Allosteric Probe-Based Colorimetric Assay for Direct Identification and Sensitive Analysis of Methicillin Resistance of Staphylococcus aureus

  • The accurate and rapid detection of methicillin-resistance of Staphylococcus aureus (SA) holds significant clinical importance. However, the methicillin-resistance detection strategies commonly require complicated cell lysis and gene extraction. Herein, we devised a novel colorimetric approach for the sensitive and accurate identification of methicillin-resistance of SA by combining allosteric probe-based target recognition with self-primer elongation-based target recycling. The PBP2a aptamer in the allosteric probe successfully identified the target MRSA, leading to the initiation of self-primer elongation based-cascade signal amplification. The peroxidase-like hemin/G-quadruplex undergo an isothermal autonomous process that effectively catalyzes the oxidation of ABTS2- and produces a distinct blue color, enabling the visual identification of MRSA at low concentrations. The method offers a shorter duration for bacteria cultivation compared to traditional susceptibility testing methods, as well as simplified manual procedures for gene analysis. The overall amplification time for this test is 60 min, and it has a detection limit of 3 CFU/ml. In addition, the approach has exceptional selectivity and reproducibility, demonstrating commendable performance when tested with real samples. Due to its advantages, this colorimetric assay exhibits considerable potential for integration into a sensor kit, thereby offering a viable and convenient alternative for the prompt and on-site detection of MRSA in patients with skin and soft tissue infections.

  • Objectives : Methicillin-resistant Staphylococcus aureus (MRSA) has a cephalosporin and beta-lactam antibiotic-resistant strains. MRSA is one of the major pathogens causing hospital infection and the isolation ratio of MRSA has gradually increased. Consequently, increased resistance to antibiotics is causing serious problems in the world. Therefore, there is a need to develop alternative antimicrobial drugs for the treatment of infectious diseases. Methods : The antibacterial activities of Ipyo-san were evaluated against 2 strains of MRSA and 1 standard Methicillin-susceptible staphylococcus aureus (MSSA) strain by using the disc diffusion method, minimal inhibitory concentrations (MIC) assay, colorimetric assay using MTT test, checkerboard dilution test and time-kill assay performed under dark. Results : The MIC of Ipyo-san water extract against S. aureus strains ranged from 1000 to $2,000{\mu}g/ml$, so we confirmed that it had a strong antibacterial effect. Also, the combinations of Ipyo-san water extract and conventional antibiotics exhibited improved inhibition of MRSA with synergy effect. We suggest that Ipyo-san water extract against MRSA has antibacterial activity so it has potential as alternatives to antibiotic agents. For the combination test, we used Triton X-100 (TX) and DCCD for measurement of membrane permeability and inhibitor of ATPase. As a result, antimicrobial activity of Ipyo-san water extract was affected by the cell membrane. Conclusions : We suggest that the Ipyo-san water extract lead the treatment of bacterial infection to solve the resistance and remaining side-effect problems that are the major weak points of traditional antibiotics.

  • Methicillin-Resistant Staphylococcus aureus (MRSA) is a cephalosporin and beta-lactam antibiotic-resistant strain. In most cases, MRSA is spread from infected patients and infection rates are growing increasingly. Furthermore, increased resistance to antibiotics is causing serious problems in the world. Staphylococcus aureus is responsible for both nosocomial and community-based infections that range from relatively minor skin and soft tissue infections to life-threatening systemic infections. Therefore, there is a need to develop alternative antimicrobial drugs for the treatment of infectious diseases. In this study, we investigated antimicrobial activity of oriental medicine prescription against MRSA. The minimum inhibitory concentration (MIC) of Sohamhyung-tang water extract against S. aureus strains ranged from 500 to 2,000 ${\mu}g/mL$, so we have it confirmed that a strong antibacterial effect. Also, the combinations of Sohamhyung-tang water extract and conventional antibiotics exhibited improved inhibition of MRSA with synergy effect. We suggest that Sohamhyung-tang water extract against MRSA have antibacterial activity, it has potential as alternatives to antibiotic agent. We suggest that the Sohamhyung-tang water extract lead the treatment of bacterial infection to solve the resistance and remaining side-effect problems that are the major weak points of traditional antibiotics.

임상검체로부터 분리된 methicillin 내성 Staphylococcus aureus의 독소 및 항생제 내성 (Toxins and Antibiotic Resistance of Methicillin-Resistant Staphylococcus aureus Isolated from Clinical Specimens)

  • 2009년 7월부터 12월까지 순천 소재 한 병원에 내원한 환자의 검체로부터 methicillin 내성 Staphylococcus aureus (MRSA) 75균주와 methicillin 감수성 S. aureus (MSSA) 24균주를 분리하였다. 분리균의 항생제 감수성 조사는 디스크 확산법을 사용하여 측정하였다. 분리균의 독소 유전자 보유는 multiplex PCR을 이용하여 장독소(enterotoxin; SE), 독성 쇼크 증상 독소 1(toxic shock syndrome toxin-1; TSST-1), 피부박탈성 독소(exfoliative toxin; ET) 및 백혈구 용해 독소(Panton-Valentine leukocidin; PVL) 유전자를 검출하였다. 분리된 MRSA 60개 균주는 1개 혹은 2개의 독소 유전자를 가지고 있으며, 22.7%의 균주가 seb, sec, seg, sei와 tst 유전자를 동시에 보유하고 있었으며 18.7%는 sec, seg, sei와 tst 유전자를 동시에 보유하고 있었다. 백혈구 용해독소를 암호하는 pvl 유전자는 검출되지 않았다. MRSA는 sec, seg, sei와 tst 유전자 보유에 높은 상관성을 보였다. MRSA 균주들은 erythromycin (분리균의 89%), gentamicin (70.7%), ciprofloxacin (69.3%), clindamycin (61.3%)과 tetracycline (58.7%)에 내성이 높은 반면, MSSA 균주들은 erythromycin를 제외한 다른 항생제에는 민감하였다. 독소 유전자 seb, sec와 tst는 tetracycline 내성과 높은 상관관계가 있었다.

Prevalence and Molecular Characterization of Methicillin-Resistant Staphylococcus aureus from Nasal Specimens: Overcoming MRSA with Silver Nanoparticles and Their Applications

  • Staphylococcus aureus is a cause of high mortality in humans and therefore it is necessary to prevent its transmission and reduce infections. Our goals in this research were to investigate the frequency of methicillin-resistant S. aureus (MRSA) in Taif, Saudi Arabia, and assess the relationship between the phenotypic antimicrobial sensitivity patterns and the genes responsible for resistance. In addition, we examined the antimicrobial efficiency and application of silver nanoparticles (AgNPs) against MRSA isolates. Seventy-two nasal swabs were taken from patients; MRSA was cultivated on Mannitol Salt Agar supplemented with methicillin, and 16S rRNA sequencing was conducted in addition to morphological and biochemical identification. Specific resistance genes such as ermAC, aacA-aphD, tetKM, vatABC and mecA were PCR-amplified and resistance plasmids were also investigated. The MRSA incidence was ~49 % among the 72 S. aureus isolates and all MRSA strains were resistant to oxacillin, penicillin, and cefoxitin. However, vancomycin, linezolid, teicoplanin, mupirocin, and rifampicin were effective against 100% of MRSA strains. About 61% of MRSA strains exhibited multidrug resistance and were resistant to 3-12 antimicrobial medications (MDR). Methicillin resistance gene mecA was presented in all MDR-MRSA strains. Most MDR-MRSA contained a plasmid of > 10 kb. To overcome bacterial resistance, AgNPs were applied and displayed high antimicrobial activity and synergistic effect with penicillin. Our findings may help establish programs to control bacterial spread in communities as AgNPs appeared to exert a synergistic effect with penicillin to control bacterial resistance.

Antibacterial activity of Chamaecyparis obtuse Extract and Profile of Antimicrobial Agents Resistance for Methicillin-Resistant Staphylococcus aureus

  • In vitro antimicrobial activities of hot water extracts of Chamaecyparis obtuse, for methicillin-resistant Staphylococcus aureus (MRSA) was compared to commonly used conventional antimicrobial agents. All MRSA was susceptible to linezolid or vancomycin, but also to erythromycin. MIC range and MIC90 to erythromycin, clindamycin, levofloxacin, tetracycline for MRSA were each 4 ㎍/mL, 2 ~ >128 ㎍/mL, ≤0.06 ~ >128 ㎍/mL, 0.25 ~ >128 ㎍/mL, 0.25~64 ㎍/mL and 4 ㎍/mL, .128 ㎍/mL, >128 ㎍/mL, >128 ㎍/mL, 64 ㎍/mL. The hot water extracts of leaf of C. obtuse had the lowest MIC range, MIC50, and MIC90 (0.125 µL/mL) for the MRSA tested, and it was possible more potent than various conventional antimicrobial agents. Screen antibacterial drug candidate with high antibacterial activity such as derivatives of C. obtuse leaf extract such as terpinen-4-ol or using combined therapy with commercialized antibacterial agents will likely be helpful in treating refractory MRSA infections.

In Vitro Activity of Taurine-5-Bromosalicylaldehyde Schiff Base Against Planktonic and Biofilm Cultures of Methicillin-Resistant Staphylococcus aureus

  • Staphylococcus aureus is a major human pathogen, implicated in both community and hospital acquired infections. The therapy for methicillin-resistant Staphylococcus aureus (MRSA) infections is becoming more difficult because of multidrug resistance and strong biofilm-forming properties. Schiff bases have attracted attention as promising antibacterial agents. In this study, we investigated the in vitro activity of taurine-5-bromosalicylaldehyde Schiff base (TBSSB) against MRSA. The minimum inhibitory concentrations (MIC) and minimum bactericidal concentration (MBC) were determined using a microtiter broth dilution method. TBSSB effectively inhibited planktonic MRSA, with an MIC of $32{\mu}g/ml$. The time-kill curve confirmed that TBSSB exhibited bactericidal activity against MRSA. TBSSB was also found to significantly inhibit MRSA biofilm formation at 24 h, especially at $1{\times}MIC$ and sub-MIC levels. Furthermore, scanning electron microscopy and transmission electron microscopy showed remarkable morphological and ultrastructural changes on the MRSA cell surface, due to exposure to TBSSB. This study indicated that TBSSB may be an effective bactericidal agent against MRSA.

Target Recognition Triggered Split DNAzyme based Colorimetric Assay for Direct and Sensitive Methicillin-Resistance Analysis of Staphylococcus aureus

  • The accurate and rapid detection of methicillin-resistant Staphylococcus aureus (MRSA) holds significant clinical importance. This work presents a new method for detecting methicillin-resistant Staphylococcus aureus (S. aureus) in clinical samples. The method uses an aptamer-based colorimetric assay that combines a recognizing probe to identify the target and split DNAzyme to amplify the signal, resulting in a highly sensitive and direct analysis of methicillin-resistance. The identification of the PBP2a protein on the membrane of S. aureus in clinical samples leads to the allosterism of the recognizing probe, and thus provides a template for the proximity ligation of split DNAzyme. The proximity ligation of split DNAzyme forms an intact DNAzyme to identify the loop section in the L probe and generates a nicking site to release the loop sequence ("3" and "4" fragments). The "3" and "4" fragments forms an intact sequence to induce the catalytic hairpin assembly, exposing the G-rich section. The released the G-rich sequence of LR probe induces the formation of G-quadruplex-hemin DNAzyme as a colorimetric signal readout. The absorption intensity demonstrated a strong linear association with the logarithm of the S. aureus concentration across a wide range of 5 orders of magnitude dynamic range under the optimized experimental parameters. The limit of detection was calculated to be 23 CFU/ml and the method showed high selectivity for MRSA.

  • Methicillin resistance Staphylococcus aureus (MRSA) is a gram-positive bacterium, the most commonly isolated bacterial human pathogen. JiYu-san is one of the natural products used to treat diseases in the folk recipe. In this study, we investigated the antimicrobial activity of EtOH 70% extracts of JiYu-san (JYS) against MRSA. The antibacterial activity of JYS against MRSA strain was evaluated using minimum inhibitory concentration (MIC), checkerboard dilution test, and time-kill assay. The effect of JYS on the immune mechanism of MRSA was confirmed through cell membrane permeability tests and energy metabolism tests, and the antibacterial activity mechanism was performed using qRT-PCR and western blot. As a result, in the antibacterial test of JYS, the MIC was measured to be 1.9~1000 ㎍/mL, and synergistic or showed a partial synergistic effect. In addition, JYS showed antibacterial activity in a combination test with DCCD or TX-100. In a study on the mechanism of action of antibacterial activity, it was found that JYS suppressed MRSA resistance genes and proteins. These results suggest that JYS has antibacterial activity and provides great potential as a natural antibiotic by modulating the immune mechanism against MRSA.

Detection of Inducible Clindamycin Resistance Genes (ermA, ermB, and ermC) in Staphylococcus aureus and Staphylococcus epidermidis

  • The aim of the present study was to survey the frequency of inducible and constitutive phenotypes and inducible cross-resistant genes by regulating the methylation of 23S rRNA (ermA, ermB, and ermC) and macrolide efflux-related msrA gene in Staphylococcus aureus and S. epidermidis strains. A total of 172 bacterial isolates (identified based on standard tests), were examined in this study. Antibiotic susceptibility was determined by the disk diffusion method, and all isolates were evaluated with respect to inducible and constitutive phenotypes. The presence of ermA, ermB, ermC, and msrA genes was investigated by a PCR assay. The constitutive resistance phenotypes showed a higher distribution among the isolates. R phenotype was detected more among S. epidermidis isolates (46.25%). ermB, ermC, and msrA genes were detected more in methicillin-resistant S. aureus (MRSA) and methicillin-resistant S. epidermidis (MRSE) isolates that had R and HD phenotypes (>77% strains). The ermA gene had the lowest frequency among MRSA, MRSE, MSSA, and MSSE strains (<14% isolates). Distribution of inducible resistance genes in MRSA and MRSE strains, and possibly other species, leads to increased constitutive resistance to erythromycin, clindamycin, and other similar antibiotics. Therefore, it can be challenging to treat infections caused by these resistant strains.
