식이 단백질과 Ca 수준이 흰쥐의 Cd 중독과정중 Metallothionein 과 조직의 형태변화에 미치는 영향 (The Effect of Dietary Protein and Calcium Levels on Metallothionein and Histopathological Changes during Cadmium Intoxication in Rats)

  • This study was performed to investigate effect of dietary protein and calcium levels on cad-mium intoxication in rats. Adult Sprague-Dawley male rate(245$\pm$21g) were blocked into 18 groups of 7 animals according to body weight Nine experimental diets different with protein(40%, 15%, 7%) and calcium (1.3%, 0.6%, 0.1%) levels were prepared. Nine groups of animals were fed each diet with 50ppm cadmium in drinking water and the other 9 groups without cadmium for 30days. Results were summarized as follows: 1) Body weight gain F. E. R(Food Efficiency Ratio) and weights of liver kidney and femur were higher in high protein groups among cadmium exposed groups. 2) Cadmium contents in liver and intestine were higher in rats fed high protein diet or low calcium diet among cadmium exposed groups. Fecal cadmium excretion was highest in high protein-high calcium diet group among cadmium exposed animals. Metallothionein contents in liver kidney and intestine were higher in animals exposed to cadmium and fed high protein diets. 3) Gel filtration chromatography of cytosolic solution showed that the higher dietary protein and calcium levels were the more cadmium was found in metallothionein fractions. 4) No gross histopathological change was seen in liver kidney and intestine of cadmium exposed rats. However a significant increase of smooth endoplasmic reticulum which was alleveated by high protein-high calcium diet was observed. Results obtained indicated that not only high protein diet but also high calcium diet showed preventive effect on cadmium intoxication by increasing the induction of metallothionein syn-thesis and decreasing the cadmium absorption.

미호종개 metallothionein 유전자의 구조 및 중금속 노출과 고온 자극에 대한 MT mRNA의 발현 특징 분석 (Gene Structure and Altered mRNA Expression of Metallothionein in Response to Metal Exposure and Thermal Stress in Miho Spine Loach Cobitis choii (Cobitidae; Cypriniformes))

  • 멸종위기 어류 미호종개(Cobitis choii)로부터 중금속해독 단백질(metallothionein) 유전자를 분리, 클로닝하고 중금속 및 고온 스트레스에 대한 전사 발현 특정을 분석하였다. 미호종개 metallothionein는 gDNA, mRNA 및 아미노산 서열 모두에서 경골 어류 MT들의 구조적 특징을 잘 보전하고 있었으며, 생물정보분석을 통해 미호종개 MT 유전자 5'-upstream 영역은 중금속 조절, 면역 반응 및 온도 반응에 관여하는 다양한 전사 조절인자들의 부착 위치들을 포함하는 것으로 관찰되었다. 카드뮴(Cd), 구리(Cu), 니켈(Ni), 망간(Mn) 및 아연(Zn)을 이용한 침지 노출 실험(0.5 및 $1.0\;{\mu}M$; 24시간)에서 미호종개 MT mRNA 발현은 구리 및 카드뮴 처리군에서 가장 많이 유도되었고($1.0\;{\mu}M$ Cu 처리군에서 최대 10배), 망간 처리군에서는 비교적 적은 양의 MT 발현이 유도된 반면(2배), 아연 및 니켈 노출 군에서는 유의적인 MT 발현의 증감이 관찰되지 않았다. 또한 미호종개 MT 전사 발현은 고온 자극 ($25^{\circ}C$로부터 $31^{\circ}C$까지 증가)에도 민감하게 반응하는 것으로 나타나, $31^{\circ}C$ 도달시점에서 $25^{\circ}C$ 초기 MT mRNA 발현 수준보다 9배 높은 mRNA 발현이 관찰되었다. 본 연구 결과는 MT 기반의 유전자 발현 분석을 이용함으로써, 향후 멸종위기 어류 미호종개의 스트레스 반응 연구에 유용한 기초 자료를 제공할 수 있다고 기대된다.

Cadmium 투여 흰쥐에 있어서 Metallothionein 합성과 항산화적 해독기구에 미치는 식이 Selenium의 영향 (Effect of Dietary Selenium of Metallothionein Synthesis and Antioxidative Detoxificantion Mechanism in Cadmium Administered Rats)

  • In order to investigate the effect of selenium (Se) on the liver damage, metallothionein synthesis and hepatic antioxidative detoxification system in cadmium(Cd) administered rats. Sprague-Dawley male rats(60\\5g) were divided into two diet groups, depending on with (CdS groups) or without (Cd groups) 0.5ppm Se supplementation and fed experimental diets ad libidum for 4 weeks. And then each group was again subdivided into five groups, depending on injection number of Cd, i.e., 0, 1, 2, 3, and 4 times of 2.5mg Cd/kg of body wt once a day. Hemoglobin concentration, hematocrit values, superoxide dismutase, glutathione peroxidase and glutathione S-transferase activite were decreased progressively with increasing number of Cd injection, but increased by the supplementation of Se. The reduced form of glutathione (GSH) contents in blood and liver and vitamin E content were decreased and oxidized form (GSSG) increased in Cd groups, but these of Se supplemented groups were not very different from controls. Cd reduced liver vitamin E content which was not restored by Se supplementation. Liver lipid peroxide values were elevated with increasing doses of Cd, but Se supplementation reduced these elevated levels. Accumulation of metallothionein in liver and kidney was increased with increasing number of Cd injection, but Se did not affect on them. Histological examination revealed that lysosomes were significantly increased and mitochondria and Golgi apparatus were enlarged by Cd, however, these changes were reduced by Se. It was concluded that Se administration promoted antioxidative detoxification and alleviated peroxidative damage in rat liver by Cd.

  • This study was performed to investigate the effect of dietary protein and calcium levels on cadmium detoxication in rats. Seventy male Sprague-Dawley rats weighing 208 $\pm$ 19 g were blocked into 10 groups of 7 animals according to body weight. Five groups were fed 15% protein-0.6% calcium diet with 100ppm cadmium in drinking water for first 15days and the other 5groups fed same diet without cadmium in drinking water for same period and served as controls. After this 15-day intoxication period, each one of cadmium intoxication and control groups were fed each of 4 kinds of detoxifying diets different with protein(40%, 15%) and calcium(1.3%, 0.6%) levels without cadmimum in drinking water for following 15 days of detoxifying period. Results were summarized as follows: 1) Food intake, body weight gain, F.E.R. and weights of liver, kidney and femur were increased by detoxifying diets and high protein diet was most effective in weight gains of liver and kidney. 2) When cadmium and metallothionein contents of initial intoxication group and those of all detoxication groups were compared, cadmium and metallothionein contents in the liver were not changed, but those in kidney increased, and those in intestine decreased markedly. 3) Only dietary protein level affected cadmium and metallothionein distribution among organs, and cadmium contents of whole blood, liver, kidney and femur were lower in high protein diet, but metallothionein contents in liver and kidney were higher in high protein diet. 4) Gel filtration chromatogram showed that most of cadmium in the cytosol was bound to metallothionein fractions in high protein-high calcium group. Results obtained indicated that high protein diet was effective in cadmium detoxication by increasing the induction of metallothionein synthesis. But high calcium diet did not play a role in cadmium detoxication.

  • 당뇨병쥐에 있어서 생체내 항산화방어계에 미치는 zinc투여의 영향을 관찰하기 위하여 비타민 E 투여군과 비교실험하였다. 혈당량은 당뇨군들이 대조군에 비해 현저히 증가되었으나 ZDM군이 DM과 EDM군에 비해 약간 감소되는 경향을 띠었다. 혈장 인슐린수준은 혈당량과 반대의 경향을 띠었다. 간조직중의 TBARS 축적량은 대조군에 비해 DM군은 2.3배 높았고 ZDM군은 DM에 비해 낮아졌으며, EDM군은 대조군과 비슷하였다. 간조직중의 환원형 glutathione(GSH) 함량은 대조군에 비해 DM군은 감소되었으나 ZDM과 EDM군은 DM군에 비해 각각 2.3, 1.7배씩 증가되었다. 산화형 GSH함량은 대조군에 비해 DM군이 증가되었고, DM군에 비해 ZDM군과 EDM군 모두 감소하였으며 GSH/GSSG비는 GSH비와 비슷한 경향이었다. 간조직중의 SOD, GSH-Px, GST활성은 모두 대조군에 비해 DM군은 유의적으로 감소되었으나 ZDM, EDM군은 DM군에 비해 증가되었다. 간장과 신장에서의 metallotruonein 함량은 모두 대조군에 비해 DM, EDM군은 증가하였고, ZDM군은 20배, 5.3배 각각 현저히 증가하였다.

Cadmium에 대한 Metallothionein 및 Free Radical Scavenger에 관한 최근의 연구 동향 (Recent Research Trend on Free Radical Scavenger and Metallothioneins on Cadmium)

  • Since the Industrial Revolution, people have enjoyed an abundant life, owing to the technological innovations of science. However, because of changes in the environment or polution to the environment, it has brought on many chronic diseases. Especially, even though it may be a small amount, if cadmium gets into a human body, because its biological half life is long, it fatally causes a kidney disease and damage to human organisms. It was reported that Metallothioneins(MT), a substance that is closely related with a free radical that comes from environmental pollution and the course of which cadmium, which causes chronic addition in the body, is formed, along with harmful metals, make the toxicity reduce. MT compound led into the body by cadmium indirectly functions as an antioxidation, supplied adequate amount of vitamin E, and suppresses the accumulation of cadmium in heart, liver and blood. Cadmium content found in Korean soil according to the test conducted from 1992 to 1996 was 0.02~0.03mg/kg in cereals, pulses, and potatoes. Free radical scavenger, finding solution for metal substance that comes from environmental pollution from food and natural substances implicates a better future for the study of food science.

Metallothinein 1E Enhances Glioma Invasion through Modulation Matrix Metalloproteinases-2 and 9 in U87MG Mouse Brain Tumor Model

  • Malignant glioma cells invading surrounding normal brain are inoperable and resistant to radio- and chemotherapy, and eventually lead to tumor regrowth. Identification of genes related to motility is important for understanding the molecular biological behavior of invasive gliomas. According to our previous studies, Metallothionein 1E (MT1E) was identified to enhance migration of human malignant glioma cells. The purpose of this study was to confirm that MT1E could modulate glioma invasion in vivo. Firstly we established 2 cell lines; MTS23, overexpressed by MT1E complementary DNA construct and pV12 as control. The expression of matrix metalloproteinases (MMP)-2, -9 and a disintegrin and metalloproteinase 17 were increased in MTS23 compared with pV12. Furthermore it was confirmed that MT1E could modulate MMPs secretion and translocation of NFkB p50 and B-cell lymphoma-3 through small interfering ribonucleic acid knocked U87MG cells. Then MTS23 and pV12 were injected into intracranial region of 5 week old male nude mouse. After 4 weeks, for brain tissues of these two groups, histological analysis, and immunohistochemical stain of MMP-2, 9 and Nestin were performed. As results, the group injected with MTS23 showed irregular margin and tumor cells infiltrating the surrounding normal brain, while that of pV12 (control) had round and clear margin. And regrowth of tumor cells in MTS23 group was observed in another site apart from tumor cell inoculation. MT1E could enhance tumor proliferation and invasion of malignant glioma through regulation of activation and expression of MMPs.

식이내 Calcium 수준이 흰쥐의 카드뮴과 납중독에 미치는 영향 (Effect of dietary Calcium Level on Cadmium and Lead Toxicity in Rats)

  • This study was performed to investigate the effect of dietary calcium level on cadmium and lead toxicity in rats. Fifty-four male rats of Sprague-Dawely strain weighing 152$\pm$12g were blocked into 9 groups according to body weight, and were raised for 30 days. Nine experimental diets different with cadmium(0%, 0.04%), lead (0%, 0.071%) and calcijm(0.5%, 1.0%, 1.5%) levels were prepared. The results are summarized as follow. Weight gain, F.E.R.(food efficiency ratio), and weights of liver, kidney and femur were lower in cadmium exposed groups than those of heavy metal free groups. Weight gain F.E.R. and ash weight of lead groups were lower than those of heavy metal free groups. But, these were increased with increasing dietary calcium level. Cadmium and lead concentrations in blood, liver, kidney and femur were lower in rats fed 1.5% calcium than 0.5% calcium diet. Fecal cadmium and lead excretions were remarkably increased in 1.5% calcium groups, and cadmium and lead retention rates were decreased in 1.5% calcium groups. Metallothionein concentrations in liver, kidney and small intestine were higher in rats exposed to cadmium and lead. Calcium content in blood, femur and daily urinary and fecal calcium excretion were decreased by cadmium and lead additions, and increased in 1.5% calcium groups. Creatinine clearance were decreased with cadmium administratino and calcium addition. In conclusion, weight gain and organ weights were decreased with cadmium or lead administration. But, cadmium administration was more toxic than lead adminstration. Cadmium or lead toxicity was alleviated by increasing dietary calcium level. Especially, lead toxicity was alleviated in proportion to dietary calcium level.

  • 생쥐에서 페르몬으로 알려진 소수성의 분자와 결합하는 major urinary proteins(MUPs)는 부분적으로 성장 호르몬(GH)의 조절을 받는다. 본 연구에서는 MT-GRF로 형질전환되어 성장호르몬이 증가된 생쥐에서 MUP의 발현을 조사하였다. 이 MT-GRF로 형질전환된 생쥐의 소변에서 MUP이 대조군보다 심하게 저하되어 나타났고, 암컷보다 수컷에서 더 저하되어 나타났다. 또한 MT-GRF를 근육에 주사한 생쥐에서도 MUP는 저하되어 나타났다. 부가하여, MT-GRF로 형질전환시킨 생쥐의 소변에서 전기영동상에서 albumin과 동일하게 이동하는 고분자의 단백질이 다량으로 관찰되었는데 신장의 사구체 여과가 손상되었음을 암시하고 있다. 이 혈장 단백질의 손실도 암컷보다 수컷에서 심하게 나타났다. 결과적으로 GRF의 과다생성은 성별에 차별화된 영향을 미치면서 GH로 형질전환된 동물모델에서 관찰되는 MUP의 발현과 사구체 기능의 변화를 동일하게 유발하고 있었다.

카드뮴 투여수준이 흰쥐의 임상적인 중독증과 정자능력 및 장기조직에 미치는 영향 (Effect of Dietary Cadmium Level on Clinical Toxicity, Sperm Capacity and Histopathological Changes in Rats)

  • 카드뮴 (Cd)의 투여가 흰쥐의 임상적인 중독 정도와 청자능력 및 장기조직에 미치는 영향을 조사하였다. 카드뮴의 임상적인 중독증 유발을 위한 시험사료는 흰쥐용 purified diet(AIN-76)에 CdCl를 이용하여 Cd 농도가 각각 0, 25, 50, 100 및 250ppm이 되도록 첨가하였다. 생후 7주령 ( 평균체중 231$\pm$14g) 된 Sprague-Dawley계 웅성 흰쥐 30 수를 체중에 따라 완전임의 배치법으로 6수씩 5군으로 나누어 8주간 사육하였다. 중체량, 사료섭취량 및 음수량은 카드뮴 첨가수준이 높을수록 유의하게 감소한 것으로 나타났으며, 장기무게에 있어서 특히 신장과 정소의 무게는 카드뮴 첨가수준이 높을수록 비대하였고 처리구간 유의한 차이가 있었다 (P<0.05). Hematocrit 치와 hemoglobin 함량 역시 Cd 첨가수준이 높을수록 감소하였고 처리구간 유의한 차이가 있었다 (P<0.05). 식이중 카드뮴의 간과 신장조직에의 평균 축적율은 간 조직이 34.1%, 신장조직이 54.3% 이었고, 저수준의 Cd 투여군이 고수준의 Cd 투여군 보다 간과 신장조직에의 축적율이 대체로 높았다. 간 조직중 metallothionein(MT) 함량은 대조군 (A) 이 30.0 $\mu\textrm{g}$/g 인데 비해 250ppm 투여군 (E) 에서는 138.6 $\mu\textrm{g}$/g 으로 카드뮴 투여수준에 따라 MT 합성은 유의한 영향을 받은 것으로 나타났다. 정자농도는 대조군에 비해 Cd 투여수준에 따라 유의한 감소 경향 (P<0.05) 이었고, 정자 활력은 비록 통계적인 차이는 없었으나 대조군에 비해 Cd 투여군에서 낮은 경향이었다. 한편 카드뮴 투여가 조직병변에 미치는 결과를 보면 육안병변의 경우 대조군에 비하여 Cd l00ppm과 250ppm 투여군의 흰쥐에서 외관상 수척되어 있었고, 특히 250ppm 투여군의 흰쥐에서 신장과 고환의 현저한 위축이 관찰되었다. 병리조직학적 검사에서 Cd 투여군에서 간상간세포의 변성, 간세포와 간세포사이의 모세혈관이 sinusoid에서의 호산성물질의 저류 및 Kupffer cell의 활성화가 관찰되었다. 신장의 경우 Cd 투여군에서 세뇨관 상피세포의 변성 및 호염성병변이 관찰되었으며 고환에서는 Cd 투여 농도에 의해서 세포변성, 정자세포수의 감소 및 거핵세포의 출현이 관찰되었다. 따라서 카드뮴에 장기간 노출은 성선조직의 변화에 의해 정자활력 및 농도의 감소가 일어나고 이로 인해 번식효율이 저하되는 한 요인으로 작용되는 것으로 사료된다.
