Pathogen infection in plants induces complex responses ranging from gene expression to metabolic processes in infected plants. In spite of many studies on biotic stress-related changes in host plants, little is known about the metabolic and phenotypic responses of the host plants to Pseudomonas cichorii infection based on image-based analysis. To investigate alterations in tomato plants according to disease severity, we inoculated plants with different cell densities of P. cichorii using dipping and syringe infiltration methods. High-dose inocula (${\geq}10^6cfu/ml$) induced evident necrotic lesions within one day that corresponded to bacterial growth in the infected tissues. Among the chlorophyll fluorescence parameters analyzed, changes in quantum yield of PSII (${\Phi}PSII$) and non-photochemical quenching (NPQ) preceded the appearance of visible symptoms, but maximum quantum efficiency of PSII ($F_v/F_m$) was altered well after symptom development. Visible/near infrared and chlorophyll fluorescence hyperspectral images detected changes before symptom appearance at low-density inoculation. The results of this study indicate that the P. cichorii infection severity can be detected by chlorophyll fluorescence assay and hyperspectral images prior to the onset of visible symptoms, indicating the feasibility of early detection of diseases. However, to detect disease development by hyperspectral imaging, more detailed protocols and analyses are necessary. Taken together, change in chlorophyll fluorescence is a good parameter for early detection of P. cichorii infection in tomato plants. In addition, image-based visualization of infection severity before visual damage appearance will contribute to effective management of plant diseases.

Purpose: The tumor microenvironment is known to be associated with the metabolic activity of cancer cells and local immune reactions. We hypothesized that glucose metabolism measured by 2-deoxy-2-($^{18}F$)fluoro-D-glucose ($^{18}F-FDG$) positron emission tomography (PET)-computed tomography (CT) ($^{18}F-FDG$ PET-CT) would be associated with local immune responses evaluated according to the presence of tumor infiltrating lymphocytes (TILs). Materials and Methods: We retrospectively reviewed 56 patients who underwent $^{18}F-FDG$ PET-CT prior to gastrectomy. In resected tumor specimens, TIL subsets, including cluster of differentiation (CD) 3, CD4, CD8, Forkhead box P3 (Foxp3), and granzyme B, were subjected to immunohistochemical analysis. The prognostic nutritional index (PNI) was calculated as: ($10{\times}serum$ albumin value)+($0.005{\times}peripheral$ lymphocyte counts). Additionally, the maximum standard uptake value ($SUV_{max}$) was calculated to evaluate the metabolic activity of cancer cells. Results: The $SUV_{max}$ was positively correlated with larger tumor size (R=0.293; P=0.029) and negatively correlated with PNI (R=-0.407; P=0.002). A higher $SUV_{max}$ showed a marginal association with higher CD3 (+) T lymphocyte counts (R=0.227; P=0.092) and a significant association with higher Foxp3 (+) T lymphocyte counts (R=0.431; P=0.009). No other clinicopathological characteristics were associated with $SUV_{max}$ or TILs. Survival analysis, however, indicated that neither $SUV_{max}$ nor Foxp3 held prognostic significance. Conclusions: FDG uptake on PET-CT could be associated with TILs, especially regulatory T cells, in gastric cancer. This finding may suggest that PET-CT could be of use as a non-invasive tool for monitoring the tumor microenvironment in patients with gastric cancer.
In this study, using a continuous behavior measurement technique, the short-term behavioral responses and tolerance limits of red seabream Pagrus major fingerlings to sudden exposure to low salinity in a controlled environment were observed. The activity of the fingerlings suddenly exposed to 21.4, 17.3, and 9.8 psu increased temporarily at the initial exposure to show irregular swimming behavior, but then recovered a stable activity pattern through rapid salinity adaptation. However, the organisms suddenly exposed to 7.3 and 4.3 psu could not withstand the salinity stress, and their swimming behavior was severely disturbed and all individuals died within 48 hours. The findings suggest that red seabream underwent a temporary salinity stress process at the beginning of the exposure to concentrations of 10.0 psu or higher. At these concentrations, osmotic control was possible within at least 11 hours, so stable metabolic activity was also possible. However, organisms suddenly exposed to concentrations below 5.0 psu exceeded the tolerance to low salinity and the sublethal limit. In red seabream exposed to this concentration range, severe behavioral and metabolic disturbances were observed, and death was observed due to osmotic control failure. In conclusion, a salinity range of 5.0 to 10.0 psu can be predicted to correspond to a concentration range in which the osmotic control ability of the red seabream fingerlings is lost, and sub-lethal reactions occur.
A splenocyte culture system supplemented with liver microsomes was developed to detect immunotoxic chemicals which require metabolic activation using cyclophosphamide as a positive standard. When liver microsomes were added to splenocyte cultures isolated from female B6C3Fl mice, the proliferation of splenocytes by lipopolysaccharide (LPS) was increased and the proliferation by concanavalin A (Con A) was decreased. However, when compared with each corresponding control, cyclophophamide was successfully activated to metabolites capable of suppressing Iymphoproliferative responses. This suppression was clearly dependent upon the amounts of microsomes added and/or the concentration of cyclophosphamide exposed. In these cultures, the proliferation of splenocytes was suppressed when the cells were exposed to cyclophosphamide on the day of culture initiation. On the other hand, microsome was responsible for the increase in LPS mitogenicity and NADPH was responsible for the decrease in Con A mitogenicity. Finally, our present culture system was compared with the hepatocyte-splenocyte coculture system which we had developed earlier. We found that the hepatocyte-splenocyte coculture was better able to activate cyclophosphamide to metabolites capable of suppressing the antibody response to sheep erythrocytes. Although our present culture system was relatively poor to activate cyclophosphamide in cultures for antibody response, it will be useful as a simple screening method to detect suppression of certain in vitro immunotoxic parameters like LPS mitogenicity by chemicals which require metabolism.
Inflammasomes are specialized signaling platforms critical for the regulation of innate immune and inflammatory responses. Various NLR family members (i.e., NLRP1, NLRP3, and IPAF) as well as the PYHIN family member AIM2 can form inflammasome complexes. These multiprotein complexes activate inflammatory caspases (i.e., caspase-1) which in turn catalyze the maturation of select pro-inflammatory cytokines, including interleukin (IL)-$1{\beta}$ and IL-18. Activation of the NLRP3 inflammasome typically requires two initiating signals. Toll-like receptor (TLR) and NOD-like receptor (NLR) agonists activate the transcription of pro-inflammatory cytokine genes through an NF-${\kappa}B$-dependent priming signal. Following exposure to extracellular ATP, stimulation of the P2X purinoreceptor-7 ($P2X_7R$), which results in $K^+$ efflux, is required as a second signal for NLRP3 inflammasome formation. Alternative models for NLRP3 activation involve lysosomal destabilization and phagocytic NADPH oxidase and /or mitochondria-dependent reactive oxygen species (ROS) production. In this review we examine regulatory mechanisms that activate the NLRP3 inflammasome pathway. Furthermore, we discuss the potential roles of NLRP3 in metabolic and cognitive diseases, including obesity, type 2 diabetes mellitus, Alzheimer's disease, and major depressive disorder. Novel therapeutics involving inflammasome activation may result in possible clinical applications in the near future.

Caballeronia sordidicola strain PAMC 26592 was isolated from Umbilicaria sp., a lichen material collected from Svalbard Archipelago in the Arctic Ocean. We report the draft genome sequence of the strain PAMC 26592, a metabolic generalist. As we have observed in previous genomic studies in the genus Caballeronia draft genomic sequences of PAMC 26592 had an assortment of genes of ecological importance and of bio-technical potentials, which include diverse metabolic genes for carbohydrates, aromatic compounds, amino acids, and vitamins, and genes for nitrogen / sulfur metabolisms, stress responses, membrane transporters, antibiotic resistance, and heavy metal resistance.

Cryopreservation is a process that preserves organelles, cells, tissues, or any other biological constructs by cooling the samples to very low temperatures. The responses of living cells to ice formation are of theoretical interest and practical relevance. Stem cells and other viable tissues, which have great potential for use in basic research as well as for many medical applications, cannot be stored with simple cooling or freezing for a long time because ice crystal formation, osmotic shock, and membrane damage during freezing and thawing will cause cell death. The successful cryopreservation of cells and tissues has been gradually increasing in recent years, with the use of cryoprotective agents and temperature control equipment. Continuous understanding of the physical and chemical properties that occur in the freezing and thawing cycle will be necessary for the successful cryopreservation of cells or tissues and their clinical applications. In this review, we briefly address representative cryopreservation processes, such as slow freezing and vitrification, and the available cryoprotective agents. In addition, some adverse effects of cryopreservation are mentioned.
Though bile acids have been well known as digestive juice, recent studies have demonstrated that bile acids bind to their endogenous receptors, including Farnesoid X receptor (FXR) and G protein-coupled bile acid receptor 1 (GPBAR1; TGR5) and serve as hormone to control various biological processes, including cholesterol/bile acid metabolism, glucose/lipid metabolism, immune responses, and energy metabolism. Deficiency of those bile acid receptors has been reported to induce diverse metabolic syndromes such as obesity, hyperlipidemia, hyperglycemia, and insulin resistance. As consistent, numerous studies have reported alteration of bile acid signaling pathways in type II diabetes patients. Interestingly, bile acids have shown to activate TGR5 in intestinal L cells and enhance secretion of glucagon-like peptide 1 (GLP-1) to potentiate insulin secretion in response to glucose. Moreover, FXR has been shown to crosstalk with TGR5 to control GLP-1 secretion. Altogether, bile acid receptors, FXR and TGR5 are potent therapeutic targets for the treatment of metabolic diseases, including type II diabetes.
This research investigates the influence of chair tilt functionality on biometric signals and user comfort, addressing the ergonomic challenges posed by modern sedentary lifestyles. Through an experimental study involving eight male participants, the impact of chair tilt on electromyography (EMG), heart rate, metabolic rate, pressure distribution, and distance between the lumbar spine and the lumbar support part of the chair was measured across different seating postures. The study utilized chairs with both synchronous and non-synchronous tilt mechanisms to explore how adjustments in chair design affect user comfort and physiological responses during prolonged sitting. Key findings suggest that chair tilt functionality can significantly reduce muscle activity and energy expenditure, enhancing user comfort and potentially mitigating health risks associated with prolonged sedentary behavior. Notably, the study revealed a preference among participants for chairs that aligned the rotational center of the tilt with the hip joint, highlighting the importance of this ergonomic feature in enhancing user comfort. Additionally, the research proposes a novel methodology for assessing seating comfort through the analysis of both biometric and physical signals, providing valuable insights for the development of ergonomic chair designs focused on user health and comfort.

The knowledge of the nutrition in animal production is difficult to assess due of difficulty to determine the amount and quality of feeds intake, mainly if forages are the most important compound in the diet. It must be able to know responses to the metabolic process in lactating dairy cows earring out feeds evaluation trials. These metabolism studies with cows, requires measurements of: I) The amount of all feeds consumed. II) Excretion of faeces. III) Excretion of urine. Taking as a basis these trials, forage and total intake, dietary digestibility and balances of nitrogen and energy can be calculated. However, these feeds evaluation experiments with animals are very time consuming and expensives. The faeces excreted by animals containt undigested residues of the diet consumed. For this reason, their analysis can be an successful tool to determine the amount and quality of feed intake and other important biological parameters. The aim of this work was to know if faeces analysis by NIR could be used to determine with enough accuracy some attributes of different lactating dairy cows diets, using a global equation, developed on an heterogeneus population. For this purpose a total of 79 faecal samples from eight grass, three grass silages and two maize silages metabolic trials, on six cows each one, were used to constitute the initial population. The calibration equations were developed to predict forage and total intake, organic matter digestibility, digestibility coefficient of crude energy and digestibility energy. The combination of several trials with different diets and animal conditions gave promising results.
