멜론괴저반점바이러스-Me의 몇 가지 특성과 멜론 품종의 저항성 선발 (Some Characteristics of Melon necrotic spot virus-Me and Resistance Screen to the Virus in Melon Cultivars)

  • Melon necrotic spot virus (MNSV)는 국내에서 재배되고 있는 멜론(Cucumis melo)에 심각한 피해를 주고 있다. 2009년 전남 나주 멜론재배단지에서 괴저반점을 나타내는 멜론 잎에서 MNSV를 분리 동정하였다. 지표식물 반응, immuno captured(IC) RT-PCR 및 염기서열 상동성 비교 등 몇몇 분석으로 이 분리주를 MNSV-Me로 명명하였다. 멜론 시판 35품종에 대한 MNSV-Me의 저항성 선발을 위하여 유묘의 자엽에 인위적 즙액 접종 방법을 이용하였다. 접종된 모든 품종의 자엽에서는 괴저반점을 형성하였다. 이들 중 25품종은 상엽으로 괴저 증상의 이행 또는 일부 개체에서는 접종 약 40일 후 줄기에 3 cm 이상의 괴저를 나타내는 감수성 반응을 나타냈다. 5품종은 상엽으로 잘 이행이 되지 않지만 줄기에 3 cm 미만의 괴저증상을 형성하는 중간 저항성을 발현하였다. 접종한 상엽으로는 바이러스 증상이 발현되지 않고, 단지 줄기에 희미한 괴저가 형성되는 얼스타이탄, 얼스럭서리, 베타리치 하계, 베타리치 및 원더플하계1호가 저항성을 나타냈다.

  • Melon necrotic spot virus (MNSV) was recently identified on watermelon (Citrullus vulgaris) in Korea, displaying as large necrotic spots and vein necrosis on the leaves and stems. The average occurrence of MNSV on watermelon was found to be 30-65% in Hapcheon and Andong City, respectively. Four isolates of the virus (MNSV-HW, MNSV-AW, MNSV-YW, and MNSV-SW) obtained from watermelon plants in different areas were non-pathogenic on ten general indicator plants, including Chenopodium quinoa, while they infected systemically six varieties of Cucurbitaceae. The virus particles purified by 10-40% sucrose density gradient centrifugation had a typical ultraviolet spectrum, with a minimum at 245 nm and a maximum at 260 nm. The morphology of the virus was spherical with a diameter of 28-30 nm. Virus particles were observed scattered throughout the cytoplasm of watermelon cells, but no crystals were detected. An ELISA was conducted using antiserum against MNSV-HW; the optimum concentrations of IgG and conjugated IgG for the assay were $1{\mu}l/ml$ and a 1:8,000-1:10,000 dilutions, respectively. Antiserum against MNSV-HW could capture specifically both MNSV-MN from melon and MNSV-HW from watermelon by IC/RT-PCR, and they were effectively detected with the same specific primer to produce product of 1,172 bp. The dsRNA of MNSV-HW had the same profile (4.5, 1.8, and 1.6 kb) as that of MNSV-MN from melon. The nucleotide sequence of the coat protein of MNSV-HW gave a different phylogenetic tree, having 17.2% difference in nucleotide sequence compared with MNSV isolates from melon.

  • A severe disease of muskmelon (Cucumis melo cv. Alsnight) grown on rockwool in a plastic house was characterized by leaf and stem necrosis followed by death of the plants. In 2001, an isolate of Melon necrotic spot virus-MN (MNSV-MN) of the genus Camovirus was identified as the causal agent of the disease on the basis of biological reactions and nucleotide sequence analyses of coat protein (CP) gene. MNSV-MN induced necrotic local lesions on mechanically inoculated leaves and systemic necrotic spots on the upper leaves of melon cvs. Alsnight, Rui III, Party, Imperial, and Seolhang. However, the inoculated leaves of watermelon and cucumber showed only necrotic lesions. DsRNAs extracted from the melon infected with MNSV-MN were separated into three components. Molecular sizes of the dsRNAs were estimated at approximately 4.5, 1.8, and 1.6 kbp. The amplified cDNA products of CP gene for MNSV-MN by RT-PCR showed approximately 1.2 kbp. The amplified DNA was digested to three fragments by MspI treatment. The cDNA of the genomic RNA of MNSV-MN was cloned and the region deduced to encode the CP was sequenced. The CP coding region, located near 3' end of the genome, consisted of 1,170 nucleotides and had the potential to encode a 390 amino acid protein. The nucleotide and amino acid sequences of MNSV-MN CP gene were 84.0-94.6% and 90.8-94.9% identical with other MNSV isolates found in the GeneBank database, respectively. This is the first report on the occurrence of MNSV in Korea.

Biological and Molecular Characterization of a Korean Isolate of Cucurbit aphidborne yellows virus Infecting Cucumis Species in Korea

  • Surveys of yellowing viruses in plastic tunnels and in open field crops of melon (Cucumis melo cultivar catalupo), oriental melon (C. melo cultivar oriental melon), and cucumber (C. sativus) were carried out in two melon-growing areas in 2014, Korea. Severe yellowing symptoms on older leaves of melon and chlorotic spots on younger leaves of melon were observed in the plastic tunnels. The symptoms were widespread and included initial chlorotic lesions followed by yellowing of whole leaves and thickening of older leaves. RT-PCR analysis using total RNA extracted from diseased leaves did not show any synthesized products for four cucurbit-infecting viruses; Beet pseudo-yellows virus, Cucumber mosaic virus, Cucurbit yellows stunting disorder virus, and Melon necrotic spot virus. Virus identification using RT-PCR showed Cucurbit aphid-borne yellows Virus (CABYV) was largely distributed in melon, oriental melon and cucumber. This result was verified by aphid (Aphis gossypii) transmission of CABYV. The complete coat protein (CP) gene amplified from melon was cloned and sequenced. The CP gene nucleotide and the deduced amino acid sequence comparisons as well as phylogenetic tree analysis of CABYV CPs showed that the CABYV isolates were undivided into subgroups. Although the low incidence of CABYV in infections to cucurbit crops in this survey, CABYV may become an important treat for cucurbit crops in many different regions in Korea, suggesting that CABYV should be taken into account in disease control of cucurbit crops in Korea.

The Incidence of Virus Diseases on Melon in Jeonnam Province during 2000-2002

  • The occurrence and relative incidence of viruses including Cucumber mosaic virus (CMV), Zucchini yellow mosaic virus (ZYMV), Papaya rings pot virus (PRSV), and Watermelon mosaic virus (WMV), Cucumber green mottle mosaic virus (CGMMV), Kyuri green mottle mosaic virus (KGMMV), and Melon necrotic spot virus (MNSV) were surveyed from main melon (Cucumis melo L.) production areas in Jeonnam province during 2000-2002. Virus disease incidences of melon cultivating fields were 0% and 11% in spring and fall 2000; 40%, 2.1%, and 8.8% in spring, summer, and fall 2001; and 6.3 % in spring 2002 in main cultivated areas in Jeonnam province, respectively. Field disease incidences of melon virus infections were 0% and 18.8% in spring and fall 2000; 50%, 38.5%, and 82.6% in spring, summer, and fall 2001; and 47.4% in spring 2002, respectively. Total of 101 melon samples showing typical disease symptoms were collected from 2000 to 2002 and tested for virus infection by RT-PCR. Potyvirus-specific DNA fragments for WMV, ZYMV, and PRSV were amplified from 46, 5, and 4 samples, respectively. MNSV specific DNA fragment was amplified from 18 samples. CMV-specific DNA fragment was detected from only 3 samples.

2007년 우리나라 주요 작물 바이러스병 발생 상황 (Survey of Viral Diseases Occurrence on Major Crops in 2007)

  • The severe damage induced by the important viruses of Rice stripe virus (RSV), Cucumber green mottle mosaic virus (CGMMV), Melon necrotic spot virus (MNSV), Tomato spotted wilt virus (TSWV) and Tomato bushy stunt virus (TBSV) was described on major crops in Korea. In 2007, the plot incidence rate of RSV was 100% on the precocious rice cultivars at the Western coastal provinces of Gyeonggido, Chungcheongnamdo, Jellabugdo and Jellanamdo, and Jejudo. RSV occurred in 2,441 ha with incidence rate of 70% over at 5 areas of Seocheon, Seosan, Boryung, Hongsung and Buyou in Chungcheongnamdo. At 4 areas of Buan, Gimje, Gunsan and Gochang in Jellabukdo, RSV occurred in 2.016 ha. CGMMV occurred on watermelon in 4.6 ha at Cheongyang area, and its outbreak was also 890 ha on oriental melon for 120 farmers with the incidence area of 23% against total cultivation areas of Seongju. MNSV was recorded firstly on watermelon in 2006 at Andong and it spread to 3 areas of Hapcheon, Gochang and Yanggu. TSWV occurred firstly at Danggin in Chungcheongnamdo in 2005. TSWV in 2006 spread to 6 areas; Taian, Hongsung and Seosan in Chungcheongnando, Namwon in Jellabukdo, and Sunchon and Kwangju in Jellanamdo. In 2007, TSWV covered 17 areas of western and southern parts; the 5 area including Taian in Chungcheongnamdo, Kwangju in Jellanamdo, Bucheon in Gyunggido, and so forth. TBSV was described firstly on table tomato at Sacheon in Kyungsangnamdo in 2004. TBSV occurred on cherry tomato at Chungju in 2006 and on table tomato at Busan area.

2007-2011 우리나라 농업현장 임상진단 요청 작물의 바이러스 감염 특성 (2007-2011 Characteristics of Plant Virus Infections on Crop Samples Submitted from Agricultural Places)

  • 농업인 등 농업현장에서 임상진단을 요청한 시료 수는 2007년부터 5년간 총 2,992건이었으며, 바이러스를 검정한 샘플 수는 13,325개이었다. 연도별 검출된 바이러스 종류는 2007년 21종, 2008년 15종, 2009년 23종, 2010년 21종, 2011년 17종이었다. 5년 동안에 우리나라에서 처음 검정된 바이러스는 2007년에는 TbLCV, 2008년에는 TYLCV, 2009년에는 INSV, RaMV, 2010년에는 BWYV이었다. 2007년부터 5년간 총 발생한 바이러스 종류는 알파파모자이크바이러스(Alfalfa mosaic virus, AMV) 등 총 40종이었다. 임상진단 요청 시료에서 가장 감염율이 높은 상위 10종의 바이러스는 CMV, TSWV, TYLCV, CGMMV, BBWV2, ZYMV, MNSV, PMMoV, WMV, PepMoV이었다. 농업현장에서 임상진단 요청한 작물의 종류는 총 53종이었으며, 상위 10위의 작물 종류는 고추, 토마토, 파프리카, 수박, 멜론, 벼, 오이, 옥수수, 무, 박이었으며, 상위 10개 작물과 20개 작물의 전체 임상진단 요청률은 각각 81.6%와 94.2%이었다. 고추에 감염된 바이러스는 BBWV2 등 8종이었으며, 검정한 샘플 가운데 바이러스가 검출된 비율은 CMV 24.6%, PMMoV 18.9%, TSWV 14.7%이었다. 2종 바이러스 복합감염 형태는 평균 감염률 14.7%의 BBWV2+CMV 등 7가지, 3종 바이러스 복합감염 형태는 평균 감염률 0.9%의 BBWV2+CMV+PepMoV 등 4가지이었다. 토마토에서 검출된 바이러스 종류는 CMV 등 6종이었으며 검정한 샘플 중 바이러스가 검출된 비율은 TYLCV 50.6%, TSWV 14.5%, TbLCV 10.9%이었다. 복합감염형태와 감염률은 CMV+TSWV 3.9%, ToMV+TYLCV 0.4%이었다. 수박에 감염된 바이러스는 CMV 등 5종이었으며 평균 검출 비율은 MNSV 37.0%, CGMMV 20.4%, ZYMV 18.1%, WMV 17.8%이었다. 수박 바이러스의 복합감염 형태와 검출비율은 CMV+MNSV 0.7%, WMV+ZYMV 5.0%이었다. 멜론에서 검출된 감염된 바이러스는 CMV 등 3종이었고 평균 검출비율은 MNSV 77.6%, CMV 5.6%, WMV 3.3%이었다. 복합감염은 CMV+ MNSV로 평균 검출비율은 13.5%이었다.

2010년 우리나라 주요 작물 바이러스병 발생 상황 (Occurrence of Virus Diseases on Major Crops in 2010)

  • 2010년 농업현장의 농업인 등이 임상진단 요청한 시료 중 채소류는 고추 등 8개 작물이었으며 이중 토마토가 31.8%, 수박 21.4%, 고추 19.5%로 바이러스병으로 인한 피해가 많은 작물이었다. 과수류는 사과 등 4작물이었으며, 포도가 63.6%, 사과 33.0%로서 임상진단 요청시료의 대부분을 차지하였다. 화훼류는 국화 등 6개 작물이었으며, 튜립 60.0%, 선인장이 20.0%로 바이러스 피해가 발생하고 있었다. 바이러스 감염시료를 분석한 결과 고추와 토마토는 CMV, BBWV2 등 6종의 바이러스가 감염되었으며, 수박은 MNSV, CMV 등 5종의 바이러스가 감염되었다. 포도는 GFkV 등 6종의 바이러스가 감염되었다. 바이러스 종류별 감염은 CMV가 8종의 채소류 중 6종의 채소류와 튜립에 감염되었다. 2010년 농업현장에서 임상진단한 시료에 감염된 바이러스는 32종이었으며, 바이로이드는 4종이었다. 토마토황화잎말림바이러스(TYLCV)는 2010년 전북 부안 등 6개 도 18개 시 군에 신규로 발생되어 2008년 처음 발생된 이후 총 58개 시 군으로 확산되었다. 토마토원형반점바이러스(TSWV)는 2010년 전북진안, 제주도의 제주 지역에서 신규로 발생하였으며, 2004년 안양 관양동 지역에 대 발생한 이후 25개 시 군에 확산되었으나, 경북 및 충북 지역에서는 현재까지 발생하지 않았다. 토마토덤불위축바이러스(TBSV)는 2010년 경남진주 지역에 신규로 발생하여 2004년 경남 사천지역에 처음 발생한 이후 경남 충북 지역의 5개 시 군에 발생 하였다.
