Anti-oxidative and anti-inflammatory effects of aerial parts of Rumex japonicus Houtt. in RAW 264.7 cells (양제엽(羊蹄葉) 메탄올 추출물의 항산화 및 항염증 효과)

  • Objectives : The aerial parts of Rumex japonicus Houtt. (RF) is used by traditional clinics to treat parasite infection in East asia. This study aims a verification of anti-oxidative and anti-inflammatory effects of RF methanol extract. Methods : Anti-oxidative effects of RF were measured by scavenging activities of DPPH, superoxide, nitric oxide (NO) and peroxynitrite radicals. And also scavenging activities of anti-oxidation in lipopolysaccharide (LPS)-treated RAW 264.7 cells were measured. The inhibitory effects against the production of inflammatory mediators including NO, prostaglandin $E_2$ ($PGE_2$), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin-$1{\beta}$ (IL-$1{\beta}$), IL-6, inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and the translocation of nuclear factor (NF)-${\kappa}B$ in LPS-stimulated RAW 264.7 cells by RF were tested. Results : RF scavenged DPPH, superoxide, NO and peroxynitrite radicals, and RF (at $200{\mu}g/m{\ell}$) reduced the inflammatory mediators definitely. Conclusions : These results indicate that RF may be a potential drug source for oxidative stress related inflammatory diseases.

NON-ENZYMATIC BROWNING REACTIONS IN DRIED SQUID STORED AT DIFFERENT WATER ACTIVITIES (마른 오징어 저장중의 수분활성과 갈변반응)

  • In the previous work(Kim et al, 1973), the quality of sun-dried Alaska pollack, Theragra chalch-ogramma, was discussed in the aspect of non-enzymatic discoloration as a function of relative humidity during storage at room temperature($20^{\circ}C$). In this paper, sun-dried squid, Ommastrephes steam pacificus was investigated at the same aspect mentioned above. Fresh squid from the whole sale market was dressed, filleted, dried for 48 hours in the open air and finally stored in the humidistat chamber. Lipid oxidation ana development of non-enzymatic browning were tested by the same methods described in the previous paper. The TBA value showed a maximum peak on 30 day storage, hereafter tended to decrease gradually. The rate of browning, however, in water soluble fraction as well as in chloroform-methanol fraction was lower at 0.34 to 0.45 water activity than at any other case, and propagation of lipid oxidation was also diminished at the above level of water activity. From the results, it is recognized that storage at Aw=0.34 to 0.45 provides higher quality stability for sun-dried squid.

Characteristics of Pt, Pt-Ru and Pt-CeO2 Catalysts Supported on Carbon Nanotubes for Methanol Fuel Cell (탄소 나노튜브에 담지된 Pt, Pt-Ru 및 Pt-CeO2 메탄올 연료전지 촉매의 특성)

  • Nanosized Pt, Pt-Ru and Pt-$CeO_2$ electrocatalysts supported on acid-treated carbon nanotube (CNT) were synthesized by microwave-assisted heating of polyol process using $H_2Cl_6Pt{\cdot}6H_2O$, $RuCl_3$, $CeCl_3$ precursors, respectively, and were characterized by XRD and TEM. And then the electrochemical activity of methanol oxidation for catalyst/CNT nanocomposite electrodes was investigated. The microwave assisted polyol process produced the nano-sized crystalline catalysts particles on CNT. The size of Pt supported on CNT was 7~12 nm but it decreased to 3~5 nm in which 10wt% sodium acetate was added as a stabilizer during the polyol process. This fine Pt catalyst particles resulted in a higher current density for Pt/CNT electrode. It was also found that 10 nm size of PtRu alloys were formed by polyol process and the onset potential decreased with Ru addition. Cyclic voltammetry analysis revealed that the $Pt_{75}Ru_{25}/CNT$ electrode had the highest electrochemical activity owing to a higher ratio of the forward to reverse anodic peak current. And the chronoamperemetry test showed that $Pt_{75}Ru_{25}$ catalyst had a good catalyst stability. The activity of Pt was also found to be improved with the addition of $CeO_2$.

Differential Expression of Laccase Genes in Pleurotus ostreatus and Biochemical Characterization of Laccase Isozymes Produced in Pichia pastoris

  • In this study, transcriptome analysis of twelve laccase genes in Pleurotus ostreatus revealed that their expression was differentially regulated at different developmental stages. Lacc5 and Lacc12 were specifically expressed in fruiting bodies and primordia, respectively, whereas Lacc6 was expressed at all developmental stages. Lacc1 and Lacc3 were specific to the mycelial stage in solid medium. In order to investigate their biochemical characteristics, these laccases were heterologously expressed in Pichia pastoris using the pPICHOLI-2 expression vector. Expression of the laccases was facilitated by intermittent addition of methanol as an inducer and sole carbon source, in order to reduce the toxic effects associated with high methanol concentration. The highest expression was observed when the recombinant yeast cells were grown for 5 days at $15^{\circ}C$ with intermittent addition of 1% methanol at a 12-hr interval. Investigation of enzyme kinetics using 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) as a substrate revealed that the primordium-specific laccase Lacc12 was 5.4-fold less active than Lacc6 at low substrate concentration with respect to ABTS oxidation activity. The optimal pH and temperature of Lacc12 were 0.5 pH units and $5^{\circ}C$higher than those of Lacc6. Lacc12 showed maximal activity at pH 3.5 and $50^{\circ}C$, which may reflect the physiological conditions at the primordiation stage.

In vitro Antioxidant Activity of Mentha viridis L. and Mentha piperita L. (박하의 in vitro 항산화활성)

  • For selecting a new candidate as functional material, this study was conducted on in vitro antioxidant activity and total phenol content of methanol and water extracts prepared from two Mentha species (M viridis L. (M spicata L.) and Mentha piperita L, Extracts of M. viridis showed more efficient scavenging activity on superoxide and DPPH ${({\alpha},{\alpha}-diphenyl-{\beta}-picrylhydrazyl)}$ radical and inhibitory activity on oxidation of human low density lipoprotein (LDL) induced by $CuSO_4$ and auto-oxidation of linoleic acid than those of M piprita. Methanol extract $(65.88%{\sim}77.59%)$ and water extract $(37.69%{\sim}87.21%)$ of M. viridis also exhibited more potent inhibitory activity on LDL oxidation than that of ${{\alpha}-tocopherol\;(28.37%{\sim}66.54%)}$ at ${1{\sim}100\;{\mu}g/ml}$ of final concentration. The total phenol contents of methanol extract and water extract of Mviridis (17.95% & 10.18%, respectively) as tannic acid equivalent were higher than those of M piperita (15.44% & 9.19%). But the yields of methanol and water extracts of M. viridis (13.3 % & 13.5%) were lower than those of M. piperita (14.1 % & 14.6%). The results implies that the extracts from M. viridis (spicata) is more useful material for industrialization as functional food than those from M. piperita.

Oxygen Reduction of PAFC Gas Diffusion Electrode with Various Pt Impregnation Methods (인산형 연료전지용 기체확산전극의 백금촉매 담지방법에 따른 산소환원 특성)

  • Pt catalyst on carbon black was prepared by colloidal method, ion exchanging method and methanol reducing method. The colloidal method has been used generally. At ion exchanging method, $H^+$ of functional group on carbon surface made by oxidation treatment was exchanged with Pt ion. At methanol reducing method, Pt was impregnated on carbon to reduce by methanol contained with surfactants. With TEM and XRD, Pt particle size impregnated on carbon by various methods was $30{\sim}50{\AA}$. Loading yield was about 100%, loading yield of ion exchanging method was 99.92% by DCP analysis and 99.87% by combustion method. Within 60 hour, current density of oxygen reduction was $460mA/cm^2$ at 0.7V(vs. RHE) at colloidal method. It was the better performance than catalyst prepared by ion exchanging, methanol reducing method. But, it was shown some decrease of performance for long operation time(after 100hour), catalyst prepared by methanol reducing method was shown stable performance.

  • In this study, we investigated the biological activity of antioxidant and antibacterial activity of citrus by-product ferment. Hot water extracts and 80% methanol extracts from citrus byproduct of ferment were screened for antibacterial activity pathogenic bacteria by paper disc method. Among the various hot water extracts and 80% methanol extracts the Bacillus subtilis showed relatively strong antibacterial activities in the order. The reducing activity on the 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical and ${\cdot}OH$ radical scavenging potential were sequentially screened, in search for antioxidant activities of citrus by-product ferment.

DPPH Radical Scavenger Activity and Antioxidant Effects of Cham-Dang-Gui (Angelica gigas) (참당귀(Angelica gigas)의 DPPH Radical 소거 활성과 항산화 효과)

  • This study was carried out to evaluate the free radical scavenging effect and antioxidant effect of Cham-Dang-Gui (Angelica gigas) on cyclophosphamide (CYP) injected rats. Rats were divided into five groups: CON (normal group), ANS (CYP-injected and normal diet group), AND (CYP-injected and normal diet and Cham-Dang-Gui-treated group), ALS (CYP-injected and low iron diet group), and ALD (CYP-injected and low iron diet and Cham-Dang-Gui-treated group). CYP (30 mg/kg) was intraperitoneally injected to rats for early 3 days. Saline or Cham-Dang-Gui was administrated orally for entire experimental period. DPPH radical scavenger activity was measured by DPPH method, it was shown higher in methanol extract (81.5%) than in water extract (66.3%) of Cham-Dang-Gui. We observed the preventive effects of Cham-Dang-Gui on lipid oxidation of liver and protein oxidation of plasma. Hepatic SOD and catalase activities were significantly higher in CYP-injected group (ANS) than CON group, but SOD activity was slightly lowered in Cham-Dang-Gui treated group than CYP-injected group (ANS). These results suggest that extract of Cham-Dang-Gui could be useful for functional materials to reduce the oxidation of lipids and protein induced by free radicals.

Antioxidant Activity of Anthraquinones and Flavonoids from Flower of Reynoutria sachalinensis

  • Bioassay-guided fractionation of methanol extract of Reynoutria sachalinensis flower using DPPH assay has led to the isolation of three anthraquinones and three flavonoids. Their structures were identified as emodin (1), emodin-8-O-$\beta$-D-glucopyranoside (2), physcion-8-O-$\beta$-D­glucopyranoside (3), quercetin-3-O-$\alpha$-L-arabinofuranoside (4), quercetin-3-O-$\beta$-D-galactopyra­noside (5), and quercetin-3-O-$\beta$-D-glucuronopyranoside (6) by comparing their physicochemical and spectral data with those published in literatures. All isolated compounds were evaluated for antioxidant activities with free radical 1, 1-diphenyl-2-picrylhydrazyl (DPPH) scavenging, superoxide radical scavenging and $Cu^{2+}$-mediated low density lipoprotein (LDL) oxidation assay. The results demonstrated that three flavonoids, 4, 5, and 6 had remarkable antioxidant activities with the $IC_{50}$ values of 64.3, 54.7, and 46.2${\mu}M$ (DPPH scavenging), the $IC_{50}$ values of 6.0, 6.7, and $4.4{\mu}M$ (superoxide radical scavenging) and the $IC_{50}$ values of 3.8, 3.2, and 5.4${\mu}M$ against LDL oxidation, respectively.

Electrocatalytic activity of the bimetallic Pt-Ru catalysts doped TiO2-hollow sphere nanocomposites (Pt-Ru@TiO2-H 나노구조체촉매의 합성 및 전기화학적 특성평가)

  • This paper describes the electrocatalytic activity for the oxidation of small biomolecules on the surface of Pt-Ru nanoparticles supported by $TiO_2$-hollow sphere prepared for use in sensor applications or fuel cells. The $TiO_2$-hollow sphere supports were first prepared by sol-gel reaction of titanium tetraisopropoxide with poly(styrene-co-vinylphenylboronic acid), PSB used as a template. Pt-Ru nanoparticles were then deposited by chemical reduction of the $Pt^{4+}$ and $Ru^{3+}$ ions onto $TiO_2$-hollow sphere ($Pt-Ru@TiO_2-H$). The prepared $Pt-Ru@TiO_2-H$ nanocomposites were characterized by transmission electron microscopy (TEM), X-ray diffraction (XRD), and elemental analysis. The electrocatalytic efficiency of Pt-Ru nanoparticles was evaluated via ethanol, methanol, dopamine, ascorbic acid, formalin, and glucose oxidation. The cyclic voltammograms (CV) obtained during the oxidation studies revealed that the $Pt-Ru@TiO_2-H$ nanocomposites showed high electrocatalytic activity for the oxidation of biomolecules. As a result, the prepared Pt-Ru catalysts doped onto $TiO_2$-H sphere nanocomposites supports can be used for non-enzymatic biosensor or fuel cell anode electrode.
