보존되어 있다. 기포의 생성이 많았으며 입자가 불균일(10~$20\mu{m}$)하였다. 번데기 농축단백질은 탈지번데기분보다 세포성이 많이 파괴되어 기포의 생성도 적어 입자의 표면적이 어느정도 넓고 부드러웠다. 또한 변형번데기농축단백질에서는 현저하게 세포성이 파괴되고 가 감소하여 좋은 조직을 형성하였으며 특히 숙시닐화한 단백질에서는 아주 부드럽고 좋은 조직을 나타내었다.

Determination and Prediction of the Amino Acid Digestibility of Sunflower Seed Meals in Growing Pigs

  • This experiment was conducted to evaluate the chemical composition and amino acid (AA) digestibility of sunflower seed meal (SFSM) and to use this data to develop prediction equations for estimating AA digestibility for growing pigs. Ten SFSM were collected from five provinces in China. Twelve barrows ($38.8{\pm}4.6kg$), fitted with ileal T-cannula were allotted into two $6{\times}6$ Latin square designs. Each of six experimental periods comprised a 5-d adaption period followed by a 2-d collection of ileal digesta. The ten test diets contained 50% SFSM as the sole source of AA. Another nitrogen-free diet was used to measure the basal endogenous losses of crude protein (CP) and AA. Chromic oxide (0.3%) was used as an inert marker in each diet. There was considerable variation (CV>10%) among the ten SFSM in chemical composition (dry matter [DM]). The concentration of CP and ether extract (EE) ranged from 29.33% to 39.09% and 0.88% to 11.33%, respectively. Crude fibre (CF), neutral detergent fibre and acid detergent fibre ranged from 21.46% to 36.42%, 38.15% to 55.40%, and 24.59% to 37.34%, respectively. There was variation among the ten SFSM in apparent ileal digestibility (AID) and standardized ileal digestibility (SID) for lysine and threonine, which ranged from 63.16 to 79.21 and 55.19% to 72.04% for AID and 67.03% to 82.07% and 61.97% to 77.01% for SID, respectively. The variation in CP and methionine ranged from 60.13% to 74.72% and 74.79% to 88.60% for AID and 66.70% to 79.31% and 77.16% to 90.27% for SID, respectively. Methionine was a good indicator to predict AA digestibility. These results indicate that conventional chemical composition of SFSM was variable (CV>10%) among the ten SFSM (DM). The results of AID, SID and prediction equations could be used to evaluate the digestibility of SFSM in growing pigs.

Apparent Digestibility of Amino Acids, Energy and Proximate Nutrients in Dietary Protein Sources for Young Pigs

  • This experiment was conducted to determine apparent ileal and fecal digestibilities of protein sources for young pigs (15.6 kg BW) which were fitted with simple ileal T-cannulas. Experimental diets included one of the followings as a sole protein source: spray-dried plasma protein (SDPP), spray-dried blood meal (SDBM), soybean meal (SBM), isolated soy protein (ISP), dried skim milk (DSM), and fish meal (FM). The digestibilities of dry matter (DM), gross energy (GE) and crude protein (CP) in DSM-based diet were the highest (p < 0.05), and those of SDBM and SBM diets were the lowest at the terminal ileum. The average ileal digestibilities of essential amino acids (AAs) were 88.3, 84.7, 84.6, 83.4, 83.0 and 77.7% for pigs fed the DSM, ISP, SDBM, SBM, SDPP and FM diets, respectively. There were no significant differences in overall apparent ileal AAs digestibilities in DSM and ISP diets. Digestibilities of essential AAs in DSM, however, were significantly higher than those of ISP (p < 0.05). Among the essential AAs, the digestibility of arginine was higher except for SDBM or DSM, than other AAs. The ileal digestibilities of lysine, methionine and threonine were higher (p < 0.05) in pigs fed SDBM or DSM diet than in those fed SBM diet. Digestibilities for SDPP or ISP diet in these AAs were similar except for threonine which was higher (p < 0.05) for SDPP diet. The apparent ileal digestibilities of lysine and methionine ranged from 79.6 (FM) to 92.5 (DSM) and 75.4 (SDPP) to 88.7 (DSM), respectively. The apparent fecal digestibilities of average essential AAs were 90.4, 89.1, 86.0, 84.4, 84.2 and 80.8% in pigs fed the DSM, ISP, SDBM, SDPP, SBM and FM diets, respectively. The fecal digestibilities of essential AAs were higher (p < 0.05) in pigs fed the DSM and ISP. Lysine digestibility in pigs fed the DSM or ISP diet was higher (p < 0.05) than that of pigs fed SBM diet. Apparent essential AAs digestibility was the highest (p < 0.05) in pigs fed DSM diet, and was the lowest (p < 0.05) in pigs fed FM diet. Digestiibilities of essential AAs and nonessential AAs were higher when measured over the total digestive tract than at the end of the small intestine except for several AAs in SDPP, SDBM and SBM diets. In conclusion, whether it is fecal or ileal, the digestibilities of proximate nutrients and AAs in pigs fed DSM and ISP diet were the highest, and those of pigs fed FM was the lowest.
