A Study on Menthol Migration Patterns in Different Mentholated Cigarettes (멘솔 첨가방법이 제품담배의 멘솔 전이형태에 미치는 영향)

  • This study was conducted to determine menthol migration from the tobacco to the filter and the change in percentage of menthol in pack for different mentholated cigarettes. Methanol was used to extract menthol from tobacco, filter and foil. A known weight of internal standard was added to each extract and then the extracts were analyzed by gas chromatography. After 4 days, 95% of the menthol applied to the foil was delivered into the tobacco and filter, but in case of menthol applied to the tobacco, the percentage of menthol that migrated from the tobacco to the filter was below 10%. After 100 days, 30-45% of the menthol in the tobacco had migrated into the filter. The amount of menthol in the pack decreased after 20 days of storage, however, the remaining menthol in cigarette treated with 0.30% propylene glycol was higher in amount than that with 0.12% propylene glycol In analysis on menthol transfer to mainstream smoke, the most efficient sample was the cigarette with menthol applied to the foil rather than to the tobacco.

  • The majority of ι-menthol is still obtained by freezing the oil of Mentha arvensis to crystallize the menthol present. This 'natural' menthol is then physically seperated by centrifuging the supernatant liquid away from the menthol crystal. But the price of natural ι-menthol has fluctuated widely so effort has been devoted to the production of ι-menthol by synthetic more readily available raw materials. In the 1970's, many researcher synthesised a new compounds with the menthol cooling effect. During this period many molecular structure designed and synthesised on concepts of correlation between structure and biological activity and the various types of molecule which give rise to cooling effect more than ι-menthol. Specially, N-alkyl-carboxamide group is substituted for the hydroxyl group in ι-menthol. Recently, the most active compounds synthesised is 4-methyl-3-(1-pyrrolidinyl)-2-[5H]-furanone. This compound is 35 times more powerful in the mouth and 512 times more powerful on the skin than ι-menthol. The cooling effect also lasts twice as long. While not yet commercially available, it is expected that these types of materials will be subjected to toxicological studies and will soon be sell on the market.

Improved Menthol Production Using Suspension Cultures of Mentha piperita with Pectinase Elicitation

  • The effect of pectinase on menthol production by Mentha piperita in shake flasks was investigated. The optimum concentration of pectinase and period of elicitation for menthol production were 15 U/1 and 9 days, respectively. Pectinase elicitation at 15 U/1 for 9 days using a Lin-Staba medium with 2,4-dichlorophenoxyacetic acid (2,4-D) enhanced menthol production 37-fold (211.5 ㎎ menthol/l) with the specific menthol concentration (menthol concentration per unit weight of cells) of 27.5 ㎎/g dry cell weight (DCW). Our results also indicate that pectinase elicitation may activate the conversion of pulegone to menthol.

Pyrolytic Behavior of $l$-Menthol ($l$-멘솔의 열분해 특성 분석)

  • This study was conducted to investigate the pyrolysis products of ι-menthol by Curie-Point pyrolysis. The pyrolysis of ι-menthol was performed at 16$0^{\circ}C$, 42$0^{\circ}C$, $650^{\circ}C$, and 92$0^{\circ}C$ by Curie-Point Pyrolyzer and their pyrolysis products were analyzed by GC/MSD. In addition, tobacco leaves added ι-menthol were pyrolyzed at the same condition in case of ι-menthol. The beginning temperature for pyrolysis formation was in the vicinity of 42$0^{\circ}C$ and the major components of the pyrolysis products identified were iso-menthol, 2-menthene, menthomenthene, and menthone. The amount of these components was increased by increasing temperature and the hydrocarbons such as hexadecene and pentadecene formed by ring cleavage were generated at 92$0^{\circ}C$. The yield of ι-menthol in pyrolysis of tobacco leaves was decreased as the temperature of pyrolysis was raised and the pyrolysis products of ι-menthol weren't identified in the pyrolysis of tobacco leaves. Also, to analyze the weight decrease, ι-menthol was analysed by thermal analyzer(TA), and then the weight decrease of ι-menthol was occurred in the vicinity of 18$0^{\circ}C$.

Study of the Migration Rate of L-menthol into Cigarette Pack and Delivery Rate to Cigarette Smoke (포갑 내에서 L-menthol의 이동과 연중 이행 효과에 관한 연구)

  • This study was carried out to investigate the transfer rate of I-menthol constituent into cigarette case and delivery rate to cigarette smoke. Test sample was used a light-type standard brand cigarette made in Korea. I-Menthol contents was measured according to A.O.A.C(1990) method by G.C and smoking conditions was a puff volume of 35$m\ell$ and 2.0s duration, taken once per minute to filter tipping paper plus 3mmwith 20 cigarettes. Migration rate of menthol into cigarette case had an interrelation with triacetin contents into filter and 6% triacetin level was most suitable. I-Menthol transferred to filter part from tobacco sheet by 20.2% in one month after cigarette manufacture, and the transfer was continued subsequently. Maximum menthol transfer in five months was 4.1% to foils and other packing materials and the loss outside of cigarette pack was also 3.9% below. The migration balance of menthol into cigarette pack and the delivery to cigarette smoke were the optimum condition when menthol was added at the ration of 30% in filter to 70% in the other parts into cigarette.

Separation of Menthol/Water Mixture with Surface-Modified Hydrophobic Membrane (표면개질한 소수성 막을 이용한 menthol/water 혼합물의 분리)

  • The surface of tube-type alumina substrate was modified with a silane coupling agent in order to modify the membrane surface with hydrophobicity. Contact angle of water drops on modified membrane was greater than $90^{\circ}$. The modified membrane was tested in pervaporation and vapor permeation for the recovery of menthol from dilute menthol/water mixture. With increasing menthol concentration in the feed at $45^{\circ}C$, permeation rate of menthol in pervaporation and vapor permeation increased from $0.039(g/m^2hr)$ to $0144(g/m^2hr)$ and from. $0.077(g/m^2hr)$ to $0.297(g/m^2hr)$ respectively. When feed concentration is 0.005(g/L) at $45^{\circ}C$, separation factor for menthol in pervaporation and vapor permeation is 20,7 and 40.5 respectively.

Production Enhancement of Menthol in Suspension Cultures of Peppermint Using Cyclodextrin (Peppermint 세포 현탁배양에서 Cyclodextrin을 이용한 Menthol의 생산성 증대)

  • The suspension cultures of Mentha piperita produce menthol which has very low solubility in water due to its hydrophobicity. This can be considered as a factor for its low production in the suspension suspension cultures. Cyclodextrin has the hydrophobic cavity inside the molecule in which menthol can be captured and allow to form a stable complex. The suspension culture of Mentha piperita showed 70% higher production enhancement in the medium containing 1.5%(w/v) $\beta$-cyclodextrin than the control. $\beta$-cyclodextrin had no adverse effect on the cell growth and showed the best result among $\alpha$-, $\beta$- and $\gamma$-cyclodextrins tested in terms of menthol production. We demonstrated that $\beta$-cyclodextrin can be used to enhance the production of menthol in the suspension cultures by capturing hydrophobic menthol into the cavity of cyclodextrin molecules.

Enhanced Solubility of Ibuprofen with Poloxamer 188 and Menthol (폴록사머 188 및 멘톨에 의한 이부프로펜의 용해도 증가)

  • To enhance the solubility of poorly water-soluble ibuprofen with poloxamer and menthol, the effects of menthol and poloxamer 188 on the aqueous solubility of ibuprofen were investigated. In the absence and presence of additives such as ethanol and poloxamer 188, the solubility of ibuprofen increased until the ratio of menthol to ibuprofen increased from 0:10 to 4:6 followed by an abrupt decrease in solubility above the ratio of 4:6, indicating that 4 parts of ibuprofen formed eutetic mixture with 6 parts of menthol. In the presence of poloxamer, the solutions with the same ratio showed abrupt increase in the solubility of ibuprofen. Furthermore, in the presence of poloxamer, the solution with ratio of 4:6 showed more than 2.5- and 6-fold increase in the solubility of ibuprofen compared with that without additives and that without menthol, respectively. The solution with menthol/ibuprofen ratio of 1:9 and higher than 15% poloxamer 188 showed the maximum solubility of ibuprofen, 1.2 mg/ml. Thus, menthol gave the greatly enhanced solubility of ibuprofen with poloxamer 188.

  • We investigated the effects of ethanol on solubilization of L-menthol solution. L-menthol solution was prepared by addition of PEG-40 Hydrogenated Castor Oil and PEG-60 Hydrogenated Castor Oil as nonionic surfactant. Ethanol was added in the solution to investigate effects of ethanol concentration on solubilization. We measured physical properties such as particle size and surface tension. The particle size and the surface tension became smaller with the increase of ethanol concentration.

Uncertainty in Determination of Menthol from Mentholated Cigarette (담배 중 멘톨 분석에 대한 불확도 측정)

  • This study was carried out to evaluate the uncertainty in the analysis of menthol content from the mentholated cigarette. Menthol in the sample cigarette was extracted with methanol containing an anethole as an internal standard, and then analyzed by gas chromatography. As the sources of uncertainty associated with the analysis of menthol, were the following points tested, such as the weighing of sample, the preparation of extracting solution, the pipetting of extracting solution into the sample, the preparation of standard solution, the precision of GC injections for standard & sample solution, the GC response factor of standard solution, the reproducibility of menthol analysis, and the determination of water content in tobacco, etc. For calculating the uncertainties, type A of uncertainty was evaluated by the statistical analysis of a series of observation, and type B by the information based on supplier's catalogue and/or certificated of calibration. Sources of uncertainty were subsequently included and mathematically combined with the uncertainty arising from the assessment of accuracy to provide the overall uncertainty. It was shown that the main source of uncertainty came from the errors in the reproducibility of menthol and water determination, the purity of menthol reference material in the preparation of standard solution, and the precision of GC injections for sample solution. The errors in sample weighing and volume measurement contributed relatively little to the overall uncertainty. The expanded uncertainty in the mentholated cigarettes, Korean and American brand, at 0.95 level of statistical confidence was $\pm$0.06 and $\pm$0.07 mg/g for a menthol content of 1.89 and 2.32 mg/g, respectively.
