In this study, the inhibition of ammonia on anaerobic digestion of butyric acid was evaluated and the potential alleviating effects of such ammonia inhibition by the addition of magnetite particles were investigated. Independent anaerobic batch tests fed with butyric acid as a sole organic source were conducted in twenty 60-mL glass bottles with 10 different treatment conditions, comprising ammonia: 0.5, 2.0, 4.0, 6.0, and 7.0 g total ammonia nitrogen (TAN)/L and magnetite particles: 0 mM and 20 mM. The increase in ammonia concentration did not cause significant inhibition on methane yield; however, a significant inhibition on lag time and specific methane production rate was observed. The IC50 in the control treatments (without magnetite addition) was estimated as 6.2654 g TAN/L. A similar inhibition trend was observed in magnetite-added treatments; however, the inhibition effect by ammonia was significantly alleviated in lag time and specific methane production rate when compared to those in the control treatments. The lag time was shortened by 1.6-46.3%, specific methane production rate was improved by 6.0-69.0%. In the magnetite-added treatments, IC50 was estimated as 8.5361 g TAN/L. This study successfully demonstrated the potential of magnetite particles as an enhancer in anaerobic digestion of butyric acid under conditions of ammonia stress.
This study aims to observe the inhibition of methane generation, the decomposition of organic matter, and the trend of outflowing leachate, using the simulated column of the anaerobic sanitary landfill structure of sulfate addition type which is made by adding sulfate to a current anaerobic landfill structure, and the simulated column of semi-aerobic landfill structure in the laboratory which is used in the country like Japan in order to inhibit methane from a landfill site among the gases caused by a global warming these days, and at the same time to promote the decomposition of organic matter, the index of stabilization of landfill site. As a result of this study, it is thought that the ORP(Oxidation Reduction Potential) of the column of semi-aerobic landfill structure gradually represents a weak aerobic condition as time goes by, and that the inside of landfill site is likely to by in progress into anaerobic condition, unless air effectively comes into a semi-aerobic landfill structure in reality as time goes by. In addition, it can be seen that the decomposition of organic matter is promoted according to sulfate reduction in case of $R_1$, a sulfate-added anaerobic sanitary landfill structure, and that the stable decomposition of organic matter in $R_1$ makes a faster progess than $R_2$. Moreover it can be estimated that $R_1$, a sulfate-added anaerobic sanitary landfill structure has an inhibition efficiency of 55% or so, compared with $R_2$, a semi-aerobic landfill structure, in the efficiency of inhibiting methane.

The effect of organic substance on its degradation rate in burial site was investigated using batch tests. Substrate were swine and cattle with the initial concentrations of 2, 4, 6, 8, and 10 g VS(volatile solids)/L, respectively. The highest methane production rates of swine and cattle were found at 2 g VS/L as 46.3 and 48.4 ml CH4/g VS.d, respectively. As substrate concentration increased, the methane production rate decreased. The inhibition constants were n and m that were estimated using nonlinear inhibition model. The values of n and m were inhibition constants of methane production rate and ultimate methane yield, respectively. The values of n and m were 4.9 and 0.6 on swine and 1.1 and 0.4 on cattle. The methane production rate was responded sensitively by increase and decrease of substrate concentration, whereas ultimate methane yield do not relatively. From a relation between n and m, inhibitory effect of substrate concentration was confirmed as uncompetitive inhibition.

Anaerobic hydrogen production was executed using mixed anaerobic sludge. pH control (pH $5.5^{\sim}6.2)$ and heat treatment of anaerobic sluge at $92^{\circ}C$ was not effective for repressing the methane formation. The addition of $nitrate(500^{\sim}2,000\;mg/L\;KNO_3)$ made it possible to repress the methane formation, which resulted in the improvement of hydrogen production. By using SEM(Scanning Electron Microscope), less methane forming microbial with spagetti shape was observed when nitrate was supplied to anaerobic reactor.
In this study, phase equilibria and formation behaviors of methane hydrate containing mono-ethylene glycol (MEG) and salts (sodium chloride, NaCl; sodium bromide, NaBr; sodium iodide, NaI) are investigated. Equilibrium conditions of methane hydrate containing MEG and salts are measured in a temperature range 272~283 K and a pressure range 3.5~11 MPa. Hydrate inhibition performance in the presence of additives can be summarized as follows: methane hydrate containing (5 wt% NaCl + 10 wt% MEG) > (5 wt% NaBr + 10 wt% MEG) > (5 wt% NaI + 10 wt% MEG). Formation behaviors of methane hydrate with MEG and salts are investigated for analyzing the induction time, gas consumption amount and growth rate of methane hydrates. There are no significant changes in the induction time during methane hydrate formation, but the addition of MEG and salts solution during hydrate formation can affect the gas consumption amount and growth rate.
As an effort to develop an alternative transportation fuel, the production of methanol from methane gas was studied using the resting cells of an obligatory methanotroph, Methylosinus trichosporium OB3b. The reaction was carried out in high concentration phosphate buffer solutions with the flask-grown cells containing the exclusively cytoplasmic methane monooxygenase (sMMO) activity. The methanol accumulation rate was observed to be 79nmo1/mg·min during the initial 4.5hr. Phosphate-dependent inhibition was found for both sMMO and methanol dehydrogenase (MDH) activities, and the inhibition constants were 185mM and 42mM, respectively. The inhibition mode was noncompetitive. Methanol was found to be very inhibitory to the sMMO activity and the inhibition constant (noncompetitive) was 21mM when propylene was used as substrate. The sMO activity in the resting cells was declined very fast and the rate became very high during the methanol production. These results indicate that the use of M. trichosporium OB3b as a biocatalyst for the methanol production is heavily dependent on the stable maintenance of the whole-cell SMO activity as well as the effective alleviation of product inhibition.

This research examined the effect of ammonia nitrogen loading rate(NVLR) on the anaerobic digestion of slurry-typed swine wastewater. The anaerobic reactor was used an upflow anaerobic sludge blanket (UASB) process. This UASB reactor was operated at a NVLR of $0.02{\sim}0.96kg{NH_4}^+-N/m^3/day$. The methane content showed the range of 73.3~77.9% during the steady state period. Free ammonia(FA) concentration increased over inhibition level as pH increase from 7.3 to 8.2. However, in consideration of methane content, methane producing bacteria (MPB) inhibition by FA and total ammonia(TA) was not observed. A stepwise increase of the NVLR resulted in a deterioration in the COD removal rate in UASB reactor. The COD removal rate were 60% for NVLR up to $0.55kg{NH_4}^+-N/m^3/day$. As the NVLR increased from 0.09 to $0.96kg{NH_4}^+-N/m^3/day$, the biogas production rate varied from 3.71 to 9.14L/d and the methane conversion rate of the COD varied from 0.32 to $0.20m^3CH_4/kg$ COD removed. Consequently, in considerations of FA concentration, COD removal rate, and $CH_4$ production rate, the UASB reactor must be operated to lower than $0.40kg{NH_4}^+-N/m^3/day$ of NVLR.

Anaerobic digestion at the temperature of $35-55^{\circ}C$ was conducted using an artificial sludge of uniform composition. The hydraulic retention time of 5 days was chosen because the temperature effect was effectively shown at a high loading. Inhibition of the methane fermentation decreased as the temperature increased. Acid fermentation was prevalent at the mesophilic and intermediate temperatures, while active methane fermentation took place at $55^{\circ}C$. Temperature not only affects activity of the microorganisms, but also affects physical and chemical properties of the sludge, Digestion inhibition was much reduced when the feed sludge was diluted, and active methane fermentation was possible at all temperatures. The digestion efficiency was governed by the organic loading rate as well as the hydraulic 10ading rate. No reduction of the digestion efficiency at $40-45^{\circ}C$, which had been referred to a critical temperature range, was observed. The digestion efficiency increased monotonically from mesophilic to thermophilic range. Improved settling properties of digested sludge was also recorded at higher temperatures.
The interaction of fibre degrading microbes and methanogens was studied using two forages, lucerne (Medicago sativa) hay and maize (Zea mays) hay, as substrate and 2-bromoethanesulphonic acid (BES) as an additive in an in vitro gas production test. Gas and methane production (ml/g dry matter) were significantly higher (p<0.05) on lucerne as compared to maize hay. Inclusion of BES in the incubation medium significantly suppressed methane emission irrespective of substrate. The population density of total bacteria, fungi, Ruminococcus flavefaciens and Fibrobacter succinogenes was higher, whereas that of methanogens was lower with maize hay as compared to lucerne as substrate. BES suppressed methanogen population by 7 fold on lucerene and by 8.5 fold on maize at 24 h incubation as estimated by real time-PCR. This suppression was accompanied by almost complete (>98% of control) inhibition of methanogenesis. The proportion of acetate decreased, whereas that of propionate increased significantly by inclusion of BES, resulting in narrowing of acetate to propionate ratio. In vitro true digestibility (IVTD) of lucerne was significantly higher as compared to maize but BES inclusion did not affect IVTD.

Two UASB reactors were operated to investigate the effect of high concentration of sulfate on anaerobic digestion of propionate using an upflow anaerobic sludge blanket (UASB) reactor. An organic loading rate of $1.2kg\;COD/m^3{\cdot}d$ and a hydraulic retention time of 1.6 d were maintained during this study. In the absence of sulfate, the UASB reactor achieved about 95% removal of chemical oxygen demand whereas in the presence of $2,000\;SO_4^{2-}mg/L$, the COD removal rate decreased to 83% due probably to the inhibition of dissolved sulfide inhibition. Interactions between the methane producing bacteria (MPB) and sulfate reducing bacteria (SRB) were measured to investigate the competition between MPB and SRB. The MPB consumed average 58% of the available electron donors at $COD/SO_4^{2-}$ ratio of 1. Propionate was consumed mainly by SRB, converting sulfate into sulfide and suppressing the methane production. The specific methanogenic activity (SMA) using acetate and propionate increased as microorganism acclimated to the substrate.
