베타 튜불린에 의한 포스포리파제 C-감마1의 활성화 (Phospholipase C-γ Activation by Direct Interaction with β-Tubulin Isotypes)

  • 포스포리파제 C-감마1$(phospholipase\;C-{\gamma}\;1;\;PLC-{\gamma}\;1)$은 활성화될 경우 세포 내의 이차전령인 inositol 1,4,5-trisphosphate$(IP_3)$와 diacylglycerol(DG)을 생성하는 중요한 세포 신호전달 분자이다. 튜불린은 미세소관과 방추사의 주요 구성 단백질로서 알파형과 베타형의 두 가지 동위형이 있는데 이들은 모든 진핵세포에 존재하면서 이형 이합체를 형성한다. 이 중 베타형 튜불린은 사람의 경우 6종의 또 다른 동위형이 존재하는 것으로 밝혀졌는데 이들은 각 조직에서 그 발현양상이 서로 다르게 나타난다. 이전의 연구에서 우리들은 $PLC-{\gamma}\;1$과 4종의 베타튜불린 동위형 즉, ${\beta}1$, ${\beta}2$, ${\beta}3$${\beta}6$이 세포 내에서 서로 결합할 수 있으며 또한 외부의 자극이 전달될 경우 이들 4종의 동위형이 $PLC-{\gamma}\;1$을 활성화시켜 준다는 사실을 보고한 바 있다. 이번 실험에서는 이전의 연구에서 조사하지 못하였던 베타 튜불린의 나머지 두 가지 동위형 즉, ${\beta}4$${\beta}5$$PLC-{\gamma}\;1$에 결합하여 $PLC-{\gamma}\;1$의 활성을 증가시켜줌으로서 세포 내에서의 신호전달계를 조절하고 있음을 확인하였다. 이 결과는 이전의 연구결과와 연관 지워볼 때, 6종의 모든 베타형 튜불린은 세포 외부의 자극이 있을 경우 $PLC-{\gamma}\;1$을 활성화시켜줌을 시사한다.

마그네슘이온이 적출한 기니피그 대동맥평활근과 흰쥐 자궁평활근의 수축성에 미치는 효과에 관한 연구 (Effect of Magnesium on the Contractility of the Isolated Guinea-Pig Aortic and Rat Smooth Muscles)

  • It is well known that extracellular Calcium plays a very important role in several steps of smooth muscle excitability and contractility, and there have been many concerns about factors influencing the distribution of extracellular Ca++ and the Ca++ flux through the cell membrane of the smooth muscle. Based on the assumption that Mg++ may also play an important role in the excitation and contraction processes of the smooth muscle by taking part in affecting Ca++ distribution and flux, many researches are being performed about the exact role of Mg++, especially in the vascular smooth muscle. But yet the effect of Mg++ in the smooth muscle activity is not clarified, and moreover the mechanism of Mg++ action is almost completely unknown. Present study attempted to clarify the effect of Mg++ on the excitability and contractility in the multiunit and unitary smooth muscle, and the mechanism concerned in it. The preparations used were the guinea-pig aortic strip as the experimental material of the multiunit smooth muscle and the rat uterine strip as the one of the unitary smooth muscle. The tissues were isolated from the sacrificed animal and were prepared for recording the isometric contraction. The effects of Mg++ and Ca++ were examined on the electrically driven or spontaneous contraction of the preparations. And the effects of these ions were also studied on the K+ or norepinephrine contracture. All experiments were performed in tris-buffered Tyrode solution which was aerated with 100% 02 and kept at 35oC. The results obtained were as follows: 1] Mg++ suppressed the phasic contraction induced by electrical field stimulation dose-dependently in the guinea-pig aortic strip, while the high concentration of Ca++ never recovered the decreased tension. These phenomena were not changed by the a - or b - adrenergic blocker. 2]Mg++ played the suppressing effect on the low concentration [20 and 40 mM] of K+-contracture in the aortic muscle, but the effect was not shown in the case of 100mM K+-contracture. 3] Mg++ also suppressed the contracture induced by norepinephrine in the aortic preparation. And the effect of Mg++ was most prominent in the contracture by the lowest [10 mM] concentration of norepinephrine. 4] In both the spontaneous and electrically driven contractions of the uterine strip, Mg++ decreased the amplitude of peak tension, and by the high concentration of Ca++ the amplitude of tension was recovered unlike the aortic muscle. 5] The frequency of the uterine spontaneous contraction increased as the [Ca++] / [Mg++] ratio increased up to 2, but the frequency decreased above this level. 6] Mg++ decreased the tension of the low[20 and 40mM] K+-contracture in the uterine smooth muscle, but the effect did not appear in the 100mM K+-contracture. From the above results, the following conclusion could be made. 1] Mg++ seems to suppress the contractility directly by acting on the smooth muscle itself, besides through the indirect action on the nerve terminal, in both the aortic and uterine smooth muscles. 2] The fact that the depressant effect of Mg++ on the K+-contracture is in inverse proportion to an increase of K+ concentration appears resulted from the extent of the opening state of the Ca++ channel. 3] Mg++ may play a depressant role on both the potential dependent and the receptor-operated Ca++ channels. 4] The relationship between the actions of Mg++ and Ca++ seems to be competitive in uterine muscle and non-competitive in aortic strip.

진피 추출물에 의한 인간유방암 MDA-MB-231 세포의 apoptosis 유도에서 ROS 및 AMPK의 역할 (Relationship between Reactive Oxygen Species and Adenosine Monophosphate-activated Protein Kinase Signaling in Apoptosis Induction of Human Breast Adenocarcinoma MDA-MB-231 Cells by Ethanol Extract of Citrus unshiu Peel)

  • 한의학에서는 진피(陳皮)라고 칭하는 귤의 껍질(Citrus unshiu peel) 추출물은 항산화, 항염증 및 항균 특성을 포함한 다양한 약리학적 효능을 갖는 것으로 알려져 있다. 최근 그들의 항암 활성에 대한 가능성이 보고되었지만 정확한 기전 연구는 여전히 미비한 실정이다. 본 연구에서는 인간 유방암 MDA-MB-231 세포를 대상으로 진피 에탄올(EECU, ethanol extract of C. unshiu peel) 추출물의 항암 효능을 평가하고 그에 따른 기전 연구를 수행하였다. 본 연구의 결과에 의하면 EECU에 의한 MDA-MB-231 세포의 증식억제는 세포사멸(apoptosis) 유도와 관련이 있었다. EECU에 의한 apoptosis는 caspase-8, -9 및 -3의 활성화와 IAPs 계열의 발현 감소에 따른 PARP의 분해와 Bax : Bcl-2 비율의 증가와 연관이 있었다. 또한 EECU는 Bid의 truncation과 함께 미토콘드리아 막 잠재력의 감소와 세포질로 cytochrome c의 이동을 촉진시켰다. 아울러 EECU는 AMPK 및 ACC의 인산화를 촉진시켰으나, AMPK 효소 활성의 저해제는 EECU에 의한 apoptosis 유도와 생존력 저하를 현저하게 억제하였다. 부가적으로 EECU는 AMPK 활성화의 상류 신호로 작용하는 활성산소종(ROS)의 생성을 촉진시켰으며, 강력한 항산화제인 NAC는 EECU에 의한 AMPK의 활성화와 apoptosis를 역전시켰다. 결론적으로 EECU는 ROS/AMPK 의존적인 내인성 및 외인성 apoptosis 경로를 활성화시킴으로써 MDA-MB-231 세포 증식을 억제하였음을 알 수 있었다.

아밀로이드 베타(amyloid beta)로 유도된 인지장애 마우스 모델에서 갑주백목(Diospyros kaki) 추출물의 인지기능 및 뇌 신경세포 보호 효과 (Protective effect of Gabjubaekmok (Diospyros kaki) extract against amyloid beta (Aβ)-induced cognitive impairment in a mouse model)

  • 본 연구에서는 갑주백목 에탄올 추출물(ethanolic extraction from Diospyros kaki (Gabjubaekmok))을 이용하여 in vitro 항산화 활성과 더불어 $A{\beta}$로 유도된 인지기능 저하를 갖는 마우스 모델에서 인지기능 및 뇌 신경세포 보호 효과를 검증하였다. 갑주백목 추출물은 양성대조군과 비교하여 우수한 ABTS 및 DPPH 라디칼 소거활성($IC_{50}=83.30$, $403.88{\mu}g/mL$) 및 MDA 생성 억제 활성($IC_{50}=62.10{\mu}g/mL$)을 보여주었고, in vitro 아세틸콜린 분해효소 억제 활성은 $312.82{\mu}g/mL$$IC_{50}$ 값을 나타내었다. 또한, MC-IXC 뇌 신경세포에 과산화수소를 처리하여 산화적 스트레스를 유발시킨 뇌 신경세포 사멸에 대해 보호 효과를 나타냈다. 갑주 백목 추출물의 인지기능 개선 효과를 확인하기 위하여 $A{\beta}$를 사용하여 인지기능 장애 마우스 모델을 수립하였으며, Y-미로, 수동 회피 및 Morris 수중 미로 실험과 같은 행동실험을 통해 인지 및 기억능력에 대한 개선 효과를 나타냈다. 이후 마우스 뇌조직에서의 아세틸콜린 함량의 증가 및 아세틸콜린 분해효소의 활성을 억제함으로써 cholinergic 시스템을 보호하였고, SOD, 환원형 GSH 및 MDA 함량 측정을 통해 항산화 시스템을 개선시켜줌을 확인하였다. 더불어 뇌 조직의 미토콘드리아에서 ROS의 생성 억제, MMP 보호 및 ATP 함량을 회복시켜주었으며, western blot 분석을 통해 $TNF-{\alpha}$와 같은 염증성 사이토카인을 억제시켜 줌으로써 면역반응에 관여하는 JNK의 인산화를 감소시키고 Akt 신호전달을 활성화시켜 세포자동 사멸화를 억제시키는 것으로 확인되었다. 마지막으로 HPLC 분석을 통해서 갑주백목의 주요 생리활성물질이 갈산으로 확인되었다. 이러한 결과를 종합하였을 때, 갑주백목 에탄올 추출물은 뇌 조직에서의 cholinergic 및 항산화 시스템 보호효과를 통해 $A{\beta}$ 처리에 대하여 학습 및 기억 능력을 개선 시킬 수 있는 천연 소재로서의 가능성뿐만 아니라 $A{\beta}$ 및 과산화수소로부터 유발된 산화적 스트레스의 환경에서 뇌신경세포를 보호함으로써 알츠하이머성 질환과 같은 퇴행성 뇌신경질환을 예방할 수 있는 고부가가치 건강기능식품 소재로서의 활용 가능성을 확인하였다.

호화 옥테닐 호박산 전분의 유화 특성 (Emulsifying Properties of Gelatinized Octenyl Succinic Anhydride Modified starch from Barley)

  • 본 연구는 OSA 전분을 열처리 한 후, 이를 이용하여 제조한 OSA 전분 에멀션의 이화학적 특성 및 계면 흡착 구조 등을 조사하였다. 에멀션 중 지방구의 크기는 OSA 전분 농도의 증가와 더불어 지속적으로 감소하여 0.2 wt% 농도에서 최소값($0.31{\pm}0.01{\mu}m$)을 나타내었고, 그 이상의 농도에서는 변화가 없었다. 에멀션의 크리밍 안정도는 OSA 전분 농도가 높을수록 증가하였으며, 0.75 wt% 이상의 첨가 농도에서 크리밍 발생에 대하여 매우 안정하였다. 에멀션 중 OSA 전분의 계면 흡착량은 0.2 wt% 첨가 농도 이상에서 농도의 증가와 더불어 증가하였으며(0.2 wt% : $1.03mg/m^2$ ${\rightarrow}$ 1.25 wt% : $5.08mg/m^2$), 이는 계면에서 OSA 전분이 다층 구조를 이루는 것에 기인된 것으로 추정하였다. OSA 전분 에멀션의 pH를 조절하였을 때 산성 지역에서 지방구의 응집에 의해 크기가 증가하였으며, 이는 상대적으로 낮은 제타 전위에 기인된 것으로 사료되었다. 터비스캔에 의한 분산 안정도 또한 pH에 영향을 받아 산성 지역에서 낮았으며, pH 7 이상에서는 높은 분산 안정도 특성을 보였다. 공초점현미경을 이용하여 열처리된 OSA 전분이 흡착된 지방구 표면을 관찰한 결과, OSA 전분은 입자 형태가 아닌 두꺼운 계면막을 형성하는 것으로 나타났다. 따라서 에멀션 형성 전에 OSA 전분을 열처리할 경우, 전분의 호화과정에서 용출된 아밀로오스와 아밀로펙틴이 지방구 표면에 막의 형태로 흡착되므로, OSA 전분 에멀션에 있어서 중요한 유화 안정화 기작은 '입체장애 안정화(steric stabilization)'인 것으로 사료되었다.

키토산올리고당의 효소적 대량생산 및 생리활성 (Large scale enzymatic production of chitooligosaccharides and their biological activities)

  • 키토산은 천연에 존재하는 풍부한 고분자 다당류이며 동시에 항균작용, 항암작용, 면역 활성 증강 작용 등 다양한 생리 기능성을 가진 생체 물질(biomaterial)이다. 그러나 키토산은 수용액에 쉽게 용해되지 않고 체내 흡수율도 매우 낮은 고분자 물질이기 때문에 그 자체로는 생체 내에서 기능을 발휘하지 못한다. 따라서 체내 흡수율이 용이하고 보다 다양한 생리 활성을 나타내는 키토산올리고당으로 섭취할 필요가 있다. 키토산으로부터 키토산올리고당을 만드는 방법은 온화한 조건에서 부반응이 없는 효소분해가 가장 이상적이다. 그러나 키토산을 분해시킬 수 있는 효소의 가격이 매우 비싸고 효소 활성이 낮아 산업적으로 효소를 이용하여 키토산올리고당을 대량 생산하는데는 경제적으로 큰 문제가 있었다. 따라서 효소 분해에 의한 키토산올리고당의 생산을 산업적으로 적용시켰을 때 대량으로 소모되는 효소의 높은 생산 비용을 경감하기 위하여 효소의 재이용을 위한 생산 시스템을 도입하였고 한외여과막과 효소 반응기를 결합시킨 한외여과막 효소반응기를 키토산올리고당의 생산에 적용시킴으로써 대량 생산이 가능해졌다. 이 시스템은 사용하는 막의 종류에 따라 원하는 분자량의 키토산올리고당을 생산 할 수 있어 분자량 크기에 따라 생체 내에서 다른 생리기능을 나타내는 키토산올리고당을 각기 목적에 맞는 제품으로 대량 생산할 수 있다. 키토산올리고당은 암세포 성장에 영향을 미치는 면역을 증진시키고 암 전이 관련 효소인 MMP-2 및 MMP-9의 발현을 저지하여 암 전이를 억제할 뿐만 아니라 암세포의 신생 혈관형성을 억제하므로 암의 예방이나 치료에 활용될 수 있다. 또한 키토산올리고당은 생체 노화를 촉진시키는 초과산화 라디칼, 히드록시라디칼, 과산화 라디칼과 같은 활성산소종을 소거시킴으로써 활성산소종에 의해 유발되는 질환도 예방한다. 더 나아가서, 키토산올리고당은 양전하를 가진 아미노기를 갖고 있어 미생물 세포막의 음전하와 결합하여 세포막의 기능을 상실시킴으로써 막을 통해 출입하는 대사에 필요한 물질들을 봉쇄한다. 이것은 미생물의 성장과 증식을 감소시키는 효과가 있어 식품분야에서도 천연 보존제로서 활용될 것으로 기대된다. 지금까지 고혈압 및 심장 질환의 치료는 레닌엔지오텐신 시스템(RAS)경로의 치료조작(mani pulator) 및 ACE 저해에 초점이 맞추어져 왔다. Captopril, Enalapril, Alcacepril 및 Lisinopril같은 합성 ACE 저해제는 고혈압 치료 및 예방을 위해서도 널리 사용되고 있으나 기침, 알레르기 반응, 맛 장애 및 피부발진과 같은 부작용을 야기시킬 위험이 있다. 그러므로 고혈압의 치료나 관리를 위한 치료제로서 자연계에 존재하는 천연성분인 키토산올리고당이 바람직한 대안으로 사용될 수 있을 것이다. 고령 사회로 진입하면서 노인의 치매 발병률은 현저하게 증가하는 추세이나 아직까지 효과가 뛰어난 치매치료제는 개발되지 않고 있는 실정이다. 키토산올리고당이 치매유발효소인 베타-세크레테이즈뿐만 아니라 아세틸콜린 에스테르가수분해효소를 저해하고, 산화에 의한 뇌세포의 손상을 저해하는 효능이 있는 것으로 밝혀져 앞으로 치매 예방이나 치료에 적극적으로 활용되어야 한다. 최근에 소비자나 환자들은 다양한 부작용을 낳는 화학적으로 합성된 의약품을 기피하는 경향이 있어 자연식품이나 천연 생리기능성 물질과 자가치료에 대한 욕구가 높아지고 있다. 그러므로 의약품에 비해 다소 효능이 낮을지라도 특정한 질병의 예방이나 치료를 위해 특수한 생리 기능성 물질의 섭취는 더욱 더 인기를 끌게 될 것이다. 따라서 항암, 항산화, 항염증, 항균, 항고혈압, 항치매, 항당뇨, 항알레르기 등 다양한 생리활성을 나타내는 키토산올리고당을 활용한 건강기능성식품(nutraceuticals), 의약품(pharmaceuticals) 및 기능성 화장품(cosmeceuticals) 등 다양한 제품이 개발될 것으로 기대된다.

고초균에 의한 생강 발효 추출물의 신경세포 보호 효과 (Neuroprotective effect of fermented ginger extracts by Bacillus subtilis in SH-SY5Y cells)

  • 본 연구에서는 생강을 효소처리하여 수용화율을 높인 후 고초균을 이용하여 발효시킨 생강발효물 (FGEs)을 제조하고, 이를 SH-SY5Y에 6-OHDA와 함께 처리하여 세포 보호 효과 및 AChE 저해 활성을 평가하였다. 그 결과, 6-OHDA로 자극된 신경세포에서 FGE를 처리한 모든 실험군에서 세포 생존율이 증가하고 LDH 농도가 감소하였다. 6-OHDA로 유발된 세포자멸사를 억제할 수 있는지 확인하기 위해 핵의 형태학적인 변화 및 caspase-3 활성을 확인하였다. FGE를 처리한 모든 실험군에서 핵의 손상 및 apoptotic body의 감소와 caspase-3 억제 활성을 확인할 수 있었다. 또한 FGE는 AChE의 활성을 유의적으로 감소시켰다. 시료 간의 활성 차이를 비교하였을 때, 생강에 효소 처리 후 고초균으로 발효한 추출물군 (E/BKG와 E/BCG)의 신경세포 보호 활성이 효소 처리하지 않은 발효생강군 (BKG와 BCG) 보다 유의적으로 크게 나타났다. 그러나 발효에 사용된 고초균 2종 간의 활성은 유사하였으며 처리군 간 유의적인 차이를 보이지 않았다. 본 연구 결과로부터 효소처리하여 수용화율을 높여 고초균으로 발효한 FGE는 신경세포 보호 및 AChE 저해 효과를 나타내어 향후 신경질환 연구를 위한 기초자료 제공 및 고부가가치 식품소재 개발에 이용 가능할 것으로 판단된다.

기장 종자 유래 추출물의 인간 유방암 MDA-MB-231 세포에 대한 세포독성에 관련된 미토콘드리아 손상-의존적 아폽토시스 유도 효과 (Apoptotic Effect of Proso Millet Grains on Human Breast Cancer MDA-MB-231Cells Is Exerted by Activation of BAK and BAX, and Mitochondrial Damage-mediated Caspase Cascade Activation)

  • 잡곡 류에 속하는 기장(Panicum miliaceum L.)의 항암 효과를 알아보기 위해, 기장의 종자를 80% 에탄올(EtOH)로 추출하였으며, 이를 감압 농축하여 건조시키고 재차 물에 녹인 후 4가지 유기용매(헥산, 메틸렌크로라이드, 에틸아세테이트 및 부탄올)로 순차적으로 추출 분획하였다. 다양한 인간 암세포에 대하여 80% 에탄올 추출물의 세포독성을 조사한 결과, 인간 유방암 세포주 MDA-MB-231에 대한 세포독성 효과가 가장 강하게 나타났다. 또한 에탄올 추출물 유래 각 유기용매 분획들 중에서 세포독성이 가장 높게 나타난 부탄올 분획을 사용하여, 유방암 세포주 MDA-MB-231에 대한 아폽토시스성 세포사멸 유도 효과를 조사하였다. 그 결과로서, BAK/BAX 활성화, 미토콘드리아 막 전위(Δψm) 손실, 미토콘드리아 시토크롬 c 방출, 카스파아제-8/-9/-3의 활성화, PARP의 분해, 그리고 TUNEL-양성 아폽토시스성 DNA 단편화와 같은 아폽토시스 반응들이 검출되었다. 한편, 인간 급성백혈병 Jurkat T 세포의 A3 클론(야생형), I2.1 클론(FADD-결손형) 및 I9.2 클론(카스파아제-8 결손형)은 부탄올 분획의 세포독성에 대해 유사한 감수성을 나타내었는데, 이는 부탄올 분획의 아폽토시스 유도 활성에는 외인성 아폽토시스 기전이 관련되지 않음을 시사한다. 흥미롭게도, 인간 정상 유방 상피세포 MCF-10A는 유방암 MDA-MB-231세포에 비해 부탄올 분획의 세포독성에 대하여 훨씬 낮은 감수성를 보였다. 이러한 연구결과는 기장 종자 유래 부탄올 분획의 인간 유방암 세포주 MDA-MB-231에 미치는 세포독성효과는 BAK/BAX 활성화에 따른 미토콘드리아 외막 손상 및 시토크롬 c 방출, 이에 수반되는 카스파아제 활성화에 의해 매개됨을 보여준다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times.
