Effects of Pear Extracts Containing Herbal Medicine (Lycii Fructus, Coicis Semen, Alimatis Rhizoma, and Astragali Radix) on Body Weight, Lipid Metabolism, and Immune Responses of Rats Fed with High Fat Diets ( I ) (배추출물과 구기자, 의이인, 택사, 황기 배합이 고지방식이로 유발된 비만 흰쥐의 체중, 지질대사 및 면역기능에 미치는 영향 ( I ))

  • Objective : This study was conducted to observe the anti-hyperlipidemic and anti-inflammatory effects, as well as the metabolic benefits, of pear extract and herbal drug mixture (Lycii Fructus, Coicis Semen, Alimatis Rhizoma, and Astragali Radix) on rats fed with a high fat diet. Methods : The animals used were male rats and the control group was fed a high fat diet only. The experimental groups were divided into four. Exp I group was fed a high fat diet with a mixture of pear extract and 3% Lycii Fructus; Exp II group was fed a high fat diet with a mixture of pear extract and 3% Coicis Semen; Exp III group was fed a high fat diet with a mixture of pear extract and 3% Alimatis Rhizoma; and Exp IV group was fed a high fat diet with a mixture of pear extract and 3% Astragali Radix for 4 weeks. Results : The body weight gain increased in all groups, but attenuated gradually in the experimental groups compared to the control group. The food intakes were significantly lower in Exp I and Exp III groups than the control group. The concentrations of serum total cholesterol (TC), HDL-cholesterol, LDL-cholesterol, and triglyceride (TG) were significantly higher in Exp II than in the control group, and lower in Exp III group than in the control group. Also the concentration of serum free fatty acid was significantly lower in the Exp III group than in the control group. In inflammatory activities, the Exp II group was significantly lower than the control group. Conclusion : The results indicated that Exp III group (administered a mixture of pear extract and Alimatis Rhizoma) most efficiently reduced fat accumulation and body weight, while the Exp II group (administered the mixture of pear extract and Coicis Semen) had the highest elevated lipid metabolism and immune activity.

Molecular Basis of Organospecific Carcinogensis by Chemical Carcinogens-Study with Breast Cancer Specific Carcinogens: DMBA as an Indirect-Acting carcinogen and NMU as a Direct-Acting cancinogen. (화학적 발암원의 조직 특이성 암유발기전 - DMBA와 NMU의 선택적 유암 발생기전을 중심으로 )

  • To study the selective organospecific carcinogenesis by the specific chemical carcinogens, the breast cancer induction model by oral administration of 7, 12-dimethylbenzanthracene (DMBA) or by intravenous injection of N-methylni-trosourea (NMU) on female rats was analyzed. In the present experiment, we compared the effexts of ages on the chemical mammary carcinogenesis by studying the metabolic system of the carcinogenic activation, detoxification or DNA damage and repair. The breast tumor incidence was significantly higher in the young rats of 50 days old than in those of one year old rats. As an index of organospecific DNA damage or repair, the in vivo covalent binding index(CBI) of the specific organs by the specific chemical carcinogens was monitored. And for the analysis of carcinogenic activation, the quantity of cytochrome P450`s was determined with the respective type-specific monoclonal antibody, while the detoxication capacity was deduced by the activity monitoring of glutathione S-transferase (GST) and peroxidase. The skin tissues of the mammary region had the highest CBI with both of DMBA and NMU at 50 days of age. And there were contrasting differences in the contents of carcinogenic activation and detoxication system: that is, the content of T.C.D.D.-inducible cytochrome P450 was high, while the activities of GST and peroxidase was low in the mammary skin tissues at tumor prevalent age. These results led us to conclude that the molecular organospecific carcinogenesis, as illustrated with mammary carcinoge-nesis by DMBA and NMU, is operated probably through the differential capacity of the target tissues in the high carcinogenic activation, low detoxication and the low DNA repair function.

Effect of Fermented Red Ginseng Extracts on Physiological Activity and Blood Glucose Level in Streptozotocin Induced Diabetic Rats (홍삼발효 추출물의 생리활성 및 streptozotocin으로 유발된 당뇨쥐의 혈당강하에 미치는 영향)

  • The purpose of this study was investigated hypoglycemic effects of fermented red ginseng extracts. We prepared non-fermented red ginseng extracts(R), fermented with Lactobacillus plantarum(RL) extracts, Saccharomycescerevisiae(RS) extracts, and L. plantarum mixed S. cerevisiae(RLS) extracts, examined composition of ginsenosides, SOD-like activity, and $\alpha$-glucosidase inhibitory activity. Ginsenoside Re was highest contents in all extracts, second was ginsenoside Rc and then ginsenoside Rb1. Concentration of these ginsenoside was showed higher in RS than in other extracts. SOD-like activity and $\alpha$-glucosidase inhibitory activity were shown higher in fermented red ginseng extracts than non fermented extracts. And activities of mixed fermentation extracts(RLS) higher than single fermentation extracts(RL, RS). Effects of blood glucose level, serum lipid profile and metabolic variables were evaluated in streptozotocin(STZ) induced diabetic rat. Experimental group was divided into 7 groups: normal control group(hereafter NC group), diabetes control group(DC group), positive control group treated with 50 mg/kg body weight of acarbose(PC group), treated with 300 mg/kg body weight of R, RL, RS and RLS extracts groups, respectively. Blood glucose level of DC group was maintained high level in all experimental period, but treated with red ginseng extracts groups was reduced the glucose level by R group 18.00%, RL group 28.07%, RS group 29.03%, RLS group 42.42%, respectively. The concentration of total cholesterol and triglyceride of fermented red ginseng extracts treated groups (RL, RS, RLS) was lower than non- fermented extracts group(R) DC and PC groups. The activity of ALT, AST in RLS treated groups were lower than other groups.

  • Carotenoids are isoprenoids with a long polyene chain containing 3 to 15 conjugated double bonds, which determines their absorption spectrum. They typically consist of a $C_{40}$ hydrocarbon backbone often modified by different oxygen-containing functional groups, to yield cyclic or acyclic xanthophylls. Much work has also been focused on the identification, production, and utilization of natural sources of carotenoid (plants, microorganisms and crustacean by-products) as an alternative to the synthetic pigment which currently covers most of the world markets. Nevertheless, only a few carotenoids (${\beta}-carotene$, lycopene, astaxanthin, canthaxanthin, and lutein) can be produced commercially by fermentation or isolation from the small number of abundant natural sources. The market and demand for carotenoids is anticipated to increase dramatically with the discovery that carotenoids exhibit significant anti-carcinogenic activities and play an important role in the prevention of chronic diseases. The increasing importance of carotenoids in the feed, nutraceutical food and pharmaceutical markets has renewed by efforts to find ways of producing additional carotenoid structures in useful quantities. Because microorganisms and plants synthesize hundreds of different complex chemical carotenoid structures and a number of carotenoid biosynthetic pathways have been elucidated on a molecular level, metabolic and genetic engineering of microorganisms can provide a means towards economic production of carotenoid structures that are otherwise inaccessible. The aim of this article is to review our current understanding of carotenoid formation, to explain the perceived benefits of carotenoid in the diet and review the efforts that have been made to increase carotenoid in certain microorganisms.

Effects of the Prunus persica Batsch var. davidiana Max. Extract on the Blood Glucose and Serum Lipid Components in Streptozotocin-Induced Diabetic Rats (돌복숭아(Prunus persica Batsch var. davidiana Max.) 추출액이 Streptozotocin 유발 당뇨성 흰쥐의 혈당 및 지질 성분에 미치는 영향)

  • The purpose of this study was designed to observe the effects of the feeding Prunus persica Batsch var. davidiana Max. extract on the improvement of the blood glucose, lipid compositions in the serum of streptozotocin(STZ)-induced diabetic rats fed the experimental diets for 5 weeks. Concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, triglyceride(TG) and phospholipid (PL) in serum were significantly higher in the STZ (55 mg/kg B.W.)-induced diabetic group (group 2) and STZ(I.P.)+Prunus persica 5.0 g% extract group(group 3) than those in the control group (group 1, basal diet + water). But the concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, TG and PL in serum were remakably lower in the group 3 than those in the group 2. In the ratio of HDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration, Prunus persica 5.0 g% extract administration group(group 3) were higher percentage than in the group 2. The activities of aspartate aminotransferase(AST), alanine aminotransferase(ALT), lactate dehydrogenase (LDH) and alkaline phosphatase(ALP) in serum were rather lower in the Prunus persica 5.0 g% extract administration group(group 3) than in the STZ- induced diabetic group (group 2). From the above results, it was suggested that the Prunus persica Batsch var. davidiana Max. were effective on the improvement of the blood glucose, lipid compositions in serum of STZ-induced diabetic rats. Moreover, in Prunus persica Batsch var. davidiana Max. was effective therapeutic regimen for the control of metabolic derangements in adult disease.

  • Insect resources have been widely recognized that seven millions of insects exhibit it own biological activity by whole body or its metabolic intermediates. In order to investigate antioxidant activity and compare the cyclooxygenase-2 promoter activity from insect extract, we tested in vitro antioxidant assays and cyclooxygenase-2 promoter assay in Coccinella septempunctata Linne and Harmonia axyridis extracts have the anti-oxidant and cyclooxygenase-2 inhibition activities, we examined the anti-oxidant assays including DPPH, FRAP and linoleic acid, ana inhibition of cyclooxygenase-2 expression using a cyclooxygenase-2 promoter-inserted stable cell line. We found that Harmonia axyridis Pallas extract had potentials to anti-oxidant activity and inhibited about 25% of cyclooxygenase-2 transcription activity. These findings indicate that Coccinella septempunctata Linne and Harmonia axyridis Pallas extracts could be an useful insect resource for agrobiotechnological purposes.

A Study on the Relations between Yangseng Level and Obesity in Industrial Workers (근로자(勤勞者)의 양생수준(養生水準)과 비만(肥滿)과의 관계(關係))

  • In this study, the researcher tried to present the groundwork to prepare the oriental medical yangseng plan for the future obesity with the comparison between yangseng level and the obesity of workers. The researcher made up the questionnaire which asks the general character, health-related character and yangseng level, projected among 560 people. All collected material was analyzed by SPSS and tested by T-test and ANOVA. 1. The general yangseng level average is 3.27, morality yangseng 3.90, sleep yangseng 3.39, mind yangseng 3.31, sex life yangseng 3.30, exercise yangseng 3.15, activities and rest yangseng 3.08, diet yangseng 2.94, seasonal yangseng 2.84. The highest is morality yangseng and seasonal yangseng is the lowest. 2. In the aspect of yangseng level: Having a spouse, Non-smoking, Regular exercising, Sufficient sleeping have higher yangseng levels. 3. Obesity related index is changing into the higher level when he/she is older, more paid, more job experience, more education background but less metabolic calory. And drinking and regular exercising have relativity with body composition analysis. 4. After comparing yangseng level with body composition analysis, we can easily find that the more visceral fat, the higher WHR has a high yangseng level. The more mineral also has a high mind yangseng and a low diet yangseng. The visceral fat level has a high yangseng level when higher morality yangseng and mind yangseng are getting higher and higher. And sex life yangseng shows that the highest yangseng level is from 9-10 visceral fat and the lowest yangseng level is from below 4 visceral fat. The higher WHR, the higher morality, mind and sleep yangseng. The heavier, the lower diet yangseng. The mind yangseng was very high when body fat rate was higher. The more body fat, the higher morality yangseng. The higher yangseng, when we have more muscle. Yangseng level and obesity of laborer has a close relationship with individual character and daily habits. Also, relevance can be easily found between yangseng level and obesity. Now the researcher came into the conclusion that we need to control over laborers' health and prevention of their obesity.

  • During sleep, relatively major respiratory physiological changes occur in healthy subjects. The contributions and interactions of voluntary and metabolic breathing control systems during waking and sleep are quite different Alterations of ventilatory control occur in chemosensitivity, response to mechanical loads, and stability of ventilation. The activities of intercostal muscles and muscles involved in regulating upper airway size are decreased during sleep. These respiratory physiological changes during sleep compromise the nocturnal ventilatory function, and sleep is an important physiological cause of the nocturnal alveolar hypoventilation. There are several causes of chronic alveolar hypoventilation including cardiopulmonary, neuromuscular diseases. Obstructive sleep apnea syndrome (OSAS) is an important cause of nocturnal hypoventilation and hypoxia. Coexistent cardiopulmonary or neuromuscular disease in patients with OSAS contributes to the development of diurnal alveolar hypoventilation, diurnal hypoxia and hypercapnia. The existing data indicates that nocturnal recurrent hypoxia and fragmentation of sleep in patients with OSAS contributes to the development of systemic hypertension and cardiac bradytachyarrhythmia, and diurnal pulmonary hypertension and cor pulmonale in patients with OSAS is usually present in patients with coexisting cardiac or pulmonary disease. Recent studies reported that untreated patients with OSAS had high long-term mortality rates, cardiovascular complications of OSAS had a major effect on mortality, and effective management of OSAS significantly decreased mortality.

Effects of Chromium on Energy Metabolism in Lambs Fed with Different Dietary Protein Levels

  • The effects of chromium (Cr), dietary crude protein (CP) level, and potential interactions of these two factors were investigated in term of energy metabolism in lambs. Forty-eight 9-week-old weaned lambs (Dorper${\times}$Small-tail Han sheep, male, mean initial body weight = 22.96 kg${\pm}$2.60 kg) were used in a 2${\times}$3 factorial arrangement of supplemental Cr (0 ${\mu}g$/kg, 400 $\mu{g}$/kg or 800 ${\mu}g$/kg from chromium yeast) and protein levels (low protein: 157 g/d to 171 g/d for each animal, or high protein: 189 g/d to 209 g/d for each animal). Blood samples were collected at the beginning and end of the feeding trial. The lambs were then sacrificed and tissue samples were frozen for further analysis. Chromium at 400 ${\mu}g$/kg decreased fasting insulin level and the ratio of plasma insulin to glucagon, but these differences were not statistically significant; in contrast, chromium at 800 ${\mu}g$/kg increased the ratio significantly (p<0.05). Protein at the high level increased plasma tumor necrosis factor $\alpha$ (TNF-$\alpha$) level (p = 0.060). Liver glycogen content was increased significantly by Cr (p<0.05), which also increased liver glucose-6-phosphatase (G-6-Pase) and adipose hormone-sensitive lipase (HSL) activity. At 400 ${\mu}g$/kg, Cr increased muscle hexokinase (HK) activity. High protein significantly increased G-6-Pase activities in both the liver (p<0.05) and the kidney (p<0.05), but significantly decreased fatty acid synthase (FAS) activity in subcutaneous adipose tissue (p<0.05). For HSL activity in adipose tissue, a Cr${\times}$CP interaction (p<0.05) was observed. Overall, Cr improved energy metabolism, primarily by promoting the glycolytic rate and lipolytic processes, and these regulations were implemented mainly through the modulation by Cr of the insulin signal transduction system. High protein improved gluconeogenesis in both liver and kidney. The interaction of Cr${\times}$CP indicated that 400 $\mu{g}$/kg Cr could reduce energy consumption in situations where energy was being conserved, but could improve energy utilization when metabolic rate was increased.

Nutrient Digestibility, Ruminal Fermentation Activities, Serum Parameters and Milk Production and Composition of Lactating Goats Fed Diets Containing Rice Straw Treated with Pleurotus ostreatus

  • The study evaluated replacement of Egyptian berseem clover (BC, Trifolium alexandrinum) with spent rice straw (SRS) of Pleurotus ostreatus basidiomycete in diets of lactating Baladi goats. Nine lactating homo-parity Baladi goats (average BW $23.8{\pm}0.4$ kg) at 7 d postpartum were used in a triplicate $3{\times}3$ Latin square design with 30 d experimental periods. Goats were fed a basal diet containing 0 (Control), 0.25 (SRS25) and 0.45 (SRS45) (w/w, DM basis) of SRS. The Control diet was berseem clover and concentrate mixture (1:1 DM basis). The SRS45 had lowered total feed intake and forages intake compared to Control. The SRS25 and SRS45 rations had the highest digestibilities of DM (p = 0.0241) and hemicellulose (p = 0.0021) compared to Control which had higher (p<0.01) digestibilities of OM (p = 0.0002) and CP (p = 0.0005) than SRS25 and SRS45. Ruminal pH and microbial protein synthesis were higher (p<0.0001) for SRS25 and SRS45 than Control, which also had the highest (p<0.0001) concentration of TVFA, total proteins, non-protein N, and ammonia-N. All values of serum constituents were within normal ranges. The Control ration had higher serum globulin (p = 0.0148), creatinine (p = 0.0150), glucose (p = 0.0002) and cholesterol (p = 0.0016). Both Control and SRS25 groups had the highest (p<0.05) milk (p = 0.0330) and energy corrected milk (p = 0.0290) yields. Fat content was higher (p = 0.0373) with SRS45 and SRS25 groups compared with Control. Replacement of BC with SRS in goat rations increased milk levels of conjugated linoleic acid and unsaturated fatty acids compared with Control. It was concluded that replacing 50% of Egyptian berseem clover with SRS in goat rations improved their productive performance without marked effects on metabolic indicators health.
