김치 용매 추출물의 항산화성 (Antioxidant Activity of Various Solvent Extracts from Freeze Dried Kimchi)

  • 동결건조한 김치를 물, 메탄올, 에칠 에테르로 각각 추출한 후 이들 용매추출물들이 지니고 있는 지질산화반응에서의 항산화력에 대하여 고찰하였다. 즉, linoleic acid mixture의 액상산화반응 조건($37^{\circ}C$)에서 과산화물 생성억제에 대한 용매별, 김치발효기간별, 농도별, 그리고 혼합용매종류별로 실험하여 추출물의 항산화능을 살펴보았다. 그 결과 물과 메탄올 추출물이 에틸에테르 추출물 보다 과산화물의 생성을 더 억제 하였다.그리고 첨가농도가 증가할수록 항산화성이 증대되었으며 발효 초일의 김치나 과숙김치보다 잘 숙성된 김치(15$^{\circ}C$에서 7일간 발효)의 추출물에 더 항산화성이 높았다. 또한 물과 메탄올의 비율을 달리 하여 추출한 경우 순수 메탄올보다 50~80% 메탄올 농도에서의 용매추출물에서 항산화효과가 높았다. 비록 75% 메탄올추출물의 항산화성은 BHA와 $\alpha$토코페롤보다는 낮았지만 동 추출물에서 계통적 분획 등에 의한 분획물의 항산화성을 계속 검토하므로서 보다 높은 항산화성 물질이 분리될 수 있음을 알 수 있었다.

Synthesis and Characterization of Tetrathiafulvalene (TTF) and 7,7,8,8-Tetracyanoquinodimethane (TCNQ) Compounds with PdX2(X=CI, NO3and Hexafluoroacetylacetonate)

  • Tetrathiafulvalene(TTF) reacts with $PdCl_2,Pd(NO_3)_2$ and $Pd(hfacac)_2$(hexafluoroacetylacetonate) in ethanol to give $(TTF)_{1.5}PdCl_2$ (1a), $(TTF)_3Pd(NO_3)_2$ (1b) and $(TTF)_4Pd(hfacas)_2$ nd (1c), respectively. $PdCl(TCNQ)_{2.5}{\cdot}CH_3OH(2a)$was obtained from the reaction of $PdCl_2$ with LiTCNQ in methanol via the partial replacement of $Cl^-$ in $PdCl_2$ by $TCNQ^-$anion, whereas the total substitution of the labile $NO_3^-$ in $Pd(NO_3)_2$ yielded pd(TCNQ)·$CH_3OH$ (2b). $Pd(hfacac)_2(TCNQ)_2\cdot3CH_3OH$ (2c) was obtained from $Pd(hfacac)_2$ and LiTCNQ in methanol. The prepared compounds were characterized by spectroscopic (IR, UV, XPS) methods and magnetic (EPR, magnetic susceptibility) studies. The powdered electrical conductivities (${\sigma}_{rt}$) of the prepared compounds at room temperature were about~$10^{-7}S{\cdot}cm^{-1}$. The effective magnetic moments were lass than the spin-only value of one unpaired electron and no EPR signals from Pd metal ions were observed in any of the compounds, indicating that the Pd ions were diamagnetic and the magnetic moments arose from$(TTF)_n$ or $(TCNQ)_n$ moieties. The experimental evidences revealed that the charge transfer had occurred form $(TTF)_n$ moiety to the central Pd metal ion in 1a, 1b and 1c. Thus the TTF donors were ions in 2a and 2b were diamagnetic Pd(II) oxidation state. In contrast, the Pd metal ion was oxidized to Pd(IV) state in 2c as a result of an addition of $TCNQ^-$anion to $Pd(hfacac)_2$ in methanol. The oxidation states of the Pd metal ions were confirmed using the x-ray photoelectron spectroscopy.

메탄올 전기산화반응 증진을 위한 PtRuW/C 촉매에서 텅스텐의 효과에 관한 연구 (Effect of Tungsten on PtRuW/C Catalysts for Promoting Methanol Electro-oxidation)

  • PtRuW/C 촉매를 Pt : Ru : W 비를 5 : 4 : 1, 2 : 1 : 1, 1 : 1 : 1, 1 : 2 : 2로 각각 합성하였다. 촉매의 조성은 EDX분석을 통해 이론값과 비슷하다는 것을 확인하였다. TEM분석과 XRD분석으로부터 3.5~5.5 nm의 균일한 입자 크기 및 결정질 분포를 가지고 있음을 확인하였다. 유효표면적, 전류밀도, 고유 활성 및 피독률과 같은 전기화학적 특성은 CO 스트리핑, 선형쓸음 전기량 측정법, 대시간 전류법 등과 같은 방법으로 분석하였다. 이러한 분석으로부터, $PtRu_2W_2/C$를 제외한 촉매는 상업촉매보다 우수한 반응성과 안정성을 가지고 있음을 확인하였다. 이 중 가장 우수한 촉매는 $Pt_5Ru_4W/C$였으며, $121.05mA{\cdot}m^{-2}$의 specific activity와 $0.01%{\cdot}s^{-1}$의 피독률을 보였다.

참당귀 Methanol 추출물의 생리활성 (Biological Activities of Methanol Extract of Angelica gigas Nakai)

  • 참당귀(Angelica gigas Nakai)를 기능성 식품소재 및 의약품의 원료로 활용하기 위하여 참당귀 methanol 추출물에 대한 항산화, 항암 및 면역활성을 조사한 결과는 다음과 같다. 참당귀의 항산화력을 알아보기 위하여 0.1, 0.5 및 1 mg/mL의 농도로 당귀의 methanol 추출물을 처리한 후 수소 공여능, 환원력 및 hydroxyl radical 활성을 측정하였다. 그 활성은 모두 추출물의 농도에 의존적으로 높게 나타났으며, 특히 수소공여능은 1 mg/mL에서 대조군과 비교했을때 50% 정도의 활성을 나타내었다. 참당귀 methanol 추출물을 SW480세포에 처리한 결과 대조군에 비하여 농도 의존적으로 암세포의 성장을 억제하였으며, 암세포에 참당귀 methanol 추출물을 24시간 처리하여 hoechst 염색한 후 형광현미경으로 관찰한 결과 핵의 응축 및 DNA 분절이 관찰되었다. 또한 참당귀 methanol 추출물이 처리된 암세포에서는 대조군에 비하여 농도에 의존적으로 caspase-3의 활성이 증가하였다. 참당귀의 methanol 추출물은 생쥐의 비장으로부터 분리한 면역세포에 대하여 대조군과 비교하여 상이한 차이를 보이지 않았으며, 또한 대식세포인 RAW 264.7에서도 $NO_2$(Nitrite)를 함량이 증가되지 않았다. 따라서 이들 결과는 참당귀 methanol 추출물은 약한 항산화 활성을 가지며, 면역 활성에는 큰 관련이 없지만, SW480 암 세포에서 caspase dependent pathway에 의한 apoptosis를 유도함으로서 암세포의 사멸하는 것으로 판단되어진다.

마른명태 저장중(貯藏中)의 수분활성(水分活性)과 갈변반응(褐變反應) (Non-Enzymatic Browning Reactions in Dried Alaska Pollak Stored at Different Water Activities)

  • In the present work, the quality stability of sun-dried Alaska pollack, Theragra chalcogramma, was discussed in the aspects of non-enzymatic discoloration as a function of relative humidity during storage at room temperature$(20^{\circ}C)$. Frozen Alaska pollack was dressed, filleted, dried for 48 hours in the open air, and finally stored in cylindrical acrylic chambers which contained saturated specific salt solutions proposed by Rockland(1960) for humidity control. The color development of the product was analyzed by spectrophotometry at 10 day-intervals during the storage. Lipid oxidation was measured as TBA value at wavelength of 538nm. And browning pigments were extracted, divided into two fractions and measured at 460nm: one was chloroform-methanol (2:1 v/v)soluble fraction attributed to lipid oxidation, and the other was water dialyzed fraction caused by so called Maillard reaction. The TBA value showed a maximum on 30 day storage, hereafter, intended to decrease gradually. On the other hand, the rate of brown pigment development in water dialyzed fractions as well as in chloroform-methanol soluble fractions was lower at 34 to 45%RH than at any other case, and propagation of lipid oxidation was also diminished at the same levels of humidity. From the facts described previously, it is recognized that storage at 34 to 45%RH provides higher quality stability for sun-dried Alaska pollack.

수산 미이용자원 중에 존재하는 항산화 물질의 검색 (Screening for the Antioxidants in Unused Marine Resources by the Polarographic Method)

  • 수산 미이용자원의 순차 용매추출물에 대한 항산화물 존재 여부 검색결과, 모자반의 경우 $IC_{50}$이 물추출구에서 $500{\mu}g/ml$, 에테르 추출구 및 메탄올 추출구에서 각각 $133,700{\mu}g/ml$로 나타났으며, 참빗풀은 에테르, 아세톤, 메탄올 추출구에서 $IC_{50}$이 각각 30, 110 및 $200{\mu}g/ml$로 나타나 대상 시료중 가장 항산화 효과가 좋았다. 미이용 수산동물 중에서는 오징어먹즙만이 물 추출구와 메탄올 추출구에서 $IC_{50}$이 각각 $390,\;100{\mu}g/ml$로 나타날 뿐 큰 항산화효과를 보이는 시료나 추출구간은 없었다. 대상 수산 미이용 동 식물에 있어서 에테르와 같은 비극성 용매 추출구에 강한 항산화 물질이 많이 용출되어 존재해 있는 것으로 나타났다.

수소-알코올연료전지를 위한 금속-산화물 나노구조제어 (Control of Metal-Oxide Nanostructures for $H_{2}-Alcohol$ Fuel Cells)

  • Due to their excellent catalytic activity with respect to methanol oxidation on platinum at low temperature, platinum nanosized catalysts have been a topic of great interest for use in direct methanol fuel cells (DMFCs). Since pure platinum is readily poisoned by CO, a by-product of methanol electrooxidation, and is extremely expensive, a number of efforts to design and characterize Pt-based alloy nanosized catalysts or Pt nanophase-support composites have been attempted in order to reduce or relieve the CO poisoning effect. In this review paper, we summarize these efforts based upon our recent research results. The Pt-based nanocatalysts were designed by chemical synthesis and thin-film technology, and were characterized by a variety of analyses. According to bifunctional mechanism, it was concluded that good alloy formation with $2^{nd}$ metal (e.g., Ru) as well as the metallic state and optimum portion of Ru element in the anode catalyst contribute to an enhanced catalytic activity for methanol electrooxidation. In addition, we found that the modified electronic properties of platinum in Pt alloy electrodes as well as the surface and bulk structure of Pt alloys with a proper composition could be attributed to a higher catalytic activity for methanol electooxdation. Proton conducting contribution of nanosized electrocatalysts should also be considered to be excellent in methanol electrooxidation (Spillover effect). Finally, we confirmed the ensemble effect, which combined all above effects, in Pt-based nanocatalsyts especially, such as PtRuRhNi and $PtRuWO_{3}$, contribute to an enhanced catalytic activity.

Nano Electrocatalysis for Fuel Cells

  • For both oxygen reduction (ORR) and hydrogen oxidation reactions (HOR) of proton electrolyte membrane fuel cells (PEMFCs), alloying Pt with another transition metal usually results in a higher activity relative to pure Pt, mainly due to electronic modification of Pt and bifunctional behaviour of alloy surface for ORR and HOR, respectively. However, activity and stability are closely related to the preparation of alloy nanoparticles. Preparation conditions of alloy nanoparticles have strong influence on surface composition, oxidation state, nanoparticle size, shape, and contamination, which result from a large difference in redox priority of metal precursors, intrinsic properties of metals, increasedreactivity of nanocrystallites, and interactions with constituents for the synthesis such as solvent, stabilizer, and reducing agent, etc. Carbon-supported Pt-Ni alloy nanoparticles were prepared by the borohydride reduction method in anhydrous solvent. Pt-Ru alloy nanoparticles supported on carbon black were also prepared by the similar synthetic method to that of Pt-Ni. Since electrocatalytic reactions are strongly dependent on the surface structure of metal catalysts, the atom-leveled design of the surface structure plays a significant role in a high catalytic activity and the utilization of electrocatalysts. Therefore, surface-modified electrocatalysts have attracted much attention due to their unique structure and new electronic and electrocatalytic properties. The carbon-supported Au and Pd nanoparticles were adapted as the substrate and the successive reduction process was used for depositing Pt and PtM (M=Ru, Pd, and Rh) bimetallic elements on the surface of Au and Pd nanoparticles. Distinct features of the overlayers for electrocatalytic activities including methanol oxidation, formic acid oxidation, and oxygen reduction were investigated.

Kinetics and Mechanistic Chemistry of Oxidation of Butacaine Sulfate by Chloramine-B in Acid Medium

  • Butacaine sulfate is an ester of p-aminobenzoic acid which has been widely used as a local anaesthetic and it is a long standing agent particularly for spinal anaesthesia. For this reason, a kinetic study of oxidation of butacaine sulfate by sodium N-chlorobenzenesulfonamide (chloramine-B or CAB) has been carried out in $HClO_4$ medium at 303 K in order to explore this redox system mechanistic chemistry. The rate shows a first-order dependence on both $[CAB]_o$, and $[substrate]_o$, and a fractional-order dependence on acid concentration. Decrease of dielectric constant of the medium, by adding methanol, increases the rate of the reaction. Variation of ionic strength and addition of benzenesulfonamide or NaCl have no significant effect on the rate. The reaction was studied at different temperatures and the activation parameters have been evaluated. The stoichiometry of the reaction has been found to be 1:2 and the oxidation products have been identified by spectral analysis. The observed results have been explained by plausible mechanism and the related rate law has been deduced.

건어육 저장중의 온도와 수분활성에 따른 지방의 산화속도 (Kinetics of Lipid Oxidation in Dried Fish Meat Stored under Different Conditions of Water Activity and Temperature)

  • In this work, lipid oxidation and the kinetics of the oxidation reaction in fried file-fish meat were investigated when sun-dried file-fish was stored under the conditions of various water activities and temperature, 35, 45, 55 and $35/55^{\circ}C$. The storage stability and the development of browning by oxidative rancidity were also discussed. Monolayer coverage value of water content in dried file-fish was $8.03\%$ at $0.21\;a_w$Lipid oxidation at $35^{\circ}C$ was developed with increasing water activity but at $45^{\circ}C$and $55^{\circ}C$ it was rapidly progressed without clear differences between water activities except $0.44\;a_w$. The rate of reaction was more sensitive to storage temperature than to water activity. Browning in methanol-chloroform fraction was developed linearly by the progress of lipid oxidation which suggested that lipid oxidation was greatly influential to the development of browning in dried fish meat. In kinetical analysis the oxidation followed a zero order reaction mechanism as a function of carbonyl value. The activation energies obtained from the Arrhenius plot ranged 9.0 to 10.8 Kcal/mol and $Q_10$ values, 1.6-1.7. Shelf-lives at the storage of 35,45 and $55^{\circ}C$ ranged 58 days to 8 days. And in the fluctuating temperature storage at $35/55^{\circ}C$, shelf-lives were 17, 16, 15 and 13 days at 0.44, 0.52, 0.65 and $0.75\;a_w$, respectively. The shelf-lives for assessed from the accelerated shelf-life test were 125, 123, 120 and 106 days at 0.44, 0.52, 0.65 and $0.75\;a_w$, respectively, in the case of storage at $25^{\circ}C$.
