Amelioration of Cognitive Dysfunction in APP/PS1 Double Transgenic Mice by Long-Term Treatment of 4-O-Methylhonokiol

  • Alzheimer's disease (AD) is the most common neurodegenerative disease without known ways to cure. A key neuropathologic manifestation of the disease is extracellular deposition of beta-amyloid peptide (Ab). Specific mechanisms underlying the development of the disease have not yet been fully understood. In this study, we investigated effects of 4-O-methylhonokiol on memory dysfunction in APP/PS1 double transgenic mice. 4-O-methylhonokiol (1 mg/kg for 3 month) significantly reduced deficit in learning and memory of the transgenic mice, as determined by the Morris water maze test and step-through passive avoidance test. Our biochemical analysis suggested that 4-O-methylhonokiol ameliorated $A{\beta}$ accumulation in the cortex and hippocampus via reduction in beta-site APP-cleaving enzyme 1 expression. In addition, 4-O-methylhonokiol attenuated lipid peroxidation and elevated glutathione peroxidase activity in the double transgenic mice brains. Thus, suppressive effects of 4-O-methylhonokiol on $A{\beta}$ generation and oxidative stress in the brains of transgenic mice may be responsible for the enhancement in cognitive function. These results suggest that the natural compound has potential to intervene memory deficit and progressive neurodegeneration in AD patients.

Comparing Intelligence Test Profiles to Assess Tourette's Disorder with Attention-Deficit Hyperactivity Disorder

  • Objectives: The study compared the intelligence test profiles of Tourette's Disorder (TD), attention-deficit hyperactivity disorder (ADHD), and TD with ADHD (TD+ADHD) groups. Methods: The Korean Wechsler Intelligence Scale for Children-third edition (K-WISC-III) and Korean Wechsler Intelligence Scale for Children-fourth edition (K-WISC-IV) were administered to 13 children and adolescents with TD, 17 children and adolescents with ADHD, and 15 children and adolescents with TD+ADHD. Each parameter was compared among the groups using the Kruskal-Wallis test. Results: The mean scores of the freedom from distractibility/working memory index (FD/WMI) and the digit span and arithmetic subtests of the TD+ADHD group were significantly lower than those of the TD group. Conclusion: According to the intelligence test results, the comorbid ADHD+TD group showed a significant decrease in working memory compared to the TD group. These findings are similar to those of previous research on cognitive functions and suggest that the TD+ADHD comorbid and TD alone groups exhibit different endophenotypes. The results also imply that WISC-III and WISC-IV, the most commonly used intelligence tests clinically, are effective in evaluating cognitive functions such as attention. Further research is required to confirm these results.

주의력결핍 과잉행동장애 아동청소년의 Korean Wechsler Intelligence Scale for Children-Fourth Edition 프로파일 : 후향적 의무기록 분석 (Korean Wechsler Intelligence Scale for Children-Fourth Edition Profiles in Child and Adolescent with Attention-Deficit Hyperactivity Disorder : Retrospective Study)

  • Objectives : The aim of this study was to investigate Korean Wechsler Intelligence profiles and specific abilities related to attention problem of children with attention-deficit hyperactivity disorder (ADHD). Methods : The Korean Wechsler Intelligence Scale for Children-fourth edition (K-WISC-IV) and Advanced Test of Attention (ATA) were administered to 91 children and adolescents (age $8.5{\pm}2.6$ years, 73 boys) with ADHD. Pearson correlation and independent t-tests were used. Results : The means of Working Memory Index (WMI) and Processing Speed Index (PSI) showed a score of low average in K-WISC-IV. WMI scores for the K-WISC-IV showed clinically significant correlations with omission errors, commission errors, and response time variability on auditory ATA. PSI scores also showed significant correlations with response time and variability on visual ATA. In addition, significantly lower digit span backward scores were observed in hyperactive-impulsive/combined subtypes compared to inattentive subtype (t=3.60, p<.001). Conclusion : Children with ADHD showed significantly lower scores in WMI and PSI which were clinically correlated with ATA scores, and hyperactive-impulsive/combined subtypes showed poorer working memory functions in WMI. Follow-up studies are proposed.

노인의 기억 재활: 이론적 개관 (Memory Rehabilitation in the Elderly: A Theoretical Review)

  • 현대 사회에서 노인 인구의 비율이 꾸준하게 증가하면서, 정상적 및 병리적 노화와 관련된 기억 결함을 다루는 인지적 재활 전략의 영향력도 차차 커지고 있다. 본 논문에서는 현재 이루어지고 있는 노인을 위한 기억 재활 기법을 개관하였다. 먼저, 기억의 노화를 이해하기 위한 이해의 틀로서 인간의 기억 체계를 고찰하였고, 정상적 노화와 알츠하이머병과 같은 병리적 노화에 따른 기억 수행에 관하여 이루어진 연구결과들을 개관한 다음, 그 효과가 입증된 기억 재활 기법들을 소개하였다. 현재까지의 기억 재활 기법들은 아직 남아있는 외현 기억 촉진, 상대적으로 손상되지 않은 암묵 기억 이용, 외부 기억보조 도구 사용에 초점을 맞춘 것이었으나, 새로운 접근으로 뇌 가소성에 기반을 둔 기억 훈련 방법의 가능성을 제안하였다.

Effects of Chongmyung-tang on Learning and Memory Performances in Mice

  • Chongmyung-tang(CMT, 聰明湯), oriental herbal medicine which consists of Polygaglae Radix(遠志), Acori Graminei Rhizoma(石菖蒲) and Hoelen(白茯神) has effect on amnesia, dementia. In order to evaluate effect of CMT on memory and learning in mice, CMT extract was used for studies. This paper describes the effects of CMT extract on memory and learning processes by using the passive and active avoidance performance tests, novel object recognition task and water maze task. The CMT extract ameliorated the memory retrieval deficit induced by ethanol in the passive avoidance responses but did not affect ambulatory activity of normal mice. These results suggest that CMT has an ameliorating effect on memory retrieval impairment. CMT extract decreased spontaneous motor activity(SMA) in the latter sessions of memory registration in active avoidance responses. These results suggest that CMT has partly transquilizing or antianxiety effects. In novel object recognition task to measure visual recognition memory, CMT-administered mice enhanced in long term memory for 1-3 days. In water maze task to measure spatial learning, which requires the activation of NMDA receptors in the hippocampus, spatial learning in CMT-administered mice was faster than in wild-type mice. These results suggest that CMT enhances memory and activates NMDA receptors.

Anti-dementia Effects of Gouteng-san and Si-Wu-Tang

  • Recently, a traditional medicine called Gouteng-san, which consists of eleven herbs, was reported to be effective in treating vascular dementia with a double-blind, placebo-controlled study. Gout-eng-san is also used for patients with vascular dementia in combination with Si-Wu-Tang. The effect of Gouteng-san and Si-Wu-Tang on deficit of learning behavior was investigated using step-down passive avoidance task in mice. Hot-water extract of Gouteng-san (1.5 and 6 g/kg, p.o.) significantly prolonged the step-down latency shortened by scopolamine. The extract of Uncaria hook (150 mg/kg, p.o.), one of the component herb of Gouteng-san, significantly prevented the decrease in the latency after scopolamine. Hot-water extract of Si-Wu-Tang (1.5 and 6 g/kg of dried herbs, p.o.) prevented dose-dependently scopola-mine-induced disruption qf learning behavior. Si-Wu-Tang also prevented the ischemia-induced deficit of learning behavior. Both hot water extract of peony and angelica (1.5 g/kg, p.o.), which are component herbs qf Si-Wu-Tang, prevented the scopolamine-induced learning behavior deficit. Scopolamine (10 uM) suppressed long-term potentiation (LTP) of population spike in the CA1 region of the rat hippocampal slices. Peoniflorin (0.1~ 1uM) extracted from paeony root significantly ameliorated scopolamine-induced inhibition of LTR These results suggest that improvement of deficit of learning behavior by Gouteng-san and Si-Wu-Tang is mediated by direct and/or indirect activation of the cholinergic system in the brain.

Biflorin Ameliorates Memory Impairments Induced by Cholinergic Blockade in Mice

  • To examine the effect of biflorin, a component of Syzygium aromaticum, on memory deficit, we introduced a scopolamine-induced cognitive deficit mouse model. A single administration of biflorin increased latency time in the passive avoidance task, ameliorated alternation behavior in the Y-maze, and increased exploration time in the Morris water maze task, indicating the improvement of cognitive behaviors against cholinergic dysfunction. The biflorin-induced reverse of latency in the scopolamine-treated group was attenuated by MK-801, an NMDA receptor antagonist. Biflorin also enhanced cognitive function in a naïve mouse model. To understand the mechanism of biflorin for memory amelioration, we performed Western blot. Biflorin increased the activation of protein kinase C-${\zeta}$ and its downstream signaling molecules in the hippocampus. These results suggest that biflorin ameliorates drug-induced memory impairment by modulation of protein kinase C-${\zeta}$ signaling in mice, implying that biflorin could function as a possible therapeutic agent for the treatment of cognitive problems.

Effect of Saenggitang on Learning and Memory Ability in Mice

  • Objective : The effect Saenggitang (GT), which has been used for amnesia, in Oriental Medicine, on memory and learning ability, was investigated. Methods : Hot water extracts (HWE) of SGT were used for the studies. In passive avoidance performances (step through test), active avoidance performances (lever press test), Motor activity, pentobarbital-induced sleep, 20 and 50 mg/100g of SGT-HWE ameliorated the memory retrieval deficit induced by 40% ethanol. Results : The SGT-HWE did not affect the ambulatory activity of normal mice in normal condition. 20 and 50 mg/100g of SGT-HWE enhanced contextual fear memory, but not cued fear memory in a fear conditioning task, which requires the activation of the NMDA (N-methyl-D-aspartase) receptor. SGT-HWE did not affect the motor activity measured by the titling type ambulometer test performed immediately and 24 hr after the administration. SGT-HWE prolonged the sleeping time induced by 50 mg/kg pentobarbital in mice and decreased SMA (spontaneous motor activity) in active avoidance performances (lever press test). Conclusion : These results indicate that the SGT-HWE have an improving effect on the memory retrieval disability induced by ethanol and may act as a stimulating factor for activating the NMDA receptor. and the SGT-HWE has a tranquilizing and anti-anxiety action.

Two key genes closely implicated with the neuropathological characteristics in Down syndrome: DYRK1A and RCAN1

  • The most common genetic disorder Down syndrome (DS) displays various developmental defects including mental retardation, learning and memory deficit, the early onset of Alzheimer's disease (AD), congenital heart disease, and craniofacial abnormalities. Those characteristics result from the extra-genes located in the specific region called 'Down syndrome critical region (DSCR)' in human chromosome 21. In this review, we summarized the recent findings of the DYRK1A and RCAN1 genes, which are located on DSCR and thought to be closely associated with the typical features of DS patients, and their implication to the pathogenesis of neural defects in DS. DYRK1A phosphorylates several transcriptional factors, such as CREB and NFAT, endocytic complex proteins, and AD-linked gene products. Meanwhile, RCAN1 is an endogenous inhibitor of calcineurin A, and its unbalanced activity is thought to cause major neuronal and/or non-neuronal malfunction in DS and AD. Interestingly, they both contribute to the learning and memory deficit, altered synaptic plasticity, impaired cell cycle regulation, and AD-like neuropathology in DS. By understanding their biochemical, functional and physiological roles, we hope to get important molecular basis of DS pathology, which would consequently lead to the basis to develop the possible therapeutic tools for the neural defects in DS.

총명탕(聰明湯)과 향부자총명탕(香附子聰明湯)의 추출물, 나노분말 제형을 이용한 치매에 관한 연구 (A Study of ChongMyungTang(CMT) and HyangbujaChongMyungTang(HCMT) on Dementia - Extract & Nano Powder Drug types)

  • Objective : This research aims to investigates the effects of CMT and HCMT on Dementia. and we also want to know the different effect of CMT, HCMT by drug types. Methods : The research is progressed by two types of experiment. one experiment is BV2 microglial cell line treated by LPS in vitro and another experiment is memory deficit mice induced by scopolamine in vivo. Results : The CMT and HCMT is effective in BV2 microglial cell line treated by LPS in vitro and in the serum of the memory deficit mice induced by scopolamine in vivo. But, there is no significant difference between CMT and HCMT extract&nano powder in experimental conclusion. Conclusions : These results suggest that the two drug types of CMT and HCMT may be effective for the prevention and treatment of Dementia. Investigation into the further study two drug types of the CMT and HCMT for Dementia is suggested for future research.
