섬개야광나무의 부위별 메탄올 추출물의 항산화 및 항염 활성 비교 (Comparison of Anti-oxidant and Anti-inflammatory Activities of Methanolic Extracts Obtained from Different Parts of Cotoneaster wilsonii Nakai)

  • Background: The Ministry of Environment, Korea recognizes Cotoneaster wilsonii Nakai as a second-class endangered plant. It is a native species that grows in Ulleung-do, Korea. To our knowledge, the bioactivity of this plant has not yet been reported. Therefore, in this study, we have reported the bioactivity of C. wilsonii Nakai. Methods and Results: The anti-oxidant activities of C. wilsonii methanolic extract were investigated in vitro. The anti-oxidant activity was evaluated using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical assay, and the total phenolic and flavonoid content were measured. The leaf methanolic extract had the highest DPPH radical scavenging activity ($IC_{50}$; $15.74{\mu}g/m{\ell}$), and it also had the highest total phenolic and flavonoid content ($220.95mg{\cdot}GAE/g$, and $36.46mg{\cdot}QE/g$ extract respectively). Through high performance liquid chromatography (HPLC) analysis, chlorogenic acid was found to be the predominant among all phenolic compounds, showing a concentration of 84.24 mg/g extract. More than 60% decrease in nitric oxide (NO) production was found in the leaf methanolic extract. Conclusions: To the best of our knowldege, this is the first report of the bioactivities of C. wilsonii. The results demonstrate that the leaf methanolic extract of C. wilsonii shows potent anti-oxidant, and anti-inflammatory activities.

Antihepatotoxic Activity of Cassia tora Leaf Extract

  • Methanolic extract of Cassia tora leaves was evaluated for its hepatoprotective activity in rats by inducing hepatotoxicity with paracetamol (acute model). The extract at a dose of 400 mg/kg orally exhibited significant protective effect by lowering the serum levels of transaminase (SGOT and SGPT), bilirubin, and alkaline phosphatase (ALP). The effects produced were comparable to that of a standard hepatoprotective agent.

A Preliminary Study on Hypocholesterolemic and Hypoglycemic Activities of Some Medicinal Plants

  • Total cholesterol level in mice with hypercholesterolemia was determined after intraperitoneal administration of the methanolic extract of some medicinal plants. From the data obtained, it was suggested that the methanolic extract of Elaeagnus crispa, Ixeris dentata, Prunus davidiana, Eriobotrya japonice, Aralia elata and Phragmites communis produced a significant hypocholesterotemic effect. In the case of the extract of Saussurea diamantiaca, on the other band, the total cholesterol level was markedly increased. The methanolic extract of Ixeris dentata, Prunus davidiana and Phragmites communes also decreased the level of blood glucose in alloxan-diabetic male albino mice while that of Eriobotrya japonica, Allium tuberosum, Houttuynia cordata and Eucommia japonica did not produce this effect.

Hepatoprotective and antioxidant activity of the methanolic extract of Mucuna pruriens Bak. roots

  • The methanolic extract of natural roots of Mucuna pruriens was screened for hepatoprotective and antioxidant effects in carbon tetrachloride ($CCl_4$) induced hepatotoxicity in Albino rats. The degree of protection was measured by estimating biochemical parameters like serum glutamate oxaloacetate transaminase, serum glutamate pyruvate transaminase, total protein and level of serum bilirubin (both total and direct). The extract also reduced $CCl_4$ induced lipid peroxidation both in-vitro and in-vivo. Hepatoprotective activity of methanolic extract at dose 300 mg/kg body weight, i.p., was comparable with standard drug Silymarin (25 mg/kg, i.p.). Furthermore, histopathological experiments were also carried out to support the study.

Antifungal Activity of Methanolic of Centella asiatica and Andrographis panicuiata

  • The antifungal activity of methanolic extracts of Centella asiatica and Andrographis paniculata leaves was observed against fourteen fungi, viz., Alternaria alternata, A. brassicae, A. brassicicola, A. solani, A. tenuissima, Cercospora blumae, Curvularia lunata, C. penniseti, and Drechslera monoceras, D. oryzae, D. turitica, Fusarium albizziae and F. udum. Different concentrations of the methanolic extract (1000, 2000, 3000, 5000, 7000, 10000ppm) were used. The effect of mixed leaf extract (1500 ppm of C. asiatica + 1500 ppm of A. paniculata) and its 1:2 ad 1:4 dilutions were also studied. The individual extracts of both the plants showed significant inhibitory effect on spore germination of all the fungi tested. F. udum, F. albizzae, D. oryzae, D. turtica, and D. monoceras were particularly sensitive to these extracts. In general, the extract of C. asiatica showed a higher inhibitory effect in all concentrations against all the fungi as compared to A. paniculata, except for A. brassicae A. solani, D. oryzae, D. penniseti and Curvularia sp. The inhibitory effect of extracts increased when they were used in combination with or without dilutions against A. brassicicola, A. solani A. brassicae, A. alternata, A. tenussima, C. blumae, C. lunata, C. penniseti and Curvularia species. Higher efficacy of active ingredient of these extracts under field condition is envisaged against plant pathogens.

Assai 열매 메탄올 추출물의 항산화 효과 (Antioxidant Effect of Assai Palm Methanolic Extract)

  • Assai 열매추출물의 자유 라디칼 소거능을 측정하기 위하여 DPPH를 이용한 자유 라디칼 소거능 실험을 수행하여 항산화 효과를 측정한 결과, assai열매추출물의 농도가 높을수록 DPPH활성이 뛰어남을 알 수 있었고, ROS를 이용하여 항산화 효과를 확인하였다. 배양된 대식세포에 assai열매추출물을 농도별로 첨가한 결과, 농도가 높을수록 과산화수소에 의해 유도된 산화적 자극이 감소하였다. 또한 세포 생존에 미치는 영향을 알고자 assai열매추출물을 농도별로 첨가하여 24시간후 세포의 형태변화를 MTT assay로 실시한 결과, 산화적 자극에 의해 발생한 세포 손상이 assai 열매추출물의 농도가 높아질수록 감소하는 것이 확인되었다. 따라서 assai열매추출물은 천연 항산화제로서의 가능성을 보였다.

Chemical Composition and Quorum Sensing Inhibitory Effect of Nepeta curviflora Methanolic Extract against ESBL Pseudomonas aeruginosa

  • Objectives: Bacterial biofilm is regarded as a significant threat to the production of safe food and the arise of antibiotic-resistant bacteria. The objective of this investigation is to evaluate the quorum sensing inhibitory effect of Nepeta curviflora methanolic extract. Methods: The effectiveness of the leaves at sub-inhibitory concentrations of 2.5, 1.25, and 0.6 mg/mL on the virulence factors and biofilm formation of P. aeruginosa was evaluated. The effect of N. curviflora methanolic extract on the virulence factors of P. aeruginosa, including pyocyanin, rhamnolipid, protease, and chitinase, was evaluated. Other tests including the crystal violet assay, scanning electron microscopy (SEM), swarming motility, aggregation ability, hydrophobicity and exopolysaccharide production were conducted to assess the effect of the extract on the formation of biofilm. Insight into the mode of antiquorum sensing action was evaluated by examining the effect of the extract on the activity of N-Acyl homoserine lactone (AHL) and the expression of pslA and pelA genes. Results: The results showed a significant attenuation in the production of pyocyanin and rhamnolipid and in the activities of protease and chitinase enzymes at 2.5 and 1.25 mg/mL. In addition, N. curviflora methanolic extract significantly inhibited the formation of P. aeruginosa biofilm by decreasing aggregation, hydrophobicity, and swarming motility as well as the production of exopolysaccharide (EPS). A significant reduction in AHL secretion and pslA gene expression was observed, indicating that the extract inhibited quorum sensing by disrupting the quorum-sensing systems. The quorum-sensing inhibitory effect of N. curviflora extract appears to be attributed to the presence of kaempferol, quercetin, salicylic acid, rutin, and rosmarinic acid, as indicated by LCMS analysis. Conclusion: The results of the present study provide insight into the potential of developing anti-quorum sensing agents using the extract and the identified compounds to treat infections resulting from quorum sensing-mediated bacterial pathogenesis.

Antileishmanial and Cytotoxic Effects of Essential Oil and Methanolic Extract of Myrtus communis L.

  • Plants used for traditional medicine contain a wide range of substances that can be used to treat various diseases such as infectious diseases. The present study was designed to evaluate the antileishmanial effects of the essential oil and methanolic extract of Myrtus communis against Leishmania tropica on an in vitro model. Antileishmanial effects of essential oil and methanolic extract of M. communis on promastigote forms and their cytotoxic activities against J774 cells were evaluated using MTT assay for 72 hr. In addition, their leishmanicidal activity against amastigote forms was determined in a macrophage model, for 72 hr. Findings showed that the main components of essential oil were ${\alpha}$-pinene (24.7%), 1,8-cineole (19.6%), and linalool (12.6%). Findings demonstrated that M. communis, particularly its essential oil, significantly (P<0.05) inhibited the growth rate of promastigote and amastigote forms of L. tropica based on a dose-dependent response. The $IC_{50}$ values for essential oil and methanolic extract was 8.4 and $28.9{\mu}g/ml$ against promastigotes, respectively. These values were 11.6 and $40.8{\mu}g/ml$ against amastigote forms, respectively. Glucantime as control drug also revealed $IC_{50}$ values of 88.3 and $44.6{\mu}g/ml$ for promastigotes and amastigotes of L. tropica, respectively. The in vitro assay demonstrated no significant cytotoxicity in J774 cells. However, essential oil indicated a more cytotoxic effect as compared with the methanolic extract of M. communis. The findings of the present study demonstrated that M. communis might be a natural source for production of a new leishmanicidal agent.

Toxicity and Safety Profiles of Methanolic Extract of Pistacia integerrima J. L. Stewart ex Brandis (PI) for Wistar Rats

  • Objectives: The goals of this research were to evaluate acute (single-dose) and sub-acute (repeated-dose) toxicity profiles of methanolic extract of Pistacia integerrima J. L. Stewart ex Brandis (PI) for Wistar rats and to assess the safety profile of PI by observing physiological changes, mortality, changes in body weight, the histopathology of body organs, the hematology and the biochemistry of the animals. Methods: The toxicity profile of PI was evaluated using Wistar rats of both sexes. Animals were divided into four groups: Group 1; control group (normal saline), Group 2; PI-1 (250 mg/kg), Group 3; PI-2 (500 mg/kg), Group 4; PL-3 (1,000 mg/kg). An acute-toxicity study in which animals received a single dose of PI extract (2,000 mg/kg) and were then observed for 14 days for changes in skin, fur, eye color, mucous membrane secretions and excretions, gait, posture, and tonic or clonic movements was performed according to guideline 425 of the Organization of Economic and Corporation Development (OECD). In the repeated-dose toxicity study (OECD - 407) animals received a daily dose of PI extract for 28 days (4 weeks). The parameters observed in this study include body weight, hematology and biochemistry of the animals. Results: In the acute toxicity study, no mortalities or changes in behavior were noted in the animals. The repeated-dose toxicity study was also devoid of any toxicity in the animals during the 28 days of testing with PI extract. The extract did not alter- the body weight, hematology or biochemistry of the animals. The methanolic extract of PI was to be found safe to the no-observed-adverse-effect-level (NOAEL) for the single-dose and repeated-dose toxicity tests in rats. Conclusion: The methanolic extract of PI was devoid of toxicity; hence, it can be used for various ayurvedic preparations and treatments of diseases.

Analgesic and Anti-inflammatory Activities of Momordica dioica Fruit Pulp

  • In the present study Momordica dioica fruit pulp was successively extracted with hexane and methanol. To the concentrated methanolic extract ethyl acetate was added and ethyl acetate soluble portion was separated. Both hexane extract (HE) and ethyl acetate soluble portion (EASP) of methanolic extract was vacuum dried to yield the respective HE and EASP. HE and EASP were evaluated for its analgesic and anti-inflammatory activities in a dose of 50 and 100 mg/kg in mice and rats. Both HE and EASP significant exhibited analgesic and anti-inflammatory activities when compare to standard drug.
